Cheonggukjang Fermented with Bacillus subtilis SCGB574 Ameliorates High Fat Diet-Deteriorated Large Intestinal Health in Rat Model

  • Cheonggukjang is a traditional fermented food in Korea, which is known to exert beneficial effects on health. In this study, we evaluated the effects of cheonggukjang fermented by Bacillus subtilis SCGB 574 (B574) on high fat diet (HFD)-deteriorated large intestinal health. Rats were fed with HFD or HFD supplemented with 10.1% cheonggukjang (B574). Fecal microbiota was analyzed based on 16S rRNA gene sequences, and the fecal and serum metabolome were measured using GC-MS. Our results showed that SCGB574 intake significantly reduced body weight, restored tight junction components, and ameliorated inflammatory cell infiltration. SCGB574 also shifted gut microbiota by increasing the abundance of short chain fatty acid producers such as Alistipes and Flintibacter, although it decreased the abundance of Lactobacillus. Serum and fecal metabolome analyses showed significantly different metabolic profiles between the groups. The top five metabolites increased by SCGB574 were i) arginine biosynthesis, ii) alanine, aspartate, and glutamate metabolism; iii) starch and sucrose metabolism; iv) neomycin, kanamycin, and gentamicin biosynthesis; and v) galactose metabolism. These results showed that cheonggukjang fermented by SCGB574 ameliorates adverse effects of HFD through improving intestinal health.

A HACCP model for By-products feed production

  • By-products has been considered lately in Total Mixed Ration (TMR) as an alternative to livestock feed around the world. This is due to the high cost of using forage as feed, less expense in exploring by-products of agriculture origin and environmental concerns with their disposal. However, by-products usually contain contaminants and the production process requires fermentation using a storage and fermentation tank. Animal feed is the start point of the food safety chain in the 'farm-to-fork' model. This necessitated a study to model a protocol that will culminate to safe feed production. Hazard analysis and critical control points (HACCP), a systematic preventive approach to food safety from biological, chemical and physical hazards in production processes that can cause the finished product to be unsafe was explored. Implementation of this model provides a mechanism that ensures product safety is continuously achieved. The entire production process of By-products feed production was evaluated using HACCP wizard software. This includes the plant layout, technical standards, storage and fermentation tank cleansing method, staff assignment, safety control method, and distribution. The potential biological, chemical, and physical hazards that may exist in every step of the production process were identified, and then critical control points (CCPs) were selected. This will ensure the safety of products made from livestock that consumes by-product feed. These includes cheese, milk, beef, etc.

Node2vec 그래프 임베딩과 Light GBM 링크 예측을 활용한 식음료 산업의 수출 후보국가 탐색 연구 (A Study on Searching for Export Candidate Countries of the Korean Food and Beverage Industry Using Node2vec Graph Embedding and Light GBM Link Prediction)

  • 본 연구는 Node2vec 그래프 임베딩 방법과 Light GBM 링크 예측을 활용해 우리나라 식음료 산업의 미개척 수출 후보국가를 탐색한다. Node2vec은 네트워크의 공통 이웃 개수 등을 기반으로 하는 기존의 링크 예측 방법에 비해 상대적으로 취약하다고 알려져 있던 네트워크의 구조적 등위성 표현의 한계를 개선한 방법이다. 따라서 해당 방법은 네트워크의 커뮤니티 탐지와 구조적 등위성 모두에서 우수한 성능을 나타내는 것으로 알려져 있다. 이에 본 연구는 이상의 방법을 우리나라 식음료 산업의 국제 무역거래 정보에 적용했다. 이를 통해 해당 산업의 글로벌 가치사슬 관계에서 우리나라의 광범위한 마진 다각화 효과를 창출하는데 기여하고자 한다. 본 연구의 결과를 통해 도출된 최적의 예측 모델은 0.95의 정밀도와 0.79의 재현율을 기록하며 0.86의 F1 score를 기록해 우수한 성능을 나타냈다. 이상의 모델을 통해 도출한 우리나라의 잠재적 수출 후보국가들의 결과는 추가 조사를 통해 대부분 적절하게 나타난 것을 알 수 있었다. 이상의 내용을 종합하여 본 연구는 Node2vec과 Light GBM을 응용한 링크 예측 방법의 실무적 활용성에 대해 시사할 수 있었다. 그리고 모델을 학습하며 링크 예측을 보다 잘 수행할 수 있는 가중치 업데이트 전략에 대해서도 유용한 시사점을 도출할 수 있었다. 한편, 본 연구는 그래프 임베딩 기반의 링크 예측 관련 연구에서 아직까지 많이 수행된 적 없는 무역거래에 이를 적용했기에 정책적 활용성도 갖고 있다. 본 연구의 결과는 최근 미중 무역갈등이나 일본 수출 규제 등과 같은 글로벌 가치사슬의 변화에 대한 빠른 대응을 지원하며 정책적 의사결정을 위한 도구로써 충분한 유용성이 있다고 생각한다.

Development of Patients Environmental Sustainability Performance in Healthcare Sector: A Conceptual Framework and Further Research Directions

  • Purpose - The purpose of this development the business model of the healthcare sector in order to promote patents satisfaction towards medical sector services improvement for the medical business model innovation to possess a competitive advantage in the medical and pharmaceutical industry. Research design, data, and methodology - Safety standard protocol from existing multidisciplinary literature is a process of theorization, which uses grounded theory methodology rather than a description of the data and the targeted phenomenon by using Jabareen (2009). The first task is to map the spectrum of food safety literature regarding the phenomenon in safety management. This process includes developing the implementation factors and other sources such as existing business models and practices into the protocol design. Results - The study suggests the conceptual framework to improve the safety management for patients' environmental sustainability performance. Conclusion - The business model may support the beneficial aspect to healthcare government's policymakers, hospital employees, and medical specialist who can apply the practical perspective of its value regarding an educational protocol. Originality/value - This study contributes to and extends our understanding of environmental sustainability performance, identifying the rationale for safety standards performance in the healthcare industry with suggested hybrid safety standards market consumer interconnector.

Production and Prophylactic Efficacy Study of Human Papillomavirus-like Particle Expressing HPV16 L1 Capsid Protein

  • To perform the prophylactic study of a vaccine derived from human papillomavirus (HPV) using Balb/c mice, we produced virus like particles consisting of HPV capsid protein L1 which has been reported to induce significant humoral and cellular immunity using various animal model systems. In order to produce HPV16 VLPs, the cDNA of L1 capsid protein in HPV type 16, obtained by polymerase chain reaction, was inserted into yeast expression vector, YEG$\alpha$-HIR525 under the control of GAL10 promoter. The transformation of YEG$\alpha$-HPV16 L1 was performed into the yeast Saccharomyces cerevisiae Y2805 by the lithium acetate method and the yeast clone expressing the highest level of L1 capsid protein of human papillomavirus type 16 was selected by Western blot analysis using anti-HPV16 L1 antibody. The purification of HPV16 VLP has been performed by the ultracentrifugation and gel-filtration methods. To validate the vaccine efficacy of the purified HPV16 VLPs and investigate the properties of HPV16 VLPs to induce humoral immunity, ELISA assay was performed. A significantly increased production of anti-HPV16 VLP antibodies was observed in sera from immunized mice. The neutralization activity of antibodies in the sera from the vaccinated mice was demonstrated by a rapid and simple assay to detect hemagglutihation inhibition activity.

즉석섭취 삼각김밥에서의 Staphylococcus aureus 위해평가 연구 (Risk assessment of Staphylococcus aureus infection in ready-to-eat Samgak-Kimbap)

  • 본 연구는 삼각김밥에서 황색포도상구균 위해평가를 통해 삼각김밥의 섭취에 따른 황색포도상구균 식중독 발생 가능성을 분석하였다. 삼각김밥에 대한 황색포도상구균 오염도를 조사한 결과 170개의 시료가 모두 황색포도상구균 음성이었으며, 확률분포 조사를 통해 확인된 삼각김밥에서의 황색포도상구균의 초기 오염수준은 평균 -3.88 log CFU/g이었다. 삼각김밥의 섭취량 및 섭취자 비율은 미생물 위해평가를 위한 50대 주요 식품의 섭취량 및 섭취패턴 자료를 바탕으로 적정 확률분포모델을 도출하였으며, 삼각김밥의 1일 평균 섭취량과 섭취자 비율은 각각 191.31 g과 0.033으로 나타났다. 유통환경에 따라 시나리오를 구성하여 최종적으로 1일 1회 삼각김밥을 섭취하였을 때 황색포도상구균에 의한 식중독 발생확률은 평균 1.44×10-10로 추정되었다. 민감도 분석에서는 황색포도상구균 오염수준, 섭취량, 편의점 보관온도, 편의점 보관시간 등의 변수가 위해도와 가장 높은 양의 상관관계를 보였다. 본 연구에서는 삼각김밥에서의 황색포도상구균 식중독 발생 가능성은 인구 100,000명이 삼각김밥을 1일 1회 섭취했을 때 0.0000144명에게서 식중독이 발생할 수 있는 것으로 나타나 계속적으로 안전한 삼각김밥이 소비자에게 유통되기 위해서는 지속적으로 삼각김밥의 안전한 제조, 매장 내 보관온도 및 시간에 대한 모니터링이 필요할 것으로 사료된다.

Development of Strain-Specific Primers for Identification of Bifidobacterium bifidum BGN4

  • Bifidobacterium bifidum BGN4 (BGN4) has many proven beneficial effects, including antiallergy and anticancer properties. It has been commercialized and used in several probiotic products, and thus strain-specific identification of this strain is very valuable for further strain-dependent physiological study. For this purpose, we developed novel multiplex polymerase chain reaction (PCR) primer sets for strain-specific detection of BGN4 in commercial products and fecal samples of animal models. The primer set was tested on seven strains of B. bifidum and 75 strains of the other Bifidobacterium species. The BGN4-specific regions were derived using megaBLAST against genome sequences of various B. bifidum databases and four sets of primers were designed. As a result, only BGN4 produced four PCR products simultaneously whereas the other strains did not. The PCR detection limit using BGN4-specific primer sets was $2.8{\times}10^1CFU/ml$ of BGN4. Those primer sets also detected and identified BGN4 in the probiotic products containing BNG4 and fecal samples from a BGN4-fed animal model with high specificity. Our results indicate that the PCR assay from this study is an efficient tool for the simple, rapid, and reliable identification of BGN4, for which probiotic strains are known.

  • The effectiveness of dose reduction resulting from the application of countermeasures for ingestion pathways after nuclear accidents was investigated together with the derivation of optimized intervention levels for Korean foodstuffs. The radioactivity in foodstuffs was predicted from a dynamic food chain model DYNACON for the date which the deposition occurs. The effectiveness of countermeasures strongly depended on radionuclides, foodstuffs and date of deposition.

  • 얼마나 빠른 환경의 변화에 잘 적응하느냐에 따라서 개인과 조직의 성패는 좌우된다. 이러한 환경의 변화에 잘 적응하기 위해서는 정보의 활용이 필수적이다. 본 연구에서는 농장경영체의 정보 활용도 향상을 위한 구체적인 방안을 제시하는데 있다. 이를 위해 설문과 인터뷰조사, 선행사례 조사 등을 실시하고 농장경영에 필요한 의사결정 단계별 정보를 도출하고 이를 제공하기 위한 데이터베이스와 모델 베이스를 제안하였다. 또한 구축된 정보를 잘 활용하는 정보 접근 방법을 병목정보 해결의 시각에서 정형검색과 비정형검색방법을 제시하였다. 이러한 가치사슬적 접근 방법은, 아직 농업분야에서는 보편적인 내용은 아니지만 여타 첨단정보산업과 서비스업에서는 경영체별로 구체화하여 비교적 유용하게 활용하고 있는 방법이다. 본 연구를 바탕으로 품목별로 의사결정 단계별 데이터베이스와 모델베이스가 구축되어 농장경영체가 잘 활용함으로서 불확실성 요인과 변동요인에 잘 대응하여 농업 생산성 향상에 기여할 수 있도록 국가 차원에서 구체적인 시스템적 접근이 필요하다고 본다.

Slaughter practices of different faiths in different countries

  • This paper reviews many aspects of ritual and traditional slaughter methods used to produce meat for human consumption in different countries. Undoubtedly, meat is an important source of nutrients that are essential for human health. The global meat market has become increasingly interested in Islamic halal and Jewish kosher slaughter, in particular because of potential market opportunities. The requirement for unstunned slaughter or reversible pre-slaughter stunning makes religiously-based methods of animal slaughter unique. This study suggests a simple framework for a halal and tayyib meat supply chain for the Muslim community that also maintains meat quality and wholesomeness from farm to table as a model for the religious slaughter of animals.
