신선편의 샐러드와 유기농 채소류의 미생물학적 품질 및 식중독 미생물 오염도 (Microbiological Quality of Fresh-Cut Produce and Organic Vegetables)

  • 본 연구는 대형 할인매장과 백화점에서 유통되고 있는 신선편의 샐러드류와 유기농 채소의 미생물학적 품질을 평가하고, 식중독 미생물 오염도를 분석하여 위생적인 신선편의 제품의 생산을 유도하고, 신선편의 샐러드류와 유기농 채소류의 미생물학적 위해평가의 기본자료로 활용하고자 수행되었다. 경기도 지역의 대형 할인매장과 백화점에서 유통되고 있는 신선편의 샐러드류와 유기농 채소류를 구입하여 총균수과 대장균군, 장내세균, 대장균을 분석하여 미생물학적 품질을 평가하였으며, S. aureus, Salmonella spp., V. parahaemolyticus, L. monocytogenes, B. cereus 등의 식중독관련 미생물의 오염도를 분석하였다. 신선편의 샐러드 중 미생물학적으로 안전한 제품은 과일샐러드 제품으로 일반세균수, 장내세균수와 대장균군의 오염 수준이 낮았다. 하지만, 새싹채소는 일반세균수, 장내세균, 대장균군 등의 오염수준이 다른 신선편의 제품보다 높았으며, 분석 시료의 37.5%에서 B. cereus가 3 log CFU/g 이상 오염된 것으로 분석되었다. 모든 신선편의 제품에서 Salmonella spp., V. parahaemolyticus, L. monocytogenes 등의 병원성 미생물은 검출되지 않았으며, S. aureus는 2개의 샐러드 제품에서 검출되었으나, 오염 수준은 기준 이하였다. 유기농 채소류 중에서는 유기농 상추가 일반세균수, 장내세균, 대장균군의 오염도가 유기농 상추와 깻잎에 비하여 높았으며, 대장균과 S. aureus는 상추와 깻잎에서만 검출되었고, 풋고추에서는 검출되지 않았다. B. cereus는 분석된 모든 유기농 채소류에서 검출되었고, 오염도는 1.2-4.0 log CFU/g 수준이었다. 병원성 미생물인 L. monocytogenes, Salmonella spp., V. parahaemolyticus는 모든 시료에서 검출되지 않았다. 유기농 채소류는 구입 후 대부분 가정에서 간단한 세척 후 바로 섭취하는 것으로, 병원성 미생물에 대한 오염도는 낮았으나, 유기농 상추의 일반세균수가 5 log CFU/g 이상으로 충분한 세척으로 균수를 낮추지 않는다면 미생물학적으로 안전한 수준은 아니었다. 따라서 섭취적 충분한 세척으로 초기 오염도를 낮추고, 저온에서 보관하며, 가능한 빠른 섭취가 필요하다고 판단되었다.

신선 채소류의 부패에 따른 세균의 다양성 변화 및 세균에 의한 채소 부패 조사 (Changes of Bacterial Diversity Depend on the Spoilage of Fresh Vegetables)

  • 채소류가 소비되기까지 약 10~30%에 이르는 양이 부패에 의해 버려지고 있다. 채소류는 수확 후 수많은 세균이나 곰팡이와 같은 다양한 미생물들에 의해 부패가 이루어진다. 채소류의 부패와 세균과의 관계를 알아보고자 세균의 다양성에 관한 조사를 하였다. 신선한 상추와 깻잎, 치커리에서의 총 호기성 균 수는 각각 $2.6{\sim}2.7{\times}10^6$, $4.6{\times}10^5$, $1.2{\times}10^6\;CFU/g$ of fresh weight이었으며, Pseudomonas spp., Alysiella spp., Burkholderia spp.와 그 외의 18개의 다양한 속들이 확인되었다. 신선한 채소류를 $28^{\circ}C$에서 일주일 동안 배양하였을 때 세균 다양성에 변화가 생겼다. 총 호기성 균의 수는 상추와 깻잎, 치커리에 대하여 각각 $1.1{\sim}4.6{\times}10^8$, $4.9{\times}10^7$, $7.6{\times}10^8\;CFU/g$ of fresh weight로 나타났으며, 이는 약 $10^2$배 정도 증가한 수치이다. 하지만 세균 다양성은 단순해져서 보다 적은 수의 세균이 분리, 동정되었다. Pseudomonas spp.가 대부분을 차지하였으며(~48%), Arthrobacter sp., Bacillus sp.가 그 뒤를 이었다. 동정된 세균 각각의 부패능을 검정하고자 무균배양한 상추에 접종하였으며, 그 결과 Pseudomonas fluorescence와 Pantoea agglomerans가 상당한 부패를 야기하였다.

절단방법이 신선절단 사과와 엽채류의 품질에 미치는 영향 (Effects of Cutting Methods on Qualities of Fresh-Cut Apples and Leafy Vegetables)

  • 신선절단 사과와 엽채류의 품질에 절단방법이 미치는 영향을 구명하기 위하여, 사과는 sharp knife나 dull knife로 절단하고, 양배추와 결구상추는 hand나 knife cutting 및 knife로 샐러드용 크기나 채썰기로 절단하고 각 절단품의 품질특성을 비교 조사하였다. 신선절단 사과의 갈변도와 과육경도는 dull knife보다 sharp knife로 절단한 경우에 유의적으로 낮고 높게 유지되었다. 반면에 신선절단 사과의 가용성 고형분 함량과 pH는 절단용 칼날의 날카로움에 따른 유의적인 차이를 보이지 않았다. 신선절단 양배추와 결구상추의 갈변도는 hand cutting보다 knife cutting에서, 샐러드용 크기보다 세절인 채썰기한 경우에서 유의적으로 낮은 수준을 나타내었다. 이로써, 신선절단품 제조시 조직손상을 크게 하는 절단방법이 사과의 갈변과 연화를, 양배추와 결구상추의 갈변을 촉진시키는 것으로 확인되었고, 고품질 제품을 위해 조직손상을 최소화할 수 있는 기술이 적용되어야 할 것으로 판단된다.

학교급식 식재료로서 지역 농산물, 고품질농산물과 신선편이 채소의 이용현황과 필요에 관한 연구 - 경기지역 초.중.고등학교를 중심으로 - (Food Materials for School Foodservices on High-quality Products, Local Products and Fresh-cut Products - Dietitian's Choice in Elementary, Middle and High Schools in Gyeong-gi Province -)

  • The use of local product foods and fresh-cut products, the dietitian's recognition, the required items and the quality for school foodservice was investigated to increase the consumption of high-quality agricultural products. Data from 578 elementary schools, 228 middle schools and 116 high schools were collected by school foodservice personnels in each regional Education Administration in Gyeong-gi province. The first criteria of buying food materials was reliability/safety for all school foodservice. The second criteria were the origin of the product and freshness/ripeness for elementary and middle schools and high school, respectively. The most important consideration for selecting a supplier was the quality of the material. The appearance/status of the food was the first factor for buying food materials. The origin and the seasonality were very important for buying farm products. Dietitians in Gyeong-gi province recognized the local products, and used them weekly. The advantages of using local products were the quick supply, freshness and safety. On the other hand, the disadvantages were the lack of information and the uncertainty of supply. Sixty eight percent of the subjects had experienced the high-quality agricultural products in school meals, but the frequency was low. The main reasons for not servicing high-quality agricultural products were the high cost and the lack of trust on the quality. Among fresh-cut products, seasonings and root vegetables were purchased most frequently, but leaf vegetables was 5.7% only. The purchase of fresh-cut products was not frequent, but usually satisfied. The advantages of fresh-cut products were the reduced workload and waste, but the reason for not using them were poor hygiene and the high price. The diverse supply system and the promotion of the local product foods would be necessary since the most food materials were purchased through suppliers. In addition, the problems related to the high cost and distrust should be resolved to expand the consumption of the high-quality agricultural products and the fresh-cut vegetables. The institutions for certifying the quality should be needed to increase the confidence of these products.

  • Vegetables and fruits purchased from several markets in Seoul from July to October in 1977 were analyzed to know the level of nitrate-and nitrite-nitrogen accumulation in relation to a public health. Radishes and chinese cabbages utilized mainly as pickled vegetables in Korea resulted in the highest concentration of nitrate-nitrogen. Some of the levels for radishes and chinese cabbages were notably high and exceeded a recommended upper limit of 300 ppm $No_{3}-N$, and thus these levels would render these samples unsafe for use. The levels in some of vegetables other than radish and chinese cabbage, e.g., spinach, lettuce, green onion, cabbage were relatively high and considered to be unsafe for use in feeding infants, where as those of green pepper, bean sprouts and parsely were very low and safe. And also the levells in fruits were very low and safe. Nitrite-nitrogen contents in all tested vegetables and fruits ranged to trace and appeared not to be accumulated in fresh vegetables and fruits. Stems and roots of radishes and chinese cabbages accumulated approximately 2 fold more nitrate-nitrogen than leaves in 5 samples of each vegetable tested.

천연 항균제처리를 병용한 과채류의 자연 저온저장기술 개발에 관한 연구 (Inhibitory Effects of Natural Antimicrobial Agenton Postharvest Decay in Fruits and Vegetables under Natural Low Temperature)

  • In order to prevent the postharvest decay and to promote the freshness retention of fruits and vegetables grapefruit seed extract(GFSE), natural microorganism control agent, was applied to the preservation of fresh fruits and vegetables. Freshfruits and vegetables treated with GFSE and stored in polyethylene film (0.1mm) at 1$0^{\circ}C$-15$^{\circ}C$ of natural low temperature low kept better qualities in color and texture than the GFSE -not- treated control. The treatment using GFSE ina 250ppm to 500ppm concentration seemed to be an effective one for the control of Botrytis cinerea isolated in red wine grapes. After 4 weeks of storage the firmness rate of cucumbers treated with the dilute GFSE was four times higher than that of non-treated ones. GFSE showed effective inhibitory action towards plant pathological bacteria and fungi which were involved in the decay of fruits and vegetables. Minimum inhibitory concentrations of GFSE towards them were in the range of 250ppm to 500ppm .Direct visualization of microbial cells and spores using electron microscopy showed microbial cells and fungal spores the function of which was destroyed by treating with the dilute solutions of GFSE. It was observed that GFSE would reduced disease damages and have bactericide & fungicide properties during the storage of such fruits and vegetables as egg plant, wild edible greens , kumquat, and kiwi fruit.

Metagenomic Approach to Identifying Foodborne Pathogens on Chinese Cabbage

  • Foodborne illness represents a major threat to public health and is frequently attributed to pathogenic microorganisms on fresh produce. Recurrent outbreaks often come from vegetables that are grown close to or within the ground. Therefore, the first step to understanding the public health risk of microorganisms on fresh vegetables is to identify and describe microbial communities. We investigated the phyllospheres on Chinese cabbage (Brassica rapa subsp. pekinensis, N = 54). 16S rRNA gene amplicon sequencing targeting the V5-V6 region of 16S rRNA genes was conducted by employing the Illumina MiSeq system. Sequence quality was assessed, and phylogenetic assessments were performed using the RDP classifier implemented in QIIME with a bootstrap cutoff of 80%. Principal coordinate analysis was performed using a weighted Fast UniFrac matrix. The average number of sequence reads generated per sample was 34,584. At the phylum level, bacterial communities were composed primarily of Proteobacteria and Bacteroidetes. The most abundant genera on Chinese cabbages were Chryseobacterium, Aurantimonadaceae_g, Sphingomonas, and Pseudomonas. Diverse potential pathogens, such as Pantoea, Erwinia, Klebsiella, Yersinia, Bacillus, Staphylococcus, Salmonella, and Clostridium were also detected from the samples. Although further epidemiological studies will be required to determine whether the detected potential pathogens are associated with foodborne illness, our results imply that a metagenomic approach can be used to detect pathogenic bacteria on fresh vegetables.

베트남의 식문화에 관한 연구 -어장문화와 일상식- (A Study on Vietnam Food Culture -Fermented Fish sauce Culture and Daily meal-)

  • This study was performed with survay in the field and literature. The result were; 1. In Vietnam, They took their meal in three times a day. They have taken Nuoc nam (fish sauce) with sliced red pepper, half a lime, etc... in every day every night. Fish Sauce was prepared with small fish (generally fresh- water fish), the same amount of salt, and stored in 8 months, and made filltered fluid. It's Nuoc mam. They have gained protein sauce from it. 2. Their basic menu were composed with rice, soup, food with marine products and meat (generally pork), various vegetables, tropical fruit. 3. Food of cereals were Com Trang (white rice), Pho (rice noodle), Chao (rice gruel), Banh Trang (rice paper), Banh mi (bread) etc... Food of meat were Ho sua, Banh bao chien (barbecued little pork), Suon Nuon (grilled pork), Cha Lua (sausage of pork) etc..., Thit be nhung (grilled beef, Dog meat, Chicken, Duck, Frog. Food of fish were generally fresh-water fish, Ca Chien (grilled fish), Canh chua (soup with sour taste), Ca chem chung (steamed fish with fragrant vegetable), Lobster, Crab, Oyster, Cuttlefish, Shellfish, etc... Food of vegetables were Doa Hanh (Kimchi with a welsh onion), Rau xao hon hop (roasted vegetables), Goi Tom (salad), Canh he dau hu (soup) etc..., and They took much food of trophical fruit, Tra (Tea), Coffee, Lua Moi (distilled liquor). 4. For example, Their Daily meal were composed of Sup Bong Ca (Soup), Heo Sua, Banh Bao Chien (barbecued little Pork), Top Hap (steamed shrimp), Cua (steamed Crab), Luon Um (bioled a fresh-water eel), Lau Thap Cam, Hai Sam Sac Nam Dong Co, Trai Cay.
