Korean red ginseng water extract produces antidepressant-like effects through involving monoamines and brain-derived neurotrophic factor in rats

  • Background: Ginseng Radix (Panax ginseng Meyer, Araliaceae) has been used medicinally to treat the brain and nervous system problems worldwide. Recent studies have revealed physiological effects that could potentially benefit cognitive performance or mood. The present study aimed to investigate the antidepressant effects of Korean red ginseng water extract (KGE) and its active component in an unpredictable chronic mild stress (UCMS)-induced animal model and elucidate the underlying mechanisms. Methods: The antidepressant potential of the UCMS model was evaluated using the sucrose preference test and open field tests. The behavioral findings were further corroborated by the assessment of neurotransmitters and their metabolites from the prefrontal cortex and hippocampus of rats. Three doses of KGE (50, 100, and 200 mg/kg) were orally administered during the experiment. Furthermore, the mechanism underlying the antidepressant-like action of KGE was examined by measuring the levels of brain-derived neurotrophic factor (BDNF)/CREB, nuclear factor erythroid 2-related factor 2 (Nrf2), and Kelch-like ECH-associated protein 1 (Keap1) proteins in the prefrontal cortex of UCMS-exposed rats. Results: KGE treatment normalized UCMS-induced depression-related behaviors. Neurotransmitter studies conducted after completing behavioral experiments demonstrated that KGE caused a reduction in the ratio of serotonin and dopamine, indicating a decrease in serotonin and dopamine turnover. Moreover, the expression of BDNF, Nrf2, Keap1 and AKT were markedly increased by KGE in the prefrontal cortex of depressed rats. Conclusion: Our results provide evidence that KGE and its constituents exert antidepressant effects that mediate the dopaminergic and serotonergic systems and expression of BDNF protein in an animal model.

Anti-wrinkle Effect by Ginsenoside Rg3 Derived from Ginseng (인삼유래 Ginsenoside Rg3에 의한 항-주름 효과)

  • The root of Panax ginseng C. A. Meyer has been used as a traditional anti-aging and anti-wrinkle agent in the Orient. However, it is still unknown which component of ginseng is effective at suppressing wrinkle formation. Recently at least twenty ginsenosides regarded as the main active ingredients of ginseng have been isolated. Among them, we examined the effect of ginsenoside Rg3 on dermal ECM metabolism to elucidate the mechanism of anti-wrinkle by ginseng. In our study, to investigate the anti-wrinkle effect of the ginsenoside Rg3, ECM component and growth factor in dennis were evaluated by ELISA assay. Ginsenoside Rg3 was found to stimulate type I procollagen and fibronectin (FN) biosynthesis in a dose-dependent manner in normal human fibroblast culture (p < 0.05, n =3), and dose-dependently enhance TGF- ${\beta}$1 level (p < 0.05, n =3). In RT-PCR analysis mRNA level of c-Jun, a member of AP-1 transcription factor, was reduced by ginsenoside Rg3 in normal human fibroblast culture. These results indicate that ginsenoside Rg3 stimulates type I collagen and FN synthesis through the changes of TGF - ${\beta}$1 and AP-1 expression in fibroblasts.

Analysis of Serum Proteom after Intravenous Injection of cultivated wild ginseng pharmacopuncture (산양산삼 증류약침의 혈맥주입 후 나타나는 혈장의 Proteom 분석)

  • Objectives : To observe the changes in the serum proteins after intravenous injection of cultivated wild ginseng pharmacopuncture. Methods : Blood was collected before and after the administration of cultivated wild ginseng pharmacopuncture and only the serum was taken. Then differences in the spots on the scanned image after carrying out 2-Dimensional electrophoresis were located and conducted mass analysis and protein identification. Results : Following results were obtained from the comparative analysis of serum proteins before and after the administration of cultivated wild ginseng pharmacopuncture. 1. 28 spots were identified before and after the administration. 2. In confirming manifestation degree, spots with more than two-times increase were 204, 1302, 2205, 3105, 7104, 8006, spots with more than one-time increase were 1101, 1505, 2013, 2403, 3009, 3010, 4002, 4009, 6704, 8101, and spots with decrease were 205, 801, 803, 3205, 5202, 6105, 6106, 7103, 9001, 9003. 3. After conducting protein identification, proteins 205, 804, 1302, 4009, 6105, 6106 are unidentified yet, and 1l01 is unnamed protein. Protein 204 is identified as complement receptor CR2-C3d, 801 as YAPl protein, 803 as antitrypsin polymer, 1505 as PRO0684, 2013 and 3010 as proapolipoprotein, 2205 as USP48, 2403 as vitamin D binding protein, 3009 as complement component 4A preprotein, 3105 as immunoglobulin lambda chain, 3205 as transthyretin, 4002 as Ras-related protein Ral-A, 4204 as beta actin, 5202 and 7104 as apolipoprotein Ll, 6704 as alpha 2 macroglobulin precursor, 7103 as complement component 3 precursor, 8006 as testis-specific protein Y, 8101 as transferrin, 9001 as (Alpha-Oxy, Beta-(Cl12g)deoxy) T-State Human Hemoglobin, and 9003 as human hemoglobin. 4. Immune protein CR2-C3d(204), which acts against microbes and pathogenic organisms, was increased by more than two-times after the administration of pharmacopuncture. 5. Antitrypsin(803), which is secreted with inflammatory response in the lungs, was reduced after the administration of pharmacopuncture. 6. Proapolipoprotein(2013, 3010) and apolipoprotein(7104), key components of the HDL-cholesterol which plays an important role in preventing arteriosclerosis, were increased after the administration of pharmacopuncture. 7. Vitamin D binding protein(DBP, 2403), protecting the lung at the time of inflammatory response, was increased after the administration of pharmacopuncture. 8. Transthyretin(TTR, 3205), which is the main protein causing familial amyloid polyneuropathy(FAP), was decreased after the administration of pharmacopuncture. 9. Ras-related protein Ral-A(4002) that controls phospholipid metabolism, cytoskeletal formation, and membrane traffic, was increased after the administration of pharmacopuncture. 10. Testis-specific protein Y(8006), which takes part in determination of the gender, was increased by more than two-times after the administration of pharmacopuncture. 11. Transferrin(8101), which balances the iron level in the body, was increased after the administration of pharmacopuncture. Conclusion : Above results support the notion that intravenous injection of cultivated wild ginseng pharmacopuncture induce changes in serum proteins and this research can be a pioneer work in finding biomarkers.

Effect of Korean red ginseng on collagen biosynthesis and MMP-I activity in human dermal fibroblast (홍삼성분이 섬유아세포의 콜라겐 생합성과 MMP-1 활성에 미치는 영향)

  • This study was carried out to develop health & functional food by using Korean red ginseng for prevention of skin wrinkles. Effects of Korean red ginseng on the collagen biosynthesis and inhibition of matrix metalloproteinase-I (MMP-1) activity in human dermal fibroblast were investigated. Crude saponin contents of Korean red ginseng water extract (WE), Korean red ginseng ethanol extracts (EE) and Korean Red ginseng purified extracts (PE) were 72 mg/g, 107 mg/g and 220 mg/g, respectively. We incubated human fibroblast cell with Korean red ginseng component by addition of l ${\mu}g/ml$, 5 ${\mu}g/ml$, 10 ${\mu}g/ml$. Amount of collagen biosynthesis was 1.86 ng/ml in control sample and 2.85 ng/ml, 2.05 ng/ml and 2.58 ng/ml in retinoic acid, EE and PE respectively. Furthermore, $ginsenoside-Rg_1$ and $ginsenoside-Rb_1$ were shown 2.01 ng/ml and 3.07 ng/ml. MMP-l activities of EE, PE, $ginsenoside-Rg_1$ and $ginsenoside-Rb_1$ were decreased to 92%, 94%, 91% and 78% respectively as compared with control. Cell proliferation were showed 84-96% in the Korean red ginseng components. The antioxidative SOD activities of the Korean red ginseng components were showed 28-69%, however it was lower than that of Vitamin C. From this results, we conclude that Korean red ginseng have a anti-wrinkle effect and $ginsenoside-Rb_1$ may be considered as a more effective component.

Effects of Repetitive using Lime Bordeaux Mixture in the Copper Concentration of the Soil and Ginseng Root (석회보르도액 반복 사용이 토양과 인삼 체내 구리농축에 미치는 영향)

  • Three type of Lime Bordeaux mixture (LBM) that has been used since 1800's for control plant disease are used for eco-friendly ginseng (Panax ginseng) disease control. But it was restricted the use in the crops cultivation in some countries of Europe recently, because there is a possibility that the copper component is concentrated in the soil and plants with using LBM containing copper. According to the concentration and number of LBM spraying treatment, it was investigated copper and other components in soil and ginseng root. In case of LBM sprayed 33 times for three years, copper concentration was increased up to 75.9 ppm in the soil. However copper concentration of ginseng root was increased with 9.9~23.0 mg/kg in comparison with 8.38~11.39 mg/kg at LBM non-treatment. It has shown that the copper components can be concentrated to in the soil if used continuously in the long term.

Effect of White and Red Ginseng Extracts on the Immunological Activities in Lymphocytes Isolated from Sasang Constitution Blood Cells (백삼 및 홍삼 추출물의 사상체질별 면역세포 활성효과)

  • The effects of white ginseng and red ginseng ethanol extracts on the proliferation, nitric oxide (NO) production and tumor necrosis factor-$\alpha$ (TNF-$\alpha$) in human Band T lymphocytes isolated from blood cell of sasang constitution were examined in vitro. White ginseng extract known as medicine suitable for soeumin constitution showed the highest immune activities such as proliferation, NO production and TNF-$\alpha$ in soeumin constitution, and followed by taeeumin and soyangin, respectively (P<0.05). However, there were immunologically insignificant differences among sasang constitution lymphocytes treated by red ginseng ethanol extract. Both white and red ginseng ethanol extracts showed significantly higher immunological activities than LPS-induced sasang constitution lymphocytes on NO production and TNF-$\alpha$ release (P<0.05). The different immunological activities of white and red ginseng extracts in sasang constitution lymphocytes might be due to the component changes during the processing of ginseng. Thus, the findings of components suitable for all constitutions from red ginseng extract could be applied as a new constitution marker.

Analysis of Aroma Pattern of Panax Species by Potable Handheld Gas Chromatograph (Potable handheld gas chromatograph(PHGC)를 이용한 인삼속(Panax species) 식물들의 향기패턴 분석)

  • This study was performed to analyze aroma pattern of Panax species (Korean Panax ginseng C.A. Meyer, Chinese Panax ginseng C.A. Meyer, Panax quinquefolium L, and Panax notoginseng F.H. Chen) by the PHGC (potable handheld gas chromatograph). Ratios of several peak areas in chromatogram of derivative parrtern were as follows. If ratio of Korean Panax ginseng was 1, Panax notoginseng was $0.030{\sim}0.674$, Chinese Panax ginseng was $0.005{\sim}0.212$ and panax quinquefolium was $0.241{\sim}0.871$. Ratios of peak area at $Rt_{20.02}$ were that if Korean panax ginseng was 1, Chinese Panax ginseng was 0.212, Panax quinquefolium was 0.343 and Panax notoginseng was 0.065. Ratios also of peak area at $Rt_{21.70}\;and\;Rt_{24.90}$ showed clear difference among aroma patterns of Panax specie cultivars. Flavor component at $Rt_{26.15}$ was not detected in Panax quinquefolium and Panax notoginseng but in Korean Panax ginseng and Chinese Panax ginseng. Ratios of peak area at $Rt_{26.15}$ were that if Korean Panax ginseng was 1, Chinese Panax ginseng was 0.185. And so habitat of Panax species cultivars was discriminated. Cultivar and habitat of dried panax species was remarkably distinguised by the chromatogram of frequency pattern, derivative pattern and visual pattern using olfactory images known as Vapor $print^{TM}$.

Maltol, an Antioxidant Component of Korean Red Ginseng, Shows Little Prooxidant Activity

  • Some antioxidant phenolic compounds exhibit prooxidant activity mainly due to their abilities to reduce $Fe^{3+}\; to\; Fe^{2+}.$ Reducing ability and prooxidant activity of maltol, an antioxidant component of Korean red ginseng, were compared with those of pyrogallol. Maltol at 2 mM did not appreciably reduce$ Fe^{3+}\; to\; Fe^{2+}$ and also failed to reduce nitroblue tetrazolium. Stimulation of hydroxyl radical mediated-deoxyribose degradation by pyrogallol was maximal at 60 .mu.M. Maltol stimulated the deoxyribose degradation to a much less extent, and a similar stimulatory effect was observed at a concentration of more than 100-fold higher than that of pyrogallol. The stimulatory effect of maltol reached a plateau over 1 mM, suggesting the removal of hydroxyl radicals by excess maltol. In bleomycin-$Fe^{3+}$-DNA assay, maltol at 2 mM produced a 2.5-fold increase of the iron-bleomycin-dependent DNA degradation over the basal value, whereas pyrogallol at 10 .mu.M accelerated DNA degradation by ca. 10-fold. Furthermore, maltol inhibited $Fe^{2+}$-stimulated DNA degradation by bleomycin. These results strongly suggested that maltol is an antioxidant with little prooxidant activity.

Bioactive Components and Volatile Compounds According to Illite Addition in Saururus chinensis Baill Cultivation (일라이트 시용량에 따른 삼백초 잎의 유효성분 및 향기성분 함량 비교)

  • This study was carried to investigate the effect of illite addition on the growth, bioactive components and volatile compounds of leaf on Saururus chinensis Baill cultivation in greenhouse. Illite addition on Saururus chinensis Baill cultivation resulted no significant effects on the aerial part. However, the root part was highest in 5% illite addition group. Crude oil and ash amount increased as illite additives was increased. Ca of inorganics and free sugars contents were highest in 5% and 20% illite addition, respectively. Hyperoside and isoquercetin of available component on leaf were highest in 5% illite addition group, 4.02mg/g and 4.31mg/g, respectively. The volatile compounds in Saururus chinensis Baill leaf cultured with illite addition amounts were isolated by solid-phase microextraction fiber (polydimethysiloxane $65{\mu}m$) and identifed by gas chromatogtaphy mass spectrometry. As the results, the 22 volatile compounds were identified from in Saururus chinensis Baill leaf and major volatile compounds were the ${\alpha}$-cadinol (18.50%), myristicin (16.46%), methyl-9-methyl-tetradecanoate (10.22%), and ${\gamma}$-muurolene (9.75%). Especially, the content of ${\alpha}$-cadinol was highest in 5% illite addition group and ${\gamma}$-muurolene on overall illite addition group was lower than no addition group.

Effect of Korean Effective Microorganisms and Seafood Amino Acid Fertilizer on the Root Quality of Panax ginseng (해양부산물 아미노산액비 및 유용미생물시용이 인삼의 품질에 미치는 영향)

  • Contents of ginsenosides 7 subordinations of two-year ginseng (fresh ginseng) is 1.27% and three-year ginseng is 2.09%, so the three-year ginseng root increased 64.9% compared to the two-year root. Compared with the comparison group, ginsenosides component content of KEM+SAF-applied group increased 24% in case of the two-year root and 20% in case of the three-year root. In vitamin C content, two-year root showed 59.4% higher and three-year root showed 37.7% higher in KEM+SAF applied group compared with the comparison group. In case of vitamin E, the two-year root indicated 5.6% higher and three-year root indicated 1.5% higher in KEM+SAF applied group compared with the comparison group, but there is no significant difference. In phytosterol three components (campesterol, stigmasterol, sitosterol), two-year root showed 25.3, 3.6, 14.1% higher for each, and three-year root showed 23.6, 6.8, 12.9% higher in KEM+SAF applied group and 14.4% was higher on average. In DPPH, two-year root indicated 34.4% higher and three-year root indicated 42.4% higher in KEM+SAF applied group compared to the comparison group. To sum up the results, KEM+SAF applied group showed (1)22% ginsenosides components content, (2)48.6% vitamin C content, (3)3.6% vitamin E content, (4)14.4% phytosterol content, (5)38.4% DPPH higher averagely compared to the comparison group.
