Oreorchis coreana Finet is threatened globally by over-collection from its natural habitats for horticultural purposes. Its rarity in nature makes this plant one of the most endangered species in Korea. In this study, we investigated the effects of sodium hypochlorite (NaOCl) on orchid seed viability and seed germination. An in vitro bioassay swelling test using immature seeds was compared with a standard chemical procedure using triphenyl tetrazolium chloride (TTC) to test seed viability. In general, the bioassay was more appropriate for estimating embryo viability after a prolonged pre-treatment (more than 1 h) in 1% NaOCl, a surface sterilant often used to enhance germination of seeds of terrestrial plants. Therefore, an efficient method for investigating in vitro swelling of immature seeds is urgently needed. We established a method for determining the viability and swelling of O. coreana seeds via in vitro examination of immature seeds. Treatment of immature seeds with 1% NaOCl for 10 min greatly enhanced the extent of swelling of immature zygote embryos when compared to untreated seeds. These data obtained here appear to be comparable to viability and swelling that occurs in O. coreana seeds via asymbiotic germination.
The seeds of Elaeocarpus serratus, a tropical underutilized fruit tree are characterized by hard seed coat and consequent poor water uptake and low germination. To improve the regeneration through seeds, various parameters such as viability of seeds, water uptake, and effect of seed mass on germination and pretreatments were performed using a completely randomized design (CRD). Tetrazolium (TZ) test was conducted using fresh, mature seeds revealed $50{\pm}2.56%$ mean viability. Seeds of different weight classes showed similar pattern of water uptake and the saturation level was achieved at 60 hrs of soaking. Seeds belong to weight class 2.6-3.5g were germinated ($12.5{\pm}1.26%$) with $175{\pm}1.75days$ (d) of mean time taken for germination (MTG). Germination capacity of seeds varied significantly among different populations and Varkala population gave $12.5{\pm}1.1%$ germination with $174.6{\pm}2.5d$ MTG. Among various seed treatments, mechanical scarification was superior in germination and significant reduction in MTG ($p{\leq}0.05$). The mechanical scarification by complete removal of seed coat resulted in $49.2{\pm}1.52%$ germination within a short period of time ($9.52{\pm}0.89d$ MTG). However, the complete removal of seed coat without damaging to embryo is a difficult task. An alternate treatment (Mechanical scarification II) by making cracks on nut faces vertically followed by soaking in distilled water for 24 hrs gave $48.4{\pm}1.73%$ germination with significantly reduced MTG ($12.14{\pm}0.56d$) over unsoaked, untreated control ($6.5{\pm}1.84%$ germination and $197.18{\pm}1.79d$ MTG; $p{\leq}0.05$). This treatment (Mechanical scarification II) is therefore recommended for E. serratus seeds as it can adopt easily and can achieve 7 fold increases in germination over control. The recorded germination through mechanical scarification is in tune with realized viability percentage of the seeds.

Cigarette filters containing Brassica Rapa L. seeds of the dark brown and round shaped were evaluated to determine the effect of seed addition on filter degradability. The seeds with germination capability under the tar/nicotine condition in the preliminary test, were put into the active carbon part of the filter(12mm) during filter rod making by the kit. The $4{\pm}2$ pieces of the seeds were put into the opened fiber bundle of the filter tow. In order to test the germination rate of the seeds, seed filters were placed either in a petri dish or test-pot in a conditioned area ($25^{\circ}C$, 70 % RH). The seed filters were placed outdoors exposed to natural conditions with the periodic water supply. The seeds in the smoked filters showed 90 % germination rate after a month under the open air condition. No significant differences in the sensory evaluation and analysis were obtained when the control sample was compared to the same cigarettes with the seeds.
The performance of 7 sugary (su) and 12 shrunken-2 (sh2) sweet com hybrids which are commercially grown in the United States was tested in Korea. The 100-seed weight of su hybrids (16.5-23.6 g) was much heavier compared to that of sh2 hybrids (10.9-17.5 g). The germination rate of su and sh2 hybrids at $25^{\circ}C$ ranged 93.3-100% and 86.7-98.9%, the emergence rate of su and sh2 hybrids in cold test ranged 78.9-97.8% and 62.2-97.8%, and field emergence rate of su and sh2 hybrids ranged 74.4-100.0% and 79.9-98.2%, respectively. In su hybrids, there was a significantly positive correlation between germination rate at $25^{\circ}C$ and emergence rate in cold test or early growth. In contrast, in sh2 hybrids seed weight was positively correlated with early plant growth, while not with the germination rate at $25^{\circ}C$ or emergence rate in cold test and field. Most sh2 hybrids produced larger and more marketable ears compared to su hybrids although there were significant differences among the hybrids in the same genotype. At harvest (24 days after pollination) soluble solids content of su hybrids (24.3-27.1 Brix %) was much higher than that of sh2 hybrids (13.8-18.0 Brix %), while total sugars of sh2 hybrids (21.4-28.6% on the dry weight basis) was much higher compared to su hybrids (2.4-15.9%). Considering germination and emergence rates, marketable ear production, and total sugar content, 'GCB 70' and 'Sweet Satin' in su hybrids and 'Ice Queen', 'Aspen', 'Sweet Magic', 'Bandit', 'Xtrasweet 82', 'Aspen', and 'Cambella 90' in sh2 hybrids performed better than other hybrids.

The study was conducted to develop ecological restoration method of damaged area in DMZ vicinities. As the material for the ecological restoration, forest soil and trees waste have been used. Forest soils were collected for experiments in Yanggu, Gangwon Province. Effect of ecological restoration was analyzed through germination experiment. Germination experiment was performed using 12 kinds of woody and herbaceous seeds. Woody and herbaceous seed germination in test pot was relatively evenly. Mixed material consisting of forest soil and trees waste seemed a possibility as the material for the restoration. The effects on seed germination were higher in the case of mixing more than 70% by volume ratio of local resources. Total number germinated individuals were different depending on the mixing ratio of soil sampling depth. Individual plants showed different trends depending on the experimental combination. Results of the woody seed germination were affected only in the soil sampling depth. Seed germination of herbaceous received a combined effect on soil sampling depth and mixing ratio.

The differences in seed germination rates of Korean Native Plants were observed depending on the harvest years, the seed companies, the storage periods, the drying methods and the plant morphological types. The seed germination rates were changed significantly by the harvest years and the seed companies in all 4 species tested. The seed germination rates were decreased after 1 year storage at $4^{\circ}C$ in all 8 species tested and three of them did not germinate at all. A rapid drying method at $35^{\circ}C$ with a convection oven was not suitable for Chrysanthemum spp., but there were no significant changes of germination rates in Aster spp. The germination rates were decreased more at seeding in soil than at lab. test with Dicotyledon but no significant changes with Monocotyledon. Especially, the germination rates of fine seed on 5mm soil covering treatment were significantly decreased.
This study was carried out to get the genetic information on the germination rate and fat acidity after 12 weeks' storage at $35^{\circ}C$. Germination rate decreased with longer storage period, while fat acidity increased. Germination rate was higher in the order of Koshihikari, Hanmaeum and Unkwangbyeo after 12 weeks' storage at $35^{\circ}C$. Fat acidity of Koshihikari and Hanmaeum was gradually increased, but that of Unkwangbyeo was dramatically increased after 4 weeks' storage at $35^{\circ}C$. The genetic mode on germination rate and fat acidity using the joint scaling test was shown to be additive-dominance gene effects. Additive([d]) effect was higher than dominance effect([h]) in all crosses. The average degree of dominance, [h]/[d], was less than 1, indicating that germination rate and fat acidity was partially dominant. Germination rate and fat acidity based on frequency distribution of 2 crosses were quantitative characters expressed by polygenes.
Viviparous germination causes yield loss and quality deterioration of rice. This study was conducted to investigate varietal differences of the viviparous germination with different days after heading (DAH) and different temperatures. In the laboratory examination, the averaged germination rate of all varieties at 45DAH and at 25DAH was 79.9%, and 27.5% under the incubation at the temperature of 3$0^{\circ}C$/2$0^{\circ}C$ (day/night) for 12 days. Andabyeo, Da- sanbyeo, and Nonganbyeo showed the lowest viviparous germination rates among the tested varieties. The shoot length of the viviparous germination measured 12days after incubation at 30/2$0^{\circ}C$ ranged from 21 to 53mm, indicating significant deterioration of rice quality. In the field test, the averaged viviparous germination rates of rice varieties at 25, 35, 45DAH with the underwater conditions for 4 days were 2.2, 6.2 and 9.2%, respectively, while their rates at 12 day after underwater conditions increased to 17.6, 44.2 and 43.8%, respectively. A variety that showed the highest viviparous germination rate at 25 and 35 and 45DAH was Heukjinjubyeo. When standing rice panicles without lodging were examined after consecutive raining for 7 days Juanbyeo showed the highest viviparous germination (45.5%), followed by Odaebyeo (16.0%), Jinbubyeo (14.5 %), Bongkwangbyeo (14.2 %) and Obongbyeo (12.6%). The viviparous germination of rice was greatly affected by variety, days after heading and temperature settings. Bongkwangbyeo, Juanbyeo, Dongjinbyeo, Hwasunchalbyeo, Naepoongbyeo and Heukjinjubyeo were classified as the most susceptible varieties to the viviparous germination in the field conditions.
In order to find out the effect of rainfall at harvesting season on germination of malting barley, the seeds sampled from Cheju island and southern part of Korea were examined. The germination rate of seeds from Cheju island where rainfall was frequent at harvesting season, ranged from 46 to 71%, even though disease infection of the seeds was not that serious as would be expected. High sugar content of seed was resulted from the degradation of carbohydrates during the harvest season. From TZ test the rainfall - affected seeds were found to be highly viable but in the state of secondary dormancy. Results of cold germination test showed that the seeds were recovered from the secondary dormancy. Therefore, it was recommended that the seeds affected by the rainfall at harvesting season should be utilized after 12 months when the dormancy period terminated. The germination percent of the seeds was significantly enhanced by prechilling and / or 1 ppm gibberellic acid treatment. Different seed lots showed different rate of germination and the degree of dormancy.
Background and objective: For effective horticultural education, it is required to develop kits for practice. In this study, we developed a horticultural kit for germination that introduces storytelling, characterization, and assembly, and investigated the educational effects of using it to study its educational differentiation and usefulness. Methods: First, we developed a seed germination kit for learning. It consists of three components: the kit main frame for hydroponics, the activity sheet for storytelling, and the instruction manual. It is made into an acrylic assembly in consideration of the ease of delivery for contactless learning. Second, we developed two programs, each using a germination kit for horticultural education (experimental group) and a general germination kit program (control group). At this time, the class consists of pre-questionnaire, the 1st and 2nd classes, observation log writing (total six times for two weeks), the 3rd class, and post-questionnaire. The experimental group and control group were separately executed. Third, the application results were compared and analyzed before and after class. As a quantification questionnaire, the effects before and after class were compared through t-test with a questionnaire on program satisfaction and attitude change toward plants. The quantitative results were analyzed through observation activity sheets and effects before and after the class were analyzed through t-test with quantification and language coding. Results: The effect of horticulture education using the developed kit was estimated by a quantification questionnaire. Both the program satisfaction and the change in attitude toward plants showed high in the experimental group, and this difference was statistically significant (p < .05). As a result of analyzing the observation paper with a qualitative questionnaire, various cognitive and affective expressions appeared in the experimental group as well as the contents of the observation paper. The number of syllables and various qualitative expressions appeared, and these differences were also statistically significant (p < .05). Conclusion: Compared to general germination activities, it was found that the germination activities using storytelling-based horticultural kits would be effective for contactless classes in the post-COVID-19 era and in increasing the educational effect of horticultural activities.
