In 2004 and 2005, Phytophthora rot on Luffa cylindrica which had not been reported in Korea occurred in the experimental field at Gyeongsangnam-do Agricultural Research and Extension Services. The disease initiated on leaves and fruits of the plant with small watersoaked dark brown spots and progressed rapidly. The causal pathogen isolated from diseased tissues was identified as a Phytophthora sp. because of aseptate mycelia and zoospores released directly from sporangia. The fungus grew well on PDA and 10% V-8 juice agar showing an arachnoid or rosaceous colony pattern. Sporangia formed abundantly in water and were conspicuously papillate, noncaducous, ovoid to globose, and sized $26\sim62\times19\sim38{\mu}m$. The fungus was heterothallic as producing sexual reproduction structures only when mated with only A2 standard mating type strain. Oogonia and oospores were spherical, smooth walled, and measured as $20\sim28{\mu}m\;and\;16\sim24{\mu}m$, respectively. Oospores were aplerotic and antheridia were amphigynous, unicellula and spherical. Chlamydospores were globose and $20\sim38{\mu}m$ in diameter. Optimum temperature for growth was around $28\sim30^{\circ}C$. The fungus caused similar symptoms on artificially inoculated plant and could be re-isolated thereby proving Koch's postulation. Based on the mycological criteria investigated in this study, the causal fungus of Luffa sylindrica rot was identified as Phytophthora nicotianae. This is the first report of Phytophthora rot of Luffa cylindrica caused by P. nicotianae in Korea.
A $Mariannaea$ fungus was isolated during investigation of an elm tree infested with unidentified beetles. Based on morphological characteristics and molecular analysis of the internal transcribed spacer rDNA sequence, the fungus was identified as $Mariannaea$$elegans$ var. $elegans$. Fungal growth was better on malt extract agar than on potato dextrose agar and oatmeal agar. Optimal temperature and pH for growth of the fungus were $30^{\circ}C$ and pH 7.0, respectively. The fungus was found to have the ability to produce extracellular enzymes such as amylase, ${\beta}$-glucosidase, cellulase, and protease. This is first report on $M.$$elegans$ var. $elegans$ in Korea.
A black scab was observed on strawberry(Fragaria ananassa) in plastic film houses around Jinju area during the winter of 2000. The disease started from leaves then moved to calyx and runner. At the beginning, the infected area started with small dark brown spots then gradually expanded. The pathogenic fungus was purely isolated from the diseased leaves, calyx and runner. The fungus was inoculated to test Koch's postulates and proved to be the causal agent of the disease. The isolated fungus grew readily on potato dextrose agar, forming dark green to dark gray colonies. The optimum temperature for mycelial growth was about $25^{\circ}C$. The diameter of growing hyphae was $3.8{\sim}5.6{\mu}m$. Conidia were ellipsoidal, ovoid or subspherical, mostly one-celled but occasionally septate. The size of conidia were $4.1{\sim}11.7{\times}3.8{\sim}5.3$ 1-cell, $9.3{\sim}18.8{\times}4.0{\sim}7.4{\mu}m$ 2-cell and formed in long branched chains on the erected conidiophores which were dark brown and variable in length between $28.8{\sim}236.2{\times}3.0{\sim}6.2{\mu}m$ in size. The fungus was identified as Cladosporium herbarum on the basis of its morphological characteristics. The black scab disease of strawberry caused by C. herbarum has not been reported in Korea previously.
For the selection and breeding of chestnut varieties resistant to the chestnut blight fungus Cryphonectria parasitica, disease resistance and susceptibility of 28 varieties widely planted and growing in Korea were evaluated by artificial inoculation of a pathogenic fungus. For this experiment, a typical virulent strain (KCPC-19) was selected. Artificial inoculation was conducted into all varieties by using two different materials and methods, i.e., bark and wood tissue sections in the laboratory and living trees in the field. In the bark and wood tissue section method, the size of necrotic area and canker development on chestnut varieties were examined and compared 4 days after inoculation. There were wide variations of chestnut varieties in disease resistance and susceptibility against chestnut blight fungus, but 3 varieties, Daebo!, Ishizuchi, and Sandae, were shown to be relatively resistant to the disease with the necrotic area of 0.95-1.03 cm2, while Arima was the most susceptible with the size of 2.0 cm2. In the living tree inoculation examined 5 weeks after inoculation, 3 varieties, Daebo, Ishizuchi, and Riheiguri, showed the higher resistance, but Tono 2 did the highest susceptibility among tested varieties.
A destructive stem rot of strawberry (cv. Akihime) occurred sporadically in farmers' fields around Daegok-Myeon, Jinju City, Gyeongnam province in Korea. The infected plants showed stem and crown rot, sometimes whole plant blighted. White mycelia spread over stems of infected clones and sclerotia formed on the old lesions near to soil surface. The fungus formed white colony on PDA and showed maximum mycelial growth and scleotial formation around 30$^{\circ}C$. The fungus usually have many narrow mycelial strands in the aerial mycelium and the width were 4.0∼10.0$\mu\textrm{m}$. The typical clamp connections were formed on the mycelium. The shape of sclerotia was globoid and 1.0∼2.8 mm in size. The fungus was isolated repeatedly from the infected tissues and identified as Sclerotium rolfsii. The fungus was inoculated to strawberry and confirmed its pathogenecity This is the first report on the stem rot of strawberry caused by Scierotium rolfsii in Korea.
본 실험은 질소 및 탄소원이 Cercospora kikuchii 균사생장에 미치는 영향과 여러 배양조건에서 생장한 C. kikuchii가 세균생장을 저지하는 정도를 알아보았다. C. kikuchii 균사생장에 적합한 질소원은 yeastextract였고, 탄소원으로는 sucrose, glucose, maltose, fructose를 첨가하였을 때 균사생장이 좋았다. C. kikuchii의 세균생장저지 실험에서 wild type은 pH 조건은 5.0에서, 질소원으로는 yeast-extract 또는 peptone.을 첨가하였을 때, 탄소원으로는 sucrose, maltose, glucose, fructose를 첨가한 배지에서 생장하였을 때 Erwinia carotovora와 Pseudomonas solanacearum의 생장을 크게 저지시켰다. 그러나 albino 돌연변이균주는 실허된 세균의 생장을 거의 저지시키지 못하였다.
A scab disease occurred on watermelon seedlings by the infection of Cladosporium cucumberinum. This is the first report demonstrating the scab disease on watermelon in Korea. The casual agent attacked all plant parts of the seedlings on the ground. Infection sites were initiated with sunken and dark green spots, and then suddenly developed to large lesions softened with gummy substances. When hypocotyl and leaf stalk of seedlings were infected and softened, upper parts of seedlings were brokin down, dried and eventually died. Conidiophores of the fungus were characterized by pale olivaceous brown color, and variable length of about 3-5 mm in width. Conidia were formed I long branched chains, and conidium was ellipsoidal, fusiform or subspherical single cell mostly without septum. Morphological characteristics of the fungus were almost identical to Cladosporium cucumerinum Ellis & Authur. The fungus was also pathogenic to cucumber, squash and oriental melon, suggesting that it is a common pathogen to cucurbits. However, the fungus was not pathogenic to bottle gourd.
The ascomycete fungus Fusarium graminearum is an important plant pathogen responsible for Fusarium head blight in small grains and ear rot on maize. This fungus also produces the estrogenic metabolite, zearalenone (ZEA) that causes estrogenic disorders in humans and animals. Previously, we developed a conditional gene expression system for this fungus using a ZEA-inducible promoter (Pzear). In the present study, four other estrogenic compounds, including ${\beta}$-estradiol, estriol, estrone, and secoisolariciresinol, were screened as possible substitutes for ZEA in this system. Among them, ${\beta}$-estradiol was able to successfully induce the expression of a gene controlled by Pzear, while estrone was only able to partially induce its expression but the other two compounds were not effective. In combination, these results demonstrate that ${\beta}$-estradiol can replace ZEA in this conditional gene expression system, thereby eliminating the need to use the more expensive reagent, ZEA, and facilitating high-throughput functional analyses of F. graminearum in future studies.

The genomic structure of the Cu/Zn superoxide dismutase (SOD1) gene from the entomopathogenic fungus, Cordyceps pruinosa was characterized. The SOD1 gene of C. pruinosa spans 947 nucleotides and consisted of four exons encoding for 154 amino acids and three introns. Four exons of the SOD1 gene are composed of 13, 331, 97 and 20 nucleotides respectively. Homology search of amino acid sequences of the SOD1 gene of C. pruinosa with another 13 fungi species showed higher sequence similarity of 69% ~ 95% and had the most highest sequence identity of 95% with Beauveria bassiana and Cordyceps militaris, which can easely infect domesticated Bombyx mori and another wild lepidopteran species in artificial or natual manner of infection. This SOD1 gene sequence showed copper, zinc and beta-barrel fold sites. Homology search showed that the Cu/Zn SOD1 gene from the entomopathogenic fungus, C. pruinosa is an orthologous gene homolog present in different species of organism whose ancestor predates the split between the relating species. In addition, C. pruinosa SOD1 gene is placed together within the ascomycetes group of fungal clade. From these results it is concluded that C. pruinosa SOD1 gene is orthologous gene having the same or very similar functions with a common evolutionary ancestor.
This study was undertaken to isolate and to identify a fungal pathogen Colletotrichum sp. KV-21 associated with apple anthracnose. Rotted Gala apples were used for the isolation of the fungus. The infected tissues were sterilized with 70% ethanol, washed with sterilized distilled water and were transferred to 50 ml containing potato broth (PDB) flasks. The peripheral hyphae of the fungal colony which developed from the infected tissues were isolated on to potato dextrose agar (PDA). On PDA plates the fungus grew well at $25^{\circ}C$ and occupied more than half of a 9 cm petri dish within 5 days. The fungal cultures on PDA were used for morphological observation and identification of the fungus. Conidiophores were produced on the gray to whitish sporodochial structures scattered on PDA plates which gave rise to conidiogenous cells. The structures of the conidia produced on PDA plates were subcylindrical to obovoid, fusoid, tapered and 4 to $6\;{\mu}m$ in size.
