Fundamental Parameter 법에 의한 만장굴용암 쌍자석주의 형광X선분포

  • Cheju Island, which was formed by volcanic activity, is an oval in its shape with the major axis 80km and the minor axis of 40km. The island holds in its heart Mt. Hanala rising 1,950m above the sea. Petrological study of this volcanic island has been made actively by Sang-Man Lee, Chong-Kwang Won and Moon-Won Li. The chronological measurements of the island by Chong-Kwan Won and Moon-Won Lee showed that it is composed of Sanbangsan trachytes and Backlokdam trachytes(25,000 year ago). These reports are based on the chemical analysis and the rediometric chronological measurements on the ground. However, there has been no reports about the inside of caves. We made an (composition) analysis of the inside of Manjang Cave by the fundamental parameter method in X-ray fluorescence spectrometry. The fundamental parameter method in X-ray fluorescence spectrometry is nondestructive analysis. and it enables us to make the values processed by a computer. The results obtained by this methods are as follows: SiO$_2$(49%), $Al_2$O$_3$(17%), Fe$_2$O$_3$(13%), CaO(8.1%), MgO(5.5%), Na2O(3.6%), TiO$_2$(2.1%), $K_2$O(0.86%), P$_2$O$_{5}$(0.28%), and MnO(0.20%) respectively. The data obtained by the fundamental parameter method in X-ray fluorescene was compared with the data provided by Chong-Kwan Won and Moon-Won Lee. Our measurement was made by K-Ar-method in cooperation with T. ITAYA. The samples are of 30,000-420,000 years ago. The composition of the values of our underground analysis with the existing values obtained by the analyses on the ground produced new data about Cehju volcanic island.d.

Bioequivalence Assessment of Roxithromycin Tablets in Healthy Korean Volunteers

  • The objective of the study was to evaluate the bioequivalency between the $Rozid^{TM}$ Tablet (Ilhwa Pharm. Co., Ltd.) as a test formulation and the $Rulid^{TM}$ Tablet (Handok Pharm. Co., Ltd) as a reference formulation. Twenty-four healthy male volunteers were administered the formulations by the randomized Latin square crossover design, and the plasma samples were determined by a high performance liquid chromatography (HPLC) with fluorescence detector. $AUC_t,\;C_{max}\;and\;T_{max}$ were obtained from the time-plasma concentration curves, and log-transformed $AUC_t\;and\;C_{max}$ and log-untransformed $T_{max}$ values for two formulations were compared by statistical tests and analysis of variation. $AUC_t$ was determined to be $63.30{\pm}25.57{\mu}g.hr/ml$ for the test formulation and $64.02{\pm}29.27mg.hr/ml$ for the reference formulation. The mean values of $C_{max}$ for the test and reference formulations were $5.07{\pm}2.14\;and\;5.53{\pm}2.60{\mu}g/ml$, respectively. The $AUC_t,\;and\;C_{max}$ ratios of the test $Rozid^{TM}$ Tablet to the reference $Rulid^{TM}$ Tablet were -1.12% and -8.32%, respectively, showing that the mean differences were satisfied the acceptance criteria within 20%. The results from analysis of variance for log-transformed $AUC_t,\;and\;C_{max}$ indicated that sequence effects between groups were not exerted and 90% confidence limits of the mean differences for $AUC_t,\;and\;C_{max}$ were located in ranges from log 0.80 and log 1.25, satisfying the acceptance criteria of the KFDA bioequivalence. The RozidTM Tablet as the test formulation was considered to be bioequivalent to the RulidTM Tablet used as its reference formulation, based on $AUC_t,\;and\;C_{max}$ values.

Effect of High Dose ${\gamma}-irradiation$ on the Physicochemical Properties of Shell Eggs during Storage (고선량 감마선 조사가 신선란의 저장 중 이화학적 특성에 미치는 영향)

  • To determine the quality change of the irradiated eggs during storage, fresh shell eggs were irradiated using $^{60}Co$ at 0, 1, 5, 10, 30 kGy and stored for 30 days. The york index, color, pH, viscosity, egg weight, and SDS-PAGE profile of the irradiated eggs were examined. During storage, york index values of the irradiated eggs and the control were decreased and the increase of dose decreased yolk index. However, the yolk index values were increased temporarily at 10 kGy and 30 kGy. The yolk color had a bright yellow with increases in dose level and there was no significant change during storage. The albumen viscosities were decreased with increases in dosage and were decreased during storage. Also, the albumen pH values of the irradiated eggs were higher than that of the control and were increased during storage. The weight losses of eggs were increased during storage and there were no significant changes by dose level. SDS-PAGE profile of the egg white proteins of the shell eggs showed the change in molecular weight distribution and had aggregation pattern as well as degradation. CD and fluorescence spectroscopy study showed changes in the secondary and tertiary structure of egg white proteins by ${\gamma}-irradiation$. Therefore, this study clearly indicates that irradiation dose of eggs should be appropriate to prevent the loss of egg qualities.

Comparison of Diagnostic Validity between Laser Fluorescence Devices in Proximal Caries (레이저 형광 원리를 이용한 우식 진단 기기의 구치부 인접면 우식 진단 능력 비교)

  • The aim of this study was to evaluate the proximal caries detecting ability and identify the optimal cut-off values of two types of laser fluorescence (LF) devices; classic type (DD) and pen type (DDpen). The number of proximal surfaces participated in this study were 164 surfaces in primary dentition and 438 surfaces in permanent dentition. Each tooth surface was sequentially assessed by two types of LF devices, and bitewing radiograph. The radiographs were classified into 3 groups in primary dentition ($pR_0$, $pR_1$, $pR_2$), and 4 groups in permanent dentition ($PR_0$, $PR_1$, $PR_2$, $PR_3$) according to the depth of caries, and used as gold standard. In primary dentition, the area under the curve (AUC) values of DD were 0.851 and 0.890, and those of DDpen were 0.883 and 0.917, respectively in enamel caries and dentin caries. In permanent dentition, the AUC values of DD were 0.762 and 0.886, and those of DDpen were 0.828 and 0.958, respectively in enamel caries and dentin caries. When detecting proximal caries in posterior teeth with LF devices, DDpen is more useful than DD in both primary and permanent dentition. However, in primary dentition, DD can also be useful to detect proximal caries.

Photophysical Parameters, Photodecomposition, Fluorescence Quenching and Convolutive Voltammetry of 7-Diethylaminocoumarin (DEAC) Laser Dye (7 Diethylaminocoumarin (DEAC) 레이져 염료의 광물리적 파라미터, 광분해, 형광 소광 및 Convolutive Voltammetry)

  • The photophysical properties of 7-diethylaminocoumarin (DEAC) laser dye have been measured in different solvents. The emission spectrum of DEAC has also been measured in cationic (CTAC) and anionic (SDS) micelles. The laser parameters have been calculated in different solvents namely acetone, dioxane, ethanol and dimethylforamide(DMF). The photoreactivity of DEAC has been studied in $CCl_4$ solvent using 366 nm light. The values of photochemical yield (${\Phi}_c$) and rate constant (k) are determined. The interaction of organic acceptors such as picric acid (PA), tetracyanoethylene (TCNE) and 7,7,8,8-tetracynoquinonedimethane (TCNQ) with DEAC are also studied using fluorescence measurements in acetonitrile ($CH_3CN$). The electrochemical investigation of (DEAC) has been carried out using cyclic voltammetry and convolutive voltammetry combined with digital simulation technique at a platinum electrode in 0.1 mol $L^{-1}$ tetrabutyl ammonium perchlorate (TBAP) in $CH_3CN$ solvent. The electrochemical parameters of the investigated compound were determined using cyclic and convolutive voltammetry. The extracted electrochemical parameters were verified and confirmed via digital simulation method.

Spectroscopic Study on Three States of Water in the Reverse Micelle Using Methylene Blue as a Probe (Methylene Blue를 이용한 역미셀에서 물의 세 가지 상태에 대한 분광학적 연구)

  • In order to find out the microscopic environmental information on the nonionic reverse micelle of Triton X-100/n-hexanol/water in cyclohexane, an absorption and fluorescence spectroscopic study has been conducted using a methylene blue(MB). The information on the microscopic states of water in the polar core of the reverse micelle has been found by investigating complex formation and solvatochromic behavior between MB and Triton X-100. As a result, it was found that there exist three states in the polar core of the reverse micelle. The measured values of $W(=[H_2O]/[Surf])$ for the three states of water are 0.71, 4.98, and 7.26, and the corresponding lifetimes of MB are $15.45 ns{\pm}0.56$, $12.27 ns{\pm}0.79$, and $8.28 ns{\pm}0.82$, respectively.

Quantitative Estimation of Binding Site Polarities of Water-Soluble Polyparacyclophanes in Aqueous Solution by Spectrofluorometry (형광광도법에 의한 수용액중 수용성 폴리파라시클로판류의 결합부위의 극성 측정)

  • The fluorescence characteristics of l-anilinonaphthalene-8-sulfonate (ANS) and 2-p-toluidinylnaphthalene-6-sulfonate (TNS) made the dyes useful probes for the determination of the polarity at the binding sites of several water-soluble polyparacyclophanes. Polyparacyclophanes used were 1,6,20,25-tetraaza[ 6.1.6.1]paracyclophane (CPM 44), 1,7,21,27 -tetraaza[7.1.7.1]paracyclophane (CPM 55). 1,7,21,27 -tetraaza-14,34-dioxa[7.1.7.1]paracyclophane (CPE 55) and 1,8,22,29-tetraaza-15,36-dioxa[8.1.8.1] paracyclophane (CPE 66). The fluorescence quantum yield, emission maximum, and half bandwidth of ANS or TNS obtained in a variety of solvent systems were plotted as a function of four kinds of empirical solvent polarity scales such as dielectric constant (D), (D-l)/(2D+1). Y and Z values. It was found that the Z-value-emission maximum $(\overline}V_F,\;cm^{-1})$ profile showed the most reliable linearity. ANS and TNS interacted with CPM 44, CPM 55, CPE 55. CPE 66. ${\alpha}-cyclodextrin$ (CyD) and ${\beta}-CyD$ in the aqueous solution, and from the emission maxima the polarities (Z-value) of their binding sites were calculated to be 92.65, 87.50, 93.35, 84.52, 94.36, and 90.48 for ANS, respectively. and 91.07, 89.68, 85.44, 86.74 and 87.6 for TNS except for ${\alpha}-CyD$, respectively.

Fluorescence-in-situ-hybridization in the Surveillance of Urothelial Cancers: Can Use of Cystoscopy or Ureteroscopy be Deferred?

  • Background: Fluorescence in situ hybridization (FISH) testing may be useful to screen for bladder carcinoma or dysplasia by detecting aneuploidy chromosomes 3, 7, 17 and deletion of the chromosome 9p21 locus in urine specimens. This study aimed to assess the sensitivity, specificity, positive and negative predictive value of FISH in a multi-ethnic population in Asia. Materials and Methods: Patients with haematuria and/or past history of urothelial cancer on follow-up had their voided urine tested with FISH. Patients then underwent cystoscopy/ureteroscopy and any lesions seen were biopsied. The histopathological reports of the bladder or ureteroscopic mucosal biopsies were then compared with the FISH test results. Results: Two hundred sixty patients were recruited. The sensitivity and specificity of the FISH test was 89.2% and 83.4% respectively. The positive (PPV) and negative predictive values (NPV) were 47.1% and 97.9%. By excluding patients who had positive deletion of chromosome 9, the overall results of the screening test improved: sensitivity 84.6%; specificity 96.4%; PPV 75.9% and NPV 97.9%. Conclusions: UroVysion FISH has a high specificity of detecting urothelial cancer or dysplasia when deletion of chromosome 9 is excluded. Negative UroVysion FISH-tests may allow us to conserve health resources and minimize trauma by deferring cystoscopic or ureteroscopic examination.

Applicability of Fluorescein Diacetate (FDA) and Calcein-AM to Determine the Viability of Marine Plankton (FDA와 Calcein-AM 방법을 이용한 해양플랑크톤 생사판별기법)

  • Ballast water is widely recognized as a serious environmental problem due to the risk of introducing non-indigenous aquatic species. In this study we aimed to investigate measures which can minimize the transfer of aquatic organisms from ballast water. Securing more reliable technologies to determine the viability of aquatic organisms is an important initiative in ballast water management systems. To evaluate the viability of marine phytoplankton, we designed the staining methods of fluorescein diacetate (FDA) and Calcein-AM assay on each target species belonging to different groups, such as bacillariphyceae, dinophyceae, raphidophyceae, chrysophyceae, haptophyceae and chlorophyceae. The FDA method, which is based on measurements of cell esterase activity using a fluorimetric stain, was the best dye for determining live cells of almost all phytoplankton species, except several diatoms tested in this study. On the other hand, although fluorescence of Calcein-AM was very clear for a comparatively longer time, green fluorescence per cell volume was lacking in most of the tested species. According to the Flow CAM method, which is a continuous imaging technique designed to characterize particles, green fluorescence values of stained cells by FDA were significantly higher than those of Calcein-AM treatments and control, implying that the Flow CAM using FDA assay could be adapted as an important tool for distinguishing living cells from dead cells. Our results suggest that the FDA and Calcein-AM methods can be adapted for use on phytoplankton, though species-specific characters are greatly different from one organism to another.

Physiological Responses of Porphyra yezoensis Ueda (Bangiales, Rhodophyta) Exposed to High Ammonium Effluent in a Seaweed-based Integrated Aquaculture System

  • Porphyra yezoensis is known to act as a biofilter against nutrient-rich effluent in seaweed-based integrated aquaculture systems. However, few studies have examined its physiological status under such conditions. In this study, we estimated the photosynthetic activity of P. yezoensis by chlorophyll fluorescence of PSII (${\Delta}F/F'm$ and relative $ETR_{max}$) using the Diving-PAM fluorometer (Walz, Germany). In addition, bioremediation capacity, tissue nutrients, and C:N ratio of P. yezoensis were investigated. The ammonium concentration in seawater of seaweed tank 4 decreased from $72.1{\pm}2.2$ to $33.8{\pm}0.4{\mu}M$ after 24 hours. This indicates the potential role of P. yezoensis in removing around 43% of ammonium from the effluents. Tissue carbon contents in P. yezoensis were constant during the experimental period, while nitrogen contents had increased slightly by 24 hours. In comparison with the initial values, the ${\Delta}F/F'm$ and $rETR_{max}$ of P. yezoensis had increased by about 20 and 40%, respectively, after 24 hours. This indicates that P. yezoensis condition improved or remained constant. These results suggest that chlorophyll fluorescence is a powerful tool in evaluating the physiological status of seaweeds in a seaweed-based integrated aquaculture system.
