The Action of Ginkgo Bibloba Extract in the Isolated Rabbit Corpus Cavernosum

  • The extract of Ginkgo bibloba (EGb) is a complex mixture of natural products from the Ginkgo leaves and clinically used for the treatment of cerebral and peripheral circulatory disturbances due to its combined activity of several vasoactive principles. In this study we investigated the action of EGb and its mechanism in isolated rabbit corporal smooth muscle to evaluate the possibility of using this material as a pharmacoerecting agent. Strips of rabbit corpus cavernosum were mounted in organ chambers to measure isometric tension. EGb began to exert an relaxing effect at 1 mg/ml in the submaximally precontracted muscle strips with phenylephrine $(PHE,\;5{\times}10^{-6}\;M)$; causing concentration-dependent relaxation with maximal effect at $3{\sim}5\;mg/ml$. That relaxation was partially inhibited by removal of the smooth muscle endothelium or by pretreatment with a NO scavenger, pyrogallol $(10^{-4}\;M)$ or the guanylate cyclase inhibitor, methylene blue $(10^{-4}\;M)$. Pretreatment with EGb (3 mg/ml) inhibited PHE- $(5{\times}10^{-6}\;M)$ or KCI- (20 and 40 mM) induced contraction of muscle strip. In calcium-free high potassium solution EGb depressed the basal tone of the depolarized muscle strip and inhibited calcium-induced contraction when $CaCl_2$ $(10^{-4}\;M)$ was added. These results suggest that EGb relaxes rabbit corpus cavernosal smooth muscle through multiple action mechanisms that include increasing the release of nitric oxide from the corporal sinusoidal endothelium, sequestration of intracytosolic calcium, and maybe a hyperpolarizing action.

은행(Ginkgo biloba L.) 종실 및 잎 추출물의 항산화 효과에 관하여 (Antioxidative Effectiveness of Extract of Nut and Leaf of Ginkgo biloba L.)

  • 탈지 은행 종실 및 잎에서 유리페놀산, 에스터형 및 불용성 페놀산을 추출하여 식용 옥수수유 기질에서 항산화 효과를 비교하고자 0.02%(w/w)의 BHA, BHT와 각 추출물 $1m{\ell}$(2% w/w)를 넣어 $45{\pm}1^{\circ}C$의 항온기에서 45일간 저장하면서 매 5일 간격으로 과산화물가, TBA가를 측정하여 다음과 같이 항산화 효과를 추정하였다. BHA, BHT와 은행 종실의 유리페놀산, 에스터형 및 활용성 페놀산, 잎의 유리페놀산, 에스터형 및 불용성 페놀산 추출물을 첨가한 시험구와 대조구의 45일 저장후 과산화물가는 각각 133, 52, 115, 190, 127, 95, 140, 121, 280meq/kg, oil이었다. 한편 같은 조건하에서 각 항산화성 물질의 TBA가는 0.153, 0.059, 0.175, 0.260, 0.187, 0.160, 0.1 74, 0.195 0.430이었다. G.C.로 분리 확인된 페놀산은 은행 종실 및 잎 공히 Vanillin, ${\rho}$-Hydroxybenzoic acid, Syringic acid, Gallic acid, Protocatechuic acid가 확인되었고 은행 종실에서는 trans-Cinnamic acid, Caffeic acid가, 잎에서는 Coumaric acid, Gentisic acid, Phloroglucinol, Pyrogallol이 각각 확인되었다. 이상의 결과를 종합해 볼 때 은행 종실 및 잎의 페놀산은 식용 옥수수유 기질에서 우수한 항산화 효과가 있는 것으로 사료된다.

  • Acaricide effect against Tetranichus urticae, Myzus persica, Spodoptera litura and Plutella xylostella with the Ginkgo biloba leaves extracts was reviewed. This result was motality 74.3%, 88.7% respectively, after tretment 24hr, 72hr about $80^{\circ}C$ water extract(GLW80, 1%) of Tetranichus urticae and the insecticide took effect showed its effect in some 50% against the other three. But, motality of Tetranichus urticae against 10% GLW80 showed its 90.6%, 98.3% after tretment 24hr and 48hr.

  • The antimicrobial effect of the Ginkgo biloba leaves extracts, Wood flavor and 1% sorbic for the total microorganics in beef examined by serial dilution method. As a result of using GLEA(T20), GLW80, Wood flavor and 1% sorbic acid by serial dilution method, it was revealed that wood flavor is most antibiotic. The decreased of wood flavor 82.80%, GLEA(T20) 73.44%, sorbic acid 66.33%, GLW80 64.36%, was compared with not treated.

  • We investigated immunoactivity of ginkgolides (GLS), the primary active constituent of Ginkgo biloba leaves, against disseminated candidiasis due to Candida albicans. This fungus is a polymorphic opportunistic pathogen. BALB/c mice were induced neutropenia by intraperitoneal (i.p.) injection of cyclophosphamide (CP) 24 hours before an i.p administration of GLS (2 mg/mouse) to the mice. Control mice received diluent (Dulbecco's phosphate saline solution; DPBS) instead of GLS. (omitted)

  • The major active components of EGb 761, extract of Ginkgo biloba leaves, include flavonoid glycosides and unique diterpenes known as ginkgolides. Ginkgolides are potent inhibitors of platelet activating factor. In this study, we investigated antiinflammatory activity of ginkgolides on the Complete Freund"s Adjuvant (CFA)-induced mice. Inhibitory Effect of Ginkgolide B on Platelet Aggregation in a cAMP- and cGMP-dependent Manner by Activated MMP-9

  • Extracts from the leaves of the Ginkgo biloba are becoming increasingly popular as a treatment that is claimed to reduce atherosclerosis, coronary artery disease, and thrombosis. In this study, the effect of ginkgolide B (GB) from Ginkgo biloba leaves in collagen (10 ${\mu}g/ml$)-stimulated platelet aggregation was investigated. It has been known that human platelets release matrix metallo-proteinase-9 (MMP-9), and that it significantly inhibited platelet aggregation stimulated by collagen. Zymographic analysis confirmed that pro-MMP-9 (92-kDa) was activated by GB to form an MMP-9 (86-kDa) on gelatinolytic activities. And then, activated MMP-9 by GB dose-dependently inhibited platelet aggregation, intracellular $Ca^{2+}$ mobilization, and thromboxane $A_2$ ($TXA_2$) formation in collagen-stimulated platelets. Activated MMP-9 by GB directly affects down-regulations of cyclooxygenase-1 (COX-1) or $TXA_2$ synthase in a cell free system. In addition, activated MMP-9 significantly increased the formation of cyclic adenosine monophosphate (cAMP) and cyclic guanosine monophosphate (cGMP), which have the anti-platelet function in resting and collagen-stimulated platelets. Therefore, we suggest that activated MMP-9 by GB may increase the intracellular cAMP and cGMP production, inhibit the intracellular $Ca^{2+}$ mobilization and $TXA_2$ production, thereby leading to inhibition of platelet aggregation. These results strongly indicate that activated MMP-9 is a potent inhibitor of collagen-stimulated platelet aggregation. It may act a crucial role as a negative regulator during platelet activation.

천안시 가로수의 생리적 특성과 형성층 전기저항치에 관한 연구1 (Studies on the Physiological Characteristics and Cambial Electrical Resistance of Street Trees in Cheonan City)

  • 천안시내에 식재된 가로수의 건강도를 진단하기 위하여 몇 가지 생리적 특성을 측정하고, 형성층 전기저항치와 생리적 특성들간의 관계를 밝히고자 하였다. 은행나무, 벚나무, 버드나무를 대상수종으로 선정하였고. 7월에 대상 수종의 하부 토양을 채취하여 탈수소효소 활성을 분석하였으며, 수관의 잎은 엽록소 함량, 질산환원 효소, 항산화효소(SOD) 활성을 분석하는데 이용하였다. 수목의 형성층 전기전항치는 5월, 7월, 9월에 측정하였다. 토양의 탈수소효소 활성은 도심지역에서 낮은 값을 나타내 토양 오염도를 반영하였으며. 엽록소 함량은 동서대로에 식재된 벚나무가 가장 낮았다. 은행나무의 질산환원효소 활성은 다른 수종보다 높았으며 도심지역에서 높은 활성을 나타냈다. 또한 항산화효소인 SOD 활성은 도심에서 낮았다. 동서대로의 벚나무는 형성층 전기저항치가 지속적으로 상승하여 활력을 상실하였음을 보여주었다. 형성층 전기저항치는 질산환원효소($r^2$=-0.566) 및 SOD활성($r^2$=0.579)과 상관을 보임으로써 수목의 건강도 및 생리적 건강 상태를 진단하는데 유용할 것으로 판단되었다.

Terpene-Strengthened Ginkgo biloba Extract as a Platelet-Activating Factor Antagonist

  • Since platelet-activating factor (PAF) is involved in inflammation, allergic response and anaphylactic shock, PAF receptor antagonists may have potential for controlling these disease conditions. The extract of the leaves of Ginkgo biloba having a higher content of terpenoids (12%) with flavonoids (24%) (YY1224) was prepared in order to obtain the increasing PAF antagonistic activity. As expected, YY1224 showed a higher PAF antagonistic binding affinity ($IC_{50}\;=\;0.09\;{\mu}g/ml$) using $[^3H]PAF$ and rabbit platelets as ligand and receptor source, compared with an $IC_{50}$ of $>\;100\;{\mu}g/ml$ by Egb 761, a standardized extract. YY1224 also showed a higher inhibitory activity against PAF-induced platelet aggregation and NO production from lipopolysaccharide-treated RAW 264.7 cells. In addition, it protected PAF-induced death in mice by oral administration at 15 mg/kg. All these results suggest that YY1224 may show favorable effects on PAF-related disorders.

은행잎 추출물 유래 Ginkgetin 및 Isoginkgetin의 융복합 특성 연구 (Physicochemical properties of Ginkgetin and Isoginkgetin extracted from Ginko leaves)

  • 은행잎의 Biflavonoid인 ginkgetin과 isoginkgetin을 화장품 소재로 이용 가능성 여부를 확인하기 위해 이를 분리하여 화학 구조 및 물리화학적 특성을 규명하고자 하였다. 음건한 은행나무 단풍잎 1 kg을 순차적으로 ethanol, n-hexane, ethylacetate 및 normal butyl alcohol로 분획 추출하여 최종 ginkgetin과 isoginkgetin을 확보하였다. 두 화합물은 모두 HPLC를 통해 99% 이상 고 순도로 나타났고 MS spectrum을 통한 분자량은 566, 분자식은 $C_{32}H_{22}O_{10}$로 동일한 이성질체적 구조를 나타냈다. UV spectrum으로 flavonoid의 골격과 NMR로 aromatic proton의 존재와 탄소 수 및 구성비를 측정한 결과 문헌에 보고된 flavonoid 화합물의 정보와 일치하였다.
