Syntheses of Pyrrolo[2,1-b]thiazole Carboxanilides and Their Selective Antifungal Activities against Rice Blast and Wheat Leaf Rust (Pyrrolo[2,1-b]thiazole 카르복스아닐라이드 유도체의 합성 및 그들의 벼 도열병균과 밀 붉은녹병균에 대한 선택적인 항균활성)

  • For development of new agrochemical fungicide, syntheses of pyrrolo[2,1-b]thiazole carboxanilide derivatives 9 and antifungal screening against 6 kinds of plant pathogens were carried out. Functionalization of carboxylic acid on C-2 into carboxanilide in the pyrrolo[2,1-b]thiazole resulted in new 20 candidates, in which are ${\alpha},{\beta}$-unsaturated carboxanilide and methyl groups that are in cis relationship. Treatment of acetoxy-1,4-thiazin with magnesium in refluxing methanol gave pyrrolo[2,1-b]thiazole ethyl ester 4 in higher yield than that of the previous report. Hydrolysis of this compound afforded the corresponding acid 5, which reacted with aniline derivatives in the presence of coupling reagent, DIC to give pyrrolo[2,1-b]thiazole carboxanilides 9. As the result of in vivo antifungal assay of 9 against rice blast, rice sheath blight, cucumber gray mold, tomato late blight, wheat leaf rust, and barley powdery mildew, some compounds showed selectively antifungal activities against the rice blast and wheat leaf rust.

Synthesis of Trifluoromethylated Dihydro-1,4-oxathiin Carboxanilides and Their Fungicidal Activity (삼불화메틸기가 포함된 디히드로-1,4-옥사티인 카르복스아닐리드 유도체의 합성과 살균 활성)

  • ${\alpha},{\beta}$-Unsaturated carboxanilides 5 with trifluromethylated dihydro-1,4-oxathiins were synthesized for the development of new agrochemical fungicide. Chlorination of trifluoromethylated ${\beta}-keto$ ester 6 followed by the reaction with 1,2-mercaptoethanol gave intermediate 1,4-oxathiane 11. Without purification of 11, substitution of hydroxy group by chlorine, followed by dehydrochlorination of 10 in the presence of triethylamine afforded trifluoromethylated dihydro-1,4-oxathiin ethyl ester 9. Acylation of the hydroxy group of the carboxylic acid 12 followed by treatment of various amines gave the corresponding trifluoromethylated dihydro-1,4-oxathiin carboxamides 5. Antifungal screening (in vivo) of the synthesized compounds against typical plant diseases, which include rice blast, rice sheath blight, cucumber gray mold, tomato late blight, wheat leaf rust, and barley powdery mildew, was carried out. Where meta position of the phenyl group was substituted with isopropoxy or isopropyl group, excellent antifungal activities against rice sheath blight and wheat leaf rust were detected.

  • [ ${\alpha},{\beta}$ ]-Unsaturated carboxamides 12 with trifluromethylated dihydro-1,4-dithiins were synthesized for the purpose of development of new agrochemical fungicide. Chlorination of trifluoromethylated ${\beta}$-ketoester 4 followed by tile reaction with 1,2-ethanedithiol gave intermediate 1,4-dithiane 9. Without purification of 9 substitution of hydroxy by chlorine followed by dehydrochlorination in the presence of triethylamine afforded trifluoromethylated dihydro-1,4-dithiin ethyl ester 7. Activation of the hydroxy of the carboxylic acid 10 obtained from the hydrolysis of 7 and then reacted with various amines gave the corresponding trifluoromethylated dihydro-1,4-dithiin carboxamides. Antifungal screening (in vivo) against typical plant diseases, Rice Blast, Rice Sheath Blight, Cucumber Gray Mold, Tomato Late Blight, Wheat Leaf Rust, and Barley Powdery Mildew of the synthesized compounds was carried out. As a result, most of the compounds shlowed weak antifungal activities and some compounds in which isopropyl group was substituted in meta of the phenyl showed antifungal activity (99%) at 250 ppm against the disease Wheat Leaf Rust.

Selection of Effective Fungicides Against Xylogone sphaerospora, a Fungal Pathogen of Cultivated Mushroom, Ganoderma lucidum (영지 노랑병 방제에 효과적인 살균제의 선발)

  • A fungal disease of the cultivated mushroom, Ganoderma lucidum, caused by Xylogone sphaerospora was epidemic throughout all cultivation areas in Korea which caused a lot of yield losses in the mushroom production. For controlling the disease, the screening of effective fungicides against the pathogenic fungus were conducted. Thirty seven commercially available fungicides were tested for their inhibitory activities on potato dextrose agar media supplemented with these fungicides at various concentrations. Twenty one fungicides significantly inhibited mycelial growth of the pathogen, Xylogone sphaerospora, but 16 fungicides had no inhibitory effect. Among these 21 fungicides, 17 fungicides also inhibited mycelial growth of Ganoderma lucidum as well, but imazalil, procymidone, triforine, and vinclozolin had no inhibitory effects. However, vinclozolin showed no inhibitory effect on mycelial growth of the mushroom even at the concentration of 50 $\mu\textrm{g}$/ml vinclozolin solution for 2 hours, and then the pathogen was inoculated. After two month-cultivation of the mushroom, over 90% of logs treated with vinclozolin without pathogen inoculation produced fruiting bodies. However, fruiting bodies were not produced form the logs inoculated with the pathogen, but not treated with vinclozolin. Fifty seven percent of logs. which were pre-treated with vinclozolin and then inoculated with the pathogen produced fruiting bodies. Based on the results, vinclozolin is effective for the control of yellow disease of the Ganoderma lucidum caused by Xylogone sphaerospora.

Chemical Control of Gray Blight of Tea in Korea

  • Screening of effective fungicides, determination of effective fungicide application time and investigation of the emergence of resistant isolates to fungicides were conducted to establish effective chemical control strategy of gray blight of tea (Camellia sinensis) in Korea. Systemic fungicides, such as bitertanol, were effective for controlling gray blight when the fungicides were sprayed within 3 days after cutting tea leaves for harvest. Sprayed immediately after cutting, contact fungicides such as chlorothalonil were also effective, but control efficacy rapidly decreased with lengthening of the intervals between cutting and spraying. Korean isolates of P. longiseta and P. theae were still sensitive to bitertanol or fluazinam. However, the isolates highly insensitive to copper hydroxide or thiophanate-methyl have already emerged in Korea. Therefore, careful attention should be paid to use of fungicides and spraying programs for the effective control of gray blight of tea in Korea.

Hepatocarcinogenic Screening of Carbendazim Using Medium-term Carcinogenicity Bioassay (중기발암성시험기법을 이용한 Carbendazim의 간발암성 검색)

  • Carbendazim, which is widely used fungicide, was investigated for rat hepatocarcinogenesis using a medium-term carcinogenicity bioassay. All rats were initially given a single dose (200mg/kg) of diethylnitrosamine (DEN) i.p. and then, starting 2 weeks later, carbendazim treatment group and positive control group received carbendazim (7 mg/kg/ day) and 2-acetylaminofluorene (2-AAF, 1%), respectively, in the diet for 6 weeks. All rats were subjected to two-thirds partial hepatectomy (PH) at week 3 and sacrificed at week 8. Carcinogenic potential was scored by comparing the number and area per cm$^2$ of induced glutathione-S-transferase placental form (GST-P) positive foci in the liver. Carbendazim had no effect in the increase of body weight, hematological and biochemical values, and the number and area of GST-P positive loci. These results suggest that this bioassay using DEN-PH method can be useful for detection of hepatocarcinogenic potentials of pesticide.

Genetic Effects of Pesticides in the Mammalian Cells: I. Induction of Micronucleus

  • In order to evaluate the mutagenic potential in animal for these pesticides which were proved to be mutagenic in the bacterial screening system with a metabolic activation in vitro, we have studied in vivo cytogenetic effects on mouse bone marrow by means of the micronucleus test. The clastogenic activity of the chemical is evaluated as the frequency of micronuclei in polychromatic erythrocytes. We have tested six pesticides, insecticides, DDVP and trichlorfon, fungicide, TMTD, herbicides, NIP and MO and growth regula색, maleic hydrazide. It was found that among the tested pesticides only TMTD exhibited minimal activity in inducing micronuclei. Organophosphorus insecticide DDVP that is the most broadly used and economically important chemical, did not increase the micronuclei frequencies in mouse bone marrow cells as with the all other pesticides tested.

  • Botrytis cinerea isolates obtained from infected plants of cucumber, tomato, and strawberry were divided into three groups (sporing, sclerotial, and mycelial types). Of which sclerotial types were the major group. There were no correlations between morphological phenotypes and responses to benzimidazole and dicarboximide fungicides. External structure of conidia of three phenotypes by scanning electron microscope was the same with verrucose surface. Mycelial type was the most virulent on fruits of eggplants. Comparative tests were carried out to examine correlations between the virulence and production of fungal enzymes such as phenol oxidases, pectin methyl esterases (PME), amylases, cellulases, ureases, ${\beta}-glucosidases$, and proteinases. There was no correlation among the phenotypes in production of phenol oxidases and ${\beta}-glucosidases$. However, there were significantly different from each other in PME, amylase, cellulase, urease, and protease activity.

Isolation and Characterization of the Colletotrichum acutatum ABC Transporter CaABC1

  • Fungi tolerate exposure to various abiotic stresses, including cytotoxic compounds and fungicides, via their ATP-driven efflux pumps belonging to ATP-binding cassette (ABC) transporters. To clarify the molecular basis of interaction between the fungus and various abiotic stresses including fungicides, we constructed a cDNA library from germinated conidia of Colletotrichum acutatum, a major anthracnose pathogen of pepper (Capsicum annum L.). Over 1,000 cDNA clones were sequenced, of which single clone exhibited significant nucleotide sequence homology to ABC transporter genes. We isolated three fosmid clones containing the C. acutatum ABC1 (CaABC1) gene in full-length from genomic DNA library screening. The CaABC1 gene consists of 4,059 bp transcript, predicting a 1,353-aa protein. The gene contains the typical ABC signature and Walker A and B motifs. The 5'-flanking region contains a CAAT motif, a TATA box, and a Kozak region. Phylogenetic and structural analysis suggested that the CaABC1 is a typical ABC transporter gene highly conserved in various fungal species, as well as in Chromista, Metazoans, and Viridiplantae. We also found that CaABC1 was up-regulated during conidiation and a minimal medium condition. Moreover, CaABC1 was induced in iprobenfos, kresoxim-methyl, thiophanate-methyl, and hygromycin B. These results demonstrate that CaABC1 is necessary for conidiation, abiotic stress, and various fungicide resistances. These results will provide the basis for further study on the function of ABC transporter genes in C. acutatum.

Controlling effect of several surfactants on barley powdery mildew caused by Erysiphae graminis (보리 흰가루병에 대한 몇 가지 계면활성제의 방제 효과)

  • Controlling effect of several surfactants on barley powdery mildew (pathogen : Erysiphae graminis) and those influence on fungicidal activity by tank-mixing with commercialized fungicides were investigated in greenhouse test with barley seedlings. While protective activity of PNPP showed 90% at $1,000{\mu}g/mL$, that of LN 13.0 91.2%, tile best among 8 surfactants used in curative test. With $500{\mu}g/mL$, LN 13.0 showed 4% of disease percentage on the first leaf of narley continuously 4 days after the curative application. Excent for PNPP, the other surfactants, including NP 13.0, LN 13.0, and PAS, showed good inhibitory activity against spore germination in water agar medium amended with those surfactants. PAS, which showed very poor effect against barley powdery mildew in greenhouse test, strongly inhibited spore germination. Fungicide of triadimefon and triforine showed no controlling activity against Erysiphae graminis in narley seedlings, but they highly controlled powdery mildew in case of addition with $500{\mu}g/mL$ of LN 13.0 and Tween 20. SP 13.0, NP 13.0, LN 13.0 and DBC showed severe phytotoxicity in first leaf of barley seedlings, but Tween 20, SLIS, PAS, and PNPP not at $1,000{\mu}g/mL$.
