Helicases are known to be a proteins that use the chemical energy of NTP binding and hydrolyze to separate the complementary strands of double-stranded nucleic acids to single-stranded nucleic acids. They participate in various cellular metabolism in many organisms. DEAD-box proteins are ATP-dependent RNA helicase that participate in all biochemical steps involving RNA. DEAD-box3 (DDX3) gene is belonging to the DEAD-box family and plays an important role in germ cell development in many organisms including not only vertebrate, but also invertebrate during asexual and sexual reproduction and participates in stem cell differentiation during regeneration. In this study, in order to identify and characterize DDX3 gene in the earthworm, Perionyx excavatus having a powerful regeneration capacity, total RNA was isolated from adult head containing clitellum. Full length of DDX3 gene from P. excavatus, Pe-DDX3, was identified by RT-PCR using the total RNA from head as a template. Pe-DDX3 encoded a putative protein of 607 amino acids and it also has the nine conserved motifs of DEAD-box family, which is characteristic of DEAD-box protein family. It was confirmed that Pe-DDX3 has the nine conserved motifs by the comparison of entire amino acids sequence of Pe-DDX3 with other species of different taxa. Phylogenetic analysis revealed that Pe-DDX3 belongs to a DDX3 (PL10) subgroup of DEAD-box protein family. And it displayed a high homology with PL10a, b from P. dumerilii.

Objectives: The c-kit gene encodes a transmembrane receptor-type tyrosine kinase, which is known to have a significant role in the normal migration and development of germ cells and melanocytes. In the previous studies of c-kit gene, c-kit expressions showed only in adenoid cystic carcinomas, lymphoepithelioma-like carcinomas and myoepithelial carcinomas, but not in others and mutation was not found in any types of salivary carcinoma. We investigate the c-kit expression which may be useful to differentiating adenoid cystic carcinomas from others, and mutation of the gene which may not be exist nor the mechanism of c-kit activation in salivary carcinomas. Material and Methods: The archival tissue samples from 42 salivary carcinomas of major and minor salivary glands were studied for c-kit expression by immunohistochemistry and gene mutation by polymerase chain reaction amplification and single strand conformational polymorphism. Results: The c-kit expressions were noted in 22/24 adenoid cystic carcinomas, 7/9 mucoepidermoid carcinomas, 2/3 acinic cell carcinomas, 3/4 malignant mixed tumors, and one undifferentiated carcinoma. The mutation of c-kit gene was found in 3/24 adenoid cystic carcinomas, 3/8 mucoepidermoid carcinomas, one acinic cell carcinoma, and 2/4 malignant mixed tumors. Conclusion: c-kit protein overexpression is seen in a variety of salivary gland carcinomas, and the mutation of the gene may be the mechanism of c-kit activation in these neoplasms.
Eik-Nes (1966) reported that the mechanism of spermatogenesis is controlled by FSH and LH and maintaned normally in scrotum terperautre which is 3-5$^{\circ}C$ lower than body termperature. But Ojeda and Ramirez (1972) have described that the abdominal testis was shrinked severely and lost its normal function in congenital cryptorchidism or surgically induced cryptorchidism. Ramirez and Sawyer (1974) reported that the compensatory hypertorphy occured in the remaining testis of unilateral castration and the scrotal testis of unilateral cryptorchidism. Cunninham et al. (1978) reported that the serum FSH levle increased after unilateral castration. Frankel and Wright (1982) reported that the serum LH level was unchanged greatly after unilateral castration. Gomes and Jain (1976) reported that the serum testosterone level increased temporarily but not varied after unilateral castration. On the other hand, Kormano et al. (1964) reported that the serum FSH level in unilateral cryptorchidism rat was unchanged in contrast with the control and Risbirdger et al. (1981) reported that the serum LH level was unchanged till 2 weeks after operation and after then increased to 77%. Kim (1984) reported that the serum testosterone level was somewhat lower than that fo control group but there was't significant different. There were many different reports on hormone levels among different investigators when the immarue rats were castrated unilaterally or induced cryptorchidism unilaterally. Liang and Liang (1970) and Cunningham et al. (1978) described that there were no true compenastory hypertrophy in the remaining testis of unilateral castration and scrotal testis of unilateral testis of unilateral cryptorchidism in rat but they grew faster than that of control. Kormano et al.(1964), Damber et al.(1976), Cunningham et al.(1978) and Karpe et al.(1981) reported that the testis weight, germinal epithelia height and seminiferous tubules diameter developed continuously and similarily in the control, the remaining testis of unilateral castration and scrotal testis of unilateral cryptorchidism increased, however, in the abdominal testis of the unilateral cryptorchidism, they were much smaller than those of other groups. In observation of the histological changes in the seminiferous epithelium of control, remaining tesis of unilateral castration and scrotal testis of unilateral cryptorchidism differentiated and developed fully(Cunningham et al., 1978). However, the abdominal testis of unilateral crytorchidism degenerated severely and only the germ cells in early stage and Sertoli cells were found in the seminiferous tubules. (Damber et al., 1976, Gomes and Jain, 1976 and Karpe et al., 1981). By electron microscopic observation, Nagano (1963) and Leason and Leeson (1970) found that the abdominal testis of unilateral cryptorchidism was thicked in boundary tissue, increased lipid droplet in the Sertoli cell, disarranged axial filament complex and increased lipid inclusions in the Sertoli cell.
Germ cell development during oogenesis, ovarian maturation and first sexual maturity in female Ruditapes philippinarum were investigated by cytological and histological observations. R. philippinarum is dioecious. During vitellogenesis, the Golgi complex, glycogen particles, and mitochondria were involved in the formation of lipid droplets and lipid granules in the cytoplasm of the early vitellogenic oocyte. In the late vitellogenic oocyte, cortical granules, the endoplasmic reticulum, and mitochondria were involved in the formation of proteid yolk granules in the cytoplasm. At this time, exogenous lipid granular substance and glycogen particles in the germinal epithelium passed into the oocyte through the microvilli of the vitelline envelope. The spawning period was once a year between early June and early October, and the main spawning occurred between July and August when seawater temperature was approximately $20^{\circ}C$. The reproductive cycle of this species can be categorized into five successive stages: early active stage(January to March), late active stage(Februaryto May), ripe stage(April to August), partially spawned stage(May to October), and spent/inactive stage (August to February). Percentages of female clams at first sexual maturity of $15.1{\sim}20.0mm$ in shell length were 52.6%(50% of the rate of group maturity was 17.83mm in length), and 100% for the clams >25.1mm.

Members of the Pumilio are the RNA binding proteins acting as translational repressors and required for germ cell development and asymmetric division. We identified chicken Pum1 and Pum2 that are similar to mouse and human in highly conserved C-terminal RNA-binding domain and eight tandem repeats. The comparative sequence analysis of Pum1 and Pum2 from fly, chicken, mouse and human shows high degree of evolutionary conservation in the homology of the peptide sequence and the structure of PUM-HD (Pumilio homology domain) with similar spacing between adjacent Pum repeats. Also, structures of chicken Pum1 and Pum2 genes are almost identical to those of mouse and human. We revealed that the expression levels of Pum1 and Pum2 were the highest in hatched female gonad among various embryonic tissues, and Pum2 expressed highly in 12-day and hatched gonad by real-time RT-PCR. These results suggest that Pum1 and Pum2 might have an effect on the development of chicken gonad.
Spermatogenesis and reproductive cycle of the razor clam, solen grandis, were investigated monthly by histological and cytological observations. Samples were collected from natural intertidal population at Oshik-do, Kunsan, Korea, for one year, beginning from January to December, 1993. solen grandis is dioecious. Morphological structures of the spermatozoon of this species ar esimilar to those of other bivalve spermatozoa having a primitive type; i.e., a small head, a cap-shaped acrosome and a short mid-piece with four mitochondria surrounding axial filament. The head of spermatozoon is approximately 2 \mu m in length and sperm tail is about 20 \mu m long. The axoneme of tail flagellum consists of nine pairs of peripheral microtubules at the periphery and a pair of central microtubules at the center. Four spherical mitochondria form the paranucleus. Spawning occures once a year between early June and July, and the main spawning was observed in July when seawater temperature reaches above 20 \circ C. The reproductive cycle of male razor clam can be divieded into fivesuccessive stages; early active (December to january), late active (January to march), mature (March to early August), partially spawned (June to July), and spent/inactive stage (August to December).

Sex differentiation of the brown croaker Miichthys miiuy (Basilewsky) is described from hatching to the 120th day post-hatching (dph) (water temperature $24^{\circ}C$). Primordial germ cells (PGCs) were observed on the 20th dph (10.4 mm total length (TL), 0.14 g body weight (BW), and began to protrude into the peritoneal cavity from the 40th dph (19.4 mm TL, 0.39 g BW). On the 65th dph (31.3 mm TL, 0.93 g BW, $1,560D^{\circ}$ (degree-days)), initial ovarian differentiation was identified by the PGCs with condensed chromatin, and their transformation into meiotic oocytes. By the 120th dph (4.60 mm TL, 1.38 g BW, $2,880D^{\circ}$), the oocytes were in the perinucleolus stage and had increased from 20 to $40{\mu}m$ in diameter. While ovaries gradually grew after sex was differentiated, testes continued to multiply from the 65th dph. On the 80th dph (37.9 mm TL, 1.39 g BW, $1,920D^{\circ}$), the beginning of testis lobule formation was indicated by the occurrence of spermatogonial cysts enveloped by somatic cells in some of the testes. On the 120th dph, the testis lobules of some of the fish contained all germ cell stages through to the spermatocytes. Therefore, the sex differentiation type of the brown croaker is identified as gonochoristic.

The low density lipoprotein receptor-related protein 5 (LRP5) highly expressed in many tissues, including hepatocytes and pancreatic beta cells, can bind to apolipoprotein E. To evaluate in vivo roles of LRP5, we generated LRP5-deficient mice. LRP5 genomic DNA was isolated from TT2 embryonic stem (ES) cells. Targeting vector was constructed to disrupt an exon 18 of the mouse LRP5 gene and transfected into ES cells. Three homologous recombinants at LRP5 locus were identified from 178 G418-resistant clones. Chimeric males generated by morula aggregation technique were mated to C57BL/6 female mice. After achieving germ-line transmission, LRP5+/- females were crossed with LRP5+/- males to obtain LRP5-deficient mice. One line of mice lacking LRP5 gene was confirmed by Southern blotting. Such knock-out mice may serve as an effective animal model to study in vivo function of LRP5 gene.
This report describes the sex differentiation of the Korean rose bitterling, Rhodeus uyekii, from hatching to 170 days post-hatch (DPH) in relation to total length (TL), body weight (BW), and integral water temperature (IWT). The growth curve of TL from just hatching to 83 DPH was $5.144e^{0.045t}$ ($R^2=0.961$; t, time), and that of BW was $2.398e^{0.086t}$ ($R^2=0.725$). Primordial germ cells (PGCs) were observed at 17 DPH (7.9 mm TL, 3.74 mg BW, $374^{\circ}C$ IWT), and thereafter began to protrude into the peritoneal cavity. At 21 DPH ($9.2{\pm}0.14mm$ TL, $4.8{\pm}0.07mg$ BW, $462^{\circ}C$ IWT), some PGCs contained condensed chromatin and oocyte were observed in meiotic prophase. In contrast to the ovaries, which grew gradually after sexual differentiation, testes began multiplying at 25 DPH (10.1 mm TL, 5.42 mg BW, $550^{\circ}C$ IWT), when testicular differentiation was first identified, and multiplied continuously thereafter. At 33 DPH (11.2 mm TL, 10.5 mg BW, $726^{\circ}C$ IWT), the developing testes contained spermatogonia that exhibited mitotic activity. No spermatocyte or sperm cell was observed until 83 DPH (18.9 TL, 48.2 mg BW, $1,826^{\circ}C$ IWT). At 170 DPH (32.5 mm TL, 270.1 mg BW, $3,740^{\circ}C$ IWT), which was the end point of this study, the mature ovaries showed germinal vesicle breakdown, while the mature testes contained observable spermatocytes and sperm cells. These results allow us to identify the sex differentiation type of the Korean rose bitterling as differentiated gonochoristic.
Early development growth and sexual phenomena of the hybrid between male spotted flounder (Verasper variegatus) and female olive flounder (Paralichthys olivaceus) were investigated. The hybrid produced was newly called the "Bumnupchi" in Korean name, "Mottled flounder" in English name. Fertilization rate, hatching rate and early survival rate of the mottled flounder were 56.9~72.3%, 86.7~92.5% and 89.1~96.0%, respectively. At 50th day and 240th day after hatching, they were 2.1$\pm0.2cm\;and\;22.9\pm2.1cm\;in\;total\;length, \;0.10\pm0.002g\;and\;179.0\pm38.5g$ in body weight, respectively. Eye position of the mottled flounder was mixed with both dextral (77.7%) and sinistral type (22.3%). Gonads were differentiated into ovarian type and testis type at 80th day after hatching, and its ratio was 1:1 (P>0.05). Most Gonads seemed to be sterile without having germ cell, whereas only 3.6% of investigated indiciduals turned out to have oocytes in the gonad. When the induction of sex reversal were attempted by daily oral administration of $17\beta$-methyltestosterone (0.5mg/kg fish) andestradiol-17$17\beta$(0.5mg/kg fish)from 40th to 100th day after hatching, both testis and ovary type were induced 81.9% and 87.7% of each hormone-treated fish, respectively.fish, respectively.
