Protective effect of methanol extract from citrus press cakes prepared by far-infrared radiation drying on $H_2O_2$-mediated oxidative damage in Vero cells

  • In the present study, a suitable drying method was developed for citrus press cakes (CPCs), which are produced as a by-product in citrus juice plants, and the protective effect of methanol extract of CPCs prepared by far-infrared radiation (FIR) drying against $H_2O_2$-induced DNA damage was evaluated versus that of freeze-dried CPCs. Methanol extract of FIR-dried CPCs exhibited comparatively good ROS scavenging activity versus the freeze-dried CPCs at the concentration of 100 ${\mu}g$/mL. The extract strongly enhanced the cell viability against $H_2O_2$-induced oxidative damage in Vero cells. Lipid peroxidation inhibitory activity of the extract from FIR-dried CPCs was comparable to that of the extract from freeze-dried CPCs. This sample also exhibited good protective effects against $H_2O_2$-mediated cell apoptosis as demonstrated by decreased apoptotic body formation in the nuclear staining with Hoechst 33342. In the comet assay, the CPC extracts exhibited strong inhibitory effects against $H_2O_2$-mediated DNA damage in a dose-dependent manner. Thus, this study demonstrated that FIR drying effectively preserves CPC as a functionally important natural antioxidant source and the FIR drying can be adapted for drying CPCs and is more economical for massive production than freeze drying.

Pharmacokinetics, Cell Toxicity, Antitumor Activity and Spleen/Blood Cell Toxicity of Aclarubicin-entrapped Liposomes (리포좀에 봉입된 아클라루비신의 약물동태, 세포독성, 항암효과 및 비장/혈구 세포독성)

  • Aclarubicin(ACL)-entrapped freeze dried liposomes were prepared using Microfludizer to attain a sustained release at targeted organs in a prolonged time so that it can reduce th e side effect and maximize the therapeutic effect. The freeze-dried liposomes were evaluated for pharmacokinetics, antitumor activity against Sarcoma 180, cytotoxicity against L1210 and A549 tumor cells, spleen toxicity and myelosuppressive action. The AUC0->8hr values were 122+/-42, 382+/-140, 419+/-171, 835+/-206 and 443+/-309mcg min/ml for free ACL. ACL-liposome formulation I, II, III and IV, respectively. Cytotoidcity of ACL-entrapped liposomes against L1210 and A549 tumor cells was 2-4 times higher than that of free aclarubicin. ACL-liposome formulation I(PC/CHOL/TA) showed the most potent antitumor activity against Sarcoma 180 in mice. The loss of body weight was much smaller with ACL-entrapped liposomes than free ACL after I.p. injection at a dose of 2 mg/kg/day. Compared to free ACL, ACL-entrapped liposomes expressed a lower and delayed spleen toxicity up to 5th day after I.v. administration. Myelosupperssion seemed to be lower with ACL-entrapped liposome of PC/PC-hydrate/CHOL/TA (formulation III) than free aclarubicin.

Insecticidal Activity and Stability by Freeze-drying of Entomopathogenic Bacteria, Photorhabdus temperata M1021 (동결건조에 따른 살충성 세균 Photorhabdus temperata M1021의 안정성과 살충성 평가)

  • In order to develop eco-friendly biopesticide, an entomopathogenic bacterium Photorhabdus temperata M1021 has been lyophilized via freeze-drying along with protective agents such as skim milk, starch, sodium alginate, glucose and sodium glutamate to protect cells from lysis. Freeze-drying powder of P. temperata M1021 containing 7% skim milk (w/v) showed highest survival rate of 63% among all the protective agents used in trials. Furthermore, the freeze-dried microbial powder showed 75% of survival rate after stored at $4^{\circ}C$ for 4 weeks at air contact conditions. Injection toxicity of the freeze-dried sample was tested against larvae of Galleria mellonella. A dose of $2.0{\times}10^1$ cells of P. temperata M1021 killed 100% of the G. mellonella larvae within 4 days after injection. Moreover, $2.0{\times}10^0$ cells caused 50% mortality within the 4 days after injection. Freeze-dried P. temperata M1021 strains exhibited effective insecticidal activity and could be a better candidate for being used as a biopesticide.

Dietary Evaluation of the Freeze-Dried Alga Isochrysis aff. galbana for Larval Surrival of the Pacific Oyster Crassostrea gigas (동결건조한 Isochrysis aff, galbana를 이용한 굴 유생사육에 관한 연구)

  • To investigate an efficient microalgal feed for lariat culture of the Pacific oyster Crassostrea gigas, we prepared three types of Isochrysis aff, galbana (T-iso) : 1) freshly Harvested feed, 2) concentrated feed and 3) freeze-dried feed. The chemical compositions and fatty acid content of these feeds were evaluated and survival rate and lipid content of oyster larvae fed by these feeds were also determined. There was no significant difference in all types of feed in the gross biochemical compositions, In the fatty acid composition, the freeze-dried feed showed a significant increase in the level of polyunsaturated fatty acid (PUFA), eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) than freshly Harvested feed, especially EPA was 7.35-fold higher than freshly Harvested feed. The survival rate of the oyster larvae was the highest when the larvae were fed with a diet of $10\%$ freeze-dried and $90\%$ concentrated feed; it was 2.1-fold higher than that fed with freshly Harvested food alone. Thereafter, the survival rate decreased with the increased substitutions of freeze-dried food, finally equalling that fed $100\%$ freshly harvested feed at the $30\%$ substitution. Larval lipid content of the oyster was also the highest when the larvae were fed with a diet of $10\%$ freeze-dried and $90\%$ concentrated feed. This increase was by 1.6-fold ver that fed $100\%$ freshly Harvested cells. Thus feed produced during slack times, on a seedling aquaculture farm, and preparedas a freeze-dried diet can be used, mixed with concentrated feed, to supply diets more efficiently and to improve the larvae survival rate of Pacific oyster.

Effect of Immune Function on the fermentation of Kimchi Intake to append Acanthopanacis cortex Extract in Balb/c Mice (오가피(五加皮) (Acanthopanacis cortex)추출물(抽出物)을 첨가한 발효김치의 급여가 생쥐의 면역작용(免疫作用)에 미치는 영향(影響))

  • This experimental study was carried out to evaluate the effects of Kimchi intake of Acanthopanacis cortex extract (APCE) supplementation on cytokine-induction and immune response in mice. To study in experiments using male Balb/c mice fed Kimchi and Kimchi of APCE supplementation (addition of 2% of total Kimchi weight) containing fed experimental diet during 2 weeks. Experimental mice were fed control diet or diet containing freeze-dried Kimchi at the level of 5%(w/w) or 5% freeze-dried Kimchi with 2% APCE supplementation. The main ingredient of Kimchi was Korean cabbage and fermentation was carried out at $4^{\circ}C$ for three weeks. Freeze-dried 2% APCE supplementation was added to Kimchi at the begining of fermentation. In order to investigate the effect of Kimchi intake of APCE supplementation (5%Kimchi-2%APCE), the following was performed; body weight, food intake, hematological parameter, serum level of mouse interleukin-4 (mlL-4) and mouse interferon-$\gamma$ (mIFN-$\gamma$ ), and, the percentage of CD3+/CD4+, CD3+/CD8+, B220+ in splenic cells. The results of final body weight, and food diet intake of two Kimchi groups were lower than those of the control group (not supplemented experimental diet). The hematology change obtained from the level of WBC (white blood cell) and platelet were not affected by feeding different dietary regiments, but the level of RBC (red blood cells) HB (hemoglobin), and spleen weight of two Kimchi groups were increased significantly than those of the control group. The serum level of IL-4 and IFN-$\gamma$ of two Kimchi groups were increased significantly than those of the control group, also enhanced the percentages of the CD3+/CD4+ and CD3+/CD8+ by 5% freeze-dried Kimchi, and 5%Kimchi-2%APCE group were 43.9 and 65.2%, and 96.0 and 208% than those of the control group, respectively. From these results, it can be concluded that Kimchi itself has an immuno-stimulatory effect and Kimchi contaning 2% APCE supplementation has the more pronounced effect in vivo system.

  • The purpose of this study is to evaluate histologically the resorption and tissue response of various resorbale membranes used for guided tissue regneration procedures, using a subcutaneous model on the dorsal surface of the rat. In this study, 12 Sprague-Dawley male rats(mean BW 150gm) were used and the commercially available materials included dense collagen membrane, freeze-dried bovine dura mater loos collagen membrane, PLA/PLGA membrane. Animals were sacrificed at 3, 6 and 8 weeks after implantation of various resorbable membranes. Specimens were prepared with Hematoxylin-Eosin stain for light microscopic evaluation. The results of this study were as follows: 1. Resorption : Loose collagen membrane group was resorbed most rapidly. Dense collagen membrane group and freeze-dried bovine dura mater group were rarely resorbed. 2. Inflammatory reactions : PLA/PLGA membrane group showed persistent and severe inflammatory reactions for 3 to 8 weeks. Moderate inflammatory reactions and the ectopic formation of calcified material were observed in dense collagen membrane group. Freeze-dried bovine dura mater group and loose collagen membrane group showed mild inflammatory reactions 3. In PLA/PLGA membrane group, multinucleated giant cells by foreign body reactions were observed. In conclusion, the resorption of freeze-dried bovine dura mater didn't happen for 3-6weeks, which showed the best bio-compatibility. Therefore, freeze-dried bovine dura mater was considered proper resorbable membrane for guided tissue regeneration.

Effects of different drying methods on fermentation characteristics and viability of cold-adaptive yeast (저온 적응성 효모의 보존방법에 따른 균체의 생존율 및 발효특성)

  • The quality characteristics of Yakju and survival rate of yeast were investigated by modifying the drying method for the cold adapted yeast strain Saccharomyces cerevisiae Y297 (SCY297). Viability and fermentation characteristics of the freeze-dried, air blast-dried, and liquid SCY297 cultures were compared after storing them at $25^{\circ}C$. In addition, 5% skimmed milk, ${\alpha}$-lactose, or trehalose was added as a protective agent for examining the effects of drying methods. During the 15-week storage period, the liquid and freeze-dried SCY297 cultures containing a protective agent showed a survival rate of 80%. However, the air blast-dried SCY297 culture showed 80% survival rate only in the skimmed milk supplemented group. Compared to the untreated cells, the acidity and amino acidity of Yakju prepared using freeze-dried or air blast-dried cultures of SCY297 increased by 2 fold and 5.7 fold respectively, while the alcohol content decreased by 5.07%. Compared to the untreated cells, the pH and amino acidity of Yakju prepared using the liquid culture of SCY297 increased by 1.5 fold and 2.5 fold respectively. Although the alcohol content decreased by 2.9%, decrease rate was lower than that observed for the freeze-dried and air blast-dried yeast cultures. Therefore, the results of this study showed that using a liquid starter culture was more advantageous than using the conventional solid culture.

Estrogen activity of Silkworm (Bombyx mori) Pupa water extract and its fractions

  • This study was conducted to evaluate the estrogen activity of silkworm (Bombyx mori) pupa extracts and their fractions. Powdered samples of freeze-dried silkworm pupa were extracted at room temperature (RT), $40^{\circ}C$, $60^{\circ}C$, $80^{\circ}C$, and $100^{\circ}C$ in water (D.W), chloroform, ethyl acetate, and methanol for 6h and then filtered (0.45 um). The extracts were then freeze-dried. The estrogenic activity of these extracts was then investigated by competition binding assays using estrogen receptor ${\alpha}\;(ER{\alpha})$ and $ER{\beta}$, and by evaluating their effects on the proliferation of the human breast cancer cell line, MCF-7. Among the extracts evaluated, water extracts prepared at RT showed the highest binding affinity to $ER{\alpha}$ ($IC_{50}$, 1.76 ug/ml) and $ER{\beta}$ ($IC_{50}$, 0.07 ug/ml). In addition, MCF-7 cells that were treated with 62.5 ug/ml of the RT extract showed the greatest increase in proliferation (2-fold; 1291.79%) when compared to control cells (659.82%). Next, the water extract that was prepared at RT (sample 1) was dissolved in D.W. and further fractionated using a Dowex 50W - 8X ($H^+$) column. The flow-through and wash were then pooled together and freeze-dried (sample 2). The bound materials were then eluted with 20 mM NaCl, after which they were applied to a Dowex 1X2 - 200 ($Cl^-$) column and washed with D.W. to remove the sodium ions. The eluants were then freeze-dried (sample 3). Of these fractions, sample 2 showed the highest binding affinity to ER{\alpha} ($IC_{50}$, 1.44 ug/ml) and $ER{\beta}$ ($IC_{50}$, 1.18 ug/ml). In addition, MCF-7 cells that were treated with sample 2 (15.6 ug/ml) showed the largest increase in growth (1159.39%) when compared to control cells (525.26%). Taken together, these results suggest that the fraction of the RT water extract of silkworm pupa referred to as sample 2 may be useful as a phytoestrogen.

Stabilities of Artificially Transconjugated Plasmids for the Bioremediation of Cocontaminated Sites

  • Here, we attempted to evaluate the activity of artificially transconjugated multiple plasmids in 'designer biocatalysts' for the bioremediation of cocontaminated sites under nonselective conditions. We observed profound losses in the percent survivals of artificially transconjugated plasmid activity ($66 - 78\%$ loss immediately after freeze-drying, $99.45 - 99.88\%$ loss by the end of 6 months storage) in reconstituted Pseudomonas sp. KM12TC. Such unpredictable high losses of this particular plasmid appeared to clearly be a deleterious effect. However, even after 6 months of storage, the cells remained able to degrade $95\%$ of phenol within 9 days, and the full effiux of$^%${73} As, as compared to that of the non-freeze-dried cells, was successfully achieved 4 to 9 days later. These results indicate that 'stable designer biocatalysts' can remain viable, even after freeze-drying and 6 months of storage.

Fatty Acid Composition and Anti-inflammatory Effects of the Freeze Dried Tenebrio molitor Larva (동결건조 갈색거저리 유충의 지방산 조성과 항염증 효과)

  • This study aimed to assess fatty acid composition and anti-inflammatory effects, such as nitric oxide(NO) production, expression of $TNF-{\alpha}$ and interleukin-6(IL-6), of Tenebrio molitor larva using RAW 264.7 cells. The content of total fatty acid in Tenebrio molitor larva was 76.14%, which was composed of oleic acid(42.12%), linoleic acid(32.67%) etc. There was no cytotoxicity at a dose level of 0.1, 1.0, 10, and $100{\mu}g/mL$ of freeze dried Tenebrio molitor larva ethanol extract(FDTEtOH) on RAW 264.7 cells. FDTEtOH significantly decreased NO production in LPS(lipopolysaccharide)-stimulated RAW 264.7 cells in a dose-dependent manner. Also, FDTEtOH dose-dependently suppressed the expression of $TNF-{\alpha}$ and IL-6. Thus, these results showed that Tenebrio molitor larva has the potential to be used as an anti-inflammatory food to improve immunity.
