수삼중 휘발성 향기성분에 관한 연구 (Studies on the Volatile Flavor Components of Fresh Ginseng)

  • Volatile flavor components of fresh ginseng (Panax ginseng C.A. Meyer.) were studied. Steam distillate of fresh ginseng was extracted with ethyl ether and the extract was separated into four fraction: neutral, phenolic, acidic and basis fractions. The ethyl ether concentrates and neutral fraction were analyzed by a combination of SE-54 fused silica capillary gas chromatography and mass spectrometry. Major flavor components of fresh ginseng were predominantly mono(n +2) and sesquiterpenes(n +3) in over two hundred constituents. Of these, 28 were newly identified in volatile flavor components of fresh ginseng by GC-MS.

  • Twenty organic chemical substances(Table 2) were isolated from untreated wastewater, as well as treated wastewater, collected at 76 companys of 9 industry group located in the basin of Youngsan River. Those organic compounds were analyzed by Gas Chromatography/Mass Spectrometry(GC/MS) and confirmed through comparison with each standard reagents. Phenol, which was not detected in the raw wastewater, was identified in the effluent of treatment facility, indicating that phenol is generated from isopropylbenzene of plant wastewater.

  • Polychlorodibenzo-p-dioxins(PCDDs) have been prepared by microsacale pyrolysis of trichlorophenates. During the pyrolysis reaction, dechlorinated dibenzo-p-dioxins were also formed by the thermolysis of PCDDs. The dechlorination pathways of PCDDs were suggested in this reaction. The identification of these products was performed using capillary column gas chromatography-mass spectrometry.

Volatile Compounds in Oyster Hydrolysate Produced by Commercial Protease

  • Volatile compounds in raw oyster and oyster hydrolysate produced with protease were compared by vacuum simultaneous steam distillation-solvent extraction/gas chromatography/mass spectrometry. Sixty-two volatile compounds were detected in both samples. Of these, 57 were positively identified, composed mainly of aldehydes(12), ketones(9), alcohols(14), nitrogen-containing compounds(9), acids(6), terpenes(4), and miscellneous compounds(8). Levels of acids decreased after hydrolysis, whereas several other compounds such as aldehydes, ketones, and nitrogen containing compounds increased. Pyrazines, found in high abundance, were only detected in oyster hydrolysate.

  • Tetra-chlorodibenzo-p-dioxins (tetra-CDDs) were prepared by microscale pyrolysis of trichlorophenates. During the pyrolysis reaction, tri-, di-, and mono-CDDs were also formed by the thermolysis of tetra-CDDs. The dechlorination pathways of tetra-CDDs were suggested for this reaction.   • A method for determination 11 endocrine disrupting chemicals of phenols in various samples was deloped. The alkylphenols, chlorophenols and bisphenol A were determined by gas chromatography/mass spectrometry-selected ion monitoring (GC/MS-SIM) followed by two work-up methods for comparison; isobutoxycarbonyl (isoBOC) derivatization method and tert-butyldimethylsilyl (TBDMS) derivatization method. Eleven endocrine disrupting chemicals (EDCs) of phenols in biological samples were extracted with acetonitrile and then acetonitrile layer was refrigerated at -60$^{\circ}C$ for 2 hours (freezing filtration).   • 본 연구는 GC/MS를 이용하여 수질, 토양 및 저질 시료 중의 4-Nitrotoluene(4-NT)을 분석하는 방법을 확립하고자 하였다. 4-NT은 수질 시료(100mL)에서 n-hexane으로 추출하였으며, 토양 및 저질 시료(10g)에서는 먼저 메탄올로 추출한 후 hexane으로 다시 추출하여 농축시켜 분석하였다. 수질 시료 중의 4-NT의 회수율은 $99.1\%$ 이상이였으며 토양에서의 회수율(recovery)은 약 $110\%$를 보였고 재현성 (relative standard deviation)은 $11.99\%$ 이하였다. 검정곡선은 상관계수$(R^2)$값이 수질과 토양 모두에서 0.994이상의 좋은 직선성을 보여주었다. 본 연구에서 채취한 수질 43개 지역, 토양 35개 지역 및 저질 11개 지역의 모든 시료에서 4-NT은 검출한계 이하로 나타났다. 이 분석방법은 환경 중에 미량으로 존재하는 4-NT의 분석과 모니터링에 유용하게 사용할 수 있는 적합한 방법이라 사료된다.

Identification of New Urinary Metabolites of Byakangelicin, a Component of Angelicae dahuricae Radix, in Rats

  • Byakangelicin, 9-(2,3-dihydroxy-2-methylbutoxy)-4-methoxy-7H- furo[3,2-g][l]benzopyran-7-one (BKG), a component of Angelicae dahuricae Radix, is considered to be an inhibitor of aldose reductase for the treatment of diabetic cataract. An analytical method for the isolation of BKG developed by high-performance liquid chromatography has been reported. No literature on the metabolism of BKG, however, has been found. With the purpose of identifying new metabolites of BKG, BKG (100 mg/kg) was orally administered to Sprague-Dawley rats via a gavage. Using a metabolic cage, urine was collected for 24 h, and the urine samples were extracted by liquid-liquid extraction. For structural identification of new urinary metabolites of BKG, various instrumental analyses were conducted by gas-chromatography/mass spectrometry, high-performance liquid chromatography/diode array detector, liquid chromatography/mass spectroscopy with thermospray interface and $^1H$ nuclear magnetic resonance spectroscopy. Two metabolites produced from the Ο-demethylation or Ο-dealkylation of BKG were newly identified, and another new but unknown metabolite was assumed to be the hydroxylated form of BKG. These results indicate that the major metabolic products of BKG are formed by Ο-demethylation or Ο-dealkylation of BKG side chains.
