We designed a single polyethylene bonner sphere with several thermo-luminescence dosimeters (TLD), for measurement of neutron energy spectrum. For the separation of the neutron dosage in the neutron-gamma mixed field, we used 21 ea TLD-600s and TLD-700s, respectively. Because, TLD-600 is sensitive to neutron and gamma rays, and, TLD-700 is sensitive only to gamma-rays, we could determine the each dose by neutron and gamma rays. The neutron response function of the bonner sphere with TLDs was calculated by MCNPX (ver. 2.5.0) Monte Carlo simulation in the energy range from $10^{-1}$ to 20 MeV. For the Cf-252 standard neutron source in KRISS, we could estimate the neutron energy spectrum by unfolding method using the response function.
When DNA double-strand breaks (DSBs) were induced in mammalian cells, many DNA damage response proteins are accumulated at damage sites to form nuclear foci called IR-induced foci. Although the formation of foci has been shown to promote repair efficiency, the structural organization of chromatin in foci remains obscure. BAF53 is an actin-related protein which is required for maintenance of chromosome territory. In this study, we show that the formation of IR-induced foci by $\gamma$-H2AX and 53BP1 were reduced when BAF53 is depleted, while DSB- activated ATM pathway and the phosphorylation of H2AX remains intact after DNA damage in BAF53 knockdown cells. We also found that DSB repair efficiency was largely compromised in BAF53 knockdown cells. These results indicate that BAF53 is critical for formation of foci by $\gamma$-H2AX decorated chromatin at damage sites and the structural organization of chromatin in foci is an important factor to achieve the maximum efficiency of DNA repair.
Background & Objectives : Rhinitis is an inflammation of nasal mucosa. The major symtoms are watery rhinorrhea, sneezing, itchy nose, and nasal obstruction. Allergic rhinitis is an immune reaction by allergen. So, we aimed to determine therapeutic effects of Acorus gramineus by observing changes in IL-4, $IFN-{\gamma}$ and the nasal mucosal tissue. Materials and Methods : Fifteen BALC/c mice were divided into three groups : m group(which ate low concentrated herbal medicine ), M group(which ate high concentrated herbal medicine) and control group. Control and experimental group were induced allergic rhinitis by Ovalbumin as the method of Levin and Vaz. Experimental group was orally administered the Acorus gramineus extract for 28days. We observed changes in IL-4, $IFN-{\gamma}$ and trans aminase(AST, ALT) in blood and nasal mucosa and submucosa. Results : There were no significant changes statistically in IL-4 and $IFN-{\gamma}$ in blood(p<0.05). And there were no hepatotoxicity with Acorus gramineus extract. Histologically, almost no inflammatory response in treatment group(m,M) against that there were inflammatory response(increased goblet cells, dilated vessels, edema of bowman's glands and injured olfactory hairs) in control group. Conclusion : According to above results, it is supposed that Acorus gramineus has no immunological effects on allergic rhinitis.

The virtual Frisch-grid method for room-temperature radiation detectors has been widely used because of its simplicity and high performance. Recently, side electrodes were separately attached to each surface of the detectors instead of covering the entire detector surface with a single electrode. The side-electrode structure enables the measurement of the three-dimensional (3D) gamma-ray interaction in the detector. The positional information of the interaction can then be utilized to precisely calibrate the response of the detector for gamma-ray spectroscopy and imaging. In this study, we developed a 3D position-sensitive 5 × 5 × 12 mm3 cadmium-zinc-telluride (CZT) detector and applied a flattening method to correct detector responses. Collimated gamma-rays incident on the surface of the detector were scanned to evaluate the positional accuracy of the detection system. Positional distributions of the radiation interactions with the detector were imaged for quantitative and qualitative evaluation. The energy spectra of various radioisotopes were measured and improved by the detector response calibration according to the calculated positional information. The energy spectra ranged from 59.5 keV (emitted by 241Am) to 1332 keV (emitted by 60Co). The best energy resolution was 1.06% at 662 keV when the CZT detector was voxelized to 20 × 20 × 10.
The gamma-gamma logging method appplying in geophysical research are presented in this paper_ The logging probe assembly was designed which permits changing the source-to-detector spacing while conditions of proceeding ${\gamma}-{\gamma}$ logging, which a collimated gamma ray source ($^{60}Co$, 0.5mCi and/or 2 mCi) is separated from the scintillation detector as shown in Fig. 2 and 3, size is 6.0 cm in diameter and 120.0 cm in long and the exposed parts are made of stainless steel pipe. The results is confirmed by the experiment performed mainly in granite rock where a slightly constant shape was obtained but sometimes was shown sharpness shape for the measured scattered gamma-ray intensity. Consequently, the experimental results are obtained an adequate intensity of scattered gamma-rays and favourable response to density change, and also very closely correspond to between core samples of the test boring and to used this method of ${\gamma}-{\gamma}$ logging in the test bore-hole of the strata.
A cDNA encoding chicken interferon-gamma (chIFN-${\gamma}$) was amplified from P34, a CD4$^{+}$ T-cell hybridoma by reverse transcription-polymerase chain reaction (RT-PCR) and cloned into pUC18. THe sequences of cloned PCR products were determined to confirm the correct cloning. Using this cDNA as probe, chicken genomic library from White Leghorn spleen was screened. Phage clones harboring chicken interferon-gamma (chIFN-${\gamma}$) were isolated and their genomic structure elucidated. The chIFN-${\gamma}$ contains 4 exons and 3 introns spanning over 14 kb, and follows the GT/AG rule for correct splicing at the exon/intron boundaries. The four exons encode 41, 26, 57 and 40 amino acids, respectively, suggesting that the overall structure of IFN-${\gamma}$ is evolutionairly conserved in mammalian and avian species. The 5’-untranslated region and signal sequences are located in exon 1. Several AT-rich sequences located in the fourth exon may indicate a role in mRNA turnover. The 5’-flanking region contains sequences homologous to the potential binding sites for the mammalian transcription factors, activator protein-1(AP-1) activator protein-2(AP-2) cAMP-response element binding protein(CREB), activating transcription factor(ATF), GATA-binding fator(GATA), upstream stimulating factor(USF), This suggests that the mechanisms underlying transcriptional regulation of chicken and mammalian IFN-${\gamma}$ genes may be similar.r.
Kamiinsamyangyoung-tang(KIT) has been widely used to treat amenorrhea and bradymenorrea caused from vital energy and blood deficit. KIT was composed of Insamyangyoung-tang, Cervi Cornu Pantotrichum and Hominis Placenta. The aim of this study is to investigate effect of KIT on Immune response and Blood formation. We investigated thymocytes, splenocytes population, ${\gamma}$-interferon, IL-2, IL-4, NO and phagocytic activity. We found that KIT had no effect on the Th and Tc cell population of thymocytes, Th cell population of splenocytes and ${\gamma}$-interferon quantity was decreased. KIT decreased the formation of Nitric Oxide from abdominal macrophage, on the other hand, it had no influence on the quantity of IL-2, IL-4.

Adaptive response induced by low dese .gamma.-ray irradiation in human cervical carcinoma cells was examined. Cells were exposured to low dose of .gamma.-ray irradiation in human cervical carcinoma cells was examined. Cells were exposured to low dose of .gamma.-ray (1-cGy) followed by high doses of r-ray irradiation (0,1,2,3,5,7 and 9Gy for chlnogenic assay or 1.5Gy for micronucleus assay) with various time intervals. Survival fractions of cells in both low dose-irradiated and unirrated groups were analyzed by clonogenic assay. Surviva fractions of low dose-irradiated in cell survival was maximum when low and high dose irradiation time interval was 4 hr. Frequencies of micronuclei which is an indicative of chromosome aberration were also enutained from survival fractions analyzed by clonogenic assay, maximum when low and high dose irradiation time interval was 4hr. Frequencies of micronuclei which is an indicative of chromosome aberration were also enumerated in both low dose-irradiated and unirradiated groups. In consiststent with the result obtained from survival fractions analyzed by clonogenic assay, maximum reduction in frquencies of micronuclei was observed when low dose radiation was given 4 hr prior to high response to subsequent high dose .gamma.-ray irradiation in human cervical carcinomal cells. Our data suggest that one of the possible mechanisms of adaptive response induced by low dose rediation is the increase in repair of DNA double strand breaks in low dose radiation-adapted cells.
Background: As an attempt to develop a strategy to improve the protective immune response to GM-CSF-secreting CT26 (GM-CSF/CT26) tumor vaccine, we have investigated whether the apoptogenic treatment of GM-CSF/CT26 prior to vaccination enhances the induction of anti-tumor immune response in mouse model. Methods: A carcinogeninduced mouse colorectal tumor, CT26 was transfected with GM-CSF gene using a retroviral vector to generate GM-CSF-secreting CT26 (CT26/GM-CSF). The CT26/GM-CSF was treated with ${\gamma}$-irradiation or mitomycin C to induce apoptosis and vaccinated into BALB/c mice. After 7 days, the mice were injected with a lethal dose of challenge live CT26 cells to examine the protective effect of tumor vaccination in vivo. Results: Although both apoptotic and necrotic CT26/GM-CSF vaccines were able to enhance anti-tumor immune response, apoptotic CT26/GM-CSF induced by pretreatment with ${\gamma}$-irradiation (50,000 rads) was the most potent in generating the anti-tumor immunity, and thus 100% of mice vaccinated with the apoptotic cells remained tumor free for more than 60 days after tumor challenge. Conclusion: Apoptogenic pretreatment of GM-CSF-secreting CT26 tumor vaccine by ${\gamma}$-irradiation (50,000 rads) resulted in a significant enhancement in inducing the protective anti-tumor immunity. A rapid induction of apoptosis of CT26/GM-CSF tumor vaccine at the vaccine site might be critical for the enhancement in anti-tumor immune response to tumor vaccine.
In the present study, the impact of Salmonella Typhimurium on cell-mediated immunity (CMI) was investigated in 5 week-old immuno divergent broiler lines selected for the high and low response to phytohemagglutinin-P. The immune response was assessed in peripheral-blood mononuclear cells (PBMCs) induced with Salmonella Typhimurium at different time intervals (0 h, 0.5 h, 2 h, 4 h, 6 h, 12 h and 24 h). The differential mRNA expression patterns of IFN-${\gamma}$, IL-2 and iNOS were evaluated by quantitative real time PCR. In-vitro production of nitric oxide (NO) was also estimated in the culture supernatant and correlated with iNOS mRNA expression. Present study showed higher production of NO in the high cell-mediated line (HCMI) as compared to the low cell-mediated line (LCMI) upon stimulation with Salmonella Typhimurium. Correspondingly, higher mRNA expression of iNOS and IFN-${\gamma}$ were observed in high response birds (HCMI); but IL-2 was down regulated in this line compared to the low response birds (LCMI). Significantly (p<0.05) higher expression of iNOS, IFN-${\gamma}$ and higher production of NO in high line indicated that the selection for PHA-P response might be employed for increasing the immune competence against Salmonella Typhimurium in chicken flocks.
