  • The mechanism of skin-sensitizing photoreactions of coumarins and furocoumarins are studied by spectroscopic, triplet quenching, and fluorescence techniques. The excited singlet mechanism is suggested for xanthotoxin-thymine/or DNA photoreactions from the results of triplet quenching studies utilizing ${\beta}$-carotene as a quencher.

  • Four furocoumarins and two dihydrofuranocoumarin glycosides have been isolated from the benzene-soluble and butanol-soluble portions of the roots of Ostericum koreanum Kitagawa, respectively. Four furocoumarins have been determined as imperatorin, isoimperatorin, oxypeucedanin and oxypeucedanin hydrate, and two dihydrofuranocoumarin glycosides determined as marmesinin and $4'-O-{\beta}-D-glucopyranosyl-3'-hydroxymarmesin$ on the basis of spectral data and physico-chemical evidence.

  • The root of Heracleum moellendorffii was extracted with methanol and extract was fractionated with n-hexane, $CHCI_3$ and n-BuOH. Repaeated column chromatography of silica gel, sephadex LH 20 and ODS led to the isolation of ten compounds from n-hexane fraction and n-BuOH fraction. On the basis of spectroscopic evidences, the structures of isolated compounds were identified as isobergapten, psoralen, angelicin, sphondin, xanthotoxin, skimmin, cichoriin, $heratomol-6-O-{\beta}-D-glucopyranoside$, scopolin and apterin.

  • Silica gel column chromatography on the ether extract from the fruits of Angelica tenuissima $N_{AKAI}$ gave three kinds of crystalline constituents. The following derivatives of furocoumarins and steroid were identified by UV, IR, NMR spectra, elemental analysis and physico-chemical tests: iso-imperatorin m.p. $108{\sim}109^{\circ}\;C_{16}H_{14}O_4$, prangolarine m.p. $103{\sim}104^{\circ}\;C_{16}H_{14}O_5$, ${\beta}-sitosterol$ m.p. $138{\sim}139^{\circ}\;C_{29}H_{50}O$.

  • Silica gel column chromatography of the ether extract of the root of Angelica Koreana $M_{AXIMOWICZ}$ (Umbelliferae) gave five kinds of crystalline products of coumarin. The following kinds of furocoumarins were identified by UV, IR, NMR spectra and physico-chemical tests. iso-imperatorin m.p. 108-$109^{\circ}$ $C_{16}H_{14}O_{4}$ oxypeucedanin m.p. 142-$143^{\circ}$ $C_{16}H_{14}O_{5}$ prangolarine m.p. 104-$105^{\circ}$ $C_{16}H_{14}O_{5}$ imperatorin m.p. 100-$101^{\circ}$ $C_{16}H_{14}O_{4}$ These analyses also showed that white needles m.p. 129-$130^{\circ}$ were likely to be a furocoumarin. Besides, the methanol extract of the root was found to contain sucrose.

  • A number of substituted furodiketochroman derivatives ($lll_{a-f}$) have been synthesized by fusion of aromatic aldehydes with 5-hydroxybergapten and 5-hydroxyisopimpinellin. On the other hand, when the reaction was carried out in a solvent, the corresponding bis-furoccumarin derivatives ($lV_{c-n}$) were obtained. The anticoagulant effect of compounds $lll_{a,b,d}$ and $lv_{b,c,f,g,i,k}$ was tested. They failed to demonstrate any significant effect. The effect of the tested compounds on the arterial blood pressure was studied. Compounds $lV_c$, $lll_d$, $lll_b$, $lV_b$, $lV_k$ and $lV_i$ showed lowering effects on the normal systolic blooc pressure of anaesthetized rats in a decreasing manner.

  • In connection with our studies on the isolation of hepatoprotective constituents from natural products. we have recently reported hepatoprotective compounds including phenolic bakuchiol. diarylheptanoids. furocoumarins. In the course of continuing efforts. the aqueous extract of the roots of Agrimonia pilosa Ledeb. (Rosaceae) was found to exhibit promising hepatoprotective activity. A. pilosa is a perennial herb distributed throughout South Korea. and its roots have been used as the hemostatic. antimalarial. and antidysenteric agent in oriental medicine. Chemical investigation of the aqueous extract of the roots of this plant. as guided by hepatoprotective active catechin (2). Compound 1 showed hepatoprotective effects on both tacrine-induced cytotoxicity in human level derived Hep G2 cells and tert-hydroperoxide-induced cytotoxicity in rat primary hepatocyles with $EC_{50}$ values of 66.2 $\pm$ 2.8 and 22.9 $\pm$ 2.6 $\mu\textrm{M}$ respectively.

  • INTRODUCTION : Psoralens are planar tricyclic furocoumarins present in numerous plants and fungi found throughout the world.' Naturally occurring and synthesized psoralen derivatives(see Figure 1) are photosensitizers of UVA especially from 320 nm to 400 nm, a range at which cellular nucleic acids and proteins are weakly absorbing if any at all. Because of their skinphotosensitizing properties, these compounds have been used in the photochemotherapy of psoriasis and vitiligo. However, undesirable side effects such as carcinoma development in hairless mice as well as possible liver damage from the use of 8-methoxypsoralen(8-MOP) have been reported. The other photobiological effects include inactivation of DNA viruses, killing and mutagenesis of bacteria, inhibition of tumor transmitting capacity of various cells, and hyperpigmentation on human and guinea pig skin. PUVA(psoralen+UVA) photochemotherapy is in fact thousands of years old, having been used in Egypt and India since B.C. 1200-2000. Photochemotherapy for a common disfiguring disease, vitiligo, was practiced in the ancient world by physicians and herbalists who used boiled extracts of the fruits of certain umbelliferous plants, e.g. Ammi majus Linnaeus in Egypt or the leguminous plants, Psoralea corylifolia L. in India. It was first described by Kuske in 19388 that photosensitization of skin by plants was related to the presence of psoralen. He identified natural psoralens in plants as photosensitizers and isolated bergapten(5methoxypsoralen) from the oil of bergamot. The scientific interest in photosensitizing psoralens, however, has grown considerably after the introduction into clinics of the psoralen photochemotherapy for the treatment of psoriasis and of other skin (abbreviation)
