Effects of Vesicular-Arbuscular Mycorrhizae on The Growth of American Ginseng (V. A. Mycorrhizae가 미국삼의 생육에 미치는 영향)

  • Newly sprouted American ginseng (Panax quinquefolium L.) seedlings were transplanted to forest pots with mycorrhizae-infested soil and grown in screenhouse for 2 years. Growth patterns, mortality rate and fresh root weight were investigated. Plants in VAM soil had lower mortality rales than control. In soils infested with two species of mycorrhizal fungi (Glomu deseyicola, frappe, Bloss and Merge and G. intraradices, Schenck and Smith), 28-35% of plants produced 3-prongs in the second season and significantly increased fresh root weight by 41 to 43%.

  • The effects of soil phosphorus level on seed emergence, seedling mortality, plant and root development of American ginseng (Panax quinquefolium L.) were evaluated in a newly planted commercial ginseng garden. Phosphorus levels were increased from 58 ppm to 100, 150 and 200 ppm with triplephosphate (0-45-0). Higher phosphate levels increased, seed emergence and reduced seedling mortality. Root length, diameter, fresh root weight and total leaflet length were not affected by phosphate levels.

  • In order to clarify effect of transplanting angle of seedling on shape and growth of ginseng root, ginseng of seedlings were transplanted with $60^{\circ}$ and $90^{\circ}$ respectively, and the root characters of 2-, 4- and 6-year-old ginseng were investigated. The length of main root with 45$^{\circ}$was above 7 cm (standard) but that of $60^{\circ}$ and $60^{\circ}$ was below 7 cm (standard). Number of good lateral root was above 2.3 ea for $45^{\circ}$, 0~$30^{\circ}$, but below 2.3 ea per plant for 60~$90^{\circ}$ respectively. Root fresh weight with $0^{\circ}$, $30^{\circ}$, $45^{\circ}$ was heavier than that with $60^{\circ}$ and $90^{\circ}$. Root disease infection rate was highest with $0^{\circ}$. There were, no significant difference in stem length, stem diameter, leaf area, comber of palmate leaves and leaflets per plant, ratio of missing plant, root diameter in 2-, 4- and 6-year-old ginseng.

Marketing of cleaned fresh ginseng and pre-packaging fumigation of 2-phenylethyl alcohol on ginseng storability (세척인삼 유통 현황과 포장전 2-phenylethyl Alcohol 훈증이 저장성에 미치는 영향)

  • The potential factors for quality loss of cleaned fresh ginseng and technology to be associated with the improvement of marketability through pre-packaging fumigation were examined. Major microorganisms isolated from fresh ginseng included Botrytis cinerea, and Erwinia sp. Others such as Cylindrocarpon sp., Fusarium spp., Pennicilium spp., Bacillus spp. were also found at relatively low frequency. The bacterial density of vacuum packaged fresh ginseng rapidly increased during simulated marketing. Little correlation between bacterial growth and package swelling was found. In order to improve packaging method of fresh ginseng, pre-packaging treatment of 2-phenylethyl alcohol (PE, 100 uL/L, 4 hr) was examined. The fumigation treatment effectively inhibited the growth of bacteria density and also effective on keeping firmness of ginseng root, especially in cortical portion. The internal gas compositions of plastic container packaged for ginseng were approximately ranged between 6 to 8% $O_2$and 3 to 4% $CO_2$. The $O_2$ level of fumigation treatment was lower than control whereas $CO_2$ level was higher. The upsurge of ethylene evolution I day after simulated marketing was found only in fumigation treatment but it returned to ordinary level at day 2. The sucrose content of 2-PE treatment was significantly reduced at 5 days after simulated marketing but reducing sugars like glucose and fructose remained at higher level. The difference in sugar levels was reduced after 10 days of simulated marketing. The decay of fresh ginseng began at the lateral or fine root, which is weak to physical damage, in general. The epidermis was more damaged. Plastic container packaging with PE fumigation could be an alternative to vacuum packaging, which allows an aerobic environment and prevents anaerobic respiration. Further study of pre-package fumigation is required to improve technology of fresh ginseng marketing.

Effects of Addition of Inorganic Germanium, GeO2 on the Growth, Germanium and Saponin Contents of Ginseng Adventitious Root in Submerged Culture (무기 게르마늄 GeO2의 첨가가 액체 배양 중 인삼 부정근의 생장과 게르마늄 및 사포닌 함량에 미치는 영향)

  • This study was carried out to determine the optimal submerged culture conditions for production of ginseng root containing germanium using plant tissue culture technology. The ginseng (Panx ginseng C.A. Meyer) .cot induced by plant growth regulators of 0.5 mg/L BAP and 3.0 mg/L NAA was cultured on SH medium and the effects of various $GeO_2$ concentrations, addition time of $GeO_2$ and pH of medium were investigated on fresh weight, saponin production and germanium accumulation in ginseng root. Optimal $GeO_2$ concentrations for fresh weight, saponin and germanium content were 10, 0 and 110ppm, respectively. When $GeO_2$ was added after 2 weeks cultivation of ginseng root, germanium content was higher than that of adding $GeO_2$ at the initial cultivation time, but saponin content and fresh weight were lower. pH 5.5 was found to be the most favorable condition for the growth of ginseng root and germanium accumulation, but saponin production was best at pH 6.0.

  • Identification of the age of ginseng root is very important in commercial market as well as in research field. However, any criterion abut it has not been clearly established yet. We studied to find a clear morphological key for identification of ginseng root\\`s age using the histochemical staining method. Fresh sections of 3, 4, 5, and 6 year old roots were stained with 1% nile blue, observed under the light microscopy, and compared each other. The number of secretory duct layers(SDL) is a useful key to confirm the age of ginseng root as follow; three-year-old root has two, four-year-old one has three, fie-year-old one has four, and six-year-old one has five resin duct layers on each cortical region of roots. Secretory ducts are thought to be formed by the vascular cambium every year. Unlike the surrounding parenchyma cells, secretory epithelial cells lack starch grains in the cytoplasm.

  • There are two kinds of commercially available ginseng root, red ginseng and white ginseng processed from fresh ginseng root Those ginsengs are primary product from fresh ginseng root and have the characteristic of keeping their original root shape Processed ginseng products are made from either red ginseng or white ginseng by way of complicated process of pulverization. Extraction. Condensation, fettering, sterilization, etc. Among them there are extracts. extract powder, powder, capsules tablets, Candy, drinks, nectar, jelly, gums. chicken soup. tonic. etc. to meet the demand for consumer's pretheronce . The 200 kinds of processed secondary products are approximately produced in the form of 20 kinds of ginseng products by about 60 domestic companies. In spite of about 213.000 million won of domestic market in 1993. it seems like that the ginseng market of the future has not a good prospects The total market sale of white ginseng in Korea has been continuously decreased since 1991 And 963 tons of white ginseng was consumed in domestic market in 1993 The domestic market sales of white ginseng in origina1 root shave. was 90, 000 million won in 1993 and market price of the fine root used as a source of processed products has not been changed in these ten years. The total market sale of red ginseng and its processed products was 58, 000 million won in 1993 9.800 mi11ion won of red ginseng in original root shape and 48.000mi11ion of processed red ginseng product. Ginseng products such as extracts, drinks, teas and tonics etc atre mostly exported to south-east Asia. And the total exports of ginseng pi.oducts (extracts, drinks teas) decreased to 54 million dollars in 1994, compared with 85 million dollars in 1992. Despite of extensive knowledge about ginseng little is still known about the development of new processed ginseng pl.oducts because of "Know-How". Some papars have presented the effects of extracting method(amounts of solvent. time. temperature, equipment. etc.) on the quality and yields of ginseng extr acts. Also. some researchers have carried out a few studies on the poriflcation of the extracts and the amounts of precipitation in the drink at variotas pH during the storage for preventinly drink from precipitation. Extract powder by irradiation and ozone treatment have been reported by some researcher for the improvement hygienic quality of ginseng products There are also some reports about the effects of ginseng components on the acid production by lactic acid bacteria or acetic acid bacteria. and alcohol production by yeast for the development of new ginseng products processed by fermentation. To make ginseng more able to contribute to the health of mankind in the future. consistent and considerable efforts should be focussed on improving the taste of ginseng and developing various new product as a health food or a function food.tion food.

  • Ginseng has been used as a medicinal herb in the world for more than two thousand years. Inspection of the quality of ginseng was made since many hundred years ago. Ginseng quality has been graded by several methodes, based on saponin contents, number of ginsenosides, shape of root and tissue elaborateness. In present. ginseng products are usually evaluated by saponin contents and number of ginsenosides. On the other hand, fresh and manufactured ginseng roots such as red. white and semi-red ginseng, Taegeuk Sam, are mostly graded by root shape such as root development and skin (epidermis) color, and tissue elaborateness. which is a conventional grading method. However, the root shape grading method has a risk of overlooking real medicinal properties of ginseng. So. both the medicinal ingredients and the conventional grading method should be considered for the proper evaluation of ginseng quality. Therefore, for the establishment of better method in evaluating ginseng quality, the relationships of root shape and useful components are required to be studied.

  • In this study, the effects of auxin and casein hydrolysate (CH) on the growth of ginseng hairy root was elucidated. Ginseng hairy root was cultured under light and dark conditions in MS solid and liquid medium with various concentrations of auxin and CH for fifty days. After harvesting the cultures, the fresh and dry weight of cultures were examined, respectively. In the MS solid culture, 1 mg/L of IBA was most effective on the growth of ginseng hairy root under the dark condition, whereas IAA and CH did not affect on the growth of ginseng hairy root. In the MS liquid culture, the growth was maintained regularly by the treatments of IAA and NAA. IBA and CH restrained the growth of ginseng hairy root.

Anticarcinogenic Effect of Ginseng Extracts Depending on the Types and Ages Using Yun's Anticarcinogenicity Test(II) (벤조피렌으로 유도된 마우스 폐선종에 대한 인삼분말의 연근별 항발암효과(II))

  • In this study, we investigeated the anticarcinogenicity of various types and ages of ginseng extracts as an extended study using Yun's anticarcinogenicity test. Fresh ginseng at 1.5, 3, 4, 5 and 6 years was dried and powdered. And white ginseng was processed in the same way that of fresh ginseng after removal fo the ginseng cortex and fine root. For red ginsneg, fresh ginseng was steamed and dried. Each ginseng powder was extracted and extracts was freeze dried. Newborn N:GP(S) mice were given a single subcutaneous injection of 0.5 mg of benzo(a)pyrene(BP). Various types and ages of ginseng extracts at 2.5mg/ml were orally administered. All the mice were sacrificed at the 9th week. The following results were obtained. In the dried fresh ginseng extract treated group, the incidence of lung adenoma induced by BP was 63.9% and its incidence was reduced to 48.3%, 52.5%, 51.8%, 47.5% and 44.1% after co-treatment with 1.5, 3, 4, 5 and 6 year-dried fresh ginseng, respectively. The incidence of lung adenoma induced by BP on the white ginseng extract treated group was 41.3% and decreased to 31.0%, 46.0%, 44.0% and 26.5% after co-treatment with 3, 4, 5 and 6 year-white ginseng, respectively. In the red ginseng extract treated group, the incidence of lung adenoma induced by BP was 47.5% and its incidence diminished to 40.7%, 35.0%, 30.1%, 30.0% and 26.3% after co-treatment with 1.5, 3, 4, 5 and 6 year-red ginseng, respectively. From the above results, we concluded that a statistically significant anticarcinogenic effect was observed in extracts of 6 year-dried fresh ginseng, 6 year-white ginseng, and 4, 5 and 6 year-red ginseng and it is suggested that the anticarcinogenicity of ginseng varies according to the types and ages Key words Ginseng extract, types and ages. anticarcinogenic, newborn mice, lung tumor.
