우리나라 학교우유급식 관리현황 및 영양사들의 인식 조사 (Investigation into Korean School Milk Program Management and Dietitians' Perception of School Milk Program Improvement)

  • This study was aimed to investigate management conditions and dietitians' perception for improvement of school milk programs. Using an online sampling method, collected data from 1,723 dietitians nationwide were analyzed. The results show that 44.4% of respondents carried out the cyclic school milk program, providing various types of dairy products either weekly or monthly. Further, 80.3% of respondents answered that preference survey results were a determining factor in their choice of school milk provider, and 55.9% said that a school steering committee made the decision on relevant facts of the school milk program. For diversification of school milk type, 60.5% of respondents wanted to maintain the current system, and 39.5% answered that milk type needed to be diversified. The respondents preferred fermented milk products, functional milk, and processed milk, in order, if school milk type is diversified. To improve perception and knowledge about milk, 66.0% of respondents thought that nutrition education by a nutrition teacher would be the most effective, and parent's letters or a nutrition newsletter would be helpful. More than half of respondents (55.0%) disagreed with a merged program for school foodservice and milk.

Effects of Germinated Brown Rice Addition on the Flavor and Functionality of Yogurt

  • This study aimed to investigate the functional and physicochemical properties of yogurt, supplemented with germinated brown rice (GBR) containing γ-aminobutyric acid (GABA), during storage. GBR was produced by soaking brown rice at 30℃, and saccharified germinated brown rice (SGBR) was produced by treating brown rice with α- and β-amylase for 1 h, at 80℃ and 60℃, respectively. Yogurt was manufactured using a commercial starter (YC-X11, CHR. Hansen, Denmark) at 37℃ for 12 h. The fatty acids and GABA contents were analyzed using GC and HPLC, respectively. The fatty acids in the cereal samples consisted of oleic, linoleic, and palmitic acid. The portion of oleic acid was the highest, at 35.65% in GBR, and 32.16% in SGBR. During germination, the oleic acid content increased, whereas linolenic and palmitic acid contents from GBR tended to decrease. Although the portion of saturated fatty acids, such as stearic and myristic acid, decreased significantly (p<0.05), that of unsaturated fatty acids, such as oleic and linoleic acid, increased with an increase in supplementation of BR, GBR, or SGBR in the yogurt. The yogurt, supplemented with cereal samples, showed a tendency of an increase in the concentration of GABA with an increase in the supplementation of the cereal samples. However, yogurt supplemented with GBR showed the highest concentration of GABA, regardless of the supplementation of the cereal samples. These results indicated that yogurt supplemented with BR, GBR, or SGBR could be a promising dairy product.

Microbiological Characteristics of Gouda Cheese Manufactured with Pasteurized and Raw Milk during Ripening Using Next Generation Sequencing

  • Gouda cheese, one of most popular cheeses in the Korea, has been produced from only pasteurized milk in Korean dairy farms. Recently, it has become legally possible to produce ripened cheese manufactured with raw milk in Korea. In the present study, we investigated the physico-chemical and microbiological characteristics of Gouda cheese manufactured with raw (R-GC) or pasteurized milk (P-GC) during manufacturing and ripening. Particularly, this study characterized the bacterial community structure of two cheese types, which are produced without pasteurization during ripening based on next generation sequencing of 16S rRNA gene amplicons. During ripening, protein and fat content increased slightly, whereas moisture content decreased in both P-GC and R-GC. At the 6 wk of ripening, R-GC became softer and smoother and hence, the values of hardness and gumminess, chewiness in R-GC was lower than that of P-GC. Metagenomic analysis revealed that the bacterial genera used a starter cultures, namely Lactococcus and Leuconostoc were predominant in both P-GC and R-GC. Moreover, in R-GC, the proportion of coliform bacteria such as Escherichia, Leclercia, Raoultella, and Pseudomonas were detected initially but not during ripening. Taken together, our finding indicates the potential of manufacturing with Gouda cheese from raw milk and the benefits of next generation sequencing for microbial community composition during cheese ripening.

후산 발효 적합 균주 선발 및 특성 (Properties of Lactic Acid Bacteria That Cause Decrease in Post-Fermentation to Apply Product)

  • Emerging studies suggest that vegetables or fruit juices deemed to be potential alternative base medium for lactic acid bacteria fermentation. Until now, limited studies have been carried out to evaluate such applications. Thus, the objective of present study is that lactic acid bacteria were evaluated for their viability at low pH, growth during storage at low temperature, and $CO_2$ formation. Furthermore, the effects of grapefruit extract with respect to cell viability, sensory ability, and organic acid production were evaluated for these strains. The probiotic properties of the strains, including acid tolerance, bile tolerance, and adhesion to human intestinal epithelial cells (HT-29 cells), prebiotic characteristics, and safety features were examined. All strains survived in MRS medium broth adjusted to pH 3.8, at $10^{\circ}C$ for 6 days, and did not produce $CO_2$ to check post fermentation. The medium of grapefruit extract fermentation by Lactobacillus plantarum CJIH 203 resulted in maximal viable counts, compared with other strains, and the extract subsequently tasted sour due to the presence of lactic acid. Lactobacillus plantarum CJIH203 was highly resistant to artificial gastric juice and intestinal juice, while Lactococcus lactis SJ09 strongly adhered to HT-29 cells. Tagatose showed the greatest ability to enhance the growth of L. plantarum SJ21, relative to the other strains. All strains were verified by safety tests such as hemolysis, gelatin hydration, and urea degradation. Therefore, these strains could be promising candidates for use in reducing excessive post-fermentation and functional products.

Effect of peanut shell extract supplementation on the growth performance, carcass characteristics and meat quality of broilers

  • The present study investigated the effects of dietary supplementation of peanut shell extract on the growth performance and physiological properties of broiler chicks. Two diet energy levels (Positive and Negative) and four additives (0.0, 0.05, and 0.1% peanut shell extract and commercial antioxidant) were factorially arranged for eight treatments. The overall weight gain of the broilers was slightly improved at 0.05% for the antioxidant treatments regardless of the diet energy levels, but there was no statistical difference among the treatments (p > 0.05). The carcass characteristics of the broilers, such as cooking loss, crude protein content, antioxidant activity, and thiobarbituric acid reactive substances (TBARS) values, were improved by the feeding diets containing the 0.05% peanut shell extract. Furthermore, it was confirmed that the dietary supplementation of peanut shell extract did not have a negative effect on the immune responses of the broilers show by the lack of statistical differences in the liver and bursa Fabricious weight and cytokine level among the treatments. From the economic analysis, dietary supplementation of peanut shell extract significantly influenced the compensatory growth and food efficiency and, in turn, led to a decrease in the duration needed to reach 1.5 kg compared to the control. These results suggest the possibility that the peanut shell extract could be used as a functional feed additive by improving the growth performance and carcass characteristics with no detrimental effects on broilers.

Cloning and Characterization of the Urease Gene Cluster of Streptococcus vestibularis ATCC49124

  • A genomic library of Streptococcus vestibularis ATCC49124 was constructed in an E. coli plasmid vector, and the urease-positive transformants harboring the urease gene cluster were isolated on Christensen-urea agar plates. The minimal DNA region required for urease activity was located in a 5.6 kb DNA fragment, and a DNA sequence analysis revealed the presence of a partial ureI gene and seven complete open reading frames, corresponding to ureA, B, C, E, F, G, and D, respectively. The nucleotide sequence over the entire ure gene cluster and 3'-end flanking region of S. vestibularis was up to 95% identical to that of S. salivarius, another closely related oral bacterium, and S. thermophilus, isolated from dairy products. The predicted amino acid sequences for the structural peptides were 98-100% identical to the corresponding peptides in S. salivarius and S. thermophilus, respectively, whereas those for the accessory proteins were 96-100% identical. The recombinant E. coli strain containing the S. vestibularis ure gene cluster expressed a high level of the functional urease holoenzyme when grown in a medium supplemented with 1 mM nickel chloride. The enzyme was purified over 49-fold by using DEAE-Sepharose FF, Superdex HR 200, and Mono-Q HR 5/5 column chromatography. The specific activity of the purified enzyme was 2,019 U/mg, and the Michaelis constant ($K_{m}$) of the enzyme was estimated to be 1.4 mM urea. A Superose 6HR gel filtration chromatography study demonstrated that the native molecular weight was about 196 kDa.

대학생의 식생활 라이프스타일에 따른 음료 섭취 실태 분석 (Intake Behavior Regarding Beverages according to Dietary Lifestyles of University Students)

  • The purpose of this study was to categorize dietary lifestyles and analyze intake behavior regarding beverages according to lifestyles of 276 university students in Yeungnam region. Frequency analysis, factor analysis, cluster analysis, reliability analysis, one-way analysis of variance, and ${\chi}^2$ tests were conducted using SPSS V.21.0. In the factor analysis, dietary lifestyles were categorized into one of five factors: health-seeking type, popularity-seeking type, safety-seeking type, economy-seeking type and convenience-seeking type. According to cluster analysis, respondents were divided into four groups: convenience-seeking group, wellbeing-seeking group, popularity-seeking group, and indifference group. Chi-square tests showed that there were significant differences in gender, grade, dwelling status, and monthly allowance according to the four dietary lifestyle groups. The level of dietary habits and percentage of correct answers related to beverages of the wellbeing-seeking group were highest among the four groups. The frequency of beverage intake was significantly different for carbonated drinks, milk and dairy products, and sport drinks according to dietary lifestyle groups. In addition, for beverage preference, there were significant differences in carbonated drinks, sport drinks, tea and functional drinks according to dietary lifestyle groups.

Complete genome sequence of Limosilactobacillus fermentum JNU532 as a probiotic candidate for the functional food and feed supplements

  • Lactic acid bacteria (LAB) have been reported to possess various beneficial properties and are commonly used as probiotics. LAB play a crucial role in milk fermentation, industrial lactic acid fermentation, and health and medicine. Limosilactobacillus fermentum isolated from fermented dairy and food products is considered as 'Generally Recognized as Safe' by FDA. Limosilactobacillus fermentum plays an important role in modulation of the intestinal microbiota, enhancing the host immune system and improving feed digestibility. We isolated a probiotic candidate that was identified and named Limosilactobacillus fermentum JNU532. In a previous report, cell-free culture of L. fermentum JNU532 exhibited anti-melanogenic and antioxidant activities. In this study, we present the complete genome assembly of the bacterial strain JNU532. The final genome consists of one circular chromosome (2,077,416 base pairs) with a guanine + cytosine (GC) ratio of 51.5%.

Pan-Genome Analysis Reveals Origin Specific Genome Expansion in Enterococcus mundtii Strains

  • Pan-genome analysis is used to interpret genome heterogeneity and diversification of bacterial species. Here, we present pan-genome analysis of 22 strains of Enterococcus mundtii. The GenBank file of E. mundtii strains that have been isolated from different sources i.e., human fecal matter, soil, leaf, dairy products, and insects was downloaded from National Center for Biotechnology Information (NCBI) database and analyzed using BPGA-1.3.0 (Bacterial Pan Genome Analysis) pipeline. Out of a total, 4503 gene families, 1843 belongs to the core genes whereas 1,762 gene families represent the accessory genes and 898 gene families depict the unique genes among all the selected genomes. Majority of the core genes belongs to the categories of Metabolism (37.83%) and Information storage & processing (29.84%) whereas unique genes belongs to the category of Information storage & processing (48.08%). Further, accessory genes are almost equally present in both functional categories i.e. Information storage & processing and Metabolism (34.34% and 32.27% respectively). Further, subset analysis on the basis of the origin of isolates exhibits presence and absence of exclusive gene families. The observation suggests that even closely related strains of a species show extensive disparity in genome owing to their ability to adapt to a specific environment.

Application of crude enzymes obtained from Pyrus pyrifolia cv. Shingo on milk proteins

  • This study investigated the activity of crude enzymes obtained from Pyrus pyrifolia cv. Shingo on milk proteins. In the milk processing industry, there is an increasing interest in the addition of functional materials to dairy products or functional peptides isolated from milk proteins. First, Pyrus pyrifolia cv. Shingo was separated into core, flesh, and peel regions, and crude enzymes were obtained from the individual regions. The activity of the obtained crude enzymes was measured using casein and gelatin agar. The crude enzyme obtained from the flesh of Pyrus pyrifolia cv. Shingo decomposed gelatin, but the activity of the crude enzymes obtained from the peel and core regions was insignificant. On the other hand, the crude enzymes obtained from the flesh and core regions of Pyrus pyrifolia cv. Shingo had a remarkable enzymatic activity in casein agar. However, the activity of the crude enzyme obtained from the peel region was insignificant. In addition, the crude enzymes obtained from the individual regions were mixed with casein to induce reactions, and the degradation patterns were investigated through electrophoresis and high performance liquid chromatography (HPLC). According to the results, the crude enzymes from Pyrus pyrifolia cv. Shingo degraded milk proteins. Thus, the results of this study can be used in studies on functionality. Additionally, it is expected that the use of pear peels and cores in the milk processing industry would greatly contribute to the reduction of food waste.
