Antioxidant Characteristics and Phytoremediation Potential of 27 Taxa of Roadside Trees at Industrial Complex Area (공단지역에서 생육하는 가로수 27종의 항산화특성과 잠재적 환경정화능력)

  • In order to screen for the best species for mitigating air pollutants by plants at an industrial complex area, we investigated antioxidant capacity, lipid peroxidation and nitrogen content in the leaves of 27 taxa of woody plants that are mostly utilized as roadside trees. Among 27 taxa, the highest value of antioxidant capacity was given by Cedrus deodara (91.4%) and the lowest one was by Firmiana simplex (56.9%). At lipid peroxidation level, little malondialdehyde (MDA) was observed in Lagerstroemia indica and Ginkgo biloba, but Platanus occidentalis, Castanoposis cuspidata var, sieboldii, Machilus thunbergii and Juniperus chinensis showed high MDA content. Antioxidant capacity of the deciduous woody plants was not significantly different in comparison with that of the evergreen ones. But MDA content of the deciduous woody plants was lower than that of the evergreen ones. The 27 taxa of woody plants appeared to be classified into four types: those of high antioxidant capacity and low lipid peroxidation, those of high antioxidant capacity and high lipid peroxidation, those of low antioxidant capacity and low lipid peroxidation, and those of low antioxidant capacity and high lipid peroxidation. The taxa included in these types are 7 (first type), 6 (second one), 8 (third one) and 6 (fourth one) taxa. first or second type species which have a high antioxidant capacity represented low nitrogen content in their leaves. However, third or forth type species which have low antioxidant capacity showed high nitrogen content in their leaves. Metasequoia glyptostroboides, Platycarya strobilacra and P. occidentalis which belong to the first or second type had extraordinarily high antioxidant capacity and high nitrogen content. Thus, three species are considered to be good phytoremediators for an industrial complex area.

Effect of $SO_2$ on Leaf Tip Blight of Ginkgo biloba caused by Coniothyrium sp. (은행나무의 잎끝마름병에 미치는 $SO_2$ 의 영향)

  • To examine the relationship between air pollution and occurrence of disease in plants, we investigated the alteration of physiology and biochemistry of Ginkgo biloba by inoculating with Coniothyrium sp. and fumigating with 0.1 ${\mu}l/l\;SO_{2-}$ Visual damage did not appear but photosynthesis, $CO_2$ use efficiency(CUE) and water use efficiency(WUE) were reduced when G. biloba was exposed to $SO_{2-}$for 7days (8 hours a day). When inoculated with Coniothyrium sp., the seventies of damage in G. biloba showed a threefold difference between $SO_{2-}$ treatment (SI) and $SO_{2-}$ free treatment (SFI) at day 42 from initial inoculation. Little difference was observed in sugar contents that may be used pathogens feed, among control, SFI and SI. In spite of the reduction in photosynthetic rate, sugar contents and CUE were maintained. WUE was enhanced 13% more at SI than SFI. The $CO_2$fixation boosted because of enhanced WUE, and thus sugar synthesis was not affected. In addition, sugar transport seems to be retarded for some internal alteration. Consequently, the severity of SI was more serious than that of SFI because Coniothyrium sp. easily invades into the physical texture of G. biloba weakened by $SO_2$ fumigation and because sugar was accumulated in leaves of G. biloba.

Foliar Fertilization Effect of Environmentally-Friendly Organic Agricultural Materials for Grape Cultivation (포도재배를 위한 친환경 유기농자재의 엽면시비 효과)

  • This study was conducted to investigate foliar treatment effects of organic agricultural materials for the environmentally-friendly cultivation of grape. The organic materials applied were chitosan, wood vinegar (pyroligneous acid), amino acid solution, and ginkgo leaf extract. All the organic materials were relatively strong acidic ranging lower than pH 4.6. when comparing with other organic materials, amino acid solution contained relatively high contents of selected plant nutrients, such as N, P, K, Ca, Mg, Cu, Fe, Mn, and Zn. As comparison of selected soil properties in the grape cultivating field, soil pH values were lower at the harvest stage than at the first stage of grape growing (before treating the organic materials), and electrical conductivity (EC) and soil organic matter content were higher at the harvest stage in the all plots. The concentrations of available phosphorus increased in most of the plot soils except in control plot (conventional treatment). The concentrations of exchangeable K decreased in the plot treated with ginkgo leaf extract and the control plot. The exchangeable Mg concentrations decreased in soils of all the plots. On the other hand, the concentrations of N and K in the grape leaves were higher with the treatments of chitosan and amino acid solution, P concentrations were higher with the applications of chitosan, wood vinegar and amino acid solution, and Ca and Mg concentrations were higher with chitosan and amino acid solution treatments, respectively, than with others. The yields of grape were higher, $1,581{\sim}1,583kg\;10a^{-1}$, in the control and wood vinegar treatment plots than others. Sugar contents of grape were not different among all the plots.

Injury Responses of Landscape Woody Plants to Air Pollutants - Visible Injury and Ethylene Production - (조경수목(造景樹木)의 대기오염물질(大氣汚染物質)에 대한 피해반응(被害反應)(II) - 엽피해(葉被害)와 Ethylene 발생량(發生量)을 중심으로 -)

  • This study was conducted to investigate sensitivity of tree seedlings to $SO_2$. Visible injury symptoms and changes of ethylene production were investigated in tree seedlings with the fumigation of $SO_2$ in gas chamber 4 hours a day for six days. The symptoms of visible injury did not appear below 0.5ppm level of $SO_2$ exposure but a change of visible injury with the passage of time appeared at 1.5 and 2.5ppm in all seedlings. With the higher the concentration and/or the longer exposure of $SO_2$ the visible injury symptoms on leaves increased in all seedlings. The sensitivity of seedlings to $SO_2$ was the highest in Liriodendron tulipifera followed by Pinus strobus, Ginkgo biloba, Pinus densiflora and Pinus koraiensis. The amount of ethylene production was more at 1.5 and 2.5ppm of $SO_2$ exposure than at 0.5ppm and the peak time of it came faster at higher levels. The amount of ethylene production was significantly different among tree seedlings. It showed a higher at production of ethylene in Liriodendron tulipifera compared to Ginkgo biloba and the ethylene production of Pinus trees to $SO_2$ were the highest in Pinus strobus followed by Pinus densiflora and Pinus koraiensis. In needle of Pinus strobus the ethylene production increased with the increasing rate of visible injury until the injury rate of 40-50% and than decreased with the increasing rate of visible injury since the rate of 50%.

  • The relations of the growth responses of plants, i.e. 4 species of crops, 12 species of roadside trees and 5 species of horticultural plants to cadmium (Cd) to which added soil were studied in pot culture. Growth in dry weight of corn, soybean, barley and wheat plant were decreased with increase of Cd concentration. Damage of corn plant caused by Cd treatment was more or less recovered when it was grown in soil with calcium, but that of other three crops was not recovered when it was with calcium. Although crop plants used here absorbed small amount of Cd through root, Cd content in the shoot was directly proportionate to the concentration of Cd treated to soil. Additions of calcium and sulfur to soil were effectively to change of soil pH, and only of calcium increased exchangeable calcium and cation exchange capacity of its soil. The chlorosis on leaves caused by Cd treatment was observed in 2 species such as Euonymus japonica and Rhododendron yedoense out of 5 species of the horticultural plants, especially at 50ppm of Cd concentration, Euonymus japonica occurred symptoms of chlorosis and difoliation, and at the higher concentration than that the symtoms were sever more and more. At 200 ppm of Cd feeble damage was observed in Pinus koraiensis and Ginkgo biloba but severe chlorosis observed in Robinia pseudoacacia and Sabina chinensis, Buxus koreana, Abies holophylla and Platanus orientalis. Nevertheless those plants had serious damage at 200 ppm of Cd, such symptom was weakened by adding calcium to soil with Cd. There were many Cd tolerant species out of the plants used in thies xperiment, such as Crassula falcata, Chrysanthemum morfolium, Hibiscus syriacus, Ligustrum ovalifolium, Liriodendron tulipeferia, Lespedeza crytobotrys.

Inhibition of cell growth and induction of apoptosis by bilobalide in FaDu human pharyngeal squamous cell carcinoma

  • Bilobalide isolated from the leaves of Ginkgo biloba has several pharmacological activities such as neuroprotective, anti-inflammatory, and anticonvulsant. However, the effect of bilobalide on cancer has not been clearly established. The main purpose of this study was to investigate the effect of bilobalide on cell growth and apoptosis induction in FaDu human pharyngeal squamous cell carcinoma. This was examined by 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide assay, nuclear 4′,6-diamidino-2-phenylindole dihydrochloride staining, DNA fragmentation analysis, and immunoblotting. Bilobalide inhibited the growth of FaDu cells in dose- and time-dependent manners. Treatment with bilobalide resulted in nuclear condensation and DNA fragmentation in FaDu cells. Furthermore, it promoted the proteolytic cleavage of procaspase-3/-7/-8/-9 with increase in the amount of cleaved caspase-3/-7/-8/-9. Bilobalide-induced apoptosis in FaDu cells was mediated by the expression of Fas and the activation of caspase-8, caspase-3, and poly (ADP-ribose) polymerase. Immunoblotting revealed that the antiapoptotic mitochondrial protein Bcl-2 was downregulated, but the proapoptotic protein Bax was upregulated by bilobalide in FaDu cells. Bilobalide significantly increased Bax/Bcl-2 ratio. These results suggest that bilobalide inhibits cell proliferation and induces apoptosis in FaDu human pharyngeal squamous cell carcinoma via both the death receptor-mediated extrinsic apoptotic pathway and the mitochondrial-mediated intrinsic apoptotic pathway.

Injury Responses of Woody Landscape Plants to Air Pollutants(I) -Sulfur and Heavy Metal Content- (조경수목(造景樹木)의 대기오염물질(大氣汚染物質)에 대한 피해반응(被害反應)(I) -엽내유황(葉內硫黃) 및 중금속(重金屬) 함량(含量)을 중심으로-)

  • This study was conducted to investigate the absorbing and/or filtering capacity of woody landscape plants growing in urban and industrial regions of Seoul and Taejon. Korea. Total sulfur content and heavy metal content were analyzed in tree foliage of Pinus densiflora, Pinus koraiensis, Ginkgo biloba. Liriodendron tulipifera, Platanus occidentalis. The total sulfur content in the leaves of woody landscape plants in Seoul and Taejon were remarkably higher than those of the tree leaves in Kwangreung. The total sulfur content of deciduous trees had 2.6 times more than that of coniferous trees. the former had better absorbing capacity of $SO_2$ gas. Especially Ginkgo biloba and Platanus occidentalis were recommanded as absorption trees of $SO_2$ because sulfur content levels of them were the highest among all species and increased until tall coloration. While coniferous trees showed no changes in total sulfur content between four seasons. deciduous trees increased in sulfur during the growing season. Pb content of all species in polluted area were higher than those in unpolluted area. In polluted area, while coniferous trees had higher absorption and accumulation of Pb, deciduous trees had higher absorption of Zn. The content of Pb, Zn and Cu in needle of Pinus koraiensis were more than those of Pinus densiflora, In Pinus koraiensis and Pinus densiflora, heavy metal content in the 2nd year needle were higher than thous in the current year needle. In Pinus densiflora, Pinus koraiensis Liriodendron tulipifera and Platanus occidentalis there was a positive correlation between total sulfur and heavy metals [(Pb+Zn+Cu)/3] content.

Effect of Elevated $CO_2$ Concentration and Temperature on the Growth Response of Several Woody Plants, Including Two Endangered Species ($CO_2$ 농도와 온도증가가 목본성 수종 3종과 멸종위기 2종의 생육반응에 미치는 영향)

  • Increasing global $CO_2$ concentration and temperature is leading to the phenomenon of global warming and impacting the growth of plants. In order to determine the effects of global warming on the woody plants of Korea, five woody species, Pinus densiflora (Korea's dominant species), Ginkgo biloba (a commonly used street tree), Quercus glauca (dominant species in sub-tropical forests), Quercus gilva and Abeliophylum distichum (both endangered species), were grown at control (ambient $CO_2$+ambient temperature) and treatment (elevated $CO_2$+elevated temperature) conditions in a glasshouse, and were monitored for their ecological response. Shoot lengths and number of leaves were measured once a month from April to October in 2010, and were again measured in November 2011. Shoot lengths of P. densiflora, G. biloba and Q. glauca were not significantly affected by elevated $CO_2$ and temperature conditions. However. those of Q. gilva and A. distichum were both higher for plants grown under treatment than for those grown under control. The number of leaves of five woody species was not significantly affected by elevated $CO_2$ and temperature. These results indicate that P. densiflora, G. biloba and Q. glauca react more favorably than Q. gilva and A. distichum (each of which are designated as endangered plants) under global warming situations.

Changes in Chlorophyll Contents of Leaves and pH of the Extracted Solutions from the Leaves of 7 Tree Species by pH Level (7개(個) 수종(樹種)의 잎이 pH수준별(水準別) 처리(處理)에 따른 엽록소(葉綠素) 함량(含量) 및 침출액(浸出液) pH변화(變化))

  • We conducted this study as a fundamental study on the response of various tree species against acid rain. The tree species used for this study were Zelkova serrata, Robinia pseudoacacia, Quercus acutissima, Prunus serrulata, Ginkgo biloba, Pinus koraiensis and Pinus densiflora. The leaves were examined for the pH changes by treatment time and the chlorophyll content into various pH solution in vitro. The results obtained were as follows ; 1. When the leaves were immersed in the solution of various pH(pH 3.0-pH 6.0) levels, the pH were changed to species specific pH ranges as pH 5.0~pH 5.5 of Z. serrata, pH 5.5~pH 6.0 of R. pseudoacacia, pH 4.5~pH 5.0 of Q. acutissima, pH 5.5~pH 6.0 of P. serrulata, pH 3.5~pH 4.5 of G. biloba, pH 3.5~pH 4.5 of P. koraiensis until 48 hours. However, in case of P. densiflora, it was difficult to find specific pH range of the species. Z. serrata, R. pseudoacacia and P. serrulata showed a little pH increasing by pH 2.0 solution treatment, while other species showed no change by the solution. 2. The amount of chlorophyll contents in Z. serrata, R. pseudoacacia and P. serrulata were decreased after immersing in the pH 2.0 solution. Chlorophyll content was almost constant in other pH levels. Other species showed almost constant chlorophyll contents in various pH levels and treatment time.

Diversity of I-SSR Variants in Gingko biloba L. Planted in 6 Regions of Korea (국내(國內) 6개(個) 은행(銀杏)나무 식재지(植栽地)에 있어서 I-SSR 변이체(變異體)의 다양성(多樣性))

  • Genomic DNAs were extracted from the leaves of 182 ginkgo trees (Ginkgo biloba L.) planted in 6 regions and subjected to the analysis of both I-SSR and RAPD markers. A total of 227 amplicon variants were generated by PCR using 15 I-SSR primers and 67 amplicons by PCR with 5 RAPD primers. Levels of genetic diversity within 6 populations were turned out to be similar (Shannon's Index, I-SSR : 0.35~0.40; mean of 0.38, RAPD : 0.31~0.38; mean of 0.35, combined : 0.35~0.40; mean of 0.37). Ranks of the level of genetic diversity estimated from I-SSR, RAPD, and combined data were not coincided each other. Majority of genetic diversity was allocated among individuals within populations (I-SSR : 94.31%, RAPD : 93.62%, combined : 93.57%), which resulted in pretty low level of population differentiation. Genetic differentiation between male and female groups was turned out to be quite low (I-SSR : 0.03, RAPD : 0.091, combined : 0.043), which slightly fluctuated when analysis was restricted to the data obtained from 3 regions where both male and female trees were sampled (I-SSR : 0.038, RAPD : 0.084, combined : 0.047). Genetic relationships among the populations, reconstructed by UPGMA, were not coincided with geographic affinity, which might be resulted from sharing of seed sources in some regions. Whereas independent cluster analyses with I-SSR data and RAPD data, respectively, reclassified by sexes revealed two sexual groups in which all the male and the female populations were clustered together, cluster analysis with combined data did not show clear sexual grouping.
