황색포도알균의 항생제 내성 (Antibiotic Resistance of Staphylococcus Aureus)

  • Staphyloccus aureus is one of the most important pathogens in clinical settings. It is also one of the leading causes of nosocomial infections and the dissemination of multiple drug-resistant strains, mainly methicillin resistant Staphyloccus aureus, and the recent emergence of a vancomycin resistant MRSA is the concern to hospital worldwide. MRSA strains have acquired multiple resistance to a wide range of antibiotics, including aminoglycosides and macrolides. $\beta$-Lactam resistance of methicillin-resistnat Staphyococcus aureus is determined by the function of penicillin binding protein 2'(PBP2') encoded by the methicillin resistance gene mec A. MRSA strains carry methicillin resistance gene mecA, encoded by a mobile genetic element designated staphylococoal cassette chromosome mec(SCCmec). MRSA clones are defined by the type of SCCmec element and the genotype of the methicilline-susceptible Staphyococcus aureus chromosome in which the SCCmec element is integrated.

Characterization and Genetic Diversity of Benzimidazole-resistant and -sensitive Monilinia fructicola Isolates in Korea

  • To characterize benzimidazole-resistant and -sensitive Monilinia fructicola populations, the fungal isolates were obtained from peach plants showing brown rot and bloosom blight. Benzimidazole-sensitive isolates did not grow on potato dextrose agar(PDA) amended with $\geq1.0{\mu}g$ active ingredient(a.i.)/ml of the fungicides. However, benzimidazole-resistant isolates grew on PDA regardless of the tested concentrations of fungicides. Benzimidazole-resistant isolates did not grow on diethofencarb-PDA, but sensitive isolates grew on the same PDA. In the nucleotide sequences of $\beta$-tubulin gene, only codon 198(GAG: glutamic acid), a target site for benzimidazole, was replaced with GCG(alanine) in all of the resistant isolates, and this substitution seems to play an important role in the development of resistance. Other interesting codons such as 165(GCT), 200(TTC), and 241(GCT) were not changed among the isolates. Benzimidazole-resistant and -sensitive isolates were clustered clearly in random amplified polymerphic DNA analyses and the results revealed that low levels of genetic diversity between benzimidazole-sensitive and -resistant isolates of M. fructicola in the investigated regions.

먹물버섯 Coprinellus congregatus에서 분열자를 사용한 형질전환 (Genetic Transformation of a Mushroom Forming Fungus Coprinellus congregatus to an Antibiotic Resistance Using Oidia Instead of Protoplast Generation)

  • 먹물버섯의 하나인 Coprinellus congregatus를 대상으로 유전자의 도입을 위한 형질전환실험에서 원형질체를 생성하지 않고 분열자 (oidium)를 사용하는 방법을 확립하였다. 분열자는 20일 이상된 CKMM 한천배지에서 생성되며, 이를 밤샘배양으로 발아를 촉진시킨 상태에서 전기천공방법으로 항생물질 basta에 대한 형질전환을 수행한 결과 10-20 형질전환체/${\mu}g$ DNA의 수율로 형질전환체를 확보할 수 있었다. 이 형질전환체들은 도입된 벡터가 염색체 상에 삽입되어 유전적으로 안정된 상태를 유지하였다.

Assicuation between Genetic Variation of the Insulin Receptor Gene and Essential Hypertension in the Korean Population

  • Essential hypertension is a multifactorial disease, and has been shown to be associated with insulin resistance. The relationship between the genetic variation of the insulin receptor (INSR) gene and essential hypertension In Korean population was investigated by the Nsi 1 restriction fragment length polymorphism (RFLP) pattern of this gene. The observed genotype frequencies of INSR gene were not deviated from those expected for the Hardy-Weinberg equilibrium (HWE), but a significant association was observed between essential hypertension and N1 allele of Nsi 1 RFLP at the INSR gene ($X^2$-test; P<0.05). Moreover, the frequency of N1 allele was significantly different between normotensives and essential hypertensives in subgroups that were not obese ($X^2$-test; P<0.05). These data suggest that the Nsil RFLP of INSR gene may be a useful genetic marker for essential hypertension in Korean population.

  • Salinity is one of the major abiotic stresses that severely affect crop production throughout the world; especially rice plant which is generally categorized as a typical glycophyte as it cannot grow in the presence of salinity. Phenotypic resistance of salinity is expressed as the ability to survive and grow in a salinity condition. Salinity resistance has, at least implicitly, been treated as a single trait. Physiological studies of rice suggest that a range of characteristics (such as low shoot sodium concentration, compartmentation of salt in older rather than younger leaves, high potassium concentration, high $K^+/Na^+$ ratio, high biomass and plant vigour) would increase the ability of the plant to cope with salinity. Criteria for evaluating and screening salinity tolerance in crop plants vary depending on the level and duration of salt stress and the plant developmental stage. Plant growth responses to salinity vary with plant life cycle; critical stages sensitive to salinity are germination, seedling establishment and flowering. We have established a standard protocol to evaluate large rice germplasms for overall performance based on specific physiological traits for salt tolerance at seedling stage. This protocol will help in identifying germplasms which can perform better in the presence of different salinity treatments based on single trait and also combination of different physiological traits. The salt tolerant germplasm can be taken forward into developing better varieties by conventional breeding and exploring genes for salt tolerance.

Gui Valley: A High Yielding Potential and Good Processing Potato Cultivar

  • The main objective of this breeding program is to develop high yielding, disease resistance and good processing potato cultivar. 'Gui Valley' is a clonal selection resulting from a cross between 'ND2471-8' and 'Cona'. It has medium plant height and light green foliage. 'Gui Valley' has medium flowering habit and light pink flowers. 'Gui Valley' is a medium maturing potato cultivar and tubers are smooth, yellow skin, light yellow flesh, long-oval tuber shape, distinct red eyes with medium depth and medium dormancy. It has high level of tuber uniformity and good keeping quality. 'Gui Valley' demonstrates resistance to potato virus Y (PVY), soft rot, but moderately susceptible to late blight and common scab. It is also resistant to most of the internal and external physiological disorders particularly dehiscence, hollow heart and internal brown spot. The specific gravity of 'Gui Valley' is significantly higher (1.097) than that of 'Shepody' (1.078). 'Gui Valley' has suitable for processing mainly French fries and chips. This cultivar has high level of tuber uniformity and capable of yielding 37.6 $t{\cdot}ha^{-1}$, which is 18.2% higher than the control potato cultivar 'Shepody' under optimum agronomical practices.

수도의 흰등멸구(Sogatella furcifera Horvath)에 대한 저항성 유전자 연관분석 (Linkage Analysis of the Resistance Genes to Whitebacked Planthopper (Sogatella furcifera Horvath) in Rice)

  • 수도에 있어서 흰등멸구에 대한 저항성 유전자를 가진 것으로 알려진 N22(Wbphl)와 ARC 10239(Wbph2)의 흰등멸구 저항성 유전자의 연관 분석을 하기 위하여 V번 연관군을 제외한 11개 연관군의 표식 유전자를 가진 semi-dwarf 초형의 검정친과 N22및 ARC10239와의 교잡 F$_2$세대에서 12개 표식 유전자의 유전양식을 조사하고 F$_3$ 세대에서 흰등멸구에 대한 저항성의 분리를 조사하여, 이들을 이용하여 연관분석을 한 결과를 요약하면 다음과 같다. 1. N22와 검정친들과의 교잡 세대 F$_2$에서 lg, d-t, g, la, nl, bl, gl 둥은 우성과 열성이 3 : 1로, Bh는 9 : 7로 이론치에 맞게 분리하였으며, Cl, gh, Lh, bc 등 4개 형질은 오차가 컸지만 대체로 이론분리비에 비슷한 분리비를 보였다. 2. ARC 10239와 검정친들과의 조합 F$_2$에서 Cl, lg, Pn, g, la, bl, gl 등은 우성과 열성이 3 : 1로, Bh는 9 : 7로 분리하였으며, gh, Lh, nl, bc 등은 이론분리에 비하여 오차가 크게 나타났다. 3. N22의 Wbph1 유전자와 ARC 10239의 Wbph 유전자는 단순우성유전자이었으며 저항성 계통, 분리계통 및 감수성 계통의 분리비는 모든 조합에서 1 : 2 : 1 의 이론치에 합당하였다. 4. Wbph1은 II번 연관군의 liguleless (lg)와 36.8%의 조환가로 연관되어 있으며 Black hull(Bl)과도 35.9%의 조환가로 연관된 것으로 나타났으며, Bh는 II번 연관군의 Ph(Phenol staining)와 보족적으로 발현됨 을 고찰하였다. 5. Wbph 2는 검정된 I,II,III,IV,VI,VII,VIII,IX,X,XI,XII번 연관군의 Cl, lg, Pn, g, gh, Lh, la, nl, bl, bc, gl, Bh와 독립적인 것으로 나타났다. 6. 이상에서 흰등멸구 저항성 유전자 Wbph2의 연관관계가 분명하지 못한 것은 사용된 Marker 유전자가 염색체상에서 저항성 유전자와 원거리에 있거나 Marker 형질의 발현특성에서 기인된 것으로 생각된다. 7. Wbph2의 연관관계는 본 실험에서 검토되지 못한 V번 연관군을 포함하여 독립성 검정에서 변이가 큰 것으로 나타난 Ⅶ, Ⅷ, Ⅹ 및 XII 번 연관군과의 조합에 대하여는 재검토되어야 할 것으로 판단된다.

Evolution of Genetic Polymorphisms of Plasmodium falciparum Merozoite Surface Protein (PfMSP) in Thailand

  • Plasmodium falciparum malaria is a major public health problem in Thailand due to the emergence of multidrug resistance. The understanding of genetic diversity of malaria parasites is essential for developing effective drugs and vaccines. The genetic diversity of the merozoite surface protein-1 (PfMSP-1) and merozoite surface protein-2 (PfMSP-2) genes was investigated in a total of 145 P. falciparum isolates collected from Mae Sot District, Tak Province, Thailand during 3 different periods (1997-1999, 2005-2007, and 2009-2010). Analysis of genetic polymorphisms was performed to track the evolution of genetic change of P. falciparum using PCR. Both individual genes and their combination patterns showed marked genetic diversity during the 3 study periods. The results strongly support that P. falciparum isolates in Thailand are markedly diverse and patterns changed with time. These 2 polymorphic genes could be used as molecular markers to detect multiple clone infections and differentiate recrudescence from reinfection in P. falciparum isolates in Thailand.

  • A direct torque control(DTC) based sensorless speed control system which employs a new closed loop flux observer is proposed. The flux observer takes an adaptive scheduling gains where motet speed is used as the scheduling variable. Adaptive nature comes from the fact that the estimated values of stator resistance and speed are included as observer parameters. The parameters of the PI controllers adopted in the adaptive law for the estimation of stator resistance and motor speed are determined by simple genetic algorithm. Simulation results in low speed region are given for comparison between proposed and conventional flux estimate scheme.

Aggressiveness in Plasmopara halstedii (sunflower downy mildew)

  • Aggressiveness was studied in seven Plasmopara halstedii (sunflower downy mildew) pathotypes: 100, 300, 304, 314, 704, 710 and 714. Aggressiveness criteria including percentage infection, latent period, sporulation density and reduction of hypocotyl length (dwarfing) were analysed in one sunflower inbred line showing a high level of quantitative resistance. Genetic relationships were detected between the seven pathotypes using 12 EST-derived markers. Pathotypes 100, 300, 304 and 314 were characterized with shorter latent period and higher sporulation density than pathotypes 710, 704 and 714. All pathotypes showed high percentage infection values and caused a large reduction in seedling size except for pathotype 314 involved in dwarfing. Pathotypes 714, 704 and 314 had an intermediary genetic position between the pathotypes 100 and 710. No correlation was detected between aggressiveness traits and EST genotypes.
