The aphid-attacking fungus Pandora neoaphidis was collected during May and June, 1998 at various localities: two localities at Kyunggi, three at Kangwon, one at Kyungbuk and one at Chungnam Provinces. Occurrence of this fungus has never been recorded in Korea. The potential host aphids are Acyrthosiphon kondoi, Myzus persicae, Lipaphis erysimi, Sitobion avenae, Aphis rumicis, Dactynotus species and unknown species. Microscopic characters of the fungus are described and illustrated. Pandora neoaphidis studied in this study has an extremely broad range in conidial size and shape even within a single specimen. Such variable size and shape are typical characters of the fungus. During microscopic examination of specimens, the resting-spore phase was not observed.
Fungi produce various secondary metabolites that have beneficial and harmful effects on other organisms. Those bioactive metabolites have been explored as potential medicinal and antimicrobial resources. However, the activities of the culture filtrate (CF) and metabolites of whiterot fungus (Schizophyllum commune) have been underexplored. In this study, we assayed the antimicrobial activities of CF obtained from white-rot fungus against various plant pathogens and evaluated its efficacy for controlling anthracnose and gray mold in pepper plants. The CF inhibited the mycelial growth of various fungal plant pathogens, but not of bacterial pathogens. Diluted concentrations of CF significantly suppressed the severity of anthracnose and gray mold in pepper fruits. Furthermore, the incidence of anthracnose in field conditions was reduced by treatment with a 12.5% dilution of CF. The active compound responsible for the antifungal and disease control activity was identified and verified as schizostatin. Our results indicate that the CF of white-rot fungus can be used as an eco-friendly natural product against fungal plant pathogens. Moreover, the compound, schizostatin could be used as a biochemical resource or precursor for development as a pesticide. To the best of our knowledge, this is the first report on the control of plant diseases using CF and active compound from white-rot fungus. We discussed the controversial antagonistic activity of schizostatin and believe that the CF of white-rot fungus or its active compound, schizostatin, could be used as a biochemical pesticide against fungal diseases such as anthracnose and gray mold in many vegetables.
The fungus gnat, Bradysia difformis, damages various crops in greenhouses and is recognized as an important pest around the world. Additionally, in the future, many other greenhouse crops will be added to the list of crops damaged by the fungus gnat. In this study, to find effective control methods for the fungus gnat, the insecticidal effect of 20 chemical synthetic insecticides was tested with the potato disc and pot treatment methods; additionally, the control effect of 16 strains of B. thuringiensis was examined with the potato disk method. The fungus gnat larvae were treated for 2 days with each of the synthetic insecticides to determine insecticidal effect using the potato disc method. The results were as follows. Among the highly insecticidal active pesticides, chlorfenapyr exhibited a 100% insecticidal activity, and fenazaquin, acetamiprid, dinotefuran, fenthion and thiamethoxam exhibited more than a 90% insecticidal activity. For the pot treatment method, chlorfenapyr exhibited a 3.3% insecticidal effect, and thiamethoxam, acetamiprid, dinotefuran, fenthion, etc. exhibited an insecticidal effect of less than 10% of the emergence rate to adult fungus gnat after 14 days of treatment. To select the B. thuringiensis strains that have an insecticidal effect on the fungus gnat, 16 strains were biologically assayed using the potato disc method. Among the 16 strains, Bt-3, Bt-8 and Bt-13 had more than a 70% insecticidal effect. The $LC_{50}$ and $LC_{95}$ values of Bt-3, Bt-8 and Bt-13 were $3.7{\times}10^5$ and $4.7{\times}10^8cfu/ml$, $1.4{\times}10^5$ and $1.1{\times}10^7cfu/ml$, and $1.4{\times}10^5$ and $1.3{\times}10^7cfu/ml$, respectively.
The fermented black tea with tea fungus (FBTF) known to cure various diseases was prepared by culturing tea fungus biomass in black tae with different kinds (sucrose, glucose, fructose and corn syrup) and concentrations (10-60% m/v) of saccharides. pH changes and total acid production of FBTF were investigated during 14 dats incubation at $5-45^{\circ}C$ The patterns of acid production in sucrose and fructose black tea solution were similar each other, and those in glucose and corn syrup black tea solution were similar each other showing that tea fungus biomass utilizes fructose than glucose more efficiently. The optimum incubation temperature for the formation and growth of tea fungus Biomass, and the acid production was $30^{\circ}C$ Low contents of total aids (0.1%-0.2%) were produced in 20% of higher concentrations of sucrose and fructose black tea solution, while 1.8% and 0.68% were produced in the 10% of sucrose and fructose black tea solutions, respectively, pH of FBTF dropped to 2.74-3.56 after 2 days of incubation of all the culture solution and Kept this level to 14 days incubation.
One hundred and thirteen healed pulmonary tuberculosis patients and 11 patients with other underlying diseases were studied for evidence of pulmonary fungal infection because of persisting hemoptysis or chronic cough. Rediological, mycological and serological investigations revealed that 54 out of 124 patients were evidently infected with one or more species of fungi. A. fumigatus was isolated from 4 out of 70 patients whose sera did not react with antigens from this fungus, while it was isolated from 43 out of 47 serological reactors to this fungus. Chest radiography showed a distinct fungus ball in a cyst of one patient and in a preformed cavity in the lung of 17 healed tuberculosis patients and two other patients. The latter two patients were infected with A.flavus. Two patients, who were under the long period of immunosuppressive therapy, apparently succumbed to invasive aspergillosia due to A.fumigatus. A single or dual infection with A. flavus, A. nidulans, A.nidulans var. latus, C. albicans, and P. boydii were noticed in some patients without mycetomal shadow on chest radiographs. Young mycelial extract (ME) of A.fumigatus detected antibody in 95.8 percent of the sera from patients infected with this fungus, while it was isolated from 43 out of 47 serological reactors to this fungus. Culture filtrate antigen, however, was comparable with ME. There was no single antigen with which all the serum specimens reacted. Fractionation of ME resulted in a loss of some activity although it excluded substances that reacted with C-reactive protein in a loss of some activity although it excluded substances that reacted with C-reactive protein. Most reactive and specific precipitinogens distributed in the fraction (FB) which was precipitable at 75 percent saturation with ammonium sulfate and eluted in a second peak in order from gel-filtration and which contained mostly proteinic components. Glycoproteins or polysaccharides rich fractions (FA and ASI) were relatively less effective in detecting antibody. Demonstration of antibody in the serum from patients using a battery of fungal antigens and of etiologically related fungi from clinical specimens are very useful laboratory procedures for the diagnosis of pulmonary fungal infection which is a common complication of tuberculosis.
Agricultural sticky traps are used for forecasting flying insects, including fungus gnat adults. Forecasting using sticky trap is an important measure to determine pesticides or environmentally friendly control means. Sticky traps of various colors can be installed easily and quickly. The yellow sticky trap is most effective for forecasting fungus gnat adults. In addition, they are used for control purposes by mass trapping. We tested the optimum installation height of yellow sticky traps to forecast and control the fungus gnat adults effectively in the cultivation of mushroom and horticultural plants. The number of captured fungus gnat adults was highest on the second floor for button mushroom and oyster mushroom, 100 cm above the floor for sawdust shiitake mushroom, 20 cm under the floor for hydroponic strawberry, and 20 cm above the floor for hydroponic tomato. This suggests that sticky trap height should be installed differently depending on the cultivation type or environment for effective forecasting and control of fungus gnat adults.
The present study was undertaken to investigate the effects of different nitrogen source and rate on the growth of Fusarium oxysporum f. vasinfectum which is known to be a noticeable fungus causing the wilt disease of both sesame and cotton in Korea. From the results of this study, It was known that different N source and rate markedly affect the growth of Fusarium oxyspsrum f. vasinfectum Among four N sourses were used in this study, nitrate-N and urea-N were appropriate N source for the growth of fungus. Above all, nitrate N was the best N source because it is utilized in more extensive range of concentration in comparison with the other N source by the fungus, On the other hand, ammonia-N is of little avail for the growth of the fungus because of the formation of unusual colonies with wavy margin and bead-like mycelial cells in addition to marked reduction of mycelial growth and B sporulation of the fungus irrespective of concentration. Judging from the formation of such an abnormal colony and bead-like mycelial cell which is known to be a characteristic of 'staling-type' growth of fungi, the effect of ammonia-N on the growth of Fusarium oxysporum f. vasinfectum is similar to that of phenoxy componnds on some other fungi previously investigated by some workers. Ammonium and nitrate also was not considered to be an appropriate source for the growth of the fungus because of the formation of colonies with slight wavy margin and appreciable reduction of mycelial growth and sporulation in higher concentration than 50meq. , although much or less masking of the irregularity of colony occurs. Therefore, ammonia N alone or any other N combined with ammonia N is of little avail for the growth of Fusarium oxysporum f. vasinfectum.
The experiments were conducted to study the distribution of wilt of strawberry caused by Fusarium in Korea, the characters of the causal fnngus and its control. The results obtained are summarized as follows. 1. Wilt of strawberry has been found in Gimhae and Samrangjin, Gyeongnam province a few years ago. This disease has been spreading year after year, and observed on farms in most of the strawberry-growing areas in Korea. 2. The fungus was isolated frequently from the crowns and petioles of diseased straw berry plants, and the fungus belonging to Fusariun oxysporum in terms of the morphological characteristics of macroconidia, microconidia, chlamydospore and conidiophore on V-8 Agar. 3. The macroconidia formation of the fungus varied remarkably with the Isolates and kinds of medium tested. However. all isolates abundantly produced macroconidia on V-8 Agar. 4. The cross-inoculation tests with several forma specialis of F. oxysporum to cucumber, tomato, watermelon, luffa, cabbage, melon and strawberry were carried out. The isolates from strawberry viz. Kodama's F. oxysporum f. sp. fragariae and S-1 of the authors were pathogenic to only strawberry. The fungus was also similar in morphology and symptoms to Kodama's and Winks' isolate of F. oxysporum f. sp. fragariae. Therefore, the fungus is identified as Fusarium oxysporum Schl. f. sp. fragariae Winks & Williams. 5. The most effective fungicides were Benomyl and Homai for inhibiting sporulation and mycelial growth of the fungus. 6 The cultivar Kurumae 35, Himiko, Senga gigana and Daehak I were resistant, whereas Hokowase, Instiate Z4, Juspa, Puget beauty and Marshall were susceptible to the fungus with artificial inoculation.
In order to investigate an etiological characteristics of the pepper anthracnose fungus infected with Collectotrichum dematium, this experiment was undertaken. Isolation percent of the fungus was 55.3% from the seed sample of Jewon and 0.3% from that of Eumsung Kun, according to seed health blotter method. Although the acervulus was similar to the known two Colletotricum species, C. acutatum and C. gloeosporioides, setae was not only straightly extended above the acervulus, but also shaped as falcate conidia ranging $12{\sim}31.2{\times}2.4{\sim}4.8{\mu}m$. Acervului of the fungus were evenly scattered on the surface of pepper seed coat, and caused seedling blight after seed germination. Pre-and post-emergence seedling blight as well as foliar lesion and fruit rot was confirmed by inoculating conidial suspension. No typical anthracnose sysmptom was obtained from soybean seedlings, welsch onion and spinach seedlings upon inoculation. Therefore, this fungus is said to the undescribed form species of the fungus of pepper in Korea (Colletotricum dematium f. sp. capsicum).
Since 1990, destructive yellow rots on Ganoderma lucidum caused by a soilborne fungus have been occurred in major cultivation areas of Korea. Incidences of the disease were 61% in Chulwon and 94% in Kanghwa area where the mushroom has been cultivated for 10 years, whereas the disease has not been found yet in new cultivation areas such as Moonkyung and Hongsung. when severely infected, inner tissues of bed-logs showed severe yellow and fruiting bodies of the mushroom was not produced. Infected tissues of bed-logs were readily distinguished from those of healthy ones by a distinctive brown border line. When the disease progressed, mycelia of Ganoderma lucidum were totally destroyed, and abundant ascocarps of the pathogen were formed inside the tissues of bed-logs showing yellowish green. The fungus derived from a single ascospore strongly lysed mycelia of Ganoderma lucidum growing on bottle media, and non-volatile components secreted by the pathogen were also highly inhibitory to mycelial growth of the mushroom fungus. The pathogen was identified as Arthrographis cuboidea based on its distinctive cultural and morphological characters. The fungus produced arthroconidia and unbrached conidiophores. The width of fungal conidia was distinctively wide as compared with the length. Colonies of the fungi were pale yellow to yellowish green on agar media. As a causal pathogen of yellow rot of Ganoderma lucidum., this fungus has not been reported yet in Korea.
