Improved Method for the Preparation of Crude Ginseng Saponin (인삼 조사포닌의 조제 방법 개선)

  • This stuffy was carried to establish a new efficient method for the preparation of edible crude ginseng saponin. The conventional butanol extraction and resin adsorption methods were compared for the contents of total crude ginseng saponin and major ginsenosides. Seventy- percent methanol extract was applied to Diaion HP-20 column and the resin was washed with Hn and eluted with absolute methanol. The methanol elute was dried in vivo and analyzed for its ginsenosides. Use of ethanol instead of methanol to make edible crude ginseng saponin gave a similar result. Butanol extraction was performed by the conventional method. The final aqueous layer from butanol extraction was passed through Diaion HP-20 column followed by elution with methanol and Diaion HP-20 passed fraction was extracted with butanol to recover remaining components, respectively, in order to determine saponin loss. TLC and HPLC qualitatively and quantitatively monitored Ginsenosides, respectively. Loss of ginsenosides was higher in butanol extraction method than in Diction HP-20 adsorption method. In addition, saponin fractions prepared by Diction HP-20 adsorption method showed higher content of each ginsenoside, showing 8.2% higher purity than that of butanol extracted fraction. From these results, we propose the resin adsorption method as a new efficient measure for the preparation of crude ginseng saponin, which is edible by using spirit instead of methanol.

A Study on Quantitative Measurements of Equivalence Ratio in Constant Volume Chamber Using UV Laser Raman Scattering (UV Laser Raman Scattering을 이용한 정적 연소기내 분사된 연료의 정량적 당량비 측정에 관한 연구)

  • Laser Raman scattering method has been applied to measure equivalence ratio of methane/air and propane/air mixture in constant volume combustion chamber. We used high power KrF excimer laser$(\lambda=248nm)$ and a high gain ICCD camera to capture low intensity Raman signal. Raman shifts and Ram cross-sections of $H_2,\;O_2,\;N_2,\;CO_2,\;CH_4\;and\;C_3H_8$ were measured precisely. Our results showed an excellent agreement with other groups. Mole fraction measurement of $O_2\;and\;N_2$ from air showed that $O_2\;:\;N_2$ = 0.206 : 0.794. We used constant volume combustion chamber and gas injector which is operated at $5\sim10barg$. Methane and propane are used as a fuel. 50 Raman signal are obtained and ensemble averaged for measurement of equivalence ratio. Our measured results showed that the equivalence ratio of fuel/air mixture is reasonable at ${\pm}5%$ error range.

Cosmological Parameter Estimation from the Topology of Large Scale Structure

  • The genus of the matter density eld, as traced by galaxies, contains information regarding the nature of dark energy and the fraction of dark matter in the Universe. In particular, this topological measure is a statistic that provides a clean measurement of the shape of the linear matter power spectrum. As the genus is a topological quantity, it is insensitive to galaxy bias and gravitational collapse. Furthermore, as it traces the linear matter power spectrum, it is a conserved quantity with redshift. Hence the genus amplitude is a standard population that can be used to test the distance-redshift relation. In this talk, I present measurements of the genus extracted from the SDSS DR7 LRGs in the local Universe, and also slices of the BOSS DR12 data at higher redshift. I show how these combined measurements can be used to place cosmological parameter constraints on m, wde.

  • The assay on microbial dehydrogenase activity by using TTC was investigated. The experimental results are summarized as follows: 1. The TF production was affected by dissolved oxygen. Dissolved oxygen in samples could be effectively removed by adding $Na_{2}SO_{3}$ as reductant prior to incubation. 2. The toxic effect on TF formation was observed at up to 0.1% of final TTC concentration when VSS concentration was 2.7g/l. As the VSS concentration increased the effect decreased. 3. It was clearly revealed that the TF generation rate decreased with increasing VSS concentration. The dilution of sludge could be applicable when VSS level is below 3.5g/l. 4. The TF production of endogenous phase (De) could be used as a measure for the viable fraction of sludge over various sludge ages. The additional TF production (Ds) due to extracellular substrate responded to De, not to VSS concentration.

  • This study was performed to evaluate contamination degree of heavy metals in sediments of Han River, compared with other nation's evaluation method. The results were as follows 1. The contamination Ratio calculation method by heavy metal concentration in differnt fraction size has a limitation to apply to all of the areas of Han river because of its characteristics of sediment. As a result, this method applied to only 4 areas of Pal Dang, Wang Sook Chon, Uk Chon, and Bul Kwang Chon, and Contmination Ratio of heavy metals in those areas were relatively low of below 3. So it's considered that those areas have less contaminated from anthropogenic contaminants. 2. The Contamination Ratio calculation method by heavy metal concentration in different areasthat of upper area to be background level-has a limitation also to apply to Han river. But it is considered that this method was relatively suitable to apply, so it should be prepared evaluation standand method for them. Contamination ratio from background level as Pal Dang area were most high in An Yang Chon. So it must be prepared purification and control measure at An Yang Chon.

Analysis of soil coarse pore fraction by major factors for evaluation of water conservation function potential in forest soil (산림토양의 수원함양기능 잠재력 평가를 위한 주요 인자별 토양 조공극률 분석)

  • As the water shortage has become a noticeable issue due to climate change, forests play an importance role as the provider of water supply service. There is, however, little information about the relationships between the factors used in the estimation of water supply service and coarse pore fraction of forest soil which determines the potential of water supply. To find out whether there would be an amelioration in the scoring system of water supply service estimation, we examined all factors except meteorological one and additionally, analyzed 4 extra factors that might be related with coarse pore fraction of soil. A total of 2,214 soil samples were collected throughout South Korea to measure coarse pore fractions from 2015 to 2020. First, the result of average coarse pore fraction of all samples showed 32.98±6.59% which was consistent with previous studies. And the results of non-parametric analysis of variance indicated that only two of eleven factors that was used in the scoring system matched the results of coarse pore fraction of forest soils. Tree canopy coverage showed no difference among categories, and slope also showed no significance at level of 0.05 in the linear regression analysis. Additionally, the applicability of 4 extra factors were confirmed, as the result of coarse pore fractions of soil samples were different for various categories of each factor. Therefore, the scoring system of water supply service of forest should be revised to improve accuracy.

  • The ultraviolet(UV)-curable materials wildly have been used as a ink and coating materials. A according to measure UV polymerization phase separation of acryl groups 2,3,4-functional monomer, a few experiments were performed gel fraction, dynamic viscosity of alkyd resin blend system. Dependence of photoiniator concentration and various monomer, alkyd resin content investigated structural changes of films cured by UV irradiation. Curing rate measured UV irradiation under $25^{\circ}C$, cell gap 7cm and film thickness 100${\mu}{\textrm}{m}$. In results, it was through that the viscoelastic properties of films cured with increasing the ratios of monomer/alkydresin contents are network polymer in properties such as viscoelasticity.

Screening of Antioxidation-related Functional Components in Brans of the Pigmented Rices (유색미 겨에 함유된 항산화 효과 관련 기능성 성분의 검정)

  • Lipid-soluble fractions extracted with methylene chloride and ethyl acetate as well as 70% ethanolic extract were made from 5 cultivars of the pigmented rice brans (DZ 78, Elwee, LK1A-2-12-1-1, LK1-3-6-12-1-1 and Killimhukmi) to determine their antioxidative activities and to measure the contents of phenolic compounds, phytic acid, together with unsaponificated compounds such as retinol, tocopherol and stigmasterol. We also attempted to characterize candidate components in charge of antioxidation and measure their relative contents in each methylene chloride and ethyl acetate fraction by using GC-MS techniques. As the results, we found a 30% higher antioxidative activity with 26-fold greater phenolic compound content of the pigmented rices as compared to those of the normal rice. Especially, in Killimhukmi, the contents of the unsaponificated compounds such as retinol, tocopherol and stigmasterol were also found to be more than 2.3-fold greater than those of normal rice. However, we could not find the overall correlations between antioxidativity of rices and their color values, or unsaponificated compounds contents. Methylene chloride fraction from Killimhukmi and LK1A-2-12-1-1, both showing strong antioxidativity, contained relatively high contents of (Z)-9-hexadecenyl ester-9-octadecanoic acid and 2-Hydroxyl-1-1(hydroxymethyl) ethyl-9-octadecanoic acid, respectively. In ethyl acetate fractions, on the contrary, 1H indene, methyl ester-tetradecanoic acid and methyl ester-hexadecanoic acid were detected as the commonly-occurring substances in the 5 cultivars of pigmented rices, implying a possible role in antioxidative action.

  • A method, so called 'successive cracking method,' for measuring residual stresses in a circular disk is proposed. In this method residual stresses are evaluated using a fracture mechanics approach, that is, the strains measured at a point on a edge of the disk as a crack is introduced and extended from the edge are used to deduce the residual stress distribution which existed in the uncracked disk. Through finite element analysis and comparative experiments with generally used sectioning method, the successive cracking method is shown to be valid, simple and effective to measure 2-dimensional residual stress distribution in a circular disk.

  • Shape memory is physical phenomenon which a platically metal is restored to its original shape by a solid state phase change by heating. TiNi alloy the most effective material in the shape memory alloy(SMA). To study(measure) recovery stress of intelligent composite. Ti50-Ni50 shape memory matrix with prestrain SMA fiber. When SMA fiber of the intelligent composite is heated over austenite starting temperature(As) by electric heating. a recovery stress are generated. The recovery stress of the intelligent composite was measured by strain gage or photoelastic experiment. Measuring method of recovery stress by photoelastic experiment was developed in this research. It was certified that photoelastic experiment was more effective and more precise than strain gage method in the measurement of recovery stress.
