A STUDY ON THE RELIABILITY OF THE OPTICAL CARIES ACTIVITY TEST (광학적 치아우식활성 검사법의 신뢰도에 관한 연구)

  • The purpose of this study was to evaluate the specificity, sensitivity, and diagnostic power of caries activity test using LED fluorescence. The subjects of this study were 55 children of $6{\sim}7$ years old. LED light were irradiated to labial or buccal surface of all teeth. Fluorescence from initial carious lesion of teeth illuminated by an LED light was observed through barrier filter and the number of teeth showing lesion, size and position of lesion were counted. Streptococcus mutans colony counting and dDfFtT rate test were also done and their correlation was compared. And then specificity, sensitivity, diagnostic power of optical caries activity test using LED light were evaluated. 1. There was positive $correlation({\gamma}=0.43)$ between LED fluorescence test and Streptococcus mutans count(P<0.05). 2. When visual examination was defined to standard testing method, the specificity, sensitivity, diagnostic power of LED fluorescence test were 100%, 76.1%, and 100%. 3. When dDfFtT rate was defined to standard testing method, the specificity, sensitivity, diagnostic power of LED fluorescence test were 88.9%, 47.8%, and 95.7%. 4. When S. mutans colony counting was defined to standard testing method, the specificity, sensitivity, diagnostic power of LED fluorescence test were 100%, 58.7%, and 100%. Considering the above results, optical caries activity test using LED light could be regarded as a practical method because of its close relationship with microbiological caries activity test.

Biological Evaluation of the Methanolic Extract of Eriobotrya japonica and Its Irradiation Effect (비파 메탄올 추출물의 생리활성 및 방사선 조사 효과)

  • Eriobotrya japonica has been used as a folk medicine for treatment of skin diseases, inflammation, coughing, phlegm, and ulcers in Korea and other Asian countries. In a search for possible bioactive agents from natural sources, we found that the methanolic extracts from various parts of E. japonica showed moderate antioxidative and antimicrobial activities in several in vitro bioassay systems. Additionally, the respective parts of E. japonica were irradiated at 20 kGy to investigate the effects of irradiation. Gamma irradiation of E. japonica extracts removed the deep greenish color without affecting its natural biological activities such as its antioxidative and antimicrobial properties. Based on these findings, the methanolic extracts of this plant source may be not affected by gamma irradiation as its bioactive constituents may be insensitive to this irradiation. Moreover, the methanolic extract of E. japonica may serve as a good natural resource for beneficial functions in food and other related industries.

Effects of Gamma Irradiation and Cooking Methods on the Content of Thiamin in Chicken Breast and Vitamin C in Strawberry and Mandarine Orange (방사선 조사와 가열처리에 의한 닭고기의 비타민 B1 및 감귤과 딸기의 비타민 C 함량의 변화)

  • In order to expand the acceptability of irradiated foods by public, substantial basic data about the change of nutrient contents during irradiation are needed. The nutrient contents and digestibility of macronutrients such as carbohydrates, proteins and lipids in foods are known to be not significantly influenced by irradiation treatment. However, some of the vitamins among micronutrients are susceptible to irradiation to a large extent depending upon the food composition, food process and storage condition. This study was conducted to investigate change of thiamin in chicken breast, and vitamin C in strawberry and mandarine orange after irradiation. The effects of irradiation at frozen or refrigerated state and the effects of cooking such as heating or micron ave on thiamin contents in chicken breast were observed. Irradiation reduced the thiamin content, however, temperature condition during irradiation was much more important factor to the loss of thiamin contents. In strawberry, vitamin C content was significantly affected by original content or the variety rattler than treatments such as irradiation, heating or microwave. These results indicated that the losses of water-soluble vitamins, especially thiamin or vitamin C, are affected by food temperature during irradiation process and variety or composition of foods rallier than irradiation itself, within an acceptable range of irradiation.

In Vitro Selection and Characterizations of Gamma Radiation-Induced Salt Tolerant Lines in Rice (방사선을 이용한 내염성 계통의 기내선발 및 특징)

  • The combination of radiation technique with an in vitro culture system was initiated to develop salt tolerant rice. We established an in vitro culture system to select tolerant lines against salt stress. NaCl tolerant cell lines were selected from the callus irradiated with gamma ray on N$_{6}$ medium with 1.5% NaCl and 2 mg/L 2,4-D. Regenerants (M$_1$) were obtained from the tolerant callus which was cultured for 30 days auxin-free medium. The M$_2$seeds were harvested from M$_1$plants on an individual plant basis. Thirty seedlings from each 450 M$_2$lines were transplanted in a field and total 5,000 M$_3$lines were harvested with an average 90 percent of fertile grain. M$_3$lines were utilized for selection of salt tolerance. Salinity-tolerant lines (225) were selected among 5,000 M$_3$lines. Of the 225 lines tested, the morphological traits of two lines (120-10 and -11) were far superior to control (Donagjinbyeo) in agromomic traits such as plant height, root length and no. of roots. Control and tolerant lines were analyzed by RAPD markers. Three polymorphic bands were presented in only tolerant lines, demonstrating a genetic difference between control and the tolerant lines. Such tolerant lines could be used as genetic resources to improve salt tolerance.e.

Effect of Gamma Irradiation on the Physiological Characteristics of Garlic Bulbs During Storage (감마선조사가 저장중 마늘의 생리적 특성에 미치는 영향)

  • This study was intended to investigate the physiological effect of gamma irradiation at 50, 100 and 500 Gy on the garlic bulbs stored at low temperature of $3\;{\pm}\;1\;^{\circ}C$ and a relative humidity of 70-80%, and room temperature of $12\;{\pm}\;5\;^{\circ}C$ and a relative humidity of 75-85%, respectively for 10 months. Irradiation treatment stimulated temporary the respiration of garlic, which was greatly affected by storage conditions later. Sprouting of garlic was effectively inhibited in the all irradiated and low temperature groups until 10 months of storage, while the nonirradiated and 50 Gy groups were partially rooted around the 4th month after storage under both conditions. Weight loss and spoilage were shown to be little affected by irradiation until the nonirradiated garlic sprouted, and at the latter stage of storage period, the optimum dose of irradiation showed a significant effect on the reduction of weight loss and spoilage in stored garlic. It was also found that irradiation for sprout inhibition did not influence the firmness of garlic cloves.

Inhibition of Photooxidation by Breakdown of ChlorophyII in Oil Model System Using Gamma Irradiation (감마선 조사된 유지 model system의 chlorophyII 제거 및 광산화 억제)

  • ChlorophyII destruction by irradiation in linoleic acid in methanol system was studied. ChlorophyII b standard (3 ppm) was added into methanol solution containing 1% of linoleic acid, and the sample was irradiated up to 20 kGy with or without nitrogen gas-bubbling. The content of chlorophyII b was analyzed by HPLC, and Hunter color value and UV-visible spectra were analyzed during 6 hr of photooxidation. The content of chlorophyll was reduced by irradiation and completely destroyed at 2.517 kGy. The model system with nitrogen gas-bubbling did not develop the lipid oxidation after irradiation or photooxidation at 20 kGy UV-visible spectra showed decreasing values of optical density by increase of irradiation doses, supporting the destruction of chlorophyII b. The results indicate that irradiation technology can be applied to reduce or eliminate the residual chlorophyII and to prolong the shelf-life of oil products.

  • The purpose of this study is to see the feasibility of the newly developed 2D dosimetry system using phosphor screen for helical tomotherapy. The cylindrical water phantom was fabricated with phosphor screen to emit the visible light during irradiation. There are three types of virtual target, one is one spot target, another is C-shaped target, and the other is multiple targets. Each target was planned to be treated at 10 Gy by treatment planning system (TPS) of tomotherapy. The cylindrical phantom was placed on the tomotherapy table and irradiated as calculations of the TPS. Every frame which acquired by CCD camera was integrated and the doses were calculated in pixel by pixel. The dose distributions from the fluorescent images were compared with the calculated dose distribution from the TPS. The discrepancies were evaluated as gamma index for each treatment. The curve for dose rate versus pixel value was not saturated until 900 MU/min. The 2D dosimetry using the phosphor screen and the CCD camera is respected to be useful to verify the dose distribution of the tomotherapy if the linearity correction of the phosphor screen improved.

Comparative Study on Human Risk by Ionizing Radiation and Pesticide as Biological Information about Environmental Disaster (환경재해에 관한 생물정보로서의 이온화 방사선과 살충제의 인체 위해성 비교 연구)

  • Environmental risk factors such as ionizing radiations, heavy metals, and pesticides can cause environmental disasters when they exist in excess. The increases in use of ionizing radiation and agricultural pesticide are somewhat related to the possibility of the disaster. The risk of radiation and pesticide was evaluated by means of the single cell gel electrophoresis (SCGE) assay on the human blood lymphocytes. The lymphocytes were irradiated with $0{\sim}2.0Gy$ of $^{60}Co$ gamma ray. Another groups of lymphocytes were exposed to various concentrations of parathion. Significantly increased tail moment, which was a marker of DNA strand breaks in SCGE assay, showed a clear dose- or concentration-response relationship. Parathion of a recommended concentration for agricultural use ($1mg {\ell}^{-1}$ ) has a strong cytotoxic effect on lymphocytes, which is equivalent to damage induced by 0.1 Gy of ${\gamma}$-ray. Furthermore, $2mg{\ell}^{-1}$ of parathion can give rise to DNA damage equivalent to that induced by 0.25 Gy at which the radiation-induced damage can start to develop into clinical symptoms. The comparative results of this study can provide an experimental basis and biological information for the prevention of environmental disaster.

Effects of Radiation on the Chromosomes in Bone Marrow Cells of the Mouse (방사선(放射線)이 생쥐 골수세포(骨髓細胞)의 염색체(染色體)에 미치는 향영(響影))

  • To study the relationships among the frequency of chromosomal aberration induced by radiation, mouse age at the time of exposure and elapsed time after the irradiation, the 40, 150 and 300 day old mice were irradiated by gamma rays to 200 rad of acute whole body doses. The bone marrow chromosomes were observed in 8 experimental groups such as 1, 2, 3, 4, 8, 16, 30, 60 and 90 days after irradiation. In the 40 day old mice the frequencies of cell with chromosomal aberration were 53.0%, 21.5%, 8.4%, 8.4%, 12.0%, 8.0%, 7.0% and 5.0% in 8 experimental groups, in the 150 day old mice 32.5%, 12.1%, 10.5%, 12.8%, 0.03%, 9.7%, 7.0% and 4.2%. The 300 day old mice showed that the frequencies of cells with chromosomal aberration were 27.7%, 13.6%, 9.0%, 9.3%, 13.3%, 14.4%, 7.5% and 9.5% in each experimental group. There were tendencies that the frequencies of cell with chromosomal aberration were higher in the young mice than the old ones within 2 days after the irradiation but in the old mice than the young ones after 4 days.

Amylose, Tocopherol, Free Sugar and Fatty Acid Content in Selected Mutant Lines of Oryza sativa cv. Shindongjin

  • To assess the potential as biofortified rice varieties, new endosperm and grain mutant lines were selected from $M_4$ generation seeds of the rice cultivar Shindongjin, which were either $\gamma$-irradiated or treated with N-methyl-N-nitrosourea(MNU) and lipid, sugar, and tocopherol content were analyzed. Amylose content in non-waxy mutants ranged from 8.8% in SM-4, a dull-type mutant, to 29.5% in SM-51, compared to 18.9% in the parental variety, Shindongjin. SM-23, a floury-type mutant, contained 0.09 ${\mu}g/g$ $\alpha$-tocopherol(40.9% of total tocopherol), was three times higher than in the parental variety. SM-32, a giant embryo-type mutant, had a 2.2-fold higher total tocopherol content, 2.1-fold higher $\alpha$-tocopherol, and 5.5-fold higher $\delta$-tocopherol content(47.3% of total tocopherol) than the parental variety(0.13 ${\mu}g/g$). Total free sugar content was elevated in all selected mutants and 1.2-8.6 times higher than in the parental variety(11.38 ${\mu}g/g$). These increased sugar levels were due to increase in sucrose concentration. SM-23(floury-type mutant) and SM-51(high amylose-type mutant) had 4.6 and 7.0 times more sugar, respectively, than the parental variety(11.38 ${\mu}g/g$). With relatively high concentrations, most mutants showed elevated fatty acid content in the SM 32(giant embryo-type) and SM-51(high amylose-type) mutants, at 124.56 and 89.59 mg/g, respectively. All selected mutants displayed valuable characteristics for the development of new varieties in rice-breeding programs.
