Targeting Cellular Antioxidant Enzymes for Treating Atherosclerotic Vascular Disease

  • Atherosclerotic vascular dysfunction is a chronic inflammatory process that spreads from the fatty streak and foam cells through lesion progression. Therefore, its early diagnosis and prevention is unfeasible. Reactive oxygen species (ROS) play important roles in the pathogenesis of atherosclerotic vascular disease. Intracellular redox status is tightly regulated by oxidant and antioxidant systems. Imbalance in these systems causes oxidative or reductive stress which triggers cellular damage or aberrant signaling, and leads to dysregulation. Paradoxically, large clinical trials have shown that non-specific ROS scavenging by antioxidant vitamins is ineffective or sometimes harmful. ROS production can be locally regulated by cellular antioxidant enzymes, such as superoxide dismutases, catalase, glutathione peroxidases and peroxiredoxins. Therapeutic approach targeting these antioxidant enzymes might prove beneficial for prevention of ROS-related atherosclerotic vascular disease. Conversely, the development of specific antioxidant enzyme-mimetics could contribute to the clinical effectiveness.

Oxidized LDL induces phosphorylation of non-muscle myosin IIA heavy chain in macrophages

  • Oxidized LDL (oxLDL) performs critical roles in atherosclerosis by inducing macrophage foam cell formation and promoting inflammation. There have been reports showing that oxLDL modulates macrophage cytoskeletal functions for oxLDL uptake and trapping, however, the precise mechanism has not been clearly elucidated. Our study examined the effect of oxLDL on non-muscle myosin heavy chain IIA (MHC-IIA) in macrophages. We demonstrated that oxLDL induces phosphorylation of MHC-IIA (Ser1917) in peritoneal macrophages from wild-type mice and THP-1, a human monocytic cell line, but not in macrophages deficient for CD36, a scavenger receptor for oxLDL. Protein kinase C (PKC) inhibitor-treated macrophages did not undergo the oxLDL-induced MHC-IIA phosphorylation. Our immunoprecipitation revealed that oxLDL increased physical association between PKC and MHC-IIA, supporting the role of PKC in this process. We conclude that oxLDL via CD36 induces PKC-mediated MHC-IIA (Ser1917) phosphorylation and this may affect oxLDL-induced functions of macrophages involved in atherosclerosis.

  • The growth of the critical size bubble by diffusion process in viscoelastic medium was treated by an integral method for the concentration boundary layer adjacent to the bubble wall. In this study, we obtained a set of the first order time dependent equations to obtain bubble radius and gas pressure inside the bubble simultaneously. The calculated final cell sizes depending on the initial saturation pressure are in close agreement with the observed ones. The governing equations developed in this study may be used in polymer processing of microcellular foams.

  • In all Rapid Prototyping(RP) processes, computer-aided design(CAD) solid model is sliced into thin layers of uniform, but not necessarily constant, thickness in the building direction. Each cross-sectional layer is successively deposited and, at the same time, bonded onto the previous layer, the stacked layers form a physical part of the model. The objective of this study is to develop a method for obtaining necessary coordinates$(x,\;y,\;\theta_x,\;\theta_y)$ to position linear hotwire of the cutting system in three-dimensional space for the Variable Lamination Manufacturing process (VLM-S), which utilizes expandable polystyrene foam sheet as part material. In order to examine the applicability of the developed method to VLM-S, various three-dimensional shapes, such as a spanner, a patterned columm, and a pyramid were made using data obtained from the method.

Optimal Inner Case Design for Refrigerator by Utilizing Artificial Neural Networks and Genetic Algorithm

  • In this paper, an artificial neural network (ANN) was employed to build a predicting model for refrigerator structure. The predicting model includes three input variables of the plaque depth (D), width (W) and interval distance(S) on the inner wall. Finite element method was utilized to obtain the data, which would be necessary for the ANN training process. Finally, a genetic algorithm (GA) was applied to find the optimal parameters that leaded to the minimum inner case deformation under operating condition. The optimal combination found is the depth(D) of 2.63mm, the width(W) of 19.24mm and the interval distance(S) of 49.38mm which leaded to the smallest deformation of 1.88mm for the given refrigerator model.

  • There is a great demand for reducing the amount of material used in mass-produced plastics parts, for material cost constitutes a large percentage of the total cost of 60%. It may be noted that the price of plastics is directly rotated to the price of petroleum. Material reduction therefore decreases the amount of oil needed for the manufacture of plastics and thus help conserve this natural resource. Therefore microcellular foaming process(MCPs) was studied for solving this problems alternatively in 1980's at M.I.T Until now in MCPs carbon dioxide gas was mainly used for microcellular foaming. Besides, Talc was used for reducing the price of plastics. Consequently, we must certificate using the Talc in MCPs according to contents of the Talc.

Numerical simulation of wave slamming on wedges and ship sections during water entry

  • The open source software OpenFOAM is utilised to simulate the water entry and hydrodynamic impact process of 2D wedges and ship hull sections. Incompressible multiphase flow solver interDyMFoam is employed to calculate the free fall of structure from air into water using dynamically deforming mesh technique. Both vertical and oblique entry of wedges of various dead-rise angles have been examined. A convergence study of dynamics as well as kinematics of the flow problem is carried out on successively refined meshes. Obtained results are presented and compared to the experimental measurements showing good agreement and reasonable mesh convergence of the solution.

  • Slopping phenomena deteriorate the productivity and the stability of operation in oxygen converter process. To predict and prevent that, it needs reliable perception of the internal state of the oxygen converter chamber. Equipment was developed to measure the dynamic and static behavior of the oxygen-blowing lance 3-dimensionally and the measured signal is analyzed in relation to slopping phenomena. It was found that the behavior of the lance is strongly correlated to the change of slag foam height and the instability of molten steel behavior. So slopping phenomena can be predicted by simultaneously analyzing the measured behavior of the lance and the control factors such as the flow rate of oxygen and the height of lance.

  • The low foamed high strengthen styrofoam samples made by dielectric heating are discussed. We used the oscillator which have the frequency of 13.6 MHz and the power of 7 kW. 3 times expanded beads by steaming method were used in our foam-molding test. Internal fusion properties and density of internal structure were improved by dielectric foaming process. At the temperature of $105-110^{\circ}C$, the internal fusion property was maximally improved.
