Abamectin 1.8% EC and emamectin benzoate 2.15% were evaluated the simultaneous control effect of both insect pests on black pine (Pinus thunbergii) against Thecodiplosis japonensis and Matsucoccus thunbergianae. Abamectin 1.8% EC was highly effective against T. japonensis and M thunbergianae, however, emamectin benzoate was highly effective against only M. thunbergianae when abamectin and emamectin benzoate were applied through trunk injection against T. japonensis and M thunbergianae at the rate of 1 ml per cm in diameter of breast height. The rate of gall formation of T. japonensis was 0% at the applied year, but 85% at the following year. However, the rate of gall formation was over 90% at the treatment of emamectin benzoate showing no difference compared with control. Abamectin 1.8% EC and emamectin benzoate 2.15% showed high control efficacy representing 89.1% and 91.4% against M. thunbergiana, respectively.
Biological and structural mechanisms of the nematode disease development in chili pepper caused by the root-knot nematode, Meloidogyne incognita, were investigated. Out of 39 chili pepper cultivars/lines tested, six were found resistant, while 33 were susceptible to M. incognita, of which a susceptible cultivar Chilseongcho and three resistant cultivar/lines CM334, 02G132 and 03G53 with different resistance degrees were selected for microscopic studies on the disease development. Gall formation was greatly reduced in the resistant cultivars/lines. Nematode penetration occurred both in the susceptible and resistant chili pepper roots; however, the penetration rates were significantly lowered in the three resistant peppers compared to the susceptible pepper cv. Chilseongcho. In the susceptible pepper, giant cells were extensively formed with no discernible necrosis around the nematode feeding sites. In the highly resistant pepper cultivar CM334, no giant cell was formed, but extensive necrosis formation was observed around the penetrating nematodes. In the other two resistant pepper lines (02G132 and 03G53), both giant cells and prominent necroses were formed, and the necrotic responses appeared to inhibit the further development of giant cells or accelerate their early degeneration. Although the nematode penetration was retarded significantly in the resistant cultivar/lines, all of the above results suggest that the disease resistance of pepper may be related to post-infectional defense mechanisms (nematode growth and development) more than pre-infectional ones (penetration and establishment). Variations in structural modifications in the resistant cultivar/lines may reflect their genetic differences related to the nematode resistance.

I conclude that this recovering phenomena is closely related with the insect fauna of this area and this characteristic insect fauna was derived from the bush formation which reveals specific bush arrangement that can be illustrated as surrounded by another bush of Quercus acutissima Carruthers. 4. As a final conclusion, I recommend mixed bush formation of chestnut tree with Quercus species to control insect population dynamics in healthy status.
This study was performed to find differences in phototaxis response of the larva, adult emergence, sex ratio and gall formation rate of Thecodiplosis japonensis Uchida et Inouye between two areas, north and south. Larvae from these two areas, Hweng-sung in north and Moo-an in south, both considered to be the sities of severe outbreak of the insect recently, were collected from the floor and transported to the entomology larboratory, Won-Kwang University in Iri, then overwintered. These larvae were used for various experimental purposes on various dates. The following results were obtained. 1. The sizes of larvae reached 2.45 in length, 0.70 in width for north, and 2.50 in length, 0.72 in width for sourth. It seems that the differents were not significant. 2. It is known that Leaving gall of larvae to the ground for the hibernation usually occurs on rainy days. For the larvae dispersion, the phototaxis response of the larvae was hypothesized. To check this, the author manipulated some different illumination intensities adjusting the distance between the glass tube in which 100 larvae were contained and electric bulb. The glass tubes were blackened all but except a small portion. The phototaxis responses between two areas seemed to be different particularly at low light intensities, 45, 145 and 1000 lux, The larvae from north assembled to the clear portion of the glass tube were more than 30 out of 100, however less than 30 in south sample regardless of the time passage. In either cases, the saturation points came after about 8 to 10 hours. If temperature units were used, the phototaxis curve= after 1 hour illumination between two areas provided significant differences. 3. The adult emergence of south area was lagged 10 days later than that of north area. The accumulated effective day-degrees of temperature for adult emergence were $934^{\circ}C$ for north area and $1180^{\circ}C$ for south area. The emerging duration of north area appeared to be in late May through the early of July and the peak emergence occurred in middle June that was approximately 15days earlier than that of south area. The sex ratio of female to male, regardless of area, was approximately 3 : 1. 4. One hundred and nine Agrobacterium isolates were recovered from 68 samples(51 plant tumor and 17 soil) that were collected from different habitats in Northern Jordan. The isolated cultures were grouped into 3 biovars based on their biochemical characteristics and biovar I, II, and III comprised a total number of 46, 41, and 22 isolates, respectively. Isolates of biovar I were obtained primarily from the diseased peach, oak and rose plants, whereas isolates of biovar II and ill were obtained mostly from apple and grape plants, respectively. Twenty-nine isolates were found to be virulent to at least one of the tested hosts such as carrots, chickpeas, garden peas and tomato plants with a response of tumor formation or tumor with roots induction. Pathological interrelations of two soil-borne diseases in cucurbits (watermelon, oriental melon, shintosa and cucumber) caused by Fusarium isolates (FI) and the root-knot nematode (RKN), Meloidogyne incognita were characterized by the fusarium disease severity index (DI), RKN gall index (GI) and eggmass index (EI) in inoculation tests using FI and RKN. Virulence of FI as determined by DI at 4 weeks after inoculation was mostly in the higher order of Fusarium proliferatum F6, F5 and Fusarium oxysporum f. sp. melonis or Fusarium oxysporum f. sp. niveum with no significant differential interactions among the cucurbits and RKN co-infection. Significant increases of DI due to RKN coinfection were noticed in watermelon and oriental melon infected with F. proliferatum isolates, suggesting the DI increase due to RKN coinfection may depend upon the virulence of FI relative to aggressiveness of RKN on the cucurbits. For the coinfection of FI and RKN, GI and EI were mostly reduced logarithmically with the increase of DI, largely more in EI than GI, in all cucurbits except for shintosa. Microscopic examination of the root tissues showed histopathological features characteristic to infection types; formation of fungal hyphae and/or spores and plant defense structures (tyloses and mucilage) in variable degrees and formation of giant cells at variable developmental stages and with variable cytoplasmic depletion or degeneration which were visualized in relations with the values of DI, GI and EI. These findings will be helpful to develop control strategies of the soil-borne disease complex based on their pathological characteristics.
Monoterpenes, phenolic substances, growth substances and total nitrogen from normal needles and infested needles attacked by Thecodiplosis japonensis were compared for Pinus densiflora, Pinus thunbergii and Pinus rigida. The results obtained in this study were as follows: 1. Major monoterpene components in P. densiflora needles were ${\beta}$-phellandrene, ${\alpha}$-pinene and those in P. rigida needles were ${\beta}$-pinene and ${\alpha}$-pinene. In P. rigida resistant to this insect, infested needles showed higher ${\alpha}$-pinene, ${\beta}$-pinene and myrcene but lower camphene, limonene and ${\beta}$-phellandrene than normal needles. 2. Orcinol, catechol ferulic acid, salicylic acid and five unknowns were detected in P. rigida needles whereas orcinol, catechol, ferulic acid and four unknowns in P. densiflora needles. 3. Tryptophan, a precursor of IAA, was detected in larvae and also in gall tissues of both P. densiflora and P. thunbergii needles. This fact shows that growth substances may involve in gall formation by Thecodiplosis japonensis. 4. Total nitrogen contents per unit needle weight were neither significantly different between P. densiflora and P. rigida, nor between normal and infested needles.
Susceptible trees of Pinus densiflora and resistant trees of Pinus rigida following pine gall midge (Tnecodiplosis japonensis Uchida et Inouye) attack were seasonally compared to examine the variation in needle growth and photosynthetic ability, respiration rate, chlorophyll contents, carotenoid and anthocyanin contents. Also, carotenoid and anthocyanin contents of larvae both from soil and from galled tissue were compared during March and September, respectively. The plantation damaged severely by this insect consisted mostly of 10-to 15-year old P. rigida and P. densiflora. The results obtained in this study were as follows: 1) The length of the infested needles of P. densiflora decreased by 48.1 percent compared with the normal needles, while that of P. rigida did 37.4 percent. 2) All of P. densiflora and P. rigida showed higher photosynthetic ability in normal needles than in infested needles. The maximum photosynthetic ability of P. densiflora was shown in mid-August, while that of P. rigida in mid-October. In contrast to that, respiration rate of infested needles was higher than that of normal needles in both species. The respiration rate of P. rigida was higher than that of P. densiflora. 3) P. rigida had higher total chlorophyll contents than P. densiflora. The total carotenoid contents tents in infested needles were higher than those in normal needles of both species. 4) Total carotenoid contents were generally higher in P. rigida than in P. densiflora during the growing season. The total carotenoid content (0.094mg/g) in larvae from soil was similar to that (0.092mg/g) in larvae from galled tissues. 5) Infested needles of both species showed higher anthocyanin contents than normal needles. Higher anthocyanin contents in galled needles were due primarily to its active formation stimulated by larval attack. Thus, reddish-brown coloration occurred only in galled needles of P. densiflora.
These studies were carried out to select somatic hybrid using selectable marker genes of Nicotiana glauca transformed by NPTII gene and Solanum tuberosum transformed by T- DNA, and to study characteristics of transformant. The results are summarized as follows. 1. Crown gall tumors and hairy roots were formed on potato tuber disc infected by A. tumefaciens Ach5 and A. rhizogenes ATCC15834. These tumors and roots could be grown on the phytohormone free media. 2. Callus formation from hairy root was prompted on the medium containing 2, 4 D 2mg/I with casein hydrolysate lg/l. 3. The survival ratio of crown gall tumor callus derived from potato increased on the medium containing the activated charcoal 0. 5-2. 0mg/I because of the preventions on the other hand, hairy roots were necrosis on the same medium. 4. Callus derived from hairy root were excellently grown for a short time by suspension culture on liquid medium containing 2, 4-D 2mg/I and casein hydrolysate lg/l. 5. The binary vector pGA643 was mobilized from E. coli MC1000 into wild type Agrobacteriurn tumefaciens Ach5, A. tumefaciens $A_4T$ and disarmed A. tuniefaciens LBA4404 using a triparental mating method with E. ccli HB1O1/pRK2013. Transconjugants were obtained on the minimal media containing tetracycline and kanamycin. pGA643 vectors were confirmed by electrophoresis on 0.7% agarose gel. 6. Kanamycin resistant calli were selected on the media supplemented with 2, 4-D 0.5mg/1 and kanamycin $100\mug$/ml after co- cultivating with tobacco stem explants and A. tumefaciens LBA4404/pGA643, and selected calli propagated on the same medium. 7. The multiple shoots were regenerated from kanamycin resistant calli on the MS medium containing BA 2mg/l. 8. Leaf segments of transformed shoot were able to grow vigorusly on the medium supplemented with high concentration of kanamycin $1000\mug$/ml. 9. Kanamycin resistant shoots were rooting and elongated on medium containing kanamycin $100\mug$/ml, but normal shoot were not. 10. For the production of protoplast from potato calli transformed by T-DNA and mesophyll tissue transformed by NPTII gene, the former was isolated in the enzyme mixture of 2.0% celluase Onozuka R-10, 1.0% dricelase, 1.0% macerozyme. and 0.5M mannitol, the latter was isolated in the enzyme mixture 1.0% Celluase Onozuka R-10, 0.3% macerozyme, and 0.7M mannitol. 11. The optimal concentrationn of mannitol in the enzyme mixture for high protoplast yield was 0.8M at both transformed tobacco mesophyll and potato callus. The viabilities of protoplast were shown above 90%, respectively. 12. Both tobacco mesophyll and potato callus protoplasts were fused by using PEG solution. Cell walls were regenerated on hormone free media supplemented with kanamycin after 5 days, and colonies were observed after 4 weeks culture.
