Epidemiological Concepts and Strategies in Breeding Soybeans for Disease Resistance

  • The epidemiology of plant disease deals with the dynamic processes of host-pathogen interactions, which determine the prevalence and severity of the disease. Epidemic processes for most foliar diseases of plants follow a series of steps: arrival of pathogens on plant surfaces, initial infection, incubation period, latent period, sporulation, dissemination of secondary inoculum, and infectious period. These complex biological processes are influenced by the environment-Man also often interfers with these processes by altering the host and pathogen populations and the environment. Slowing or halting any of the epidemic processes can delay the development of the epidemic, so that serious losses in yield due to disease do not occur. It is generally recognized that the most effective and efficient method of minimizing disease damage is through the use of resistant cultivars, particularly when other methods such as fungicide applications are not economically feasible-Populations of plant pathogens are not genetically uniform nor are they necessarily stable. Cultivars bred for resistance to current populations of a pathogen may not be resistant in the future due to selection pressures placed on the pathogen populations. Understanding population development and genetic variability in the pathogen, and knowledge of the genetics of resistance in the plant should help in developing breeding strategies that wi1l provide effective and stable disease control through genetic resistance. Changes in Sensitivity Levels of Botrytis cinerea Populations to Benzimidazole, Dicarboximide, and N-Phenylcarbamate Fungicides (잿빛곰팡이병균(Botrytis cinerea)의 Benzimidazole계, Dicarboximide계 및 N-phenylcarbamate계 살균제에 대한 감수성 변화)

  • Three three hundred and ninety seven isolates of Botrytis cinerea were isolated from infected plants of strawberry, tomato and cucumber from several areas in Korea during 1994∼1996 and the resistance of these isolates against some fungicides were examined. The isolation frequency of phenotypes resistant to carbendazim, procymidone, and diethofencarb were found to be 69.9 43.7, and 31.8%, respectively. The isolates were divided into six phenotypic groups; SSR, SRR, RSS, RRS, RSR and RRR, representing sensitive (S) or resistant (R) to benzimidazole, dicarboximide, and N-phenylcarbamate fungicides in order. The percentage of six phenotypes were 28.2, 2.0, 27.2, 41.0, 0.9 and 0.8%, respectively. On the basis of the mycelial growth inhibition (%) B. cinerea isolates were divided into three classes (class 1; 0∼50%, class 2; 51-99%, class 3; 100% inhibition) on carbendazim and three classes (class 1; 0∼75%, class 2; 76∼99%, class 3; 100% inhibition) on procymidone and the mixture of carbendazim+diethofencarb, respectively. Changes in sensitivity levles to carbendazim and carbendazim+diethofencarb were affected by introduction and increasing ratio of the use of diethofencarb.

Discussions on the Efficacy, Phytotoxicity and Residue of Organo-Arsenic Fungicides (유기비소제의 약효 및 약해와 잔류에 관한 종합적 고찰)

  • Since rice high yielding practices with blast resistant varieties induced severe and earlier infestation of sheath blight recently, it is strongly suggested to spray at least 10 days earlier than the present recommendation and more than 2 sprays from early July to early August seems to be essential for the efficient control of sheath blight in the future. Among organo-arsenic fungicides, MAFe (Neo Asozin) is known as the most effective and economic as well as with the advantages of free phytotoxicity and safe residure on rice, and applications of the mixed formulation with other fungicide and insecticide is suitable for the one-hit control of rice diseases and insect pests in connection with cooperative mass control system. Present level of arsenic $(As_2O_3)$ residue in .ice is summarized as 0.2ppm to 0.65 ppm in highest average, and there are more possibilities of extensive MAFe fungicide uses for the efficient and economic control of this disease. During the past few years, the acreage of sheath blight control has been extended several times while the increase of the fungicides used for sheath blight showing about 10 times in quantity and mort than 15 times in amount, and also the fungicide cost estimated for the control is differ from 1 to 12 billion 1on by the use of MAFe or antibiotic fungicide in a year.

Response to metalaxyl of Phytophthora capsici isolates collected in 2005 and 2006 (2005년과 2006년에 채집한 고추 역병균(Phytophthora capsici)의 Metalaxyl에 대한 약제 반응)

  • It was the investigated the response to metalaxyl of Phytophthora capsici isolates collected in 2005 and 2006. With effective concentrations ($EC_{50}$) of metalaxyl causing 50% growth inhibition, resistance baseline was determined as more than $1.0\;{\mu}g\;mL^{-1}$. Based on the resistance baseline, isolation frequency (%) of P. capsici resistant to metalaxyl was 29.0% and 218% in 2005 and 2006, respectively. Among the isolates of P. capsici obtained in 2006, the isolation was variable; 33.3% in Chungnam, 26.3% in Chungbuk and 11.1% in Gyeongbuk. Two isolates of metalaxyl-sensitive (MS) and too isolates of metalaxyl-resistant (MR) P. capsici were selected and then used to investigate the activity of metalaxyl to their development stages. Even though there was a difference in mycelial growth inhibition by metalaxyl between MS and MR isolates, the fungicide was not active or nearly to sporangium germination, zoospore release, and zoospore germination of both MS and MR isolates. However, the fungicide showed weak activity against sporangium germination and zoospore release of P. capsici, not related with its resistance. Also, it was not inhibitory to zoospore germination of both resistant and sensitive isolates. In a greenhouse test, it showed 100% of control value against P. capsici 06-86 sensitive to metalaxyl, when it was applied by soil-drenching at $25\;{\mu}g\;mL^{-1}$. However, 06-130 and 16-155 resistant to metalaxyl showed less than 20% of control value.

Cross-resistance of Colletotrichum acutatum s. lat. to Strobilurin Fungicides and Inhibitory Effect of Fungicides with Other Mechanisms on C. acutatum s. lat. Resistant to Pyraclostrobin (Strobilurin계 살균제에 대한 고추탄저병균의 교차저항성과 Pyraclostrobin 저항성균에 대한 다른 기작 살균제의 억제 효과)

  • Colletotrichum acutatum s. lat. 20JDS8 sensitive and 20CDJ6 resistant to pylaclostrobin were used to investigate the cross-resistance with fungicides belonging to the strobilurins and the characteristics of fungicidal controlling activities with different mechanisms against the isolate resistant to the fungicide. The resistant isolate of 20CDJ6 also showed the resistance to azoxystrobin, trifloxystrobin, and kresoxim-methyl, suggesting that there is a cross-resistance relationship. All fungicides with different action mechanisms inhibited mycelial growth of both susceptible and resistant isolates of C. acutatum s. lat., but their disease control effects in fruits were different according to the fungicides. The disease control effect of isopyrazam against 20JDS8 and 20CDJ6 was very low, and fluazinam showed a control effect of 91.9% and 88.1% against 20JDS8 and 20CDJ6 only when it was treated before inoculation by spraying spore suspensions on pepper fruits without wounds. Tebuconazole and prochloraz effectively inhibited not only the mycelial growth of 20JDS8 and 20CDJ6 on potato dextrose agar medium, but also disease incidence in red pepper fruits. As a result of this study, C. acutatum s. lat. 20CDJ6 resistant to pyraclostrobin showed cross-resistance with other strobilurin fungicides. In addition, we think that fluazinam, tebuconazole, and prochloraz can be recommended as alternative fungicides for the control of red-pepper pyranthracnose pathogens resistant pyraclostrobin. However, fluazinam can be effective only if it is treated protectively before the occurrence of the disease.

Design, Synthesis and Antifungal Activities of Novel Strobilurin Derivatives Containing Pyrimidine Moieties

  • Strobilurins are one of the most important classes of agricultural fungicides. To discover new strobilurin derivatives with high activity against resistant pathogens, a series of novel ${\beta}$-methoxyacrylate analogues were designed and synthesized by integrating substituted pyrimidine with a strobilurin pharmacophore. The compounds were confirmed and characterized by infrared, $^1H$ nuclear magnetic resonance, elemental analysis and mass spectroscopy. The bioassays indicated that most of the compounds (1a-1h) exhibited potent antifungal activities against Colletotrichum orbiculare, Botrytis cinerea Pers and Phytophthora capsici Leonian at the concentration of 50 ${\mu}g/mL$. Exhilaratingly, compound 1d (R=3-trifluoromethylphenyl) showed better antifungal activity against all the tested fungi than the commercial strobilurin fungicide azoxystrobin.

Occurrence of Multiple Resistant Isolates of Botrytis cinerea to Benzimidazole and N-phenylcarbamate Fungicides (Benzimidazole계 및 N-phenylcarbamate계 살균제에 다중 저항성인 잿빛곰팡이병균의 발생)

  • 1994년, 1995년에 주요 채소 작물에서 분리한 잿빛곰팡이병균(Botrytis cinerea)의 방제 살균제에 대한 저항성을 조사한 결과, 1994년 분리된 713균주 중 610균주(85.6%)가 benzimidazole계에 저항성(Ben\ulcorner)이었고, 249균주(34.9%)는 dicarboximide계 살균제에 저항성(Pro\ulcorner)이었으며, benzimidazole계 및 N-phenyl carbamate계 두약제 모두에 저항성인 균주는 분리되지 않았다. 1995년에는 현재까지 520균주를 분리하였는데, 그중 Ben\ulcorner이 317균주(61%)이며 Pro\ulcorner은 214균주(41.2%)이었고, Ben\ulcorner이며 NPC\ulcorner인 균주(Ben\ulcorner+NPC\ulcorner)는 15균주(2.9%)로 나타났다. 이러한 저항성 균주의 최소 억제 농도는 carbendazim 1,000 $\mu\textrm{g}$/ml 이상, diethofencarb 1,000 $\mu\textrm{g}$/ml 이상이었다. 또 Ben\ulcorner+NPC\ulcorner인 15균주 중 3균주는 dicarboximide계 살균제인 procymidone에도 저항성이었으며, 한 균주를 제외한 모든 Pro\ulcorner 균주는 Ben\ulcorner로 나타났다.

Chemical Control of Gray Blight of Tea in Korea

  • Screening of effective fungicides, determination of effective fungicide application time and investigation of the emergence of resistant isolates to fungicides were conducted to establish effective chemical control strategy of gray blight of tea (Camellia sinensis) in Korea. Systemic fungicides, such as bitertanol, were effective for controlling gray blight when the fungicides were sprayed within 3 days after cutting tea leaves for harvest. Sprayed immediately after cutting, contact fungicides such as chlorothalonil were also effective, but control efficacy rapidly decreased with lengthening of the intervals between cutting and spraying. Korean isolates of P. longiseta and P. theae were still sensitive to bitertanol or fluazinam. However, the isolates highly insensitive to copper hydroxide or thiophanate-methyl have already emerged in Korea. Therefore, careful attention should be paid to use of fungicides and spraying programs for the effective control of gray blight of tea in Korea.

Morphology, Molecular Phylogeny, and Fungicide Sensitivity of Phytophthora nagaii and P. tentaculata in Korea

  • Phytophthora species, classified under Oomycota, cause significant damage to various crops and trees. The present study introduced Phytophthora species, P. nagaii and P. tentaculata, new to Korea, which pose notable risks to their respective host plants. Our research provided a comprehensive description of these species taking into account their cultural features, morphological characteristics, and molecular phylogenetic analysis using the internal transcribed spacer rDNA region and cytochrome c oxidase subunit mtDNA genes (cox1 and cox2) sequences. In addition, this study first evaluated the sensitivity of P. nagaii and P. tentaculata to five anti-oomycete fungicides, finding both species most responsive to picarbutrazox and P. tentaculata resistant to fluazinam. The data can guide targeted treatment strategies and offer insights into effective control methods. The findings expand our understanding of the diversity, distribution, and management of Phytophthora species in Korea.

Antifungal Properties of Streptomyces bacillaris S8 for Biological Control Applications

  • Soybean (Glycine max), a crucial global crop, experiences yearly yield reduction due to diseases such as anthracnose (Colletotrichum truncatum) and root rot (Fusarium spp.). The use of fungicides, which have traditionally been employed to control these phytopathogens, is now facing challenges due to the emergence of fungicide-resistant strains. Streptomyces bacillaris S8 strain S8 is previously known to produce valinomycin t through a nonribosomal peptide synthetase (NRPS) pathway. The objective of this study was to evaluate the antifungal activity of S. bacillaris S8 against C. truncatum and Fusarium sp., assessing its efficacy against soybean pathogens. The results indicate that strain S8 effectively controlled both above-ground and underground soybean diseases, using the NRPS and NRPS-related compound, suggesting its potential as a biological control in plant-microbe interactions. These findings underscore the pivotal role of the stain S8 in fostering healthy soybean microbial communities and emphasize the significance of microbiota structure studies in unveiling potent biocontrol agents.
