Crude oils are essential source of energy. It is majorly found in geographical locations beneath the earth's surface and crude oil is the main factor for the economic developments in the world. Natural crude oil contains unrefined petroleum composed of hydrocarbons of various molecular weights and it contains other organic materials like aromatic compounds, sulphur compounds, and many other organic compounds. These hydrocarbons are rapidly getting degraded by biosurfactant producing microorganisms. The present study deals with the isolation, purification, and characterization of biosurfactant producing microorganism from oil-contaminated soil. The ability of the microorganism producing biosurfactant was investigated by well diffusion method, drop collapse test, emulsification test, oil displacement activity, and blue agar plate method. The isolate obtained from the oil contaminated soil was identified as Bacillus subtilis. The identification was done by microscopic examinations and further characterization was done by Biochemical tests and 16SrRNA gene sequencing. Purification of the biosurfactant was performed by simple liquid-liquid extraction, and characterization of extracted biosurfactants was done using Fourier transform infrared spectroscopy (FTIR). The degradation of crude oil upon treatment with the partially purified biosurfactant was analyzed by FTIR spectroscopy and Gas-chromatography mass spectroscopy (GC-MS).

In order to develope the in vitro short term screen-ing method for carcinogen, we studied a purification method for thymine glycol in oxidaized DNA. Thymine glycol (5,6-dihydroxy-5, 6-dihydrothymine) is the major stable radiolysis poduct in thymine by chemical oxidants and ionzing radiation and it is a useful biomarker among oxidized DNA adducts, related with carcinogenests. Standard thymine glycol was prepared by oxidation of 〔$^3$H〕 thymine with KMnO$_4$ followed by purification with HPLC-LSC system and it was assayed by TLC and gas chromatography-MSD. 〔$^3$H〕 DMA adducts was isolated from E. coli (wild type ) treated with oxidative agents such as benzo(a)pyrene, adriamycin, aflatoxin B$_1$ and KBrO$_3$. These oxidative agents generated free radicals in cells by oxidative metabolism. As a result, thymine glycol was produced in cultured E. coli by four chemicals. This result shows that this methodology should be useful tool in screening oxidative carcinogen.

Carbon nanotubes(CNTs) are largely synthesized on Fe catalysts by catalytic chemical vapor deposition. The various shapes and compositions of these nanostructure CNTs were obtained by controlled parameters such as the reaction temperature, gas-mixing ratio. The influence of these parameters is investigated, together with observations of the produced materials after the purification processes. A diameter of CNTs, range from 2 to 10 nm, closely correlated with the size of the catalyst particle found attached to the tube end. The yield of CNTs was estimated to be 88.5 % and the purities of CNTs thus obtained were more than 80 %. The experimental results were documented with field emission scanning electron microscopy and raman spectroscopy and transmission electron microscopy, both before and after the purification.

The purpose of this study is to develop venturi type after-burner in order to obtain pollutant reduction effect and find the best stable combustion condition. For this purpose, through a flow analysis, the shape of venturi type was made and flame holder locations were also decided by measuring chemical species at before and after the after-burner. Also, various chemical species concentration were measured at changing the induced air rates and the oxygen for oxygen enrichment for the solution the problems of much oxygen flow rate and the flame stability range. As results of this study, a flow distribution and the purification effect was excellent at venturi contraction 0.5 and flame holder location 12mm below the center of Venturi throat. On the purification characteristics, we found that pollutants reduction was effective when area ratio and oxygen are increased. But there are suitable quantities due to the flame shape change and combustion efficiency.

A fuel handling process combined with a pressure swing adsorption system (PSA) was simulated to produce pure hydrogen with a purity greater than 99.97%. The simulation consists of two parts. The fuel processing part consisting of reformer and water-gas shift reaction was simulated with Aspen plus®, and the hydrogen purification part consisting of PSA was simulated with Aspen Adsorption®. In this study, the effect of reformer temperature and pressure on the total hydrogen production yield was investigated. Simulations were performed over a temperature range of 700 to 1,000℃ and a pressure range of 1 to 10 bar. The total hydrogen production yield increased with increasing temperature and decreasing pressure. The maximum hydrogen yield was less than 50% in the simulation and will be lower in the real process.
The removal efficiencies by elastic fire gas mask of toxic gases CO, HCl, HCN, and $SO_2$ produced by a fire have a key role in saving lives. The elastic fire gas mask comprises a visible window, elastic hood, gas purification canister, and air vent. It does not have hair or neck thongs, which makes it easy to use and put on quickly. This research examined the removal efficiency of toxic gases by such a mask. The removal efficiencies for CO with a background concentration of 2505.0 ppm were 99.99 and 99.98% after 3.5 and 8.5 min, respectively. The residual CO concentration was drastically increased after 8.5 min. The removal efficiencies for HCl, HCN, and $SO_2$ with background concentrations of 1003.0, 399.0, and 100.3 ppm, respectively, were 100% after 20 min.
To date, carbon and nitrogen co-doped photocatalysts (CN-$TiO_2$) for environmental application focused mainly on the aqueous phase to investigate the decomposition of water pollutants. Accordingly, the present study explored the photocatalytic performance of CN-$TiO_2$ photocatalysts for the purification of indoor-level gas-phase aromatic species under different operational conditions. The characteristics of prepared photocatalysts were investigated using X-ray diffraction, scanning emission microscope, diffuse reflectance UV-VIS-NIR analysis, and Fourier transform infrared (FTIR) analysis. In most cases, the decomposition efficiency for the target compounds exhibited a decreasing trend as input concentration (IC) increased. Specifically, the average decomposition efficiencies for benzene, toluene, ethyl benzene, and xylene (BTEX) over a 3-h process decreased from 29% to close to zero, 80 to 5%, 95 to 19%, and 99 to 32%, respectively, as the IC increased from 0.1 to 2.0 ppm. The decomposition efficiencies obtained from the CN-$TiO_2$ photocatalytic system were higher than those of the $TiO_2$ system. As relative humidity (RH) increased from 20 to 95%, the decomposition efficiencies for BTEX decreased from 39 to 5%, 97 to 59%, 100 to 87%, and 100 to 92%, respectively. In addition, as the stream flow rates (SFRs) decreased from 3.0 to 1.0 L $min^{-1}$, the average efficiencies for BTEX increased from 0 to 58%, 63 to 100%, 69 to 100%, and 68 to 100%, respectively. Taken together, these findings suggest that three (IC, RH, and SFR) should be considered for better BTEX decomposition efficiencies when applying CN-$TiO_2$ photocatalytic technology to purification of indoor air BTEX.

Pd-Ag alloy membranes have attracted a great deal of attention for their use in hydrogen purification and separation due to their high theoretical permeability, infinite selectivity and chemical compatibility with hydro-carbon containing gas streams. For commercial application, Pd-based membranes for hydrogen purification and separation need not only a high perm-selectivity but also a stable long-term durability. However, it has been difficult to fabricate thin, dense Pd-Ag alloy membranes on a porous stainless steel metal support with surface pores free and a stable diffusion barrier for preventing metallic diffusion from the porous stainless steel support. In this study, thin Pd-Ag alloy membranes were prepared by advanced Pd/Ag/Pd/Ag/Pd multi-layer sputter deposition on the modified porous stainless steel support using rough polishing/$ZrO_2$ powder filling and micro-polishing surface treatment, and following Ag up-filling heat treatment. Because the modified Pd-Ag alloy membranes using rough polishing/$ZrO_2$ powder filling method demonstrate high hydrogen permeability as well as diffusion barrier efficiency, it leads to the performance improvement in hydrogen perm-selectivity. Our membranes, therefore, are expected to be applicable to industrial fields for hydrogen purification and separation owing to enhanced functionality, durability and metal support/Pd alloy film integration.

Steam methane reforming is currently the most widely used technology for producing hydrogen, a clean fuel. Hydrogen produced by steam methane reforming contains impurities such as carbon monoxide, and it is essential to undergo an appropriate post-purification step for commercial usage, such as fuel cells. Recently, membrane separation technology has been gaining great attention as an effective purification method; in this study, we evaluated the feasibility of using commercial polysulfone membranes for biogas upgrading to separate and recover hydrogen from a hydrogen/carbon monoxide gas mixture. Initially, we examined the physicochemical properties of the commercial membrane used. We then conducted performance evaluations of the commercial membrane module under various conditions using mixed gas, considering factors such as stage-cut and operating pressure. Finally, based on the evaluation results, we carried out simulations for process design. The maximum H2 permeability and H2/CO separation factor for the commercial membrane process were recorded at 361 GPU and 20.6, respectively. Additionally, the CO removal efficiency reached up to 94%, and the produced hydrogen concentration achieved a maximum of 99.1%.

Gibberellin(GA) 3-$\beta$ hydroxylation is very important for the shoot elogation in the higher plants, since only 3$\beta$-hydryoxylated GAs promote shoot elogation in several plants. Fluctuation of 3$\beta$-hydryoxylase activity was examined during seed maturation using two cultivars of , P. vulgaris, Kentucky Wonder (normal) and Masterpiece (dwarf). Very immature seeds of both cultivars contain high level of 3$\beta$-hydroxylase activity (per mg protein). Both cultivars showed maximum of enzyme activity (per seed) in the middle of their maturation process. Gibberellin 3$\beta$-hydroxylase catalyzing the hydroxylation of GA20 to GA1 was purified 313-fold from very early immature seeds of P. vulgaris. Crude soluble enzyme extracts were purified by 15% methanol precipitation, hydrophobic interaction chromatogrphy, DEAE ion exchange column chromatography and gel filtration HPLC. The 3$\beta$-hydroxylase activity was unstable and lost much of its activity duting the purification. The molecular weight of purified enzyme was extimated to be 42, 000 by gel filtration HPLC and SDS-PAGE. The enzyme exhibited maximum activity at pH 7.7. The Km values for [2.3-3H] GA20 and [2.3-3H]GA9 were 0.29 $\mu$M and 0.33 $\mu$M, respectively. The enzyme requires 2-oxoglutarate as a cosubstrate; the Km value for 2-oxoglutarate was 250 $\mu$M using 3H GA20 as a substrate. Fe2+ and ascorbate significantly activated the enzyme at all purification steps, while catalase and BSA activated the purified enzyme only. The enzyme was inhibited by divalent cations Mn2+, Co2+, Ni2+, Cu2+, Zn2+, Cd2+ and Hg2+. Effects of several GAs and GA anaogues on the putrified 3$\beta$-hydroxylase were examined using [3H]GA9 and GA20 as a substrates. Among them, GA5, GA9, GA15, GA20 and GA44 inhibited the enzyme activity. [13C, 3H] GA20 was converted by the partially purified enzyme preparation to [13C, 3H]GA1, GA5 and GA6, which were identified by GC-MS, GA9 was converted only GA4, GA15 and GA44 were converted to GA37 and GA38, respectively. GA5 was epoxidized to GA6 by the preparation. This suggests that 3$\beta$-hydroxylation of GA20 and epoxidation of GA5 are catalyzed by the same enzyme in P, vulgaris.
