방사무늬김(Porphyra yezoensis)에서 추출한 Porphyran 효소 분해물의 화학적 결합 특성 (Structural Features of Enzymatic Hydrolysate of Porphyran Isolated from Porphyra yezoensis)

  • Enzymatic hydrolysate of porphyran from Porphyra yezoensis was prepared by treatment with ${\beta}$-agarase. The hydrolysate was fractioned into molecular sizes of <3, 3-30, and 30-300 kDa using an ultrafiltration membrane. The membrane fractions were further separated into neutral and anionic fractions using Dowex $1{\times}8$ ion exchange chromatography. After hydrolysis of porphyran with ${\beta}$-agarase, 23.2% of the starting porphyran was recovered as a neutral fraction of low-molecular weight (<3 kDa), and 28.9% remained as an enzyme-resistant anionic fraction of high molecular weight (>300 kDa). Desulfation of porphyran and $^{13}C$-NMR analysis of the anionic fraction of low molecular weight (<3 kDa) showed that the anionic fraction has a backbone consisting of 3-linked ${\beta}$-D-galactose units alternating with either 4-linked a-L-galactose 6-sulfate or 3, 6-anhydro-a-L-galactose units. These results indicate that porphryan is a copolymer of two moieties, about 25% of which are composed of neoagarose moieties and 75% as anionic moieties.

택사(Alismatis Rhizoma) Hemagglutinating Protein의 정제와 특성 (Purification and Characterization of Hemagglutinating Protein from Rhizome of Alisma orientale)

  • 택사로부터 황산암모늄 분별 침전, DEAE-cellulose ion exchange chromatography, Sephadex G-150 chromatography 등의 방법을 이용하여 렉틴을 분리, 정제하였다. 정제한 렉틴의 분자량은 gel filtration법에 의해 측정한 결과 90,500 dalton이었으며, SDS polyacrylamide gel 전기영동을 실시한 결과 subunit 분자량은 각각 42,000, 27,000, 22,500 dalton으로 나타나므로 택사 렉틴이 heterotrimer임을 알았다. 정제된 택사 렉틴은 사람 적혈구의 경우 모든 혈액형에 대하여 응집현상을 나타내었으며 돼지, 쥐 및 개 등의 적혈구에 대해서도 모두 응집현상이 나타나 혈구 비특이성임을 보여주었다. 또한 이 렉틴은 sialic acid, glucose, ribose, sucrose, lactose, galactose 등 당에 의해서, 그리고 $Hg^{2+},\;Fe^{2+},\;Cu^{2+}$ 이온 등에 의해 적혈구 응집력이 저해되었다.

Streptococcus faecium M74 · LC에 의한 L-α-Glycerophosphate Oxidase의 생산 (Production of L-α-Glycerophosphate Oxidase by streptococcus faecium M74 · LC)

  • L- $\alpha$ -glycerophosphate oxidase(GPO)는 L- $\alpha$-glycero-phosphate를 산화시켜 dihydroxyacetone phosphate와 과산화수소의 생성을 촉매시켜 주는 효소로서 혈중에 존재하는 triglyceride의 양을 측정하는 kit를 개발하는데 이용할 수 있으며 본 연구 결과로 종균을 개발 및 확보하였으며, 이 종균의 사용으로 GPO의 발효공정기술을 확립하였고, 또한 이들 조효소로부터 순수한 효소를 생산할 수 있는 정제공정 기술을 확립하였다. 먼저 ATCC에서 네 가지 균주와 KCTC에서 세 가지 균주, 그리고 본 연구실에서 분리한 Streptococcus faecium $M_{74}$.LC 균주 등 8가지 균주의 성장상태를 비교하고 GPO의 역가를 측정 해 본 결과, 배양액 1 L당 ATCC 19634는 65 units, ATCC 12755는 60 units, S. faecium $M_{74}$. LC는 67 units로 S. faecium $M_{74}$.LC가 제일 높은 역가를 생산하였다. 또한 발효조에서 배지의 양을 3 L로 하여 배양온도는 $37^{\circ}C$, 교반속도는 300 rpm, 통기량은 0.5 L/min, 17시간 배양하였을 때 가장 많은 양의 균체가 생성되었으며, GPO의 생산량도 가장 많았다(256 units/L). 이 때 배지 조성은 glucose 0.1%, glycerol 0.2%, tryptone 1.0%, yeast extract 1.0% 및 $K_2HP0_4$0.5%로 하여 배양하였고 GPO의 정제공정은 염투석 분획과 이온 교환수지 공정으로 대별할 수 있으며, 이온교환수지는 DEAE-cellulose 칼럼을 이용한 정제수율은 약 47%를 얻을수 있었다.

영양상담이 제2형 당뇨병 환자의 혈당과 식사요법에 대한 지식과 실천에 미치는 영향 (Effects of Nutrition Counseling on Diabetes Management in Type 2 Diabetes Mellitus Patients)

  • The purpose of this study was to investigate the effects of nutrition counseling on diabetes management by determining changes in anthropometry and blood components as well as knowledge and practice of diet therapy and nutrient intake in 34 (male 11, female 23) type 2 diabetes patients. The knowledge and the practice of diet therapy, drinking, smoking and exercise were analyzed by questionnaires. Dietary nutrient intake were obtained from the patients by the 1 day 24-hr recall. Blood glucose level and blood pressure were measured before and 3 months after the treatment. The results are summarized as follows: Average weight (p<0.05) and body mass index (p<0.05) were significantly lower post-counseling. Fasting blood glucose levels (p<0.01) and postprandial-2hour blood glucose levels (p<0.01) were also significantly lower post-counseling. In lifestyle changes for self-management the patients showed significantly higher exercise habits post-counseling (p<0.01). Regarding their level of diet knowledge, they showed significantly higher levels post-counseling in six items such as importance of diet therapy for diabetes (p<0.001), principles of diet therapy (p<0.001), nutrient composition of foods (p<0.01), carbohydrate composition of foods (p<0.001), the prescribed calories (p<0.001) understanding food item and exchange units of cereals, grains (p<0.001) and fruits, juices (p<0.001). Regarding their diet practices, the patients showed significantly higher levels of practice post-counseling in keeping within permitted meal size (p<0.001), using food exchange lists (p<0.001), keeping exact meal times (p<0.01), and controling sweet foods (p<0.001). Protein (p<0.05), animal lipid (p<0.05), and vitamin C (p<0.05) intakes were significantly higher post-counseling.

Purification and Characterization of α-Galactosidase from Lactobacillus salivarius subsp. salivarius Nam27

  • Lactobacillus salivarius subsp. salivarius CNU27 possessed a high level of ${\alpha}$-galactosidase activity. Purified ${\alpha}$-galactosidase was obtained after sonication of harvested cell pellet followed by DEAE-Sephadex A-50 and Mono Q anion exchange chromatography. The specific activity of the purified enzyme was 8,994 units/mg protein which is 17.09 times higher than that in crude extract. The native enzyme was a monomer with a molecular mass of 56,397.1 dalton. The optimum temperature and pH for the enzyme were $40^{\circ}C$ and 6.0, respectively. The enzyme was stable between 25 and $50^{\circ}C$. However, ${\alpha}$-galactosidase activity was lost rapidly below pH 4.5 and above pH 8.5. The enzyme activity decreased to 6.73% and 4.30% of the original activity by addition of $Cu^{2+}$ and $Hg^{2+}$, respectively. Other metal compounds did not affect the enzyme activity significantly. The enzyme liberated galactose from melibiose, raffinose, and stachyose. The rate of substrates hydrolysis was measured by HPLC. Raffinose, stachyose and melibiose were completely decomposed after 24 hr at $40^{\circ}C$.

Molecular Typing in Public Health Laboratories: From an Academic Indulgence to an Infection Control Imperative

  • Using three Austrian case studies, the variegated applications of molecular typing in today's public health laboratories are discussed to help illustrate preventive management strategies relying on DNA subtyping. DNA macrorestriction analysis by pulsed field gel electrophoresis has become the gold standard for subtyping of food borne pathogens like listeria, salmonella, campylobacter and Bacillus cereus. Using a Salmonella Mbandaka outbreak from the year 2010 as example, it is shown how the comparison of patterns from human isolates, food isolates, animal isolates and feed isolates can allow to identify and confirm a source of disease. An epidemiological connection between the simultaneous occurrence of tuberculosis in cattle and deer with cases of human tuberculosis due to Mycobacterium caprae in 2010 was excluded using mycobacterial interspersed repetitive units variable-number tandem repeats subtyping. Also in 2010, multilocus sequence typing with nonselective housekeeping genes, the so-called sequence based typing protocol, was used to elucidate connections between an environmental source (a hospital drinking water system) and a case of legionellosis. During the last decades, molecular typing has evolved to become a routine tool in the daily work of public health laboratories. The challenge is now no longer to simply type microorganisms, but to type them in a way that allows for data exchange between public health laboratories all over the world.

식용달팽이 β-Galactosidase의 정제와 생화학적 특성 (Purification and Characterization of the β-Galactosidase from Edible Snail)

  • 달팽이 내장을 마쇄, 추출, 염석 및 투석 후 얻은 조효소액을 두 번의 이온교환 크로마토그래피 및 두 번의 겔여과 크로마토그래피를 거쳐 최종적으로 얻은 $\beta$-galactosidase의 비활성도는 18.8 units/mg protein이었고, 정제도는 31.3%배, 수율은 20.8%이었다. $\beta$-Gallactosidase의 분자량을 측정하기 위해 겔 여과와 전기영동을 실시한 결과, native molecular weight가 약 144,000으로, SDS-PAGE 결과, 약 72,000으로 나타나 이 효소는 동일한 polypeptide로 구성된 dime로 추측되었으며, 등전점은 pI 4.1이었다. $\beta$-Galactosidase의 최적 pH와 온도는 각각 pH 3.0과 6$0^{\circ}C$로 측정되었으며, pH 2.0~8.0에서 80%이상의 효소 활성을 나타내었고, 온도 30~5$0^{\circ}C$에서 안정되었다. 모든 금속이온과 fructose, glucose, galactose, maltose 및 xylose는 $\beta$-galactosidase의 저해제로 작용하였다.

해양 미생물Algibacter lectus AS-3으로부터 agarase의 분리 및 특성 (Purification and Characterization of Agarase from Marine Bacterium, Algibacter lectus AS-3)

  • Agar로부터 oligosaccharides를 제조하기 위하여 효소 (agarase)를 이용한 분해법이 시도되면서 agarase의 균주의 분리 및 유전자에 대한 많은 연구가 수행되고 있다. 본 연구에서는 해양으로부터 agarase를 생성하는 균주를 분리 동정하고 균주의 특성을 조사하였다. 분리된 균주는 AS-3균주는 16S rDNA 염기서열분석에 의하여 Algibacter lectus AS-3로 동정되었다. 정제된 효소의 최적 활성 조건 및 agar 분해산물의 항산화활성을 조사하였다. 분리된 균주의 최적배양조건은 marine broth 2216에서 온도 $27^{\circ}C$, pH 7일 때 가장 균주의 생육이 높았다. Agarase 효소는 salt 침전, ion exchange와 gel filtration chromatography에 의해 9.6 units/mg으로 6.9배 정제되었다. 최적 효소활성은 온도 $40{\sim}50^{\circ}C$, pH 7일 때 나타났다. Agar 첨가한 배지에 agar 분해균을 접종한 후 분해산물의 시간에 따른 항산화활성은 12시간 배양 후 62%의 가장 높은 전자소거능 (EDA)을 나타내었다.

Carbohydrate counting 을 이용한 제2형 당뇨병 환자의 식사 관리 (The Meal Management of Korean Type 2 Diabetes Patients Using Carbohydrate Counting)

  • Carbohydrate(CHO) counting is a meal planning approach used with diabetic patients that focuses on carbohydrate as the primary nutrient affecting post-prandial glycemic response. However, it has not been used in meal management of diabetic patients in Korea. CHO counting can be used by clients with type 1 and 2 diabetes. The purpose of the study was to determine the barriers to utilize the CHO counting when three levels of CHO counting were educated to type 2 diabetic patients who started continuous subcutaneous insulin infusion (CSⅡ) therapy by nutrition lectures and counseling. And the CHO-to-insulin ratios were determined for the individual patients who followed the carbohydrate counting as a meal management, and the factors to influence the CHO-to-insulin ratios were selected through the stepwise regression analysis. Twenty- four subjects were received three lectures, and one or two nutritional counseling for a month. The average age of the subjects was 50.7 years, and the duration of diabetes was 9.4 years. Their body mass index (BMI) was 21.5 kg/$m^2$. The difficulties of using CHO counting were 1) confusing the CHO exchange system to diabetic food exchange system, 2) lack of basic nutrition and not distinguishing nutrients such as CHO, fat and calorie, and 3) lack of motivation to make effort to count and record the amount of carbohydrates eaten. Nutritional counseling replenished the nutrition education and made patients practice CHO counting. Average CHO-to-insulin ratios at breakfast, lunch and dinner were 4.1$\pm$3.3, 2.9$\pm$2.6 and 2.9$\pm$3.0units/23g of CHO, respectively. CHO-to-insulin ratios were influenced by gender, age, BMI, post-prandial blood glucose levels and post-prandial c-peptide levels. The effective education and nutritional counseling of CHO counting can make CHO counting applicable to type 2 diabetic patients as meal management for improving glycemic control with less hypoglycemic episode.

발아 옥수수 amylases의 정제 및 특성 (Partial Purification and Some Properties of Amylases from Germinating Corn(Zea mays L.))

  • 발아중인 옥수수에서의 전분의 가수분해효소인 amylases의 종류를 규명하고자 황산 암모늄염석법, DEAE-Sephadex A-50을 이 용한 이온교환칼럼법과 Sephadex G-100 Gel filtration chromatography 방법으로 정제하였으며 전분가수분해 효소는 3개의 peak가 나타났으며, 이를 각각 모아서 정제한 결과 각각의 비활성은 70.47(units/mg), 62.98(units/mg), 80.39(units/mg)으로 저단백식품의 하나인 옥수수임에도 불구하고 높은 활성을 나타내었다. 이것은 발아중 가수분해되는 전분체내에 이 효소의 작용이 커졌음을 유리당의 양이 증가하였음으로 알 수 있었다. 또한 고속액체 크로마토그라피를 이용하여 분석한 결과 3종류의 amylases중에 amylases(I)은 $\alpha$-amytotetrose의 종류로 밝혀졌으며, amylase(II)와 (III)는 각각, 주로 maltotetrose의 단위로 가수분해하는 전분 분해 효소이나. 서로 생물학적 성격에서 약간씩의 차이를 보이므로 같은 종류는 아닐 것으로 사료되었다.
