IDENTIFICATION OF GENES EXPRESSED IN LOW-DOSE-RATE γ-IRRADIATED MOUSE WHOLE BRAIN

  • While high-dose ionizing radiation results in long term cellular cytotoxicity, chronic low-dose (<0.2 Gy) of X- or ${\gamma}$-ray irradiation can be beneficial to living organisms by inducing radiation hormesis, stimulating immune function, and adaptive responses. During chronic low-dose-rate radiation (LDR) exposure, whole body of mice is exposed to radiation, however, it remains unclear if LDR causes changes in gene expression of the whole brain. Therefore, we aim to investigate expressed genes (EGs) and signaling pathways specifically regulated by LDR-irradiation ($^{137}Cs$, a cumulative dose of 1.7 Gy for total 100 days) in the whole brain. Using microarray analysis of whole brain RNA extracts harvested from ICR and AKR/J mice after LDR-irradiation, we discovered that two mice strains displayed distinct gene regulation patterns upon LDR-irradiation. In ICR mice, genes involved in ion transport, transition metal ion transport, and developmental cell growth were turned on while, in AKR/J mice, genes involved in sensory perception, cognition, olfactory transduction, G-protein coupled receptor pathways, inflammatory response, proteolysis, and base excision repair were found to be affected by LDR. We validated LDR-sensitive EGs by qPCR and confirmed specific upregulation of S100a7a, Olfr624, and Gm4868 genes in AKR/J mice whole brain. Therefore, our data provide the first report of genetic changes regulated by LDR in the mouse whole brain, which may affect several aspects of brain function.

Solar CO2-Reforming of Methane Using a Double-Layer Absorber (더블 레이어 흡수기를 이용한 고온 태양열 메탄-이산화탄소 개질반응)

  • Solar reforming of methane with CO2 was successfully tested with a direct irradiated absorber on a parabolic dish capable of 5kWth solar power. And the new type of double-layer absorber - the front layer, porous metal foam which absorbs the radiation and transfers the heat from material to gas, and the back layer, catalytically-activated metal foam - was prepared, and its activity was tested by using electric furnace. Ni was applied as the active metal on the gamma-Al2O3 coated Ni metal foam for the preparation of the catalytically-activated metal foam layer. Compared to conventional direct irradiation of the catalytically activated metal foam absorber, this new type of double layer absorber is found to exhibit a superior reaction and thermal storage performance at the fluctuating incident solar radiation. In addition, unlike direct irradiation of the foam absorber, double layer absorber has better thermal resistance, which prevents the emergence of cracks caused by mechanical or thermal shock. The total solar power absorbed reached up to 3.25kW and the maximum CH4 conversion was almost 59%.

Browning end Color Characteristics in Mushrooms (Agaricus bisporus) As Influenced by ionizing Energy (버섯의 갈변 및 색도에 대한 전리에너지의 영향)

  • Browning and color characteristics of stored mushrooms (Agaricus bisporus)following ionizing irradiation were investigated in connection with quality deterioration. The phenolic compounds of stored mushrooms showed a gradual decreasing tendency, while extractable browning pigments apparently rose from around 3days of storage under the conditions of $9{\pm}1^{\circ}C,\;80{\pm}7%$ RH and packaging with a corrugated paper box wrapped up in PE. ${\gamma}-irradiation$ at 2 to 3 kGy resulted in a significant reduction of their changes. Immediately after treatment, irradiated mushrooms were more discolored, i.e. a lower Hunter L value and higher Hunter a and b values than control. However, the subsequent storage for 15 days resulted in a preventive influence of ionizing energy on mushroom discoloration. This beneficial effect of ionizing energy was somewhat higher in the pilei than in the stipes of mushrooms and was found to increase lineally with increasing doses up to 3 kGy.

  • A neutron activation method has been developed for the simultaneous determination of chromium, iron, lanthanum, scandium and zinc in river-water samples. The sample is sealed in the silica ampoule without pretreatment and irradiated for a week at a thermal neutron flux of $1{\times}10^{13}n{\cdot}cm^{-2}{\cdot}sec^{-1}$. After cooling for about two days, the elements in the sample are sequentially extracted at different pH by 0.1M oxine-chloroform solution. The organic layers are checked by Gamma-ray spectrometry with $″3\;{\times}\;3″$ NaI (T1) detector connected to a 800-channel pulse hight analyzer. The ppb concentration of the elements in most of river-water samples could be determined by this method. The tracer study for the quantitative separation of the elements was also carried out.

The Radioprotective Effects of Rhizomata Herbs (대표적 근경류 생약의 방사선 방호효과)

  • We performed this study to determine the effect of Baizhu (Atractylodes japonica), Chuanxiong (Cnidium officinale), Shanyao (Discorea japonica) and Shengma (Cimicifuga heracleifolia), as Oriental rhizomata herbs, on jejunal crypt survival, endogenous spleen colony formation and apoptosis in jejunal crypt cells of mice irradiated with high and low dose of ${\gamma}$-radiation. Shengma was effective in intestinal crypt survival(p<0.05). The frequency of radiation induced apoptosis was also reduced by pretreatment with Chuanxiong and Shengma(p<0.05). Although the mechanisms of this effort remain to be elucidated, these results indicated that Shengma might be a useful radioprotector, especially since it is a relatively nontoxic natural product.

  • Effects of irradiation on the microbial growth and physicochemical properties of garlic powder were investigated during 3 months storage. Total bacteria and coliform group of garlic powder were $4.74{\times}10^{4}$ and $5.0{\times}10^{3}$ per g, respectively and irradiation of 5 kGy and 7 kGy could sterilize coliform group and total bacteria, respectively. $D_{10}$ value of total bacteria was 3.34 and no microorganisms were grown in 5 to f kGy irradiated groups after 3 months storage at $30{\pm}1^{\circ}C$. Moisture, sugars and pH of garlic powder were not remarkably changed during storage but pyruvic acid content was slightly decreased with storage period. Color difference of garlic powder after 3 months storage could not be distinguished by naked eye, but a slight change was recongized by the mechanical measurement.

The Radioprotective Effect of Kuei-Pi-Tang as a prescription of Traditional Chinese Medicine in Mice (생쥐에서 귀비탕 및 구성단미의 방사선 방호효과)

  • In this study, we evaluated the radioprotective effects of Kuei Pi Tang and its ingredients, as a prescription of traditional Oriental medicine. The jejunal crypt survival, endogenous spleen colony formation, and apoptosis in jejunal crypt cells were investigated in mice irradiated with high and low dose of ${\gamma}$ rays. In the mice administered with Kuei Pi Tang extract before irradiation, the jejunal crypts were protected significantly(p<0.005) and the frequency of radiation induced apoptosis was reduced(p<0.05). In the experiment on the effects of the ingredients of Kuei Pi Tang, the results indicated that the extract of Dangui(Angelica sinensis), Suanzaoren(Zizyphus valgaris), Rensan (Panax ginseng), Fuling(Poria cocos) and Muxiang(Saussurea lappa) might have a major radioprotective effect. Although the mechanisms of this inhibitory effect remain to be elucidated, these results indi cated that Kuei Pi Tang might be a useful radioprotector, especially since it is a relatively nontoxic natural product.

  • The radurization and radicidation effects on mixed condiment with Co-60 gamma irradiation at 3-9kGy and the physicochemical aspects of irradiated samples stored at $30^{\circ}C$ for three months were investigated. The initial microbial loads of the sample were $7.5{\times}10^5/g$ in total bacterial count and $1.2{\times}10^2/g$ in coliforms, respectively. The irradiation treatment of below 9 kGy could decontaminate the sample and $D_{10}$ value of the bacteria contaminated was shown to be 1.94 kGy. The chemical components associated with the quality of the sample were little affected by the irradiation at 3-6kGy. The sensory evaluation showed that the high sterilizing dose caused appreciably the change in overall flavors of mixed condiment.

Study on Diminution of Pesticide Residues in Citrus Fruits by γ-Irradiation (감마선 조사에 의한 감귤의 잔류농약 저감화 연구)

  • The citrus fruits were irradiated at different doses (25~200 kGy) to remove pesticides such as diuron, dichlorbenil, trifluralin, dimetoate, chlorothalonil, fenitrothion, bromacil, phenthoate and norflurazon, and the rates of degradation were detected by using gas chromatography electron capture detector (GC-ECD). The limit of detection (LOD) was in the range of 0.007~0.105 ppm. The limit of quantification (LOQ) was in the range of 0.024~0.352 ppm, and the recoveries of 9 pesticides were in the range of 71.5~106.5%. The results demonstrated that the ${\gamma}$-irradiation was effective method to remove pesticides such as diuron, dichlorbenil, trifluralin, dimetoate, chlorothalonil, fenitrothion, and norflurazon. However, the bromacil and phenthoate showed lower removal rates in the rate of 40.5 and 54.8%, respectively, at 200 kGy irradiation.

Physiological Characterization of Mono-Cotyledonous Model Plant by Ionizing Irradiation (단자엽 모델 식물의 이온화 에너지원에 따른 생리 활성)

  • The present study has been performed to compare the physiological analysis of monocot model plant (rice) in response to ionizing irradiations (cosmic-ray, gamma-ray, and Ion beam). Ionizing radiations were implanted into monocot model plant (rice) seed. After irradiation, the seeds were planted in the plastic pots for a growth period of one month. Thereafter, the morphological and physiological characteristics including malondealdehyde (MDA) and chlorophyll content, activities of antioxidant enzymes in irradiation samples were investigated. We are confirmed that the activity level of MDA and chlorophyll content were not changed by ionizing irradiation samples. However, the free radical contents were increased in all irradiated plants. And the activities of SOD, POD, and APX were significantly increased by irradiation compared with non-irradiation plant.
