Purification and Properties of ${\gamma}-Cyclodextrinase$ from Bacillus megaterium(KFCC 11855) (Bacillus megaterium이 생산하는 ${\gamma}-cyclodextrinase$의 정제와 특성에 관한 연구)

  • The experiment was carried out to purify and to investigate the properties of the cyclodextrinase produced from Bacillus megaterium KFCC 11855. The enzyme was partially purified with $(NH_4)_2SO_4$ and chromatography on DEAE-trisacryl, Ultrogel AcA 34, DEAE-trisacryl and Ultrogel HA. The optimum temperature and pH of the purified enzyme were $60^{\circ}C$ and 6.0, respectively. The enzyme was stable at the temperature of $45^{\circ}C$ below and at the pH range of $6.0{\sim}9.0$, respectively. The Km value for ${\gamma}-cyclodextrin$ was 0.903 mM. The enzyme activity was increased by $Mg^{2+}$ and $Mn^{2+}$, but decreased by $Hg^{2+}$ and $Cu^{2+}$. The enzyme degraded ${\gamma}-cyclodextrin$ but not ${\alpha}-cyclodextrin$. The degree of ${\beta}-CD$ degradation by the enzyme was very low. The decomposed products of ${\gamma}-cyclodextrin$ by the enzyme were mainly glucose, maltose and a little amount of maltotriose.

The Changes of Casein of Cheese Base Treated with Enzyme during Ripening (Enzyme의 처리에 의한 Cheese Base의 숙성중 Casein의 변화)

  • This experiment was carried out to investigate the changes of casein of cheese base treated with substitute enzyme during ripening. The cheese base without enzyme treatment(control, D)and cheese base treated with only calf rennet(A), cheese base treated with mixed enzyme(calf rennet :porcine pepsin 1:1, B), cheese base treated with only porcine pepsin(C) were manufactured. The changes of casein were analyzed by means of HPLC and electrophoresis as experimental parameters during ripening. Gel filtration(HPLC) of casein by Superose 12 column in Cheddar cheese showed 5 fractions immediately after manufacturing and 8 fractions after six months ripening. Though D showed no difference in number of fraction(4 fraction) during 8 weeks ripening, A, B, C have represented the change of fraction number 4 to 5, 4 to 7, 4 to 8, respectively. As the mixing ratio of porcine pepsin increased, higher degradability of casein appeared. After 8 weeks ripening, electrophoresis of casein in cheese base showed three bands as an ${\alpha}$$_{s1}$casein from A and five bands from B, C. In case of D one major band and two minor bands were appeared as an ${\alpha}$$_{s1}$-casein. As the additional level of porcine pepsin increased the concentration of ${\beta}$-casein band decreased. however, that of ${\gamma}_1$ ${\gamma}_2$-casein band increased and para-${\kappa}$-casein band appeared from A, B, C, except D.

  • A reliable and well-characterized dosimetry system which is traceable to the international measurement system, is the key element to quality assurance in radiation processing with cobalt-60 gamma rays, X-rays, and electron beam. This is specifically the case for health-regulated processes, such as the radiation sterilization of single use medical devices and food irradiation for preservation and disinfestation. Polyethylene is considered to possess a lot of interesting dosimetric characteristics. In this work, a detailed study has been performed to determine the dosimetric characteristics of a commercialized high-density polyethylene (HDPE) film using Fourier transformed infrared spectrometry (FTIR). Correlations have been established between the absorbed dose and radiation induced infrared absorption in polyethylene having a maximum at 965 cm-1 (transvinylene band) and 1716 cm-1 (ketone-carbonyl band). We have found that polyethylene dose-response is linear with dose for both bands up to1000 kGy. For transvinylene band, the dose-response is more sensitive if irradiations are made in helium. While, for ketone-carbonyl band, the dose-response is more sensitive when irradiations are carried out in air. The dose-rate effect has been found to be negligible when polyethylene samples are irradiated with electron beam high dose rates. The irradiated polyethylene is relatively stable for several weeks after irradiation.

N400 Event-related Potential and Gamma Band Activities during Visual Perception of Korean/English Words (한글 및 영어 단어의 시각적 인지 시 N400 사건관련 뇌전위 및 감마대역 활성화)

  • The observations of difference and similarity in brain activities involved in processing different languages have fundamental importance in cognitive neuroscience. The purpose of this study was to investigate the difference and similarity in temporal brain activation patterns due to the language difference during visual perception of Korean and English words under priming. Especially, we tried to find the difference in evoked spectral power in gamma-band, which is known to reflect feature binding. The stimulation was visually presented as word pairs belonging to same or different categories so that N400 event-related potential(ERP) was evoked. Average ERP analysis and spectral analysis of gamma-band activity(GBA) were performed on 12 normal Korean subjects. Several ERP components such as P1, N1, N400, and P600 could be identified consistently, and the differences in N1, N400, and P600 were observed. From the spectral analysis, we found that the evoked GBA(eGBA) was significantly larger for English at ${\sim}100$ ms poststimulus. The latency of the eGBA was also considerably delayed for English. Overall, the results on the ERP components and eGBA analyses seem to be commensurate with subjects' familiarity of each language, and the difficulty of perceiving words of each language. The methods of this study can also be applied for clinical purposes considering that the language-related processing can be greatly altered for the patients with neurological or psychiatric diseases.

  • The interaction between pyronine G(PG) and surfactants has been examined by the absorption and fluorescence spectroscopy. The optimum condition for ${\gamma}$-band in PG-AOT system was found when S/D(concentration of surfactant / concentration of PG) was in the vicinity of 80. The disassociation of stacking conformation occured gradually at S/D>80. In PG-cationic and nonionic surfactant systems the absorptivity of monomeric PG increased. With the increment of the concentration of NaCl in PG-AOT system, the absorptivity of ${\gamma}$-band decreased at S/D<10 but, increased remarkably at S/D>20. PG-AOT system with the addition of NaCl showed greater dissociation than that without the addition of NaCl.

Trapping centers due to native defects in the $CdIn_2S_4$ films grown by hot wall epitaxy

  • $CdIn_2S_4$ (110) films were grown on semi-insulating GaAs (100) by a hot wall epitaxy method. Using photocurrent (PC) measurement, the PC spectra in the temperature range of 30 and 10 K appeared as three peaks in the short wavelength region. It was found that three peaks, A-, B-, and C-excitons, correspond to the intrinsic transition from the valence band states of ${\Gamma}_4(z),\;{\Gamma}_5(x),\;and\;{\Gamma}_5(y)$ to the exciton below the conduction band state of ${\Gamma}_1(s)$, respectively. The 0.122 eV crystal field splitting and the 0.017 eV spin orbit splitting were obtained. Thus, the temperature dependence of the optical band gap obtained from the PC measurement was well described by $E_g$(T)=2.7116eV - $(7.65{\times}10^{-4}\;eV/K)T^2$/(425+T). But, the behavior of the PC was different from that generally observed in other semiconductors. The PC intensities decreased with decreasing temperature. This phenomenon had ever been reported at a PC experiment on the bulk crystals grown by the Bridgman method. From the relation of log $J_{ph}$ vs 1/T, where $J_{ph}$ is the PC density, two dominant levels were observed, one at high temperatures and the other at low temperatures. Consequently, the trapping centers due to native defects in the $CdIn_2S_4$ film were suggested to be the causes of the decrease in the PC signal with decreasing temperature.

F-GAMMA with KVN

  • The F-GAMMA (FERMI-GST AGN Multi-frequency Monitoring Alliance) project is a program for the monthly monitoring of the broad-band spectra of currently about 90 selected Fermi-GST AGNs. F-GAMMA utilizes several facilities in cm, mm, sub-mm, infrared and optical bands, achieving an unprecedented coverage for the study of the spectral evolution of powerful relativistic jets in AGNs. The KVN joined the F-GAMMA project in May 2011, aiming to monitor flux density at 22 and 43 GHz. We present the preliminary results of flux density variability, evolution of spectral index, and modulation index.

  • Let ${\gamma}$(G) be the domination number of a graph G. It is shown that for any $k {\ge} 0$ there exists a Cartesian graph bundle $B{\Box}_{\varphi}F$ such that ${\gamma}(B{\Box}_{\varphi}F) ={\gamma}(B){\gamma}(F)-2k$. The domination numbers of Cartesian bundles of two cycles are determined exactly when the fibre graph is a triangle or a square. A statement similar to Vizing's conjecture on strong graph bundles is shown not to be true by proving the inequality ${\gamma}(B{\bigotimes}_{\varphi}F){\le}{\gamma}(B){\gamma}(F)$ for strong graph bundles. Examples of graphs Band F with ${\gamma}(B{\bigotimes}_{\varphi}F) < {\gamma}(B){\gamma}(F)$ are given.

  • Lipid peroxidation induces functional deterioration of cell membrane and induces cell death in extreme cases. These phenomena are known to be related generally to the change of physical properties of lipid membrane such as decreased lipid order or increased water penetration. Even though the electric property of lipid membrane is important, there has been no report about the change of electric properties after lipid peroxidation. Herein, we demonstrate the molecular energy band change in red blood cell membrane through peroxidation by air-based atmospheric pressure DBD plasma treatment. Ion-induced secondary electron emission coefficient (${\gamma}$ value) was measured by using home-made gamma-focused ion beam (${\gamma}$-FIB) system and electron energy band was calculated based on the quantum mechanical Auger neutralization theory. The oxidized lipids showed higher gamma values and lower electron work functions, which implies the change of surface charging or electrical conductance. This result suggests that modified electrical properties should play a role in cell signaling under oxidative stress.
