Lactic Acid Fermentation in Soymilk by Single and Mixed Cultures of Lactobacillus Casei and Kluyveromyces fragilis (Lactobacillus casei IFO 3012 와 Kluyveromyces fragilis KFCC 35458의 혼합배양에 의한 두유의 젖산발효)

  • Lactobacillus casei IFO 3012 and Kluyveromyces fragilis(KFCC 35458) were cultured together in Soymilk to investigate the growth characteristics and the conditions suitable for acid Production. L. casei produced more amount of acid rapidly when cultured with K. fragilis in soymilk than when cultured singly. The optimum conditions for acid production by the mixed cultures of L. casei and K. fragilis were achieved with a temperature of $35-37^{\circ}C$, a 1:5-1:9(O.D 660) ratio of L. casei to K. fragilis at inoculum, a 1.0 level of sucrose fortification or a 2.0% level of skim milk powder fortification and a culture time of 24hr. Under these conditions the amounts of acid produced by the single culture of L. casei and the mixed cultures with K. fragilis were 0.31% and 0.44% in soymilk, 0.43% and 0.97%, respectively, in soymilk fortified with 1.0% level of sucrose. These indicate that the amount of acid produced by mixed cultures is about 1.42 fold greater in soymilk and about 2.26 fold greater in soymilk fortified with 1.0% level of sucrose than that produced by the single culture of L. casei. The amount of acid produced in soymilk fortified with 2.0% level of skim milk powder was 1.0 level for both of the single culture of L. casei and the mixed cultures of L. casei and K. fragilis after 24hr incubation. In soymilk fortified with skim milk power less than 1.5 the mixed culture with K. fragilis showed higher content of acid than the single culture of L. casei only.

A Study on the Bioavailability of Calcium in Eggshell Powder Fortified Ramen in the Growing Rats (성장기 쥐에서 난각분 강화라면의 칼슘 이용성에 관한 연구)

  • This study has investigated the bioavailability of calcium in eggshell powder (ESP) fortified ramen. Wean-ling SD rats were maintained for 2 weeks on 0.05$\%$ low calcium diet then assigned randomly to one of 5 groups. Animals were provided for 4 weeks one of 3 different experimental diets containing 0.15$\%$ calcium as $CaCo_{3}$, NFDM, or ESP fortified ramen. Two control groups were fed either 0.05$\%$ calcium or general ramen containing 0.08$\%$ calcium. The body weight, diet intake, food efficiency ratio (FER), bone growth, calcium contents of bones, and apparent absorption were measured. Experimental results show that regardless of calcium sources 0.15$\%$ calcium groups increased body weight, weight and length, calcium content, and the strength of two bones (tibia and femur) significantly compared to 0.05$\%$ calcium group. The apparent absorption rate of calcium also showed similar results supporting the bioavailability of ESP fortified ramen is not inferior to either NFDM or $CaCO_{3}cdot$ The results indicate that ESP fortified ramen is a proper mediate for calcium fortification in Korea.

Effects of Various Polysaccharides on the Physicochemical Properties of the Dextran Culture Containing Carrot Juice Residue Obtained from Submerged Culture Using Leuconostoc citreum S5 (다당류 첨가에 따른 당근박 함유 Leuconostoc citreum S5 발효물의 물성변화)

  • The physicochemical properties of dextran culture fermented by Leuconostoc citreum S5 were evaluated by the addition of various polysaccharides and heat-treatment. The consistency of dextran culture increased with the addition of carrot juice residue (CJR) in submerged culture, resulting in the highest consistency of 150 $Pa{\cdot}s^n$ and viable cell counts of $2.36{\times}10^9$ CFU/mL at 20% level of CJR. The dextran culture showed the pseudoplastic behavior and its consistency was greatly increased with the addition of various polysaccharides at 2% level. Addition of glucomannan indicated the highest consistency of 1275 $Pa{\cdot}s^n$ and their heat-treatment resulted in the increase of consistency except for glucomannan. After heat-treatment, the fermented dextran culture containing CJR fortified with gellan gum and carrageenan showed great change in rheology, indicating the highest consistency and hardness value resulted in the great increase of elastic and viscous moduli. The dynamic viscoelastic properties of dextran culture were greatly increased by the heat-treatment after fortification of various polysaccharides. Thus, the consistency and viable cell counts of dextran culture were increased with the addition of CJR. The rheological properties of dextran culture were manipulated by the fortification of various polysaccharides and heat-treatment.

Characterization of the Functional Properties of Soy Milk Cake Fermented by Bacillus sp.

  • The mucilage production and tyrosine content in soy milk cake (SMC) fermented by Bacillus firmus NA-1, Bacillus subtilis GT-D, and B. subtilis KU-A was improved by fortification with 10% defatted soybean flour. The fibrinolytic activity and consistency of the SMC were drastically increased by solid-state fermentation for 1 day. However, the consistency of the fermented SMC gradually decreased during fermentation for 3 days. Furthermore, the tyrosine content of the freeze-dried powder of SMC fermented by three Bacillus sp. was 9 times higher than that of unfermented SMC. The soybean proteins, including the 7S and 11S subunits, were partially digested during alkaline fermentation, producing lower molecular-weight peptides. The fibrinolytic enzyme produced in SMC fermented by B. firmus NA-l and B. subtilis KU-A exhibited higher thermal stability than that of B. subtilis GT-D fermentation. The powder obtained from B. subtilis GT-D fermentation had an ${\alpha}$-amylase activity and lower consistency compared to those of B. firmus NA-1 and B. subtilis KU-A. In addition, this powder contained 6.3% moisture content, 27% crude protein content and 9 units of fibrinolytic activity and proteolytic activity.

Dried Sugarcane Press Residue as a Potential Feed Ingredient Source of Nutrients for Poultry

  • Sugarcane press residue (SPR), a byproduct from the sugar industry was evaluated for it's nutrient and energetic quality in broilers and layers. The composition of SPR included (% DM): CP-11.76 (methionine-2.21, cystine-1.05, lysine-4.85, threonine-5.48% of CP), EE-7.87 (palmitic acid-30.3, stearic acid-4.1, oleic aicd-17.2, linoleic acid-38.0, linolenic acid-5.4% of EE), CF-10.08, TA-21.08 (Ca-3.87, P-1.10, Mg-0.95%, Fe-3500, Mn-284, Zn-113, Cu-61.5, Co-5.0 ppm and AIA-4.93%) and NFE-48.35% indicating that SPR is a valuable source of both organic and inorganic nutrients for poultry. The metabolic trials revealed the average ME of SPR as 749, 842 and 1,270 kcal/kg, respectively in broilers and 844, 936 and 1,031 kcal/kg in layers, at 10, 20 and 30% inclusion levels, respectively. Further, the fortification of SPR incorporated diets with biotechnological products viz., lipid utilizing agents (lipase and lecithin) or NSP degrading enzymes and their combination did not improve the ME content of such diets.

Seismic Performance Evaluation of Unreinforced Masonry Buildings Retrofitted by Strengthening External Walls (외부벽체 강도증진형 보강이 적용된 비보강 조적조 건물의 내진성능평가)

  • Nonlinear static analysis and preliminary evaluation were performed in this study to evaluate the seismic performance of unreinforced masonry buildings subjected to various soil conditions based on the revised Korean Building Code. Preliminary evaluation scores and nonlinear static analyses indicated that all buildings were susceptible to collapse and did not reach their target performance. Therefore, retrofit of those building models was carried out through a systematic procedure to determine areas to be strengthened. It was possible to make most building models satisfy performance objectives through the reinforcement alone of damaged external shear walls. However, the application of a preliminary evaluation procedure to retrofit design was found to be too conservative because all the retrofitted building models verified with nonlinear static analysis failed to satisfy performance objectives. Therefore, it is possible to economically retrofit unreinforced masonry buildings through the fortification of external walls if a simple evaluation procedure that can efficiently specify vulnerable parts is developed.

Surrounding rock pressure of shallow-buried bilateral bias tunnels under earthquake

  • By means of finite element numerical simulation and pseudo-static method, the shallow-buried bilateral bias twin-tube tunnel subject to horizontal and vertical seismic forces are researched. The research includes rupture angles, the failure mode of the tunnel and the distribution of surrounding rock relaxation pressure. And the analytical solution for surrounding rock relaxation pressure is derived. For such tunnels, their surrounding rock has sliding rupture planes that generally follow a "W" shape. The failure area is determined by the rupture angles. Research shows that for shallow-buried bilateral bias twin-tube tunnel under the action of seismic force, the load effect on the tunnel structure shall be studied based on the relaxation pressure induced by surrounding rock failure. The rupture angles between the left tube and the right tube are independent of the surface slope. For tunnels with surrounding rock of Grade IV, V and VI, which is of poor quality, the recommended reinforcement range for the rupture angles is provided when the seismic fortification intensity is VI, VII, VIII and IX respectively. This study is expected to provide theoretical support regarding the ground reinforcement range for the shallow-buried bilateral bias twin-tube tunnel under seismic force.

  • As the forefront of structural design method, capacity spectrum method can be applied conveniently, and through this method, deformation demand of structure can be considered. However, there is no research for the seismic application in the structure of sheet pile retaining wall to report. Therefore, focusing on laterally loaded stabilizing sheet pile wall, which belongs to flexible cantilever retaining structure and meets the applying requirement of capacity spectrum method from seismic design of building structure, this paper studied an approach of seismic design of sheet pile wall based on capacity spectrum method. In the procedure, the interaction between soil and structure was simplified, and through Pushover analysis, seismic fortification standard was well associated with performance of retaining structure. In addition, by comparing the result of nonlinear time history analysis, it suggests that this approach is applicable.

  • Purpose: COVID-19 infections have been erupting in places of worship, long-term care facilities, and call centers in Korea since January 2020. This study aims to diagnose and present an infection control system solution for long-term care facilities where at-risk elderly individuals are actively engaged in communal life. Methods: We conducted comparative analyses of infection control systems between long-term care facilities and medical institutions respective of relevant laws and this study's evaluation system. Results: To prepare for future infectious diseases, it is necessary to establish a long-term care facility infection control system and strengthen the standards thereof, to strengthen long-term care facility evaluation standards and to newly establish medical charges for infection control. Conclusion: Systematic procedure fortification and financial support provisions are necessary for infection control at long-term care facilities.

A study on the determination of residual Antibiotics and Synthetic Antibacterial Agents in Meat(III) Simultaneous Gas Chromatography/Mass Spectrometry Analysis of Erythromycin and Tylosin (식육중의 잔류 항생.항균제의 검정에 관한 연구(III) Macrolide계 항생물질인 Erythromycin과 Tylosin의 Gas Chromatography/Mass Spectrometry 동시분석)

  • In an attempt to quantitate and qualitate residual antibiotics and antibacterial agents n meat simultaneously, we studied a gas chromatogrphy-mass spectrometry (GC/MS) analysis. For a simultaneous analysis of macrolide antibiotics such as erythromycin and tylosin in meat, the homogenization with MeOH, defatting with n-hexane, extraction with CHCl3, elution with CHCl3 : MeOH=2:1 from Sep-Pak silica cartridge, acid gydrolysis, back extraction with CHCl3, and quantitation by selected ion monitoring(SIM) mode after trimethylsilyl derivatization were performed. The recoveries of erythromycin and tylosin (CV,%) at 10 ppm fortification level were 90.59(4.89) and 45.91(0.20) , and the detection limits of those were 0.02 and 2.0 $\mu\textrm{g}$/g beef, respectively. From these results, the developed analytical method using GC/MS-SIM mode allows excellent detection and quantitation of residual macrolide antibiotics in meats, using complementary method with bio-assay.
