Effects of Fruits and Stems of Opuntia ficus-indica on Blood Glucose and Lipid Metabolism in Streptozotocin-induced Diabetic Rats (손바닥선인장의 열매와 줄기가 Streptozotocin으로 유발된 당뇨 쥐의 혈당 및 지질대사에 미치는 영향)

  • This study was conducted to examine the effects of fruits and stems of Opuntia ficus-indica var. saboten Makino on water intake, feed intake, body weight, blood glucose level and glucose tolerance in streptozotocin (STZ)-induced diabetic rats. Forty Sprague-Dawley male rats were divided into non-diabetic control (NC), diabetic control (DC), 8% Opuntia fruit (DOF), 5% Opuntia stem (DO-5) and 10% Opuntia stem (DO-10) groups. Fruits and stems of Opuntia ficus-indica were freeze-dried and ground before use in the experiment. Animals were fed experimental diet for 3 weeks. DOF, DO-5 and DO-10 groups showed lower water and feed intake as well as less body weight loss than DC group. The fasting blood glucose levels were 100 mg/dL for NC and 379 mg/dL for DC. Fasting glucose level of DOF was a significantly low level of 28% (p<0.05), whereas DO-5 and DO-10 had a decrease of 5% and 9% compared to DC. As for the glucose tolerance test, the highest blood glucose levels for NC and DC-10 group were observed at 30 minutes after glucose injection while those of DOF and DO-5 groups were after 60 minutes. DOF and DO-5 plasma insulin level improved. Plasma total cholesterol, triglyceride, non-esterified fatty acid (NEFA) and LDL-cholesterol concentrations were also lower in DOF, DO-5 and DO-10 groups, although HDL-cholesterol level was only slightly affected by experimental diets compared to DC. These results suggest that the feeding of Opuntia ficus-indica fruits and stems improved blood glucose and lipid metabolism in STZ-induced diabetic rats.

Viability Assessment of Fresh and Frozen-thawed Dog Spermatozoa by Flow Cytometry (Flow Cytometry에 의한 개 신선정액과 동결정액의 생존성 분석)

  • This study was performed to examine the correlations among dog sperm viabilities evaluated by flow cytometry, by microscopic evaluation (ME), by carbo-xifluorescein diacetate and propidium iodide (CFDA/PI) and by hypoosmotic swelling (HOS) test. Semen were collected from 5 dogs ranging in age from 2 to 4 years. Each ejaculate was divided into 3 aliquots and different proportions of freeze-killed cells were added to each aliquot (1:0, 1:1 and 1:3). In the other experiment, semen was extended with Sweden extender containing 5% glycerol and equex STM paste, and frozen using liquid nitrogen vapor. Fresh and frozen-thawed dog sperm viability were assessed by flow cytometry using PI staining method. The accuracy of flow cytometry was evaluated by comparing with other classic assessments, microscopic evaluation, epifluorescence microscopic analysis using CFDA/PI, and HOS test. High correlations of sperm viabilities were found among flow cytometry, epifluorescence evaluation, HOS test (p<0.01) in fresh semen. Especially, sperm viability assessed by HOS test was highly correlated with viability by flow cytometry in all the ratios of live and dead spermatozoa, 1:0, 1:1 and 1:3 (p<0.01). The viability evaluated by ME were significantly correlated with that by flow cytometry in ratios of 1:0 and 1:3 (p<0.05) however, there was no significance in ratio of 1:1. The viability evaluated by C/p were highly correlated with that by flow cytometry in ratio of 1:0 and 1:1 (p<0.01) and significantly correlated in ratio of 1:3 (p<0.05). In frozen-thawed spermatozoa, the viability determined by HOS test was considerably correlated with that by flow cytometry (p<0.01). There was significant correlation between the viabilities by ME and by flow cytometry (p<0.05). But the viability evaluated by CFDA/PI was not correlated with viability by flow cytometry. The result from this study validate the use of flow cytometry as a precise method for assessing the viability of fresh and frozen-thawed dog spermatozoa.

INTRINSIC NMR ISOTOPE SHIFTS OF CYCLOOCTANONE AT LOW TEMPERATURE (저온에서의 싸이클로옥타논에 대한 고유동위원소 효과)

  • Several isotopomers of cyclooctanone were prepared by selective deuterium substitution. Intrinsic isotope effects on $^{13}C$ NMR chemical shifts of these isotopomers were investigated systematically at low temperature. These istope effects were discussed in relation to the preferred boat-chair conformation of cyclooctanone. Deuterium isotope effects on NMR chemical shifts have been known for a long time. Especially in a conformationally mobile molecule, isotope perturbation could affect NMR signals through a combination of isotope effects on equilibria and intrinsic effects. The distinction between intrinsic and nonintrinsic effects is quite difficult at ambient temperature due to involvement of both equilibrium and intrinsic isotope effects. However if equilibria between possible conformers of cyclooctanone are slowed down enough on the NMR time scale by lowering temperature, it should be possible to measure intrinsic isotope shifts from the separated signals at low temperature. $^{13}C$ NMR has been successfully utilized in the study on molecular conformation in solution when one deals with stable conformers or molecules were rapid interconversion occurs at ambient temperature. The study of dynamic processes in general requires analysis of spectra at several temperature. Anet et al. did $^1H$ NMR study of cyclooctanone at low temperature to freeze out a stable conformation, but were not able initially to deduce which conformation was stable because of the complexity of alkyl region in the $^1H$ NMR spectrum. They also reported the $^1H$ and $^{13}C$ NMR spectra of the $C_9-C_{16}$ cycloalkanones with changing temperature from $-80^{\circ}C$ to $-170^{\circ}C$, but they did not report a variable temperature $^{13}C$ NMR study of cyclooctanone. For the analysis of the intrinsic isotope effect with relation to cylooctanone conformation, $^{13}C$ NMR spectra are obtained in the present work at low temperatures (up to $-150^{\circ}C$) in order to find the chemical shifts at the temperature at which the dynamic process can be "frozen-out" on the NMR time scale and cyclooctanone can be observed as a stable conformation. Both the ring inversion and pseudorotational processes must be "frozen-out" in order to see separate resonances for all eight carbons in cyclooctanone. In contrast to $^1H$ spectra, slowing down just the ring inversion process has no apparent effects on the $^{13}C$ spectra because exchange of environments within the pairs of methylene carbons can still occur by the pseudorotational process. Several isotopomers of cyclooctanone were prepared by selective deuterium substitution (fig. 1) : complete deuterium labeling at C-2 and C-8 positions gave cyclooctanone-2, 2, 8, $8-D_4$ : complete labeling at C-2 and C-7 positions afforded the 2, 2, 7, $7-D_4$ isotopomer : di-deuteration at C-3 gave the 3, $3-D_2$ isotopomer : mono-deuteration provided cyclooctanone-2-D, 4-D and 5-D isotopomers : and partial deuteration on the C-2 and C-8 position, with a chiral and difunctional case catalyst, gave the trans-2, $8-D_2$ isotopomer. These isotopomer were investigated systematically in relation with cyclooctanone conformation and intrinsic isotope effects on $^{13}C$ NMR chemical shifts at low temperature. The determination of the intrinsic effects could help in the analysis of the more complex effects at higher temperature. For quantitative analysis of intrinsic isotope effects, the $^{13}C$ NMR spectrum has been obtained for a mixture of the labeled and unlabeled compounds because the signal separations are very small.

Effects of Various Mulberry Products on the Blood Glucose and Lipid Status of Streptozotocin-induced Diabetic Rats (오디의 가공형태에 따른 Streptozotocin 유발 당뇨쥐의 혈당 및 지질의 상태에 미치는 영향)

  • The study aim was to investigate the effects of three types of mulberry products on the blood glucose and lipid statuses and peroxidative state under diabetic condition. The three types were mulberry liquor prepared by adding 30% ethanol to the crushed fresh fruit, mulberry wine and vinegar by fermentation. For diet experiment the mulberry liquor (M-Liquor), wine (M-Wine), and vinegar (M-Vinegar) were prepared as powders by freeze-drying of the respective product and were added to the diet at the level of 1% and mulberry fruit powder (M-Powder) at the level of 5%. Sprague- Dawley female rats weighing $150{\pm}10\;g$ were randomly assigned to one normal group, and five diabetic groups induced by one intraperitoneal injection of streptozotocin (STZ) at the level of 50 mg/kg. The normal and diabetic control (DM-Control) groups were fed diet without the mulberry products. During twenty-one days of experimental diet, blood glucose was maintained at a low level in the M-Liquor group compared with the DM-Control group. However, serum insulin level was higher in both M-Liquor and M-Vinegar groups after the experimental diet period. Serum levels of total cholesterol and triglyceride (TG) were lower in M-Liquor but HDL-/total cholesterol ratios were higher in the four M groups. The TG liver level was lower in M-Powder and M-Vinegar groups but the cholesterol level was lower in M-Powder than in the DM-Control group. Serum levels of thiobarbituric acid reactive substances were not different among the groups but the liver levels of these substances were lower in the four M groups than in DM-Control. Serum GOT and GPT levels were not changed by the mulberry products. These results indicated that mulberry liquor is the most effective among the four mulberry products for normalizing blood glucose and lipid status and that all the mulberry products were effective in enhancing antioxidant defense in the diabetic state.

Physicochemical Properties of the Mung Bean (Phaseolus aureus L.) as Biohealth Functional Substance (기능성 소재로서 녹두(Phaseolus aureus L.)의 이화학적 특성)

  • This study on the physicochemical property of physiological activity substance in mung bean (Phaseolus aureus L.) was performed for the use as an functional food materialization. The proximate composition in the vacuum freeze dried mung bean was carbohydrate 57.20±0.29%, crude protein 26.40±0.69%, moisture 9.90±0.16%, crude ash 3.54±0.43%, and crude fat 2.96±0.26%, respectively. The vitamin content of mung bean was vitamin B5 0.62±0.013 mg/100 g, vitamin E 0.17±0.001 mg/100 g, vitamin B1 0.13±0.016 mg/100 g, and β-carotene 87.37±0.754 ㎍ RE/100 g, respectively. The mineral content of mung bean was potassium (K) 12,428.55±147.55 mg kg-1, magnesium (Mg) 2,053.32±14.13 mg kg-1, calcium (Ca) 1,966.40±14.53 mg kg-1, sodium (Na) 1,063.99±7.75 mg kg-1, iron (Fe) 63.77±0.98 mg kg-1, and manganese (Mn) 14.67±0.22 mg kg-1. The compositions of fatty acid were saturated fatty acid 29.23±0.03%, monoenes 20.30±0.04%, and polyenes 50.46±0.06%. Protein bound amino acid content of mung bean was 21.75±0.24 g%. And major amino acids were glutamic acid 3.93±0.03 g%, aspartic acid 2.68±0.03 g%, respectively. The composition of free amino acid of mung bean was 336.77±8.66 mg%, and major free amino acids were arginine, glutamic acid, asparagine, and aspartic acid. As a results of these experiment, Mung bean could be used a natural resouce and functional biohealth food substance.

Variation of Image Analysis Results for Determining the Characteristics of the Air-Void System on Hardened Concrete (콘크리트 공극구조 분석을 위한 화상분석결과의 변동성 분석)

  • Recently, the cases of freeze-thaw distress on concrete pavement in domestic have been reported periodically. Hereupon, the necessity to establish the standard of spacing factor came to the fore. The test method for analyzing spacing factor is provided in the standard of ASTM C 457. Since researchers tend to judge study results subjectively, the results should be revised throughly. Image analysis program for determining the characteristics of air-void system on hardened concrete identify air void through the difference of a color. The pixel intensity values used in this program have a significant effect on the analysis results. This study compared the automated void count by varying pixel intensity values with the manual void count in order to determine the optimum range of pixel intensity values. Also, this study analyzed the air-void characteristics on eight kinds of concrete mixtures. In this study, it was confirmed that the variation of void counted manually was around 10% from the results of round robin test, and that the optimum range of pixel intensity values is around 80~90. And it was also confirmed that air content (as a whole) was increased generally and spacing factor was decreased by increasing air-entrainer content. But some concrete mixtures showed a tendency that air content was constant and spacing factor was decreased by increasing air-entrainer content. This causes the air entrained by air-entrainer has more influence on spacing factor than air content. Also, the deviation of spacing factor by cutting position of concrete specimen was about 30~100${\mu}m$ because of the limit of 2-D image analysis. The additional study about variation of spacing factor by cutting position of concrete specimen will be performed later.

Cytotoxic Effect of X-irradiation of Mouse Tumor Cells in the Presence of Korean Ginseng Extract (마우스 종양세포의 세포독성에 미치는 인삼 추출액과 방사선조사의 병용 효과)

  • Purpose : We already reported the results that aqueous extract of Korean ginseng roots showed a marked cytotoxicity. In this study, we investigated whether combined ginseng product with X-irradiation increase the cytotoxicity of tumor cells than X-irradiation or not. Materials and Methods : Fifty gram of Korean ginseng powder mixed with 1 L of distilled water was extracted with reflux flask under condition of $100^{\circ}C$ for 5 hrs. This aquaous ginseng extract was filtered, centrifuged and then was freezed under condition of $-90^{\circ}C$ for 16-18 hrs. The freezing extract was dried with freeze drier, and then diluted. X-irradiation was given to tumor cells by 6 MeV linear accelerator. The cytotoxicity of ginseng in vitro was evaluated from its ability to reduce the clonogenecity of fibrosarcoma (FSa II) cells. In X-irradiation alone group, each 2, 4, 6 and 8 Gy was given to tumor cells. In X-irradiation with ginseng group, 0.2 mg/mL of ginseng extract was exposed to tumor cells for 1 hour before X-irradiation. Results : The yield for 50 g of ginseng extract which was treated with freezing drier was 3.13 g($6.3\%$). Cytotoxicity In vitro was measured as survival fraction which was judged from the curve, at ginseng concentration of 0.001, 0.01, 0.1 and 1 mg/mL were $0.89\pm0.04$, $0.86\pm0.06$, $0.73\pm0.01$ and $0.09\pm0.02$, respectively. Survival fraction at X-irradiation alone of 2, 4, 6 and 8 Gy were $0.81\pm0.07$, $0.42\pm0.08$, $0.15\pm0.02$, $0.03\pm0.01$, respectively. But, suwival fraction in combined group of X-irradiation and ginseng (0.2mg/ml) at each same radiation dose were $0.28\pm0.01$, $0.18\pm0.03$, $0.08\pm0.02$, $0.006\pm0.002$, respectively (p<0.05). Conclusion : The yield for ginseng extract which was treated with freezing drier was $6.3\%$. Cytotoxicty of Fsa 11 in combined ginseng with X-irradiation group was increased than that of X-irradition alone group, and its enhancing effect seemed to be added.

Quality Characteristics of Soybean Curd Mixed with Freeze Dried Onion Powder (동결건조 양파분말을 첨가한 두부의 품질특성)

  • Soybean curd was mixed with onion powder to develop new foods, and changes in quality characteristics were investigated. The moisture content of onion soybean curd rose as the proportion of onion powder increased Whiteness (as measured by the L value) was high in soybean cud admixed with 0.1% (w/v) onion powder, Redness (the a value) was not significantly altered (the readings were 1.03-1.54) on addition of various onion powder concentrations. Yellowness (the b value) was similarly unaffected (readings 13.00-13.93) when various levels of onion powderwere added. Free sugar analysis showed that glucose was high in soybean curd (67.22 g/100 g) admixed with 0.1% (w/v) onion powder, The main organic acid was tartaric acid, and control organic acids included citric and oxalic acids at high levels. The major free amino acids were L-arginine, ${\gamma}-Amino-n-butyric$ acid, L-histidine, L-glutamic acid, L-serine, L-tyrosine and L-threonine, and amino acid content were high in soybean curd admixed with 0.2% (w/v) onion powder. Major phenolic compounds of onion soybean curd were quercitrin, protocatechuic acid and caffeic acid. The hardness of onion soybean curd was similar to that of the control when onion powder was added to 0.1% or 0.2% (w/v), and decreasedmore onion powder was added. Organoleptic qualities dropped as onion powder levels increased. In summary, onion powder addition to soybean curd is optimal at the 0.2% (w/v) level..

Antioxidant Activities of Extracts from Fermented Mulberry (Cudrania tricuspidata) Fruit. and Inhibitory Actions on Elastase and Tyrosinase (꾸지뽕열매 발효 추출물의 항산화 특성과 Elastase 및 Tyrosinase 저해활성)

  • We evaluated the nutritional value of 70% ethanol extracts (EE) from fermented cudrania tricuspidata fruit (ECT: EE of C. tricuspidata, EFCT: EE of fermented C. tricuspidata, EFCTL: EE of fermented C. tricuspidata by B. licheniformis, EFCTS: EE of fermented C. tricuspidata by B. subtilis) by determined the total polyphenol and flavonoid contents, antioxidant activities, and inhibitory actions on elastase and tyrosinase. The yields of freeze-dried powder of ECT, EFCT, EFCTL, and EFCTS were 54.22%, 54.43%, 57.51%, and 57.23% (each w/w), respectively. The color of $L^*,\;a^*,\;b^*$ values decreased of fermented cudrania tricuspidata. The fermented EFCTL and EFCTS had relatively higher contents of total polyphenol and flavonoid than those of other extracts. ABTS radical scavenging ability were 94.61%, 95.85%, 94.36%, and 96.69%, respectively. SOD (superoxide dismutase)-like activities were in the order EFCT (32.60%) > EFCTS (27.10%) > EFCT (23.30%) > ECT (22.00%), and nitrite scavenging activity was in order of EFCTS (51.18%) > EFCTL (45.61%) > EFCT (41.93%) > ECT (27.76%), respectively. Ferrous ion chelating activity of EFCTL (67.34%) and EFCTS (60.36%) was significantly the highest, whereas ECT (52.34%) and EFCT (51.73%) had not different significantly. Xanthine oxidase, elastase and tyrosinase inhibitory activities at 0.5% (w/v) solutions of EFCTL and EFCTS were somewhat higher than those of non-fermented ECT. In conclusion, we provide experimental evidence that extracts of fermented cudrania tricuspidata of B. subtilis and B. licheniformis exhibited higher antioxidant activities and inhibitory actions on elastase and tyrosinase compared with non fermented cudrania tricuspidata.

The Effect of Platelet Derived Growth Factor - BB Loaded Chitosan/Calcium Metaphosphate on Bone Regeneration (혈소판유래성장인자를 함유한 Chitosan/Calcium Metaphosphate의 골조직재생효과에 관한 연구)

  • Chitosan is biodegradable natural polymer that has been demonstrated its ability to improve wound healing, and calcium metaphosphate(CMP) is a unique class of phosphate minerals having a polymeric structure. In this study, chitosan/CMP and platelet derived growth factor(PDGF-BB) loaded chitosan/CMP sponges were developed, and the effect of the sponges on bone regeneration and their possibility as scaffolds for bone formation by three-dimensional osteoblast culture were examined. PDGF-BB loaded chitosan/CMP sponges were prepared by freeze-drying of a mixture of chitosan solution and CMP powder, and soaking in a PDGF-BB solution. Fabricated sponge retained its 3-dimensional porous structure with $100-200\;{\mu}m$ pores. The release kinetics of PDGF-BB loaded onto the sponge were measured in vitro with $^{125}I-labeled$ PDGF-BB. In order to examine their possibility as scaffolds for bone formation, fetal rat calvarial osteoblastic cells were isolated, cultured, and seeded into the sponges. The cell-sponge constructs were cultured for 28 days. Cell proliferation, alkaline phosphatase activity were measured at 1, 7, 14 and 28 days, and histologic examination was performed. In order to examine the effect on the healing of bone defect, the sponges were implanted into rat calvarial defects. Rats were sacrificed 2 and 4 weeks after implantation and histologic and histomorphometrical examination were performed. An effective therapeutic concentration of PDGF-BB following a high initial burst release was maintained throughout the examination period. PDGF-BB loaded chitosan/CMP sponges supported the proliferation of seeded osteoblastic cells as well as their differentiation as indicated by high alkaline phosphatase activities. Histologic findings indicated that seeded osteoblastic cells well attached to sponge matrices and proliferated in a multi-layer fashion. In the experiments of implantation in rat calvarial defects, histologic and histomorphometric examination revealed that chitosan/CMP sponge promoted osseous healing as compared to controls. PDGF-BB loaded chitosan/CMP sponge further echanced bone regeneration. These results suggested that PDGF-BB loaded chitosan/CMP sponge was a feasable scaffolding material to grow osteoblast in a three-dimentional structure for transplantation into a site for bone regeneration.
