Anti-fatigue effect of a beverage mixture containing red ginseng and Prunus mume fruit vinegar on high-intensity exercised rats (홍삼과 매실식초 혼합 음료의 고강도 운동을 실시한 흰쥐에서 항피로 효과)

  • New types of vinegar drinks are being processed and developed using various raw materials. In this study, a beverage containing a mixture of 0.25% red ginseng extract and 5% Prunus mume fruit vinegar (RPV) was evaluated for its anti-fatigue effect on high-intensity exercised rats. RPV administration markedly enhanced running endurance and significantly decreased fatigue-related serum biomarkers, such as inorganic phosphate, ammonia, and L-lactate, compared to the other groups. In addition, RPV administration increased glycogen contents in the liver and muscles and decreased creatine kinase activity in the serum and muscles. RPV administration also remarkedly increased the activity of lactate in the muscles. Furthermore, HPLC analysis revealed that main organic acids in RPV were acetic acid, malic acid, and citric acids. Overall, the results indicate that RPV improved fatigue recovery in exhausted rats, thus proving a promising material of functional food to attenuate fatigue.

Verification of Whitening Activity of Inonotus obliquus Extracts in B16F10 Melanoma Cells (차가버섯(Inonotus obliquus) 추출물의 B16F10 멜라노마 세포에서의 미백활성 검증)

  • In this study, the various whitening activities of Inonotus obliquus were assessed for potential use as functional cosmetic materials. When measuring electron donating ability to confirm the antioxidant ability of I. obliquus extract, increased activity was observed as the concentration increased, and it was found an outstanding antioxidant capacity of 82.1% at a 1,000 ㎍/ml concentration. Also, the tyrosinase inhibitory effect, related to a whitening effect, was found to have inhibitory activity that increased in a concentration-dependent manner. The results of verifying the viability of melanoma cells (B16F10) using an MTT assay showed cell viability of more than 80% at concentrations below 100 ㎍/ml. Therefore, cell-related experiments were conducted at 25, 50, and 100 ㎍/ml concentrations. By measuring protein expression related to melanin synthesis via treating B16F10 cells with I. obliquus extract, it was confirmed that protein expression was inhibited in all factors, depending on the concentration. TRP-1 and MITF appeared by 40.1% and 64.2% amount of expression, respectively, at 100 ㎍/ml concentrations, and tyrosinase and TRP-2 were verified as having better protein expression inhibition than arbutin. In measuring mRNA expression related to melanin synthesis by treating B16F10 cells with I. obliquus extract, it was confirmed that mRNA expression was suppressed as the concentration increased. Accordingly, it was confirmed that I. obliquus extract has excellent whitening activity and could be used as a cosmetic material.

  • Biorenovation is a biotransformation method that converts the structure of chemical compounds and natural product through biocatalytic metabolism of microorganism and could enhance biological effectiveness and mitigate cytotoxicity compared to its substrates. Althaea rosea L. has been used as oriental medicine and is known for physiological efficacies such as antiurolithiatic, anti-inflammatory, and anti-cancer activities. A. rosea L. callus, the plant tissue grown to protect its wound, has been reported to have antioxidant and whitening effects. However, mechanisms of its other activity such as inflammation have not yet been investigated. In this study, we extracted A. rosea L. callus (AR) and produced biorenovated AR (ARBR), and then analyzed anti-inflammatory effect in Lipopolysaccharide-induced RAW 264.7 macrophage at 50, 100, 200 ㎍/mL of ARBR. As a result of inhibition test of nitric oxide production, it was found that ARBR was superior to AR without apparent toxicity. Furthermore, ARBR significantly inhibited production of prostaglandin E2, inducible nitric oxide synthase, cyclooxygenase-2 and pro-inflammatory cytokines including Tumor necrosis factor-α, Interleukin-6, Interleukin-1β in a concentration-dependent manner. In conclusion, we suggest that ARBR could regulate the excessive inflammatory response to an appropriate level and be a promising material for functional cosmetics and pharmaceuticals.

Changes in oral health status and function among older adults through professional oral care and rehabilitation exercises (전문적인 구강관리와 재활운동을 통한 노인의 구강건강 상태와 기능의 변화)

  • Objectives: To develop a program that combines the oral health promotion program for the elderly and to investigate the effect of improving oral function by applying the oral health promotion program to elderly people using welfare facilities. Methods: From June to October 2022, a total of 29 elderly individuals utilizing welfare facilities in the Gyeongnam region were recruited. The control group underwent surveys and objective oral health assessments. The experimental group participated in an oral health promotion program, consisting of oral functional rehabilitation exercises and expert oral health management, conducted once a week for 8 weeks at the facilities. Objective oral health assessments were conducted before, during (at 4 weeks), and after (at 8 weeks) the intervention period for both groups. Results: As a result of the oral health promotion program, it was confirmed that there was an improvement effect in bad breath (p<0.001), saliva secretion amount (p<0.001), orbicularis oculi muscle (p<0.001), and plaque index (p<0.001). Conclusions: A revitalization plan is needed so that the efficient oral health promotion program for the elderly can be reflected as a systematic and professional community care oral health management program.

  • In the past, retailers secured customer loyalty by offering convenient locations, unique assortments of goods, better services than competitors, and good credit policy. All this has changed. Goods assortments among stores have become more alike as national-brand manufacturers place their goods in more and more retail stores. Service differentiation also has eroded. Many department stores have trimmed services, and many discount stores have increased theirs. Customers have become smarter shoppers. They don't pay more for identical brands, especially when service differences have diminished. In the face of increased competition from discount storess and specialty stores, department stores are waging a comeback war. Growth of intertype competition, competition between store-based and non-store-based retailing and growing investment in technology are changing the way consumers shop and retailers sell. Different types of stores-discount stores, catalog showrooms, department stores-all compete for the same consumers by carrying the same type of merchandise. The biggest winners are retailers that have helped shoppers to be economically cautious, simplified their increasingly busy and complicated lives, and provided an emotional connection. The growth of e-retailers has forced traditional brick-and-mortar retailers to respond. Basically brick-and-mortar retailers utilize their natural advantages, such as products that shoppers can actually see, touch, and test, real-life customer service, and no delivery lag time for small-sized purchases. They also provide a shopping experience as a strong differentiator. They are adopting practices as calling each shopper a "guest". The store atmosphere should match the basic motivations of the shopper. If target consumers are more likely to be in a task-oriented and functional mindset, then a simpler, more restrained in-store environment may be better. Consistent with this reasoning, some retailers of experiential products are creating in-store entertainment to attract customers who want fun and excitement. The retail experience must deliver value to turn a one-time visitor into a loyal customer. Retailers need a tool that measures the full range of components that define experience-based value. This study uses an experiential value scale(EVS) developed by Mathwick, Malhotra and Rigdon(2001) which reflects the benefits derived from perceptions of playfulness, aesthetics, customer "return on investment" and service excellence. EVS is useful to predict differences in shopping preferences and patronage behavior of customers. EVS consists of items measuring efficiency, economic value, visual appeal, entertainment value, service excellence, escapism, and intrinsic enjoyment, which are subscales of experiencial value. Efficiency, economic value, service excellence are linked to the utilitarian shopping value. And visual appeal, entertainment value, escapism and intrinsic enjoyment are linked to hedonic shopping value. It has been found that consumers value hedonic experiences activated from escapism and attractiveness of shopping environment as much as the product quality, price, and the convenient location. As a result, many department stores, discount stores, and other retailers are introducing differential marketing strategy based on emotional/hedonic values. Many researches suggest that consumers go shopping not only for buying products but also for various shopping experiences. In other words, they seek the practical, rational value as well as social, recreational values in the shopping process(Babin et al, 1994; Bloch et al, 1994). Retailers may enhance buyer's loyalty to store by providing excellent emotional/hedonic value such as the excitement from shopping, not just the practical value of buying good products efficiently. We investigate the effect of perceived shopping values on the emotional experience and store loyalty based on the EVS(Experiential Value Scales) developed by Holbrook(1994), Mathwick, Malhotra and Rigdon(2001). This study assumes that the relative effect of shopping value dimensions on the responses of shoppers will differ according to types of stores and analyzes the moderating effect of store type(department store VS. discount store) on the causal relationship between shopping value dimensions and store loyalty. Emprical results show that utilitarian values of shopping experience and hedonic value of shipping experience give the positive effect on the emotional response of consumers and store loyalty. We also found the moderating effect of store types. The effect of utilitarian shopping values on the attitude toward discount store is higher than the effect of utilitarian shopping values on the attitude toword department store. And the effect of hedonic shopping value on the emotional response to discount store is higher than on the emotional response to department store. The empirical results reflect on the recent trend that discount stores try to fulfill the hedonic needs of consumers as well as utilitarian needs(i.e, low price) that discount stores traditionally have focused on

In Vitro Anticancer and Antioxidant Effect of Solvent Extracts from Tuna Dried at Low Temperature Vacuum. (저온진공건조 참치추출물의 in vitro 항암 및 항산화 효과)

  • We investigated the inhibitory effects of solvent extracts from dried tuna on the growth of cancer cell lines (HT1080 human fibrosarcoma and HT-29 human colon cancer cells) and $H_2O_2$-induced oxidative stress. Inhibitory effects of acetone with methylene chloride (A+M) and methanol (MeOH) extracts on the growth of HT1080 and HT-29 cancer cells increased in a dose dependent manner (p<0.05). The inhibitory effect was more significant on the growth of HT1080 cells, and A+M extracts had a higher inhibitory effect compared to MeOH extracts. The treatments of hexane, 85% aq. methanol, butanol and water fractions significantly inhibited the growth of both cancer cells (p<0.05). Among the fractions, hexane and 85% aq. methanol fractions showed higher inhibitory effects. In order to determine the protective effect on $H_2O_2$-induced oxidative stress, a DCHF-DA (dichlorodihydrofluorescin diacetate) assay was conducted. All fractions, including crude extracts of dried tuna, appeared to significantly reduce the levels of intracellular reactive oxygen species (ROS) with dose responses (p<0.05). Among the fractions, BuOH and 85% methanol fractions showed a higher protective effect on the production of lipid peroxides. These results indicate that the consumption of tuna may be recommended as a potent functional food for preventing cellular oxidation and cancer.

Effect of Enzyme Treatment on Functional Properties of Nectarine Beverage (효소처리에 의한 천도복숭아 음료의 기능성 증진)

  • Nectarine beverage treated with cellulase and pectinase enzymes was measured for mineral contents, total flavonoids, and free amino acids and DPPH radical scavenging effect, nitrite scavenging effect. Total flavonoid contents of the no treatment, treated with pectinase, with cellulase, and with both measured 0.146 mg/mL, 0.167 mg/mL, 0.148 mg/mL and 0.171 mg/mL, respectively. DPPH was measured as 13.42% with no treatment and more than 28.98% with enzyme treatments. Nitrite scavenging effect with no treatment was 79% at pH 1.2. Whereas, it was measured above 90% while treated with enzymes at pH 1.2. And also, the nitrite scavenging effect was slightly higher at pH 3.0, pH 4.0 and pH 6.0 than no treatment. Results of free amino acids analysis revealed that, aspartic acid, serine, alanine, $\gamma$-aminobutylic acid, and glutamic acid were present with the amount ranging from 86.71% to 94.14% from total detected free amino acids. Ornithine and taurine were also observed from the beverages. The mineral contents, nitrogen element (T-N) of enzyme treatment of nectarine beverages were measured slightly higher than T-N of no treatment, however, the $P_2O_5$ was similar. Moreover, CaO, MgO and $K_2O$ in the beverages were measured above 45 mg/L, 85 mg/L and 2,133 mg/L, respectively.

Studies on the Antibacterial Activity of Enzymatic Hydrolyzates of Lactoferrin Derived from Bovine Colostrum (유우 Lactoferrin 효소가수분해물 항균 활성에 관한 연구)

  • The investigative research on the mammalian milk purely consisted of the physiological quality of lactoferrin was conducted to reveal the antimicrobial ativity of specifically functional foods with antibiotic characteristics as a basic data in food manufacturing. Bovine lactoferrin were isolated from raw milk samples, and was digested with pepsin, trypsin and chymotrypsin. It was necessary then to separate and purify lactoferrin from bovine raw milk, and in order to analyze the antimicrobial activity of the enzyme-treated bovine lactoferrin in their required quantitative fraction. Afterwards Escherichia coli and Staphylococcus aureus was incubated in it. It was that investigated to enzyme-treated fractions molecular weight and the peptide fragment with antimicrobial effect. 1. The purity of enzyme-treated bovine lactoferrin(BLF) was tested by SDS-PAGE. As a results of 12% SDS-PAGE assay, pepsin-treated LF did not exhibited band until if reaches 14 KDa, while trypsin and chymotrypsin treated LF, known to contain the non-digestive lactoferrin exhibited band at a molecular weight of 33 KDa. 2. Bovine lactoferrin was sucessfully purified through the use of Sephadex G-50 Column. In order to assay LF through the Sephadex G-50 column chromatography, the digestive bovine lactoferrin (BLFs) was eluted with a linear gradient of 0.05% Tris-HCl. When the gel-filtration analysis, pepsin, trypsin and chymotrypsin treatments of BLF fragments was showed 2, 3, and 2 peak, respectively. The results of the HPLC analysis confirmed that had a non-digestive lactoferrin receptor, and trypsin and chymotrypsin treated BLFs has an antimicrobial effect. 3. To measure the strength of the antimicrobial effect of enzyme treated lactoferrin it was compared to the antimicrobial activity taking place at the incubated Escherichia coli and Staphylococcus aureus. This might explain the resistance of the microorganisms for peptide fragment. The pepsin-treated of bovine lactoferrin was markedly reduced by incubation of the cells. Trypsin-treated of BLF was similar to chymotrypsin-treated of BLF. However, trypsin and chymotrypsin treatments of BLFs were showed the antimicrobial effect until eight hours incubation for native bovine lactoferrin. Therefore the enzyme-treated lactoferrin have an antimicrobial effect even non-digestive lactoferrin. 4. The digestive bovine lactoferrin fragments assay was carried out by the use of Sephadex G-50 column chromatography and SDS-PAGE. The pepsin and chymotrypsin-treated fragments has a low molecular weight and trypsin-treated lactoferrin was only showed a band. It was described that characteristics of digestive protein. It appeared that there may be a relation between virulence and resistance to enzyme-treated BLF.

Comparison of Biological Activity between Nelumbo nucifera G. Extracts and Cosmetics Adding Nelumbo nucifera G. (백련(Nelumbo nucifera G.) 추출물 및 화장품에 첨가 시 생리활성 비교)

  • The solvent extracts of Nelumbo nucifera G. were investigated for antioxidant activities, whitening and anti-wrinkle effects to apply as a functional ingredient in cosmetic products. For their industrial application, the cosmetic products were also prepared with advanced formulation techniques such as W/O/W multiple emulsion. Total phenolic and flavonoids contents increased in Nelumbo nucifera G.-Leaf (NN-L). The electron donating ability of Nelumbo nucifera G.-Flower (NN-F) or Nelumbo nucifera G.-Leaf (NN-L) extracts were above 85% at a concentration of 500 ppm. The superoxide dismutase (SOD)-like activity of Nelumbo nucifera G. (NN-L) extracts was about 60% at a concentration of 1,000 ppm. The xanthine oxidase inhibitory effect of NN-L extract was higher than that of NN-F and NN-S extracts. The tyrosinase inhibitory effect, which is related to skin-whitening, was 36% in NN-F at 1,000 ppm. For anti-wrinkle effect, the elastase inhibition activity of NN-L was about 30% at 1,000 ppm. The results of stability test showed that W/O/W multiple emulsion (ME) containing Nelumbo nucifera G. extracts. The electron donating ability of the ME containing NN-F and NN-L were about 60% at a concentration of 100 ppm. The superoxide dismutase (SOD)-like activity of the ME containing NN-L was 30% at 1,000 ppm. The tyrosinase inhibitory effect, which is related to skin-whitening, was 34% in the ME containing NN-F at 1,000 ppm. In anti-wrinkle effect, the elastase inhibition activity of the ME containing NN-L was about 55% at 1,000 ppm.
