Effects of acute dibutyl phthalate administration on hepatic lipid peroxidation and gamma-glutamyl transferase activity in mice (마우스에서 dibutyl phthalate 급성 투여가 간 지질과산화와 gamma-glutamyl transferase 활성에 미치는 효과)

  • Dibutyl phthalate (DBP) is used extensively in the plastic industry and has been known as an endocrine disruptor. Present study was undertaken to examine whether DBP can induce oxidative stress in mice. In this study, oxidative stress was measured in terms of the modification of lipid peroxidation and gamma-glutamyl transferase (GGT) activity. The serum toxicity index, level of lipid peroxidation and triglyceride (TG), and activity of GGT were measured in male ICR mice after a single administration of DBP (5 g/kg, po). DBP did not alter serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), creatinine, glucose and cholesterol level. However, the treatment with DBP was found to significantly increase the level of lipid peroxidation in liver and lung. The TG content and activity of GGT in the liver of DBP-exposed animals was also increased. These results indicate that DBP can induce mild oxidative stress in mice. The GGT activity is considered to be increased as one of the adaptive defense mechanisms to oxidative stress induced by DBP.

Evaluation of Clinical Usefulness of Gamma Glutamyl Transferase as a Surrogate Marker for Metabolic Syndrome in Non Obese Adult Men (비만하지 않은 성인 남성에서 대사증후군의 대리 표지자로서 감마 글루타밀 전이효소의 임상적 유용성 평가)

  • This study was to evaluate the usefulness of gamma glutamyl transferase (GGT) as a surrogate marker predicting metabolic syndrome. 7,155 non obese men over the age of 20 were studied as subjects. The criteria for diagnosing MetS were the National Cholesterol Education Program - Third Adult Treatment Panel (NCEP-ATP III). The risk of developing MetS according to GGT was conducted logistic regression analysis, and the ROC (receiver operating characteristic) curve was obtained to confirm GGT ability to predict the risk of MetS. Regardless of age and body mass index, MetS had a 7.09 times higher risk of onset in the fourth quartile than in the first quartile of GGT (p<0.001). The AUC (area under the curve) of GGT for the diagnosis of MetS was 0.715, and the cutoff value of GGT was 40.0 U/L, the sensitivity was 65.0%, and the specificity was 70.2%. Therefore, GGT is considered to be a useful diagnostic index for diagnosing MetS.

The Alteration of % Carbohydrate-Deficient Transferrin and Gamma-Glutamyl Transferase Levels of Alcohol-Dependent Inpatients according to Age and Sex (알코올의존으로 입원한 환자에서 나이와 성별에 따른 퍼센트 탄수화물-결핍트랜스페린과, 감마-글루타밀전이효소 변화 양상의 차이)

  • Objectives This study sought to investigate the relationship between age, sex and alterations in levels of % carbohydrate-deficient transferrin (%CDT) and gamma-glutamyl transferase (GGT) in patients admitted with alcohol dependence. Methods The study retrospectively enrolled 187 patients who were diagnosed with alcohol dependence according to the Diagnostic and Statistical Manual of Mental Disorders-Fourth edition (DSM-IV) and were admitted into a closed ward in Hallym University Hangang Sacred Heart Hospital from 2009 to 2012 and Hallym University Kangnam Sacred Heart Hospital from 2012 to 2017. Demographic factors (age, sex) and biochemical markers [%CDT, GGT, mean corpuscular volume (MCV), aspartate transferase (AST), alanine transferase (ALT)] were collected by reviewing medical records. Alterations in the levels of %CDT and GGT in different groups for each demographic factor were compared after correcting for confounding variables (age, initial %CDT, GGT, MCV, AST, ALT). Results Decreased %CDT and GGT were observed during the period of abstinence after admission. The normalization period for %CDT increased with age, while the normalization period for GGT was longer in female patients. Conclusions These results suggest that alcohol-dependent patients that vary in age have different alterations in %CDT, while different sexes have different alterations in GGT. Age and sex can be potential indicators of treatment response after abstinence in patients with alcohol dependence. Further studies are needed to evaluate the relationship between these factors with regards to physiological and hematological changes in alcohol dependence.

Behavioral factors associated with serum gamma-glutamyl transferase activity in a male rural population (농촌 지역 남성들의 혈청 gamma-glutamyl transferase 활성도와 관련된 행태적 요인에 관한 연구)

  • Although serum gamma-glutamyl transferase(GGT) has been widely used as a marker of alcoholic hepatic dysfunction, little is known as to behavioral correlates in the normal population. To examine the association between serum GGT activity and some behavioral factors in male rural population, data un health examination in a rural population (248 males aged 40 years and older) was analyzed Multiple linear regression and analysis of convariance were used to control the effect of confounding factors. Adjusted average differences in the level of serum GGT according to body mass index(BMI: $kg/m^2$) and alcohol intake(ml/day) were statistically significant(p=0.051 0<0.001 respectively). Serum GGT activity for BMII$\geq$25 was significantly higher than for BMI<25 in non-drinkers(p=0.007), but not significantly different in drinkers(p=0.892). Alcohol intake was significantly associated with elevated serum GGT activity for both BMI$\geq$25 and BMI<25(p<0.001, p=0.002 respectively). These findings suggest that alcohol drinking, obesity in non-drinkers are important factors associated with serum GGT in male rural population.

  • A ninhydrin positive substance(formed during germination of Mung Bean) which on a 2-dimensional chromatography(BuOH-HAc, and$ PhOH-NH_2$ gave a spot above glutamic acid was isolated by cutting out the appropriate spot on the paper and extracting the paper with water. Hydrolysis of this substance with $2N H_2SO_4\;or\;3N\;HCl\;at\;120^{\circ}C$ for 15 hours gave the spot of mainly glutamic acid, alanine and very faint of glycine and cysteine on a paper chromatography. It is suggested that the reaction of ${\gamma}$-glutamyl-cysteinecyl-glycine + L-alanine in the presence of ${\gamma}$-glutamyle transferase ${\rightarrow}$ ${\gamma}$-glutamyl-alanine + cysteinyl-glycine, takes place in germinated Mung Bean.

Comparison of the Serum Gamma-Glutamyl Transferase and Serum Uric Acid Levels according to the Obesity Types Based upon on the Body Mass Index in Korean Men (성인 남성에서 체질량지수로 분류한 비만 유형에 따른 혈청 감마 글루타밀 전이효소와 혈청 요산농도의 차이 비교)

  • This study examined the relationship between the serum GGT (gamma-glutamyl transferase) and uric acid level in obese male adults. A total of 14,283 adult males aged 20 years or more, who visited the health examination center of Gyeonggi Regional General Hospital from January 2017 to August 2018 and underwent a physical examination, were enrolled in this study. The obesity criteria were defined by the Asia-Pacific regional standards. Abdominal obesity was defined as a male waist circumference of more than 90 cm. An increase in the serum uric acid and serum CGT levels of the male subjects was defined as 7.0 mg/dL or more and 56 IU/L or more, respectively. The results showed that the serum GGT and uric acid levels were higher in the overweight and obese groups than in the normal weight group. In the obese group, the serum GGT and uric acid were significantly higher in the patients with abdominal obesity. On the other hand, there was no difference compared to the low body weight group. The overweight and obesity groups showed a higher risk of elevated serum GGT and hyperuricemia than the normal weight group, but a low body weight did not affect the serum GGT elevation and hyperuricemia. Overall, the serum GGT and uric acid levels are useful for evaluating overweight and obesity in adult males.

Effects of Enzyme Addition to Broiler Diets Containing Varying Levels of Double Zero Rapeseed Meal

  • Maize-soybean meal diets with 0, 100, 200 and 300 g/kg double zero rapeseed meal ('00' RSM) with and without an enzyme mixture (xylanase, pectinase, cellulase) at a level of 1.6 g/kg were evaluated with 624 day-old broiler chicks for 5 weeks. The birds were randomly allocated to eight dietary treatments with three replicates of 26 birds each. Average daily gain (ADG) and feed intake (FI) were recorded weekly and ileal viscosity, organ weights, serum enzyme activity, hormonal profile and hematological parameters were measured at the end of week 5. Average daily gain during the weekly periods was significantly influenced by the dietary level of '00'RSM (p<0.01). Inclusion of '00' RSM improved the ADG up to day 28 with the increased level; beyond that time no improvement was recorded when compared to control groups. However, ADG from 1-35 days was significantly different between 300 g/kg inclusion level of '00' RSM and the control diet. Inconsistent decline in feed intake and feed conversion ratio was observed up to day 21 and the trend was reversed thereafter. The proportion of '00' RSM in the diet had a significant ($p{\leq}0.05$) influence on thyroid weight but had no effect on the relative weights of liver and heart, serum enzyme activities (${\gamma}$-glutamyl transferase, alanine amino transferase and aspartate amino transferase), thyroid hormones ($T_3$ and $T_4$), hemoglobin level and hematocrit. Significant improvement in ADG was recorded during the 2nd week of age with the addition of enzyme, whereas for all other periods, including the whole period of the trial, higher but non-significant ADG was observed. FI and FCR were not affected by the addition of enzyme but there was a numerical reduction in FCR during the whole period. The addition of enzyme reduced the ileal viscosity at all levels of '00' RSM inclusion. The results suggest that '00' RSM can be included up to 300 g/kg in broiler diets without any adverse effects on health and performance. The addition of commercial enzyme mixture containing xylanase, pectinase, cellulase to broiler diets containing '00'RSM has some effect on growth rate and feed conversion efficiency.

Surgical Correction of Peritoneopericardial Diaphragmatic Hernia with Hepatic Necrosis in a Himalayan Cat (히말라얀 고양이에서 간괴사를 동반한 복막심낭횡격막 허니아의 외과적 치료)

  • A five months old, female Himalayan cat was referred to the Veterinary Medical Teaching Hospital at Konkuk University, because of suspecting peritoneopericardial diaphragmatic hernia (PPDH). After consecutive examination, the patient was diagnosed as PPDH and hepatic dysfunction. In spite of medical therapy for a month, the serum chemistry profiles for liver enzymes (aspartate transferase: 469 U/L, alanine transferase: above analysis, gamma-glutamyl transferase: above analysis) did not decrease to the normal range. In operation, some of liver was necrotized and was adhesive to diaphragm, these were gently dissected from thoracic structures, and resected. After debriding, the edges of the defect of diaphragm were closed with a simple continuous suture pattern. One month after operation, the cat had normal condition, recovery was uncomplicated, and the serum chemistry profiles for liver enzymes decrease to the normal range.

Domestic review of Korean medical treatment on Carbon Tetrachloride-induced Hepatotoxicity - Focusing on Treatment of Korean Herbal Medicine (본초학적 접근을 통해 본 국내에서 연구된 한약재의 사염화탄소로 유발된 간손상에 대한 간보호 효과)

  • The purpose of this study is to inspect trends of the paper of Korean medical treatment on Carbon Tetrachloride-induced Hepatotoxicity and try to establish the future direction for development of Korean herbal medicine. We reviewed 79 papers which had been published from 1981 to 2010 in journals published in Korea. According to these studies, Carbon Tetrachloride-induced hepato-celluar degeneration and necrosis induced to increase in serum aspartate amintransferase (AST), alanine aminotransferase (ALT), Alkaline phosphatase (ALP), ${\gamma}$-Glutamyl transferase (${\gamma}$-GTP), lactate dehydrogenase (LDH), bilirubin, blood urea nitrogen (BUN) and albumin levels. In biochemical analyses, antioxidant enzymes such as superoxide dismutase (SOD), Glutathione S-transferase (GST) and catalase in hepatic tissue were remarkably incresed by Carbon Tetrachloride treatment. We found that some of the herbal extracts have a protective effect against Carbon Tetrachloride-induced Hepatotoxicity. More studies of oriental medicinal herbs are required for developing a treatment of hepatotoxicity.

Contrast Media Side Effects Prediction Study using Artificial Intelligence Technique (인공지능 기법을 이용한 조영제 부작용 예측 연구)

  • The purpose of this study is to analyze the factors affecting the classification of the severity of contrast media side effects based on the patient's body information using artificial intelligence techniques to be used as basic data to reduce the degree of contrast medium side effects. The data used in this study were 606 examiners who had no contrast medium side effects in the past history survey among 1,235 cases of contrast medium side effects among 58,000 CT scans performed at a general hospital in Seoul. The total data is 606, of which 70% was used as a training set and the remaining 30% was used as a test set for validation. Age, BMI(Body Mass Index), GFR(Glomerular Filtration Rate), BUN(Blood Urea Nitrogen), GGT(Gamma Glutamyl Transgerase), AST(Aspartate Amino Transferase,), and ALT(Alanine Amiono Transferase) features were used as independent variables, and contrast media severity was used as a target variable. AUC(Area under curve), CA(Classification Accuracy), F1, Precision, and Recall were identified through AdaBoost, Tree, Neural network, SVM, and Random foest algorithm. AdaBoost and Random Forest show the highest evaluation index in the classification prediction algorithm. The largest factors in the predictions of all models were GFR, BMI, and GGT. It was found that the difference in the amount of contrast media injected according to renal filtration function and obesity, and the presence or absence of metabolic syndrome affected the severity of contrast medium side effects.
