Pheophytin Content and Cytotoxicity of Silkworm Feces Against Jurkat Cells According to Dry Method and Storage Period

  • In order to find potential anticancer agents, we extracted pheophytin from silkworm feces according to various dry and storage methods such as sun dry, shade dry, fresh freezing dry and freezing dry after freezing storage (for 1∼3 years). The pheophytin extracts, mainly 10-hydroxypheophytin a, little b, of various storage silkworm feces were analyzed by reversed-phase high-performance liquid chromatography with photodiode array and fluorescence detection. The content of those pheophytih in old silkworm for 3 years (freezing storage and freezing dried in use, or freezing dried and cold storage) was better than others. The cytotoxicity of the pheophytin extracts and ethanol extracts of various storage silkworm feces were measured using Jurkat cells originated from human leukemia, using dye uptake assay (MTT) in order to find effective photodynamic therapeutic agents. The anticancer activity of those pheophytin extracts in various storage methods showed little difference among them. But ethanol extracts of fresh freezing dried silkworm in the current year was good cytotoxic activity than those of any other silkworm feces. With regards to these results, fresh ethanol extracts of silkworm feces were better than old ones. On the other hands, the pheophytin extracts of old silkworm feces contained the highest percentage of pheophytin content and showed good cytotoxicity against cancer cells by changing the pheophytin into pheophobide in the degradative process.

The Effects of Freezing and Supplementation of Molasses and Inoculants on Chemical and Nutritional Composition of Sunflower Silage

  • This study was conducted to determine the effects of freezing and supplementation of molasses (M), lactic acid bacteria (LAB) and LAB+enzyme mixture on chemical and nutritional composition of sunflower silage (SF). Sunflower crops were harvested (at about $29.2%{\pm}1.2%$ dry matter) and half of fresh sunflower was ensiled alone and half was frozen (F) at $-20^{\circ}C$ for 7 days. Silage additives were admixed into frozen SF material. All samples were ensiled in glass jars with six replicates for 90 days. The treatments were as follows: i) positive control (non-frozen and no additives, NF), ii) negative control (frozen, no additives, F), iii) F+5% molasses (FM), iv) F+LAB (1.5 g/tons, Lactobacillus plantarum and Enterococcus faecium, FLAB); v) F+LAB+enzyme (2 g/tons Lactobacillus plantarum and Enterococcus faecium and cellulase and amylase enzymes, FLEN). Freezing silage increased dry matter, crude ash, neutral detergent fiber, and acid detergent lignin. The organic matter, total digestible nutrient, non-fiber carbohydrate, metabolizable energy and in vitro dry matter digestibility were negatively influenced by freezing treatments (p<0.05). In conclusion, freezing sunflower plants prior to ensiling may negatively affect silage quality, while molasses supplementation improved some quality traits of frozen silage. Lactic acid bacteria and LAB+enzyme inoculations did not effectively compensate the negative impacts of freezing on sunflower silage.

  • Pellet and straw methods in canine semen freezing are compared with respect to motility, viability and acrosome demage of sperm during each of the two major processing steps, to prior-freezing and to frozen-thawing. Senen was extended with a tris-buffered egg yolk contained 4% glycero1 Pellet freezing in the hole of dry ice and straw freezing on the surface of liquid nitrogen were carried out, respectively. In the comparison of two freezing methods, the straw freezing method with 42.7% in motility. 49.2% in viability and 0.186 acrosome score after thawing seems to be superior to the pellet freezing method with 31.2%, 34.5% and 0.314%, respectively.   • The purpose of this study is to provide basic data for utilization in environment-friendly and economically outstanding CSG construction method by physical and mechanical properties of CSG materials including characteristics of uniaxial compressive strength, microscopic structure and freezing and thawing resistance in accordance with the cement content and curing time of the cement, and size of specimen. In this study, specimens with cement content of 4, 6, 8 and 10% of the total weight were, and, in order to examine the characteristics of the sizes of specimen, specimens with ${\Phi}50{\times}100mm$, ${\Phi}100{\times}200mm$ and ${\Phi}150{\times}300mm$ were manufactured to assess the features including compressive strength, microscopic structure, freezing and thawing, and degree of wet-dry. As results, it was found that with greater size specimen or contents of cement in the specimen, compressive strength, freezing and thawing resistance, and wet-dry resistance increase. Moreover, reactive products for each size of specimen were examined and it was possible to verify that some typical needle structured ettringite was generated due to blending of cement through microscopic structure analysis such as SEM and EDS analysis.

  • Evaluation of the asphalt mixture modified with crumb rubber modifier(CRM) was performed to estimate possibility of using it as a paying material. OACs(optimum asphalt content) of CRM modified asphalt mixtures by dry process and wet process were determined by Marshall mix design and Wheel tracking test and moisture susceptibility test by freezing and thawing were carried out with CRM modified asphalt mixtures at OACs. The results from these tests, resistance of permanent deformation of CRM modified asphalt mixtures were superior to one of AP-5 while showing very low resistance of moisture sensitivity by freezing and thawing. This means that CRM modified asphalt mixtures are very sensitive to freezing and thawing. However, CRM modified asphalt mixture with anti-stripping material showed high improvement to resistance of moisture susceptibility by freezing and thawing. Therefore, it is recommended that when CRM mixtures were used in domestic, CRM modified asphalt mixtures should be with prevention against freezing and thawing resistance by moisture susceptibility.

  • The round muscle of Korean cattle and squid muscle were cooked with various methods which were followed digestibility test by use of pepsin in-vitro, determination of amino nitrogen in the course of digestion procedure by using Formol method (AOAC) and influence of ether treatment for preminary test also examined. The results obtained were summarized as follows: 1. The order of digestibility values were demonstrated as follows: In case of beef, it was autoclaving, frying, raw, freezing, roasting, boiling and in case of squid muscle, it was raw, autoclaving, boiling, freezing, dry heating and roasting. 2. The amounts of amino nitrogen for beef and squid muscle were increased in proportion to digestibility value. 3. There were no significances in the digestibility between treating with ether and none of any treatment of beef and squid muscle in raw condition.

  • Korea has four distinct seasons, showing hot and humidity in summer and cold weather lasted in winter. Domestic research on earth work has been developed according to the seasonal characteristics, and most of research topics have focused on the effect of freezing-thawing on the performance of geo-materials. However, the previous research was performed on the ground compacted at room temperature and therefore, the effect of the sub-zero temperature at the time of construction was not fully investigated. The ground characteristics compacted at freezing temperature can be different from those at room temperature and show different characteristics of strength and deformation caused by freezing and thawing. Therefore, the compaction tests on sandy materials were conducted under various temperature at $-3^{\circ}C$ and $-8^{\circ}C$ with various fine contents of 0%, 5%, 10% and 15% in weight fraction. The effectiveness of soil compaction at below-freezing temperatures were compared with the compaction at room temperature at $18^{\circ}C$ in terms of the maximum dry unit weight and optimum water contents. Based on the test results, the maximum dry unit weight tends to decrease with the freezing temperature and the relative compaction at $-8^{\circ}C$ can not be satisfied with general specification standard.

Evaluation on Surface Scaling and Frost Resistance for concrete Deteriorated due to Cyclic Freezing and Thawing with Inherent Chloride

  • The purpose of this study is to evaluate freezing-thawing and surface scaling resistance in order to examine the frost durability of concrete in a chloride-inherent environment. The mixing design for this study is as follows: 3 water binder ratios of 0.37, 0.42, and 0.47; 2-ingredient type concrete (50% OPC concrete and 50% ground granulated blast-furnace slag), and 3-ingredient type concrete (50% OPC concrete, 15% fly ash, and 35% ground granulated blast-furnace slag). As found in this study, the decrease of durability was much more noticeable in combined deterioration through both salt damage and frost damage than in a single deterioration through either ofthese; when using blast-furnace slag in freezing-thawing seawater, the frost durability and surface deterioration resistance was evaluated as higher than when using OPC concrete. BF 50% concrete, especially, rather than BFS35%+FA15%, had a notable effect on resistance to chloride penetration and freezing/expansion. It has been confirmed that surface deterioration can be evaluated through a quantitative analysis of scaling, calculated from concrete's underwater weight and surface-dry weight as affected by the freezing-thawing of seawater.

Freezing-Thawing Resistance of Concrete Using Steel Slag as Coarse Aggregate (제강슬래그를 굵은 골재로 이용한 콘크리트의 동결융해 저항성)

  • In this study, freezing-thawing resistance of concrete using steel slag as coarse aggregate(steel slag concrete) from Gwangyang Iron Co. was estimated to offer basic data for utilization of much more steel slag. Freezing-thawing test of concrete using crushed stone as coarse aggregate(crushed stone concrete) whose compressive strength and air contents are as close as possible to those of the steel slag concrete was performed. Because they are main two factors that affect of freezing-thawing resistance. The test was carried out up to 400 cycles according to KS F 2456. The compressive strength and weight of two concretes were measured and compared. As a result, the freezing-thawing resistance of steel slag concrete curing in water was almost the same with that of crushed stone concrete. But the resistance of steel slag concrete curing in air dry condition was weaker than that of crushed stone concrete. Also, the steel slag concrete which has more than 60% of W/C ratio showed much more surface degradation when compared to crushed stone concrete.
