Immunocytochemical Study on the Enteroendocrine Cells in the Gastrointestinal Tracts of the Korean Snakes (한국산 뱀목 위장관 점막이 장내분비세포에 대한 면역세포화학적 연구)

  • This study attempts to investigate several enteroendocrine cells in the gastrointestinal epithelia of the Korean snakes (Dinodon rufozonatum rufozonotum Rhabdophis tigrina tigrina, Enhydris rufodorsata, Agkistrodon blomhoffii brevicaudus, Agkistrodon saxatilis, Agkistodon calginosus). For a light-microscopical examination of immunocytochemistry, the paraffin sections (5 $\mu$ m) of tissue specimens taken from the various parts of the gastrointestinal tract were stained immunocytochemically by PAP procedure with 10 antisera. The frequency of enteroendocrine cells per unit area (mm$^2$) of each mucosa were counted and the shapes of the cells were observed. In Dinodon rufozonatum rufozonatum, Rhobdophis tigrina tigrina, Enhydris rufodorsata, Agkistrodon saxatilis and Agkistrodon caliginosus, cholecystokinin (CCK)-8, gastrin, pancreatic polypeptide (PP) and serotonin cells were observed. But the freuqency of these immunoreactive cells differ trom each portion of gastrointestinal tracts of all species, respectively. In Agkistrodon blomhoffii brevicaudus, CCK-8, gastrin and serotonin cells were observed. CCK-8 and serotonin cells were found in whole gastrointestinal tracts and gastrin cells were observed in pylorus and mucosa of small intestine. The frequency of these cells was different from each portion. The shapes of CCK-8, gastrin, PP and serotonin cells were pyramidal or oval and closed type in stomach. A large number of these cells were spindle in shape and open type in small intestine and anterior pant of large intestine, whereas some cells were closed type. In posterior part of large intestine and rectum, these cells were oval in shape and closed type.

  • The gastrointestinal tract of three Percida, Lateolabrax japonicus, Epinephelus septemfasciatus and Mugil cephalus, was investigated immunocytochemically for the occurrence of somatostatin-. seotonin-, gastrin-, pancreatic polypeptide(PP)-, cholecystokinin-8(CCK-8)- and glucagon-immunoreactive cells. In Lateolabrax japonicus and Epinephelus septemfasciatus, five endorcrine cell types, such as somatostatin-, serotonin-, gastrin-, PP- and CCk-8-immunoreactive cells were demonstrated. In Mugil cephalus, however, six endocrine cell types, such as somatostatin-, serotnin-gastrin-, PP-, CCK-8- and glucagon-immunoreactive cells were detected. Somatostatin- and serotonin-immunoreactive cells were detected in the gastric mucosa of all species. Glucagon-immunoreactive cells were found only in the gastric mucoas of Mugil cephalus. In the pyloric caeca, PP-and CCK-8-immnuoreactive cells fo all species. gastrin-immunoreactive cells of Epinephelus septemfasciatus and Mugil cephalus, and serotonin-immunoreactive cells of Epinephelus septemfasciatus were demonstrated. In the intestinal mucosa of all species, gastrin-, PP- and CCK-8-immunoreactive cells were detected, and in the intestinal mucosa of Epinephelus septemfasciatus serotonin-immunoreactive cells were also detected. The frequency of these immunoreactive cells differs from each portion of the gastrointestinal tract of all species.

The Effects of Maitland Orthopedic Manual Physical Therapy on the Digestive System

  • The purpose of the present study is to apply Maitland orthopedic manual physical therapy to patients to examine the effects of the therapy on the digestive system through serum gastrin tests that can identify the motility of the digestive system and dyspepsia symptoms and can provide basic data for internal medicinal physical therapy. Maitland orthopedic manual physical therapy was implemented on 20 subjects in total, with 10 in a dyspepsia group and 10 in a control group, for 20 minutes per day, three days per week for three weeks, and the following results were obtained. In the control group, serum gastrin values significantly decreased between before and after treatment(p<.01), and among questionnaire items regarding dyspepsia, those regarding epigastric pain and belching showed significant decreases in these symptoms(p<.05). In the dyspepsia group, serum gastrin values significantly decreased between before and after treatment(p<.01) and all questions regarding dyspepsia showed significant decreases in all symptoms(p<.01). According to the analysis, the dyspepsia group decreased significantly more than the control group in serum gastrin values and all dyspepsia symptoms, except for belching(p<.001). Through the present study, it was identified that Maitland orthopedic manual physical therapy can improve dyspepsia symptoms and is an effective treatment method for internal diseases, such as dyspepsia, by improving gastric motility to become close to the normal state.

Regional distribution and relative frequency of the gastrin, secretin and pancreatic polypeptide-immunoreactive cells in the gastrointestinal tract of the fetus of Korean native goat (한국재래산양 태자의 위장관에 있어서 gastrin, secretin 및 pancreatic polypeptide 면역반응세포의 분포 및 출현빈도에 관한 연구)

  • The regional distributions and relative frequencies of the gastrin, secretin and pancreatic polypeptide(PP)-immunoreactive cells in the gastrointestinal tract of the fetus(180 days of gestation) of Korean native goat were studied with immunohistochemical(ABC) methods. Gastrin-immunoreactive cells were detected in fundus, pylorus and duodenum and these cells were most predominant in pylorus. Secretin-immunoreactive cells were observed in pylorus, duodenum and ileum. PP-immunoreactive cells were restricted to fundus. These immunoreactive cells were situated in surface epithelium and mucosal gland regions. The regional distribution and relative frequency of PP-immunoreactive cells was somewhat different to the adult Korean native goat. Immunoreactive cells in the surface epithelial regions were open typed cells which were spindle shaped cells but closed typed cells which were round or/to spherical shaped cells were observed in the mucosal gland regions.

Mitigating Effect of Yukmijihwang-tang on Antral Gastritis and Duodenitis (생쥐의 Gastrin 조절을 통한 六味地黃湯의 날문위염과 십이지장염 완화효과)

  • Objectives This study investigated the mitigating effect of Yukmijihwang-tang on antral gastritis and duodenitis by regulating gastrin release in mice. Methods : The normal group comprised mice with no inflammation; the control group comprised mice with ethanol-induced gastroduodenal inflammation. The sample group included inflammation-induced mice treated with Yukmijihwang-tang. Results Based on morphology and histochemistry findings, many hemorrhagic erosions were observed in the control group, while significantly fewer erosions were seen in the sample group. Immunohistochemistry findings showed that the distributions of gastrin, substance P, HSP70, NF-κB p65, and iNOS were considerably lower in the Yukmijihwang-tang -treated group than in the control group. Conclusions The findings suggest that Yukmijihwang-tang has a potent mitigating effect on antral gastritis and duodenitis in mice.

Immunoreactive Cells of Gastrin and Histamine and the Lymphocytes of Spleen and Thymus in Mice Stomach (화담청화탕 열탕액이 생쥐 위의 Gastrin, Histamine 면역반응세포와 비장, 흉선의 Lymphocytes에 미치는 영향)

  • To examine effects of Whadamcheongwha-tang(WDCWT) extract, the mice investigated the gastrin and histamine secreting cells of the stomach by immunohistochemical method, and the lymphocytes of the spleen and thymus by flow cytometry after the oral administration of WDCWT extract(0.2ml/day) and omeprazole (1mg/day) for 7, 14 and 21 days. The result are as follows; 1. When WDCWT extract was administrated for 7 days, in result, gastrin secreting cells were unchanged. When omeprazole was administrated for 7 and 14 days, gastrin secreting cells were slightly increased than that of normal control group. When WDCWT extract was administrated for 21 days, in result, gastrin secreting cells were significantly increased 1.9 times than that of normal control group. When omeprazole was administrated for 21 days, gastrin secreting cells were increased 1.96 times than that of normal control group. 2. When WDCWT extract and omeprazole were administrated for 7 days, in result, histamine secreting cells were unchanged. When WDCWT extract was administrated for 21 days, in result, histamine secreting cells were significantly increased 1.9 times more than that of normal control group. When omeprazole was administrated for 21 days, in result, histamine secreting cells were increased 2.1 times compared with normal control group. 3. When WDCWT extract administrated for 7, 14 and 21 days, in result, splenic Band T lymphocytes, especially T/sub H/ lymphocytes were significantly increased compared with normal control group, and thymic T/sub H/ lymphocytes were also increased in WDCWT administrated group for 14 days. The results suggest that WDCWT extract inhibit a gastric acid secretion in mice stomach, and is useful in the treatment of the hyperacidity and gastric ulcer.

The Change of Serum Gastrin Concentration with Oculo-acupuncture in Dogs (개에 있어서 안침(眼針)에 따른 혈청 가스트린 농도의 변화)

  • This study was performed to clarify the effect of oculo-acupuncture (OA) on change of serum gastrin concentration. Twenty-three clinically healthy mongrel dogs were used in this study. This study was comprised of two experiments. In experiment 1, the effect of OA (stomach/spleen region) with comparison of non-a cup oint was examined. On the other hand, the effect of OA (stomach/spleen region) with comparison by body acupuncture (BL-21) was also examined in experiment 2. Serum gastrin levels were measured on before, 0, 30, 60 and 120 minutes after treatment in both non-acupoint and stomach/spleen region groups, and BL-21 The results showed that significant decreases in serum gastrin level was detected on 30 mins(p<0.05) after OA when compared with that of non-acupoint group. Serum gastrin concentrations with OA at stomach/spleen region group were significantly decreased on 60 mins (p<0.05) and 120 mins (p<0.01) compared with those of the AP at BL21 group. In conclusion, it was demonstrated that OA at stomach/spleen region had significantly decreased serum gastrin levels in dogs and OA at stomach/spleen region decreased serum gastrin concentration more than that body acupuncture at BL-21.
