Growth and Fermentation Characteristics of Saccharomyces cerevisiae NK28 Isolated from Kiwi Fruit

  • The influences of glucose concentration, initial medium acidity (pH), and temperature on the growth and ethanol production of Saccharomyces cerevisiae NK28, which was isolated from kiwi fruit, were examined in shake flask cultures. The optimal glucose concentration, initial medium pH, and temperature for ethanol production were 200 g/l, pH 6.0, and $35^{\circ}C$, respectively. Under this growth condition, S. cerevisiae NK28 produced $98.9{\pm}5.67$ g/l ethanol in 24 h with a volumetric ethanol production rate of $4.12{\pm}0.24g/l{\cdot}h$. S. cerevisiae NK28 was more tolerant to heat and ethanol than laboratory strain S. cerevisiae BY4742, and its tolerance to ethanol and fermentation inhibitors was comparable to that of an ethanologen, S. cerevisiae D5A.

Effect of Citrus macroptera Fruit Pulp Juice on Alteration of Caspase Pathway Rendering Anti-Proliferative Activity against Ehrlich's Ascites Carcinoma in Mice

  • Citrus macroptera (Rutaceae) has long been used in folk medicine in Bangladesh. Considering the folkloric context, this study was aimed to scrutinize anti-proliferative activity of C. macroptera fruit pulp juice (CMFPJ) against Ehrlich's ascites carcinoma (EAC). The anti-proliferative capacity of CMFPJ was investigated and confirmed primarily using MTT assay. In vivo anti-proliferative aptitude of CMFPJ was investigated with 25, 50, and 100 mg/kg/day intraperitoneal (i.p.) treatment. Anti-proliferative efficacy of CMFPJ was assessed based on EAC growth inhibition. CMFPJ inhibited EAC growth in vitro in a dose-dependent manner. And the percentages of in vivo EAC growth inhibition were 19.53, 49.2, and 68.9% at 25, 50, and 100 mg/kg CMFPJ respectively. CMFPJ significantly induced expression of apoptosis regulatory genes caspase-8, caspase-9, cytochrome-c, and caspase-3. This considerable anti-cancer activity was perhaps due to combinatorial effect of lectin, polyphenols, and flavonoids present in CMFPJ.

Comparisons of Growth and Fruit Quality of Citrullus lanatus cv. Mudeungsan and Citrullus vulgaris cv. Dalgona Grown in Fertigation and Soilless Culture (무등산수박과 달고나수박의 관비재배와 양액재배에 있어서 생육 및 과실품질의 비교)

  • This experiment was conducted to compare with the growth and fruit quality of Citrullus lanatus cv. Mudeungsan and Citrullus vulgaris cv. Dalgona grown in fertigation and soilless culture. Seeds were sown on April 16 and transplanted on April 24, 1998. Cultural systems used in fertigation and soilless culture beds using mixed substrate, coir dust (50%) and perlite (50%). In the plot of fertigation system, the number of leaves of cv. Dalgona were increased more than that of cv Mudeungsan, but leaf area shown inversed trend. Plant growth shown greater in soilless culture than those of fertigation culture. In regardless of cultural systems, soluble solid content in fruit was higher in the cv. Dalgona than that of cv. Mudeungsan, but fruit fresh weight was greater in cv. Mudeungsan compare to the cv Dalgona. Nitrate content in petiole sap of watermelon in regardless of cultivars and cultural systems was 11.4∼13.4mg/gFW on 15 days after transplanting, and then increased to 17.1∼20.6mg/gFW on the fruit growth stage. Phosphorous content was 3.7∼5.7mg/gFW in the early growth stage while decreased to 0.6∼1.1mg/gFW from maturing stage to harvesting stage. Potassium content was increased to 5.8∼6.6mg/gFW in the early growth stage while decreased to 4.0∼4.8mg/gFW from pollination stage to harvesting stage. Calcium content in spa petiole of watermelon was higher in soilless culture as 3.4∼4.1mg/gFW than 2.5∼3.5mg/gFW of fertigation culture, but calcium content in fertigation culture during maturing stage was higher than that of soilless culture. The tendency of magnesium uptake was higher in fertigation culture than that of soilless culture, and was similarly absorbed in the range of 0.9∼1.3mg/gFW in regardless of cultural method after pollination. It was demonstrated that cv. Mudeungsan can be adapted to soilless culture and improved the fruit quality. Consequently, hydroponic possibility for year round culture in the greenhouse was recognized.

Effect of Artificial Pollination, Pollination Time, and Pollen Bulking Agent on Seed Formation and Fruit Quality in the Shelter Greenhouse Cultivation of Kiwifruit (비가림 시설재배에서 인공수분, 수분시기 및 화분증량제가 참다래의 종자형성과 과실특성에 미치는 영향)

  • This study was carried out to determine the effect of the artificial pollination on fruit set, seed formation and fruit quality in kiwifruit and to confirm the effective pollination time for optimal fruit set in the shelter greenhouse cultivation. Also, the effect of artificial pollination practice by the pollen diluent solution was tested. By artificial pollination, fruit set, seed formation, and fruit quality was improved both in the open field condition and in the shelter greenhouse cultivation. In order to find the effective pollination period (EPP), artificial pollination was carried out every day until 7 days after full bloom. Number of seeds and fruit set rate and fruit growth of kiwifruit were no significantly different until 4 days after full bloom (DAFB), but, those of kiwifruit from 5 DAFB significantly decreased. Consequently, the effective pollination period of kiwifruit in the shelter greenhouse was 4 days after full bloom. Also, artificial pollination by the pollen diluent solution was showed the similar fruit set and fruit quality compare to that by lycopodium powder. Accordingly, artificial pollination by the pollen diluent solution should be very effective practice for labor saving in kiwifruit cultivation.

Development of Fruit Structure in 'Fuji' Apples (사과 '후지' 과실의 조직 발달)

  • The fruit structure of 'Fuji' apples from full bloom to maturing was observed from 1997 to 1998. Cell division period of the fruit was found to be 4 to 5 weeks after full bloom. Vascular bundles in the inner part of the fruit skin which were not described in the books illustrating apple fruit structure was observed, as they were tentatively named as outer vascular bundles (OVB), and another vascular bundles were also observed newly in periphery of locules, as they were tentatively named as inner vascular bundles (IVB). In the observation of the inner epidermis (IE) in the inner part of the locules on 2 days prior to full bloom, the guard cells were observed and these were disappeared in the observation made 2 days later, i.e. on full bloom. The formation of fruit skin was observed at the microscope 65 days after full bloom and the number of cell which organized the fruit skin did not change from this time to maturation period. Tannin which is mainly in the fruit skin changing from continuously during fruit growth, specially the tannin of epidermis disappeared completely 100 days after full bloom stage, and then constituted again. Starch was not almost found out in cell division period of fruit from full bloom stage after this time it constituted much at flesh part and decreased at maturation period. Epidermis was developed by uniform cells of a layer the cell of epidermis constituted irregularity after pigmentation stage, the organization of fruit was not close because the very big intercellular space constituted at hypodermis and flesh structure.

Variation of Plant and Fruit Characteristics in Capsicum frutescens.

  • Among the 6500 accessions of the pepper germplasm which conserved at RDA Genebank, 319 accessions were investigated charateristics such as their growth habit, flowering and fruit shape in Capsicum frutescens. All accessions were characterized for their 14 qualitative and 16 quantitative characters. Results revealed that both qualitative and quantitative characters exhibited wide variation among the studied germplasm. The hypocotyl color, which investigated in juvenile stage, was divided in three types such as green, purple, and purple with green, and most of the accessions showed in purple. Flowering starts from 7 May, and most accessions were starting to flower from 27 May to 25 June. The fresh color of the mature pepper were red, yellow, orange, Etc. Fruit shape were mostly elongated, while exhibited ball shape and round shape were a few. The fresh weight of fruits ranged from 0.2 to 42.3, and the diameter of fruit showed from 4.2 to 45.1 mm, respectively. The phenotypic diversity exists in Capsicum frutescens. provide valuable information to improve agronomic traits in pepper breeding program.

Amelioration of non-irrigated stress and improvement of sweet pumpkin fruit quality by Kushneria konosiri endophytic bacteria

  • This study examined the impact of two bacterial strains, H05E-12 and H05R-04, on alleviating non-irrigation-induced stress and its subsequent effects on the fruit productivity of sweet pumpkin plants. When subjected to non-irrigation-induced stress, the lipid peroxidation, proline, total phenol, and total soluble sugar content significantly decreased in plants treated with either H05E-12 or H05R-04 compared to the control. In a greenhouse experiment under non-irrigated conditions, H05E-12-treated plants exhibited higher stomatal conductance than the control, although there was no significant change in the soil plant analysis development(SPAD) value due to treatment. Upon re-watering, an increase in fruit diameter was observed in H05E-12-treated plants, and the L-ascorbic acid content in the fruit also showed a significant increase compared to the control. The H05E-12 strain was identified as Kushneria konosiri. To the best of our knowledge, this is the first report detailing the beneficial effects of K. konosiri on the alleviation of non-irrigation-induced stress and the promotion of plant growth in sweet pumpkin plants.

Effects of Incorporation of Green Manure Crops on the Growth of Watermelonand Soil Nitrate Nitrogen Concentration (풋거름작물의 토양환원이 수박의 생육 및 토양의 질산염 농도에 미치는 영향)

  • BACKGROUND: In this study, we evaluated the effects of soil incorporation of hairy vetch (HV) or ryeas a green manure on the growth and yields of watermelon and soil nitrate nitrogen in a green house.. METHODS AND RESULTS: HV and rye were cultivated for 151 days after sowing on October 30th and incorporated into soil before transplanting watermelon. The amount of N added by soil incorporation of HV and rye were 79 kg/ha and 88 kg/ha, respectively. Five different N treatments for each of HV and rye were included as follows: green manure, green manure with urea at 25%, 50%or 75%, and 100% ureafor the N recommendation rate. The growth and fruit yield of watermelon were not different among the treatments of both HV and rye. Soil nitrate N content at both HV and rye treatments decreased continuously with the lapse of days after planting (DAP) and was lowest at 75 DAP: 44 mg/kg and 52 mg/kg the for the HV and rye treatment without urea, respectively. CONCLUSION: These results indicate that the N mineralized from the soil incorporated HV or rye accounts for an important portion of N available for the growth and fruit yield of watermelon. It can be suggested that the green manures, comparable to ureacould ensure the yield of watermelon, if soil nitrate N content isabove 40 mg/kg by soil incorporation of HV and rye during watermelon cultivation. However, further studies on the relationship between soil nitrate N content during cultivation periods and the fruit yield of watermelon are required.

The Effect of Treatment of Woody Charred Materials on the Growth and Components of Tomato and Chinese Cabbage (목질탄화물의 처리가 토마토와 배추의 생육 및 체내성분에 미치는 영향)

  • A research was conducted to investigate the effect of treatment of woody charred materials such as wood vinegar, charcoal, and mixture of wood vinegar and charcoal on the growth and components of tomato and chinese cabbage. The effect of treatment of woody charred materials on the growth and components of tomato the research findings are as follows. The total number of soil microorganisms increased in the groups treated with woody charred materials compared with the control. The fruit number, fruit length, fruit diameter, fruit weight, hardness and sugar contents of tomato did not show significant difference in all plots treated with woody charred materials, but the plot treated with wood vinegar + charcoal showed relatively higher values in general. Vitamin C contents were shown higher in the plots treated with woody charred materials than the control, and among the treatment plots. The effect of treatment of woody charred materials on the growth and components of chinese cabbage the research findings are as follows. The soil chemical components did not change before and after the treatment of woody charred materials. The total number of soil microorganisms increased in the treatment plots compared with the control, The leaf length, leaf width, inner leaf number, plant weight, and head weight of Chinese cabbage treated with woody charred materials were in general larger than the control. Among the treatment plots, the plot treated with wood vinegar + charcoal showed slightly higher values. The contents of minerals in chinese cabbage such as Ca, Fe, K, Mg and Mn were higher in the treatment plots than the control.

Effects of the Dietary Supplementation of Fermented Cactus Fruit (Opuntia ficus-indica) Fluid on the Growth of Red Sea Bream, Pagrus major (손바닥선인장 열매 발효액 첨가사료가 참돔의 성장에 미치는 영향)

  • Two feeding experiments were conducted to investigate the effects of fermented cactus fruit (Opuntia ficusindica) fluid (FCFF) as a feed additive to a commercial diet on the growth of red sea bream, Pagrus major, and to determine an effective dose. FCFF was prepared by mixing crushed cactus fruit with a starch solution and commercially available microorganisms for 2 weeks at room temperature. Three triplicate groups of red sea bream had initial mean weights of 84.1 g (Exp-1) and 5.1 g (Exp-2) and were fed experimental diets containing 0%, 1%, and 5% FCFF for 2 months (Exp-1) and 0%, 0.2%, 0.5%, and 1% FCFF for 3 months (Exp-2), respectively. In experiment 1, the mean body weight of fish fed the diet containing 1% FCFF was significantly higher (P<0.05) than that of fish fed the diet without FCFF. The survival rate was highest in fish fed the diet containing 1% FCFF, although the difference was not statistically significant. The feed gain ratio (FGR), specific growth rate (SGR), and condition factor (CF) values of fish fed the diet containing 1% FCFF were higher than those of fish in the other dietary groups that received lower levels of FCFF. The daily feeding rate (DFR) of fish fed the diet containing 1% FCFF was slightly lower but not statistically different than the DFR values of fish in the other dietary groups. In experiment 2, the final mean body weight of fish fed the diet containing 1% FCFF was significantly (P<0.05) higher than the mean weight of fish in the control group. The FGR, SGR, and CF values of fish fed the diet containing 1% FCFF were better than the values from fish in the other dietary groups that received lower levels of FCFF, although the differences were not statistically significant. However, the DFR of fish fed the diet containing 1% FCFF was lower than those of fish in the other groups. These results suggest that FCFF could be used as a feed additive in commercial fish food and a preferable level of supplementation is at least 1.0% in fingerling and young red sea bream.
