Screening of Bioactive Compounds from Edible Mushroom and Production of Anti-osteoporosis Osteoclast Differentiation Inhibitor (몇 가지 주요 식용버섯의 생리기능성 물질 탐색과 파골세포 분화 저해물질의 생산)

  • For development of new bioactive compounds from main edible mushrooms, we determined some physiological functionalities of water extracts from mushrooms. Among water extracts from some mushroom fruiting bodies, water extracts from Pleurotus ostreatus showed 73.2% of anti-hypertensive angiotensin I-converting enzyme inhibitory activity and 73.3% of anti-gout xanthine oxidase inhibitory activity, respectively. Fibrinolytic activity was also showed 21.5 mm of clear zone in water extract of Lyophyllum cinerascens. However, the other physiological functionalities were very weaked except 40.3% of antioxidant activity in Lentinus lepideus. Furthermore, the water extracts of Pleurotus eryngii and Lyophyllum cinerascens showed high anti-osteoporosis osteoclast differentiation inhibitory activity. However, the water extract from Lentinus lepideus and Pleurotus ostreatus were not detected any osteoclast differentiation inhibitory activity.

Occurrence of Rhizina Root Rot in a Black Pine (Pinus thunbergii) Forest Located at the Western Coastal Area in Korea and Its Spreading Patterns (서해안 곰솔림에서의 리지나뿌리썩음병 발생 및 확산 유형)

  • At the survey of June, 2002, total 294 dead frees were found in 20 ha of Black pine (Pinus thunbergii) forest located in the western coastal region of Korean peninsula. The dead trees were scattered over the 27 place as groups of about ten trees each. As a result of the field survey on the relationship between the conditions of dead trees and the occurrence of fruiting bodies of Rhizina undulata around the dead and/or dying trees, from June 2002 to August 2004 in the four plots, the occurrence of infected trees was observed as a shape of an irregular concentric circle from the first infected tree and R. undulata was found mainly around the dead tree. Because there was no observation of any other insects and pathogens which would kill trees, the cause of tree death in groups was considered owing to R. undulata. From the analysis of the physical and chemical proper ties of the soil collected from the damaged areas, the pH of soil was between 4.6 and 5.8 and the contents of soil nutrients were very low. Any "fire" trace was not found at all the 27 damaged places in the area, Taean, Chungcheongnam-Do, which are generally known as an important factor to initiate development of the disease. Therefore, further examination is needed to verify precisely about other environmental factors related with the group dying of the Black pines in this area beside 'fire'.

  • The fruiting bodies of Ganoderma lucidum extracted by water and ethanol extraction. we are analyzed antioxidant effects and cancer cell growth inhibition rate. ASI 7004, 7014 was higher antioxidant effects than Trolox, BHA as control. Generally, the rest of strains was higher antioxidant effects than ABTs as control. Water extracts and ethanol extracts was treated to the Liver cancer cell(HepG2) and stomach cancer cell(AGS). Inhibition activities of Liver cancer cell(HepG2) is a high in D.W. extracts of ASI 7002, 7011, 7014, 7020. Inhibition activities of Liver cancer cell(HepG2) is a high in EtoH extracts of ASI 7011, 7019. Inhibition activities of Stomach cancer cell(AGS) is a high in D.W. extracts of ASI 7001, 7002, 7019, 7020. Inhibition activities of Stomach cancer cell(AGS) is a high in EtoH extracts of ASI 7001, 7002.

Influence of Temperature on the Bacterial Community in Substrate and Extracellular Enzyme Activity of Auricularia cornea

  • Temperature is an important environmental factor that can greatly influence the cultivation of Auricularia cornea. In this study, lignin peroxidase, laccase, manganese peroxidase, and cellulose in A. cornea fruiting bodies were tested under five different temperatures ($20^{\circ}C$, $25^{\circ}C$, $30^{\circ}C$, $35^{\circ}C$, and $40^{\circ}C$) in three different culture periods (10 days, 20 days and 30 days). In addition, the V4 region of bacterial 16S rRNA genes in the substrate of A. cornea cultivated for 30 days at different temperatures were sequenced using next-generation sequencing technology to explore the structure and diversity of bacterial communities in the substrate. Temperature and culture days had a significant effect on the activities of the four enzymes, and changes in activity were not synchronized with changes in temperature and culture days. Overall, we obtained 487,694 sequences from 15 samples and assigned them to 16 bacterial phyla. Bacterial community composition and structure in the substrate changed when the temperature was above $35^{\circ}C$. The relative abundances of some bacteria were significantly affected by temperature. A total of 35 genera at five temperatures in the substrate were correlated, and 41 functional pathways were predicted in the study. Bacterial genes associated with the membrane transport pathway had the highest average abundance (16.16%), and this increased at $35^{\circ}C$ and $40^{\circ}C$. Generally, different temperatures had impacts on the physiological activity of A. cornea and the bacterial community in the substrate; therefore, the data presented herein should facilitate cultivation of A. cornea.

Biosynthesis of Organic Germanium Using Cordyceps militaris (동충하초를 이용한 유기게르마늄의 생산)

  • In the present study, an attempt has been made to produce a high quality medicinal mushroom Cordycops militaris supplemented with organic Ge. Cordycops militaris was cultivated in SDAY liquid medium containing yeast extract 10 g, peptone 10 g, glucose 40 g per liter and chrysalis media supplemented with inorganic Ge at 100, 500, 1000, and 5000 ppm concentrations. The greatest organic Ge production of 442.4 ppm/g and 284 ppm/g were observed in SDAY liquid and Chrysalis media cultures supplemented with 100 ppm inorganic Ge respectively. Similarly, 4,509.7 ppm/g and 1,058 ppm/g of organic Ge were obtained from liquid and chrysalis media cultures supplemented with 5,000 ppm and 1,000 ppm inorganic Ge, respectively. In addition, higher concentration of organic Ge was obtained in mycelia than fruiting bodies. These results indicate that the concentration of organic Ge increase with decreasing inorganic Ge concentration in the medium. This is the first report on production of high valuable Cordyceps militaris contained with organic Ge.

Occurrence and Identification of the Fungus Causing Yellow Rot on Ganoderma lucidum (불로초버섯 노랑썩음병의 발생과 병원균의 분류 동정)

  • Since 1990, destructive yellow rots on Ganoderma lucidum caused by a soilborne fungus have been occurred in major cultivation areas of Korea. Incidences of the disease were 61% in Chulwon and 94% in Kanghwa area where the mushroom has been cultivated for 10 years, whereas the disease has not been found yet in new cultivation areas such as Moonkyung and Hongsung. when severely infected, inner tissues of bed-logs showed severe yellow and fruiting bodies of the mushroom was not produced. Infected tissues of bed-logs were readily distinguished from those of healthy ones by a distinctive brown border line. When the disease progressed, mycelia of Ganoderma lucidum were totally destroyed, and abundant ascocarps of the pathogen were formed inside the tissues of bed-logs showing yellowish green. The fungus derived from a single ascospore strongly lysed mycelia of Ganoderma lucidum growing on bottle media, and non-volatile components secreted by the pathogen were also highly inhibitory to mycelial growth of the mushroom fungus. The pathogen was identified as Arthrographis cuboidea based on its distinctive cultural and morphological characters. The fungus produced arthroconidia and unbrached conidiophores. The width of fungal conidia was distinctively wide as compared with the length. Colonies of the fungi were pale yellow to yellowish green on agar media. As a causal pathogen of yellow rot of Ganoderma lucidum., this fungus has not been reported yet in Korea.

Interorder Hybridization between Pleurotus ostreatus and Elfvingia applanata by Protoplast Fusion (원형질체(原形質體) 융합(融合)에 의한 느타리버섯과 잔나비걸상버섯의 이목간(異目間) 교잡(交雜))

  • Interorder somatic hybrids were obtained by protoplast fusion between Pleurotus ostreatus in the order Agaricales and Elfvingia applanata in the order Aphyllophorales. The fusants were classified into stable heterokaryons and spontaneously segregated heterokaryons. Hyphae of all fusion products except two strains did not form clamp connections. Out of them, two clamped and three clampless fusants produced mature fruiting bodies by light-dark cycle on sawdust rice bran medium. All of these basidiocarps had clamp connections. Three fusants were analysed with the distribution of progenies and segregation of genetic characters by random spore analyses. The genetic markers were shown to segregate and recombine in the first generation of monospores isolated from basidiocarps. Phenotypes of a large number of auxotrophic progenies were not detected in the two clamped fusants. The aberration ratio of segregants indicated the gene interaction resulting from different genome structure between distantly related species. The polymerase chain reaction (PCR) was adopted for the detection of somatic hybrids nuclear DNA. Four fusants showed a positive results in three kinds of primers. The prominent reaction products are represented by new bands in primer # 87 and # 125. Out of four fusants, two somatic hybrids had non-parental mtDNA patterns when digested with EcoR1 and HindIII. Comparison of somatic hybrids, tissue culture isolates(TC) and multispore germination isolates(MS) were made using esterase isozyme analysis. It is apparent that somatic hybrids had a minor banding patterns which are quite different from those of parents.

Optimization for Effective Bioproduction of Natural (-)-1-Octen-3-ol by Lipoxygenase and Hydroperoxide Lyase from Agaricus bisporus (Agaricus bisporus의 Lipoxygenase와 Hydroperoxide Lyase를 이용한 천연 (-)-1-Octen-3-o1 생산 공정의 최적화)

  • One of the most important volatile aroma compounds responsible for mushroom flavor is 1-octen-3-ol. To meet the demand for natural mushroom flavor, a study was needed for the production of natural chiral specific (-)-1-octen-3-ol that has higher flavor intensity than synthetic chiral mixtures of (+), and (-)-1-octen-3-ol. The biosynthesis of (-)-1-octen-3-ol was achieved by an aerobic oxidation using lipoxygenase (LOX) and hydroperoxide lyase (HPOL) isolated from commercially available mushrooms in Korean market. Safflower oil from Uiseong, Gyeongsangbuk-do, that contains $\geq75\%$ of linoleic acid, was hydrolyzed using lipase. The recovered linoleic acid was biotransformed to stereo-specific 10-hydroperoxy linoleic acid by LOX. 10- hydroperoxy linoleic acid was further cleaved to (-)-1-octen-3-ol by HPOL. A commercial bioprocess for the production of (-)-1-octen-3-ol was developed using a 5-liter jar fermenter with fruiting bodies of Agaricus bisporus harvested from Buyeo, Chungcheongnam-do. The maximum production of (-)-1-octen-3-o1 was achieved at $4^{\circ}C$, pH 6.5 and 800 rpm yielding 748 mg/kg of mushroom.

Ectomycorrhizal Mushroom Occurrence around the Fairy Ring of Tricholoma matsutake at a Pine-Mushroom Forest (송이 발생림의 송이 균환 주변에 출현하는 외생균근성 균류)

  • The occurrences of ectomycorrhizal fungi were investigated in a pine-mushroom forest at Hongcheon, Korea. The fairy rings of Tricholoma matsutake were figured by the sporocarp places of T. matsutake for three years $(1995{\sim}1997)$, and the occurrences of other ectomycorrhizal fungi were surveyed with x and y dimensions for two years $(1996{\sim}1997)$. The diameters of fairy rings of T. matsutake ranged from 2m to 10m, which indicated that the age of the fairy rings as $10{\sim}50$ years when we consider that the growth of the fairy ring used to show about 10 cm per year. Russula bella, R. sororia, R. delica and Cantharellus minor were the major species occurred on the site during the survey period, and each species occupied 16.0%, 12.8%, 12.4% and 7.0% of total mushroom occurrence, respectively. From the results, we could conclude that the surveyed stand was a productivity-declining forest from the view point of pine-mushroom production. In addition, Amanita pantherina, Suillus bovinus, Ramaria flaccida and Laccaria amethystea were considered to be the indicator species for declining of pine-mushroom productivity since some fruiting bodies of the species appeared around the fairy ring of T. matsutake.

Breeding and cultural characteristics of a newly bred Lentinula edodes strain, 'Bambithyang' (표고 신품종 '밤빛향'의 육성 및 특성)

  • A new cultivar, 'Bambithyang', was bred from monokaryotic strains of Sanbaekhyang and SANJO 707ho using the mono-mono hybridization method. Its incubation period was determined to be 100 days and its optimum temperature for fruit body flushing, 11-20℃. The mushrooms had a convex cap (diameter, 69.6 mm; thickness, 15.2 mm) and stipes in their center. The color of their upper sides was chestnut brown; their gills were arranged in a ripple style and had medium density. Their scales were cream-colored and fully distributed; the stipes were cream-colored and cylindrical (thick upper part). They also had cream colored fluffs and sporadic fruiting bodies. Zone lines were present when they were cultivated with mother and father varieties. Their stipe lengths and caps were 9% shorter and 16.9% thicker, respectively, than those of Sanbaekhyang.
