The objective of this study was to investigate the responsiveness of hypophysis and gonads to synthetic GnRH among noncycling Murrah buffalo heifers at 24 months of age. The plasma FSH, LH, estradiol and progesterone levels were measured in blood samples collected at 1 hour before and upto 18th day subsequent to the administration of GnRH ($(200 {\mu}g)$) or saline (2 ml). The pretreatment levels of plasma FSH, LH estradiol and progesterone among GnRH treated heifers (N = 6) were $11.55{\pm}0.57ng/ml$, $0.68{\pm}0.06ng/ml$, $19.84{\pm}0.82pg/ml$ and $0.45{\pm}0.07ng/ml$ respectively. A quick elevation of FSH (p < 0.01) and LH (p < 0.05) within 5 min of GnRH administration was observed in all the heifers. The peak FSH ($74.97{\pm}18.63ng/ml$) and LH ($3.09{\pm}0.54ng/ml$) level was obtained at 30 min of GnRH administration. The elevated level of plasma estradiol on 5th to 18th day, FSH on 7th to 9th day (n = 3) and the progesterone on 13th to 18th day (n = 2) of GnRH injection was obtained. The study indicates that gonads of buffalo heifers at 24 months of age are responsive of GnRH induced gonadotropin release for folliculogenesis and luteal tissue formation
Protease nexin-1 (PN-1)은 활성화 자리에 serine기를 갖는 단백질 분해 효소 즉, 트롬빈, 트립신, 플라스미노겐 활성화 효소 등의 작용을 억제한다. 본 연구에서는 흰쥐의 Sprague-Dawley계통을 이용하여 조직별 mRNA발현여부 및 정도를 조사하였다. PN-1의 발현이 나타난 조직은 뇌 (전뇌, 후뇌), 심장, 간, 폐, 난소, 난관 등이다. 이들 중 유전자 발현이 가장 높은 조직은 암컷의 전뇌(forebrain) 였다. 특히, 생식기관들 중에서는 암컷의 난소와 난관에서만 발현이 관찰되는 등 PN-1 유전자는 성별에 따라 서로 다르게 발현됨이 확인되었다 이러한 결과들로 미루어 PN-1은 여포 형성과정과 초기배 형성과정 등의 생식 및 발생작용과 관련이 있을 것으로 생각된다.
포유류 원시난포의 분화에는 뇌하수체에서 분비되는 gonadotropins 외에도 다양한 성장인자들 뿐 아니라 스테로이드호르몬 등이 관여하며, 난황낭에서 기원된 생식세포들과 중신에서부터 유입되는 기질세포들의 복잡한 상호작용에 의해 이루어진다. 특히 사람의 경우 태아기에 분화가 시작된 원시난포들은 성장을 개시한 후 배란이 되거나 혹은 폐쇄되어 난소에서 제거되는데, 일부 원시난포는 성장이 개시되기까지 50년 이상 원시난포의 상태로 유지된다. 그러나 원시난포의 분화 및 성장정지, 성장개시의 기전에 관하여는 현재까지 정확하게 밝혀져 있지 않다. 본 논문에서는 태아 및 신생아의 난소에서 gonadotrorin의 수용체, 성장인자와 성장인자 수용체, 그리고 스테로이드호르몬 수용체의 발현을 조사하였다. 이 결과들을 바탕으로 사람 원시난포의 형성, 성장정지를 조절하는 기전 그리고 원시난포의 성장개시에 영향을 미치는 요인을 알아보았다.
This study was conducted to determine how the isolation method of the porcine preantral follicles influenced the following follicular growth in vitro. Mechanical and enzymatical isolations were used for retrieving the follicles from prepubertal porcine ovaries, and in vitro-growth of the follicles and the expression of folliculogenesis-related genes were subsequently monitored. The enzymatic retrieval with collagenase treatment returned more follicles than the mechanical retrieval, while the percentage of morphologically normal follicles was higher with mechanical retrieval than with enzymatic retrieval. After 4 days of culture, mechanically retrieved, preantral follicles yielded more follicles with normal morphology than enzymatically retrieved follicles, which resulted in improved follicular growth. The mRNA expression of FSHR, LHR Cx43, DNMT1 and FGFR2 genes was significantly higher after culture of the follicles retrieved mechanically. These results suggest that mechanical isolation is a better method of isolating porcine preantral follicles that will develop into competent oocytes in in vitro culture.
Intact germinal vesicle (GV) arrest and release are essential for maintaining the fertility of mammals inducing human. Intact germinal vesicle release, maturation of oocytes is maintained by very complex procedures along with folliculogenesis and is a critical step for embryonic development. Cyclic guanosine monophosphate (cGMP) has been suggested a key factor for meiotic arrest but so far its mechanisms are controversy. In this study we examine the effects of cGMP on germinal vesicle breakdown in cumulus-enclosed oocytes and denuded oocytes. Spontaneous maturation was inhibited by a cGMP agonist, 8-Br-cGMP with concentration dependent manners both in cumulus-enclosed oocytes and denuded oocytes. The inhibitory effect was more severe in denuded oocytes than cumulus-enclosed oocytes. The Rp-8-Br-cGMP and Rp-pCPT-8-Br-cGMP did not severely block GVB compared to 8-Br-cGMP. The spontaneous GVB inhibitory effects were different by the existence of cumulus. Based on them it is suggested that the cumulus modulates the role of cGMP in GV arrest.
Growth differentiation factor 9 (GDF9) and bone morphogenetic protein 15 (BMP15) are oocyte-specific growth factors that regulate many critical processes involved in early folliculogenesis and oocyte maturation. In this study, effects of GDF9 and BMP15 treatment during in vitro maturation of porcine oocytes upon development after parthenogenetic activation were investigated. Neither GDF, BMP15 alone nor in combination affects the number and viability of cumulus cells or the rates of oocyte maturation and blastocyst development. However, the treatment of GDF9 on porcine oocytes increased the number of trophectodermal (TE) cells of blastocysts derived from activated oocytes (P<0.05). The treatment of BMP15 increased the cell numbers of both inner cell mass (ICM) and TE cells (P<0.05). The treatment with the combination of GDF9 and BMP15 further increased the numbers of ICM and TE cells, compared with GDF9 or BMP15 treatment alone (P<0.05). In conclusion, the treatment of GDF9 or BMP15 (or both) enhanced the quality of blastocysts via the increased number of ICM and/or TE cells.
Transition of the resting primordial follicle to the growing primary follicle is a critical process for female reproduction, but its mechanism is poorly understood. The present study was conducted to investigate gene expression profile at the primordial-primary follicle transition process. We isolated total RNA of female mouse ovary at day1 (contains only primordial follicles) and day5 (contains primordial and primary follicles) and synthesized cDNA using annealing control primers (ACP; Seegene, Inc., Seoul, Korea). ACP provides annealing specificity and sensitivity to the template and allows to identify only authentic differentially expressed genes (DEGs). We used total 80 ACPs for PCR, observed PCR products on 2% agarose gel, cloned 42 DEGs using TOPO TA cloning vector, sequenced, and analyzed by BLAST search. Sequences of 34 clones significantly matched database entries while 4 clones were novel and 4 clones were EST. Two of 34 genes were specifically expressed only in day 5 ovaries (Sui1-rs1, Apg3p/Aut1p-like), and rest of 32 genes were expressed in both stages but were differential in amount. Differential expression was confirmed using semiquantitative RT-PCR, and there was no false positive. Anx11 and Pepp2-pending were highly expressed genes in day1-, while BPOZ, Ches1, Kcmf1, NHE3, Nid2, Ninj1, SENP3 and Survivin were highly expressed genes in day5-ovary. List of genes would provide insight for further study of mechanism regulating primordial-primary follicle transition.
SOHLH2 is a novel germ cell-specific transcription factor that is crucial for folliculogenesis in the ovary and spermatogenesis in the testis. SOHLH2 represents a candidate gene for infertility with premature ovarian failure. We analyzed whether mutations in the SOHLH2 gene in 98 Korean women with premature ovarian failure. The sequence analysis identified six novel SNPs (c.431-41G>C, c.656A>T, c.1000+27C>T, c.1000+33G>T, c1258-106G>A, and c.2094+ 11T>C) from Korean patients with premature ovarian failure. The c.656A>T found in exon 7 results in change of an amino acid, tyrosine to phenylalanine. Functional mutations in SOHLH2 gene are rare in Korean women with premature ovarian failure.
난포 내 난자를 둘러싸고 있는 과립세포는 난자를 위한 성장상태 및 난포의 발달에 중요하다. Solute carrier family 유전자는 스테로이드 호르몬, 다양한 약물, 몇몇 다른 기질을 유입시킨다. 연구에서 획득한 서로 다르게 발현하는 유전자들 (DEGs) 중 일부를 in situ hybridization을 통해 분석하였다. 분석한 결과 SLC23A3과 SLC39A10이 난소에서 높게 발현하였다. SLC39A10 유전자는 원시난포에서 높게 발현하였고, SLC23A3은 일차, 이차 난포에서 높게 발현하였다. 특히 성장하는 난포의 과립막 세포에서 발현하였다. SLC23A3과 SLC39A10은 원시난포와 일차난포에서 다르게 발현하는 것은 각 난포의 분리를 통해 좀 더 확인해야 할 것이다. 본 연구에서는 유전자 발현 정보를 통해 원시난포의 개시와 성장을 위한 전환에 관여하는 기전을 이해하는데 기초정보와 난포발달 촉진을 위한 난소기능부전의 기전을 규명하는데 정보를 제공할 것으로 기대된다.
Recently, production of transgenic animal by nuclear transfer has been known as a useful method. The production of cloned offspring derived from nuclear transfer depends upon a variety of factors such as species, donor cells type and cell cycle, and source of recipient ova. Therefore, we attempted a different transgenic methods using follicular granulosa cells (GCs). In general, ovulated GCs undergoes lutenization and transformation in vitro which might defective effects on developmental potential. In order to avoid the GCs transformation in vitro culture system, we introduced a direct injection of retrovirus into the follicles and then collected them mechanically from ovaries of 6-8 week-old ICR mice. Retrovirus vector constructed with pLN $\beta$ EGFP was injected into the follicles. The follicles are cultured in $\alpha$ -MEM supplemented with human FSH, LH and ITS in Costar Transwell dish for 4 days. Survival rate of virus injected follicles was 52.1% (12/23) and expression rate of EGPP gene was 33.3% (4/12). In this study, we found GCs performed transgenesis in our culture system. In addition, the GCs in follicle may be developed in vivo like environment rather than in vitro environment. Thus, the use of GCs as donor cells may be useful in the nuclear transfer for cloning of genetic modification. Therefore, these results suggest that follicular GCs can be transfected by viral vector during folliculogenesis in vitro.
