유중의 유우에 인삼박 급여가 유생산 및 우유품질에 미치는 영향 (The Influence of Paaax Ginseng Meal on the Milk Production and Milk Quality of Lactating Milk Cow)

  • To investigate the feeding value of the concentrated feedstuff including the Korean ginseng meal instead of wheat bran, Holstein milk cows were fed by this feedstuff (10kg/day) for 80 days, and the amounts of milk production and quality of milk were checked. The results are as follows: 1. Milk production of cows fed by the concentrated feedstuff including the ginseng meal was increased as averaged as 1.25kg/day compared with those of controlled cows during same period(SA) and before feeding treatment (SB). Significant different of milk production was found between cows treated and cows of SB plot, but there was no significant difference of milk production between cows treated and cows of SA plot. 2. Milk fat content was significantly increased as moth as 0.22%, 0.69% and 0.455% by the feeding of ginseng meal compared with that of SB Plat and that of SA Plot, and as average value, respectively. 3. Solid not fat (SNF) of milk from cows fed by the ginseng meal was also significantly increased as much as 0.845%, 1.339% and 1.0925 tempered with these of cows before treatment (SB) and cows controlled (SA), and as average value, respectively. 4. Specific gravity of milk produced from cows treated was higher as average as 0.0055 than those of controlled plots (SA and SB). Significant difference of specific gravity was found between cows treated and cows of SA plot, but there was no significant difference between tows treated and cods of SB plot. 5. Acidity of milk produced from cows treated was lower as averaged as 0.0045 than those of controlled plots (SA and SB). Significant difference between cows treated and cows of SA plot was found, but there was no significant difference between cows treated and cows treated and cows of SB plot.

수삼, 백삼 및 홍삼의 효과에 대한 비교 연구 흰쥐의 혈청내 몇가지 성분 함량 및 과산화지질생성에 미치는 영향 (Comparative Studies on the Effects of Fresh, White and Red Ginseng Their Effects on the Content of Several Components in Rat Serum and the Production of Lipid Peroxide in Rat Liver)

  • Seventy percent ethanol extracts were prepared from 6-year-old fresh, white and red ginseng cultivated in Kangwha, and administered orally into rats for the comparison of their effects. And their effects on the production of lipid peroxide in the livers of ethanol-administered rats were measured. Red ginseng administered group showed the largest body weight increase. However, fresh, white, red ginseng extract administered group showed no significant change in concentration of total lipid, triglyceride and free cholesterol in serum. White and red ginseng extract administeration decreased blood-sugar levels significantly. Lipid peroxide content in livers of white and red ginseng administered groups was decreased significantly. Red ginseng administered group showed greater decrcase in lipid peroxide content than that of white ginseng administred group. The increase of lipid peroxide content in the livers of ethanol-administered group was inhibited by administration of fresh, white, and red ginseng extract. And the strongest-inhibitory action was observed in red ginseng administered group. Therefore, it is supposed that red ginseng has the most powerful antiaging effect.

  • Ginseng Is renowned for both its medicinal and herbal uses and successful cultivation of Panax ginseng in Asia and Panax quinquefolium in North America has until recently taken place in the native geographical ranges of the plants. As a consequence of the potential high capital return and anticipated increases in consumer consumption, commercial cultivation of American ginseng now occurs well outside the native range of the plant in North America. In fact, the region of greatest expansion of cultivation is in the semi-arid interior region of British Columbia, Canada. Linked with this expansion is the potential domination of the ginseng industry by agricultural corporations. In the interior of British Columbia, the native deciduous forest environment of eastern North America is simulated with elevated polypropylene shade and a surface covering of straw mulch. The architecture of these environments is designed to permit maximum machinery usage and to minimize labor requirements. Further, with only a four- years growth cycle, plant densities in the gardens are high. In this hot, semi-arid environment, producers believe they have a competitive advantage over other regions in North America because of the low precipitation rates. This helps to minimize atmospheric humidity such that the conditions for fungal disease development are reduced. If soil moisture level become limited, supplemental water can be provided by irrigation. The nature of the radiation and energy balance regimes of the shade and many environments promotes high soil moisture levels. Also, the modified environment redlines soil heating. This can result in an aerial environment for the plant that is stressful and a rooting zone environment that is suloptimal. The challenge of further refining the man modified environment for enhanced plant growth and health still remains. Keywords Panax ginseng, Panax quinquefolium, cultivation, ginseng production.

EFFECTS OF PANAX GINSENG SAPONINS ON CHEMICAL MEDIATOR RELEASE FROM AIRWAY SMOOTH MUSCLE IN ACTIVELY SENSITIZED GUINEA PIG

  • It has been reported that ginseng is effective in the central nervous system, immune system, and the strong inflammatory responses. However, there has been no research report yet about the effect of ginseng on allergic hypersensitivity reactivity. To confirm the ginseng effects on the release of mediators(histamine. leukotrienes etc.) which cause the hypersensitivity reactivity and inflammatory response, we used actively sensitized guinea pig airway tissues by utilizing the superfusion technique. In this procedure. the contractile response and mediators released after antigen stimulation of sensitized tissues, and IgG and IgE antibody products were measured in sera of immunized animals. Then the results of the controll group were compared to those of ginseng pretreatment groups. In the total saponin(TS) and panaxatriol(PT) pretreatment, histamine release decreased by $20\%$ in the tracheal tissues after active sensitization by ovalbumin(OVA, 10mg/kg), but in the lung parenchyma, histamine release decreased by $40\%.$ Panaxadiol(PD) significantly decreased histamine release by $40\%$ in the both tissues after active sensitization. TS, PT and PD of ginseng poorly blocked leukotrienes (LTs) and prostagrandin $D_2(PGD_2)$ release(less than $10\%$). Ginseng TS and PT had no effect on the serum IgG antibody production by ovalbumin, whereas PD significantly increased serum IgG antibody contents(approximately by 2 times). However, $IgG_1$ antibody products in the serum of guinea pig actively sensitized with ovalbumin after PD pretreatment were decreased, compared to that with ovalbumin alone. IgE antibody production by passive cutaneous anaphylaxis(PCA) titer in the TS pretreatment increased 3 times more than in the absence of TS(PCA titer by PT was not detected). These studies show that some ginseng saponins can in part act to inhibit mediator release in antigen - induced airway smooth muscle by inducing the IgG antibody production which has been changed in the specificity.

한국인삼 양액재배시 배지의 영향 (Effect of Substrate on the Production of Korean Ginseng(Panax ginseng C.A. Meyer) in Nutrient Culture)

  • 한국의 인삼재배시 연작장해로 인한 생산량 감소가 문제가 되고 있다 이를 극복하기 위하여 본 연구에서는 한국인삼 재배를 위하여 양액재배 시스템을 적용하였다. 최적의 양액재배용 배지를 선발하기 위하여 모래, TSK-2 (S+T), 피트모스 (P), 재활용 암면 (RR)과 입상 암면 (GR)등의 다양한 배지를 단독 및 혼합처리구로 사용하였다. RR배지의 경우 전반적인 물리화학적 특성은 기존에 보고된 인삼배지의 토양특성과 유사하였다. S+T배지는 가밀도가 인삼용 토양보다 높았다 인삼의 지상부 생체중은 RR배지와 S+T배지에서 높았다. 인삼 뿌리의 생체중 및 건물중은 RR배지에서 가장 높았다. 인삼의 뿌리의 품질적 측면에서 vitamin C함량은 양액재배로 생육시킨 인삼이 토양에서 생육한 인삼보다 높았으나, crude saponin과 ginsenosides 함량에서는 차이를 나타내지 않았다.

Ginseng Saponin Prevents the LPS-induced TNE-$\alpha$ Production in Mice

  • 사포닌은 인삼의 약리작용을 매개하는 인삼의 주요 성분이다. 인삼의 뿌리는 여러 가지 독소에 의하여 유발되는 독성에 대한 보호작용이 있는 것으로 알려지고 있다. 본 연구에서는 세균독소(lipopolysaccharides)에 의하여 유도되는 종양괴사인자 생성에 미치는 인삼 사포닌의 영향을 살펴보았다. 독소를 투여한 생쥐의 혈청중 종양괴사인자의 농도는 상승하는데, 인삼 사포닌 50 mg/kg을 독소와 함께 복강으로 투여한 경우는 독소만 투여하였을 때에 비하여 혈정 중 종양괴사인자의 농도가 현저히 낮았다. 이러한 인삼 사포닌의 효과는 사포닌의 용량, 투여방법과 투여시간에 영향을 받았는데, 사포닌을 경구로 투여하거나 독소를 주사한 후에 투여하였을 때에는 종양괴사인자 농도에 유의성 있는 차이를 나타내지 않았다. 이러한 결과는 인삼이 간 손상에 대한 보호작용이 있을 가능성을 시사하였고, 사포닌이 이러한 인삼의 보호효과를 매개하는 활성 성분임을 의미한다.

Nonsaponin fraction of Korean Red Ginseng attenuates cytokine production via inhibition of TLR4 expression

  • Background: Ginsenosides of Korean Red Ginseng extracts (RGE) and its saponin components suppress secretion of inflammasome-mediating cytokines, whereas the nonsaponin fraction (NS) of RGE oppositely stimulates cytokine secretion. Although direct exposure of NS to macrophages in mice induces cytokine production, oral administration of NS has not been studied in inflammasome-related disease in animal models. Methods: Mice were fed RGE or NS for 7 days and then developed peritonitis. Peritoneal cytokines were measured, and peritoneal exudate cells (PECs) were collected to assay expression levels of a set of toll-like receptors (TLRs) and cytokines in response to NS ingestion. In addition, the role of intestinal bacteria in NS-fed mice was assessed. The effect of preexposure to NS in bone marrow-derived macrophages (BMDMs) on cytokine production was further confirmed. Results: NS ingestion attenuated secretion of peritoneal cytokines resulting from peritonitis. In addition, the isolated PECs from NS-fed mice presented lower TLR transcription levels than PECs from control diet-fed mice. BMDMs treated with NS showed downregulation of TLR4 mRNA and protein expression, which was mediated by the $TLR4-MyD88-NF{\kappa}B$ signal pathway. BMDMs pretreated with NS produced less cytokines in response to TLR4 ligands. Conclusion: NS administration directly inhibits TLR4 expression in inflammatory cells such as macrophages, thereby reducing secretion of cytokines during peritonitis.

Total Saponin from Korean Red Ginseng Inhibits Thromboxane A2 Production Associated Microsomal Enzyme Activity in Platelets

  • Ginseng, the root of Panax ginseng Meyer, has been used frequently in traditional oriental medicine and is popular globally. Ginsenosides, which are the saponins in ginseng, are the major components having pharmacological and biological activities, including anti-diabetic and anti-tumor activities. In this study, we investigated the effects of total saponin from Korean red ginseng(TSKRG) on thrombin-produced thromboxane $A_2$ ($TXA_2$), an aggregating thrombogenic molecule, and its associated microsomal enzymes cyclooxygenase (COX)-1 and $TXA_2$ synthase (TXAS). Thrombin (0.5 U/mL) increased $TXA_2$ production up to 169 ng/$10^8$ platelets as compared with control (0.2 ng/$10^8$ platelets). However, TSKRG inhibited potently $TXA_2$ production to the control level in a dose-dependent manner, which was associated with the strong inhibition of COX-1 and TXAS activities in platelet microsomes having cytochrome c reductase activity. The results demonstrate TSKRG is a beneficial traditional oriental medicine in platelet-mediated thrombotic diseases via suppression of COX-1 and TXAS to inhibit production of $TXA_2$.

Effect of Inoculum Size on Biomass Accumulation and Ginsenoside Production by Large-Scale Cell Suspension Cultures of Panax ginseng

  • Cell growth and ginseng saponin production by large-scale suspension (bioreactor) cultures of Panax ginseng were investigated under various inoculum sizes. Cell growth was low at an inoculum size of 40 g FW/L, and the maximum cell growth was obtained with increasing inoculum size up to 100 g FW/L. The cell density of 333 g FW/L and 12.7 g DW/L was obtained at inoculum size of 100 g FW/L after 30 days of cultivation. Maximum saponin production of $4.40\;\cal{mg/g}$ DW was achieved at 60 g FW/L of inoculum size. Thus, inoculum size 60 g FW/L was suitable for optimum biomass accumulation as well as saponin production during bioreactor cultivation of ginseng suspension cells.

인삼의 구증구포에 의한 Ginsenoside의 성분변화 및 BACE-1 억제효과 (Conversion of Ginsenosides by 9 Repetitive Steamings and Dryings Process of Korean Ginseng Root and Its Inhibition of BACE-1 Activity)

  • Red ginseng possibly has new ingredients converted during steaming and dry process from fresh ginseng. Kujeungkupo method which means 9 repetitive steamings and dryings process was used for the production of red ginseng from 6-year old ginseng roots. Saponin was extracted from each red ginseng produced at the 1st, 3rd, 5th, 7th, and 9th during the steaming and drying treatment, and we analyzed saponin content with TLC. Minor saponins, such as ginsenoside-Rg3, -Rh2, compound K, and F2, increased as the process time of steaming and drying, but major saponins (ginsenoside-Rb1, -Rb2, -Rc, -Rd, -Re, -Rf, -Rg1) were decreased. Major saponins were yet observed almost at the 1st process, then degraded as the increasing time of steaming and drying process. Especially, ginsenoside-Re and -Rg were observed as considerable amount after the 1st treatment, but there were no trace of them after the 9th treatment. Ginsenoside-Rg1, -Rb2, and -Rb1 were also reduced remarkedly by 96.6%, 96%, and 92.3%, respectively. Minor saponins were increased significantly, especially for ginsenoside-Rg3 and ginsenoside-F2. These results suggest that Kujeungkupo method is the very useful method for the production of minor ginsenoside-Rg3 and -Rh2.
