Effect of Germinated Brown Rice Concentrate on Free Amino Acid Levels and Antioxidant and Nitrite Scavenging Activity in Kimchi

  • In this study, we investigated the effect of adding a 1, 3, or 5% solution of germinated brown rice concentrate (GBRC) to fermented kimchi. During fermentation, the concentration of free amino acids and essential amino acids increased with increasing concentrations of GBRC. In particular, higher levels of free amino acids were associated with a sweet taste compared with controls. The ${\gamma}$-aminobutyric acid (GABA) content of kimchi containing the 5% GBRC solution was 3 times higher than that of controls. The total phenolic compound content (130 mg%) did not change significantly in the control group, but increased in 10 mg% increments as the GBRC concentration rose from 1 to 3 to 5%. 2,2-diphenyl-1-picryhydrazyl (DPPH) free radical scavenging activity and superoxide radical scavenging activity also increased with the GBRC concentration, with maximum activity during the ripe stage with GBRC measured at 79 to 82% compared with controls (30 to 71 %). The nitrite scavenging activity was 10% higher with GBRC compared with controls and was highest when the pH was 1.2. These results showed that the addition of GBRC is effective in improving the function of kimchi.

Preparation of High GABA-Enriched Yeast Extract by Non-Saccharomyces Yeasts Isolated from Korean Traditional Fermented Soybean Product

  • High GABA-enriched yeast extract, for various nutritionally and pharmaceutically important functional foods, was prepared using a novel isolate of Debaryomyces hansenii JBCC541. Under optimized conditions, GABA conversion rates are significantly enhanced up to 7.55 g/l by D. hansenii JBCC541, increasing their synthesis yield 40 times. The total amino acid content of the prepared yeast extract was 10733.86 mg/l (257.36 mg/g), consisting of alanine, lysine, glutamine, leucine, and valine as the primary amino acids. The GABA content was significantly enhanced up to 6790 mg/l (162.80 mg/g) in the presence of glutamic acid, with approximately 10-fold higher GABA production. Flavor amino acids were also highly enhanced, indicating that the prepared yeast extract might be useful for preparing various functional and sensuous foods. Our results were promising as a GABA-enriched yeast extract preparation tool ensuring a suitable food material level with the potential for functionally enhanced food industrial applications.

Sleep-Induction Effects of GABA Coated Rice from Fermentation of Mono Sodium Glutamate (Mono Sodium Glutamate 발효로 얻은 GABA 코팅 쌀에 의한 수면유도 효과)

  • Gamma-aminobutyric acid (GABA) is a non-protein amino acid widely present in organisms, which has shown several important physiological functions such as neurotransmission, hypotension induction, as well as diuretic and tranquilizer effects. They have also been extensively used in food industry. GABA contents in the grain and brown rice are about 1~4 mg/100 g and 4~8 mg/100 g, respectively but it is difficult to expect physiological activity from such low amounts of natural food intake. We investigate the effects of GABA-coated rice on the secretion of melatonin and serotonin which both have been used as sleep inductive compounds. As a result, the secretion of melatonin and serotonin from mice were found to be $3.578{\pm}0.158pg/mL$, $5.918{\pm}0.169ng/mL$ respectively. The melatonin and serotonin in mice increased significantly up to the 8.7 and 22.8 times respectively, when compared to that of the rice, but there was no cumulative effects. Due to the continuous intake of GABA-coated rice, which was developed as a functional food nutrient, inductive effects of melatonin and serotonin from general rice could be achieved and also the similar effect as taking up directly 25 mg/mL of GABA could be obtained.

Studies on the Amino Acid and Fatty Acid Compositions in the Seed and Pulpy Substance of Feral Peach (Prunus persica Batsch var. davidiana Max.) (야생 돌복숭아 씨와 과육의 아미노산 및 지방산 조성에 관한 연구)

  • Amino acid and fatty acid compositions of the physiological activity substance in the seed and pulpy substance of feral peach (Prunus persica Batsch var. davidiana Max.) were analyzed for the use as an biohealth functional processed products. The proximate compositions in the vacuum freeze dried seed and pulpy substance of feral peach were carbohydrate 63.92% and 75.11%, crude protein 27.85% and 12.77%, moisture 3.61% and 4.69%, crude fat 1.21% and 4.80%, crude ash 3.41% and 2.63%, respectively. Total amino acid contents in the protein of feral peach seed were 3,444.35 mg%, and the major amino acids were aspartic acid(681.10 mg%), glutamic acid(495.48 mg%), alanine(283.66 mg%), serine(251.36 mg%), proline(229.80 mg%), lysine(192.31 mg%) and leucine(191.34 mg%), respectively. Total amino acid contents in the protein of feral peach pulpy substance were 1,064.02 mg%, and the major amino acids followed aspartic acid(250.15 mg%), glutamic acid(129. 63 mg%), lysine, proline, leucine, alanine and serine, in a decreasing order. The richest total amino acid content contained in feral peach seed and pulpy substance was aspartic acid, followed by glutamic acid. The amount of free amino acids of feral peach seed were 6,215.34 ms%, and the major free amino acids were glutamic acid(827.25 mg%), threonine, valine and $\beta-aminobutyric$ acid, respectively. Free amino acid contents of pulpy substance were 683.82 mg%, and the major free amino acids were glutamic acid(339.49 mg%), serine proline, alanine and $\gamma-amino-n-butyric$ acid. Especially, in the case of glutamic acid, it was highest. The compositions of major total fatty acid in the lipid feral peach (Prunus persica Batsch var. davidiana Max.) seed and pulpy sabstance were linoleic acid($C_{18:2}$, n-6) and linolenic acid($C_{18:3}$, n-3), particularly.

  • Endosulfan, one of organochlorine insecticides, is $\gamma$-aminobutyric acid (GABA) antagonist. In sufficient dose, this pesticide lower the seizure threshold and produce CNS stimulation, with resultant seizures, respiratory failure, and death. In patients with endosulfan intoxication, the first manifestation of toxicity is largely a generalized seizure without prodromal signs or symptoms. So the management of airway and seizure control are essential for survival and prognosis of intoxicated patients. We report two cases of acute endosulfan poisoning who manifest 'status epilepticus' similarly, but have different prognosis.

  • Anatomical and physico-chemical properties of some nutrient and tonic crude drugs have been estimated and compared. Resin duct and druse crystal were observed more in Acanthopanax Cortex than in Ginseng Radix. Bast fiber bundle and solitary crystals were abandantly present in Glycyrrhizae Radix. Mucilage crude drugs contained mucilage ducts and bundles of raphides as common components and their arrangements of vascular bundles were different. TLC analysis showed different patterns of saponin, flavonoid, ${\gamma}-aminobutyric$ acid and sitosterols.

  • The present study was designed to evaluate central inhibitory effects of an essential oil from Acori graminei Rhizoma (AGR), the dry rhizomes of Acorus gramineus Solander (Araceae) upon fragrance inhalation (aroma therapy). Preinhalation of an essential oil of AGR markedly delayed the appearance of pentylenetetrazole-induced convulsion. Furthermore, the inhalation of an essential oil of AGR impressively inhibited the activity of ${\gamma}$-aminobutyric acid (GABA) transaminase, a degradating enzyme for GABA as inhalation period is lengthened.   • Intravenous midazolam has been frequently used for the relief of anxiety in dental treatment. This is likely the result of the sedative and anterograde amnestic properties of midazolam that are mediated through $\gamma$-Aminobutyric acid agonism. Unfortunately, Paradoxical reactions to midazolam include agitation, talkativeness, confusion, disinhibition, aggression,violent behavior, act of self-injury and need for restraints. These occur in less than 1% of all patients receiving midazolam, may occur at variable times after administration and are difficult to predict and diagnose. Two women with severe anxiety for dental treatment experienced paradoxical reactions associated with the use of intravenous midazolam. We are reviewed the management and prevention of paradoxical reactions and its different etiology.

  • Anxiety and anxiety disorders are related to many neurotransmitters, such as norepinephrine, serotonine, dopamine, glutamate, and Gamma-aminobutyric acid (GABA). GABA, the main inhibitory neurotransmitter of the CNS, is known to counterbalance the action of the excitatory neurotransmitters and control anxiety. GABA acts on 3 GABA receptor subtypes, $GABA_A$, $GABA_B$, and $GABA_C$. $GABA_A$ and $GABA_c$ receptors are oligomeric transmembrane glycoproteins composed of 5 subunits that are arranged around a central chloride channel. $GABA_B$ receptor comprises two 7-transmembraneis-spanning proteins that are coupled to either calcium or potassium channel via G proteins. This article highlights neurobiological interactions between anxiety and GABA system.
