APPLYING ELITIST GENETIC ALGORITHM TO RESOURCE-CONSTRAINED PROJECT SCHEDULING PROBLEM

  • The objective of this research study is to develop the permutation-based genetic algorithm for solving the resource-constrained project scheduling problem in construction engineering by incorporating elitism into genetic algorithm. A key aspect of the algorithm was the development of the elitist roulette selection operator to preserve the best individual solution for the next generation so the improved solution can be obtained. Another notable characteristic is the application of the parallel schedule generation scheme to generate a feasible solution to the problem. Case studies with a standard test problem were presented to demonstrate the performance and accuracy of the algorithm. The computational results indicate that the proposed algorithm produces reasonably good solutions for the resource-constrained project scheduling problem.

한국 연안에 서식하는 문절망둑의 지리적 분포와 유전적 거리 (The Geographical Distribution and Genetic Distance of Yellowfin Goby (Acanthogobius flavimanus) off the Coast of Korea)

  • A total of 64 individuals of Acanthogobius flavimanus, which inhabit the coast of Korea, were collected from 8 regions from July to August 2023. A haplotype network and a phylogenetic tree were created. The genomic DNA of the target fish species was compared and analyzed with the genomic DNA of four regions in Japan downloaded from the National Center for Biotechnology Information (NCBI). In the haplotype network of Acanthogoboius flavimanus, Eocheong-do (EC) and Goseong (MAJ) exhibited low genetic similarity with other regions in Korea and Japan. The Phylogenetic tree showed that the population of MAJ exhibited differences in genetic structure compared to populations in other regions of Korea and Japan, indicating a distant relationship. Most marine organisms are known to migrate and spread via ocean currents, which is the most crucial factor promoting gene flow through larvae between populations. The haplotype of Acanthogobius flavimanus in MAJ differs from the haplotypes in Korea and Japan. The population in MAJ is believed to have limited genetic exchange due to the North Korea Cold Currents. We identified haplotype patterns based on the geographical distribution of Acanthogobius flavimanus off the coast of Korea and inferred that ocean currents have some influence on genetic distances.

  • The purpose of this study is to optimally allocate the human resources to tasks while minimizing the total daily human resource costs and smoothing the human resource usage. The human resource allocation problem (hRAP) under consideration contains two kinds of special constraints, i.e. operational precedence and skill constraints in addition to the ordinary constraints. To deal with the multiple objectives and the special constraints, first we designed this hRAP as a network problem and then proposed a Pareto multistage decisionbased genetic algorithm (P-mdGA). During the evolutionary process of P-mdGA, a Pareto evaluation procedure called generalized Pareto-based scale-independent fitness function approach is used to evaluate the solutions. Additionally, in order to improve the performance of P-mdGA, we use fuzzy logic controller for fine-tuning of genetic parameters. Finally, in order to demonstrate the applicability and to evaluate the performance of the proposed approach, P-mdGA is applied to solve a case study in a hotel, where the managers usually need helpful automatic support for effectively allocating hotel staff to hotel tasks.

인삼 토양 미생물 Cellulosimicrobium sp. Gsoil 235의 배지조성에 따른 Ginsenoside $Rb_1$ 전환 (Conversion of Ginsenoside $Rb_1$ by Ginseng Soil Bacterium Cellulosimicrobium sp. Gsoil 235 According to Various Culture Broths)

  • 인삼 근권에 존재하는 토양 미생물중 esculin agar법을 이용하여 $\beta$-glucosidase를 생산하는 균주를 분리하고, 다시 ginsenoside $Rb_1$을 선택적으로 분해하는 균주 Gsoil 235를 선발 및 동정하였다. 16S rRNA 염기서열을 sequencing한 후, genebank에서 가장 가까운 type strain을 결정하여 유연 관계를 분석한 결과 Cellulosimicrobium 속의 funkei ATCC BAA-$886^T$(AY501364)와 99.7% 일치하는 균주임을 확인하였다. TSB, LB, NB등 3종류의 배지에서 균의 생장은 접종 후 12-24 시간에서 가장 잘 자라며, TSB>LB>NB의 순으로 잘 자라는 것을 알 수 있었다. ginsenoside $Rb_1$과 8, 24, 48시간 동안 반응시킨 후 TLC로 분석한 결과 NB>LB>TSB순으로 $Rb_1$ 분해 활성이 뛰어나 배지의 생장과 대조적인 결과를 얻었다. 반응시간이 증가할수록 Rd를 포함한 minor ginsenoside의 생성이 증가하였으며, 특히 다른 배지에 비해 균주 생장속도가 상대적으로 낮은 NB는 48시간 후 $Rb_1$을 거의 분해하여 강한 효소 활성을 확인할 수 있었다.

Reference Gene Screening for Analyzing Gene Expression Across Goat Tissue

  • Real-time quantitative PCR (qRT-PCR) is one of the important methods for investigating the changes in mRNA expression levels in cells and tissues. Selection of the proper reference genes is very important when calibrating the results of real-time quantitative PCR. Studies on the selection of reference genes in goat tissues are limited, despite the economic importance of their meat and dairy products. We used real-time quantitative PCR to detect the expression levels of eight reference gene candidates (18S, TBP, HMBS, YWHAZ, ACTB, HPRT1, GAPDH and EEF1A2) in ten tissues types sourced from Boer goats. The optimal reference gene combination was selected according to the results determined by geNorm, NormFinder and Bestkeeper software packages. The analyses showed that tissue is an important variability factor in genes expression stability. When all tissues were considered, 18S, TBP and HMBS is the optimal reference combination for calibrating quantitative PCR analysis of gene expression from goat tissues. Dividing data set by tissues, ACTB was the most stable in stomach, small intestine and ovary, 18S in heart and spleen, HMBS in uterus and lung, TBP in liver, HPRT1 in kidney and GAPDH in muscle. Overall, this study provided valuable information about the goat reference genes that can be used in order to perform a proper normalisation when relative quantification by qRT-PCR studies is undertaken.

  • 인삼사포닌 ginsenoside는 인삼의 주요한 약리성분으로 인삼을 경구투여 시 major 사포닌의 생체 내에서의 흡수는 매우 낮아 인삼사포닌의 약효를 증대시키기 위해서 기존에 많이 존재하는 major 사포닌을 상대적으로 흡수도 잘 되며 약효도 더 뛰어난 minor 사포닌으로의 전환이 요구된다. 본 연구는 김치에서 분리한 ${\beta}-glucosidase$ 활성균주 L. brevis LH8이 분비하는 효소를 이용하여 ginsenoside Rd를 compound K로 전환시켰다. L. brevis LH8의 효소액은 반응 온도 $30^{\circ}C$에서 효소활성이 가장 좋았고 $35^{\circ}C$이상에서는 활성이 급격히 저하되었으며, pH $6.0{\sim}12.0$ 사이에서 효소활성이 가장 좋았고, pH 5.0 이하 및 pH 13.0 이상에서는 활성도가 떨어지는 것을 관찰할 수 있었다. 또한 ginsenoside Rd는 반응 48시간부터 ginsenoside F2로 전환되기 시작하였으며, 반응 72 시간 이후에는 대부분 compound K로 전환되었다.

백삼, 발효인삼, 홍삼 농축액의 이화학적 특성 (Physico-Chemical Characteristics of White, Fermented and Red Ginseng Extracts)

  • 본 연구에서는 시중에 유통되고 있는 백삼 농축액(white ginseng extract, WG), 발효인삼 농축액(fermented ginseng extract, FG), 홍삼 농축액(red ginseng extract, RG)에 대해 고형분, 점성도, 회분, 조지방, 조단백, 탄수화물, 나트륨, 열량 등을 분석하였다. 백삼 농축액, 발효인삼 농축액, 홍삼 농축액의 고형분 조사결과, 발효인삼 농축액, 백삼 농축액, 홍삼 농축액 순으로 발효인삼 농축액이 가장 높았으나, 점성도는 홍삼 농축액에서 가장 높게 나타났다. 백삼 농축액, 발효인삼 농축액, 홍삼 농축액 모두 온도를 높일수록 점성도는 떨어지는 경향을 보였으며 $40^{\circ}C$ 이상 가열 시에는 점성도의 차이가 크게 나타나지 않았다. 회분과 조단백질의 함량은 발효인삼농축액이 가장 높았으며, 조지방은 홍삼농축액에서 가장 높게 나타났다. 탄수화물 함량은 세 농축액에서 큰 차이 없이 유사한 결과를 보였으며, 나트륨 함량은 백삼농축액이 가장 높았다. 그러나 열량에 있어서는 백삼 농축액, 발효인삼 농축액, 홍삼 농축액에서 큰 차이가 없는 것으로 조사되었다. 현재 활발히 연구 중인 백삼 농축액, 발효인삼 농축액, 홍삼 농축액 각각의 이화학적 특성을 분석하여 앞으로의 연구 및 신제품 개발에 이용할 수 있는 결과를 얻을 수 있었다. 이를 토대로 항산화, 면역활성, 혈관이완작용에 미치는 영향을 규명하고 인삼농축액, 발효인삼농축액, 홍삼농축액의 효능 차이를 비교 평가하는 실험이 이루어져야 할 것이다.

Screening of key miRNAs related with the differentiation of subcutaneous adipocytes and the validation of miR-133a-3p functional significance in goats

  • Objective: Adipocyte differentiation is regulated by a variety of functional genes and noncoding RNAs. However, the role of miRNAs in lipid deposition of goat white adipose tissue is still unclear. Therefore, this study revealed the miRNA expression profile in goat subcutaneous adipocytes by sRNA-seq. Methods: The miRNA expressed in goat subcutaneous preadipocytes and the mature adipocytes were sequenced by sRNA-seq. The differentially expressed miRNAs (DEm) were screened and gene ontology (GO) and Kyoto encyclopedia for genes and genomes (KEGG) analyses were performed. Gain-of-function and loss-of-function combined with oil red O staining, Bodipy staining, and quantitative reverse-transcription polymerase chain reaction (qPCR) were utilized to determine the effect of miR-133a-3p on adipocyte differentiation. Results: A total of 218 DEm were screened out. The target genes of these DEm were significantly enriched in GO items such as biological regulation and in KEGG terms such as FAK signaling pathway and MAPK signaling pathway. qPCR verified that the expression trend of miRNA was consistent with miRNA-seq. The gain-of-function or loss-of-function of miR-133a-3p showed that it promoted or inhibited the accumulation of lipid droplets, and CCAAT enhancer binding protein α (C/EBPα) and C/EBPβ were extremely significantly up-regulated or down-regulated respectively (p<0.01), the loss-of-function also led to a significant down-regulation of peroxisome proliferator activated receptor gamma (PPARγ) (p<0.01). Conclusion: This study successfully identified miRNAs expression patterns in goat subcutaneous adipocytes, and functional identification indicates that miR-133a-3p is a positive regulator of the differentiation process of goat subcutaneous adipocytes. Our results lay the foundation for the molecular mechanism of lipid deposition in meat-source goats from the perspective of miRNA.
