Purpose: Lingual nerve (LN) damage may be caused by either tumor resection or injury such as wisdom tooth extraction, Although autologous nerve graft is sometimes used to repair the damaged nerve, it has the disadvantage of necessity of another operation for nerve harvesting. Moreover, the results of nerve grafting is not satisfactory. The nerve growth factor (NGF) is well-known to play a critical role in peripheral nerve regeneration and its local delivery to the injured nerve has been continuously tried to enhance nerve regeneration. However, its application has limitations like repeated administration due to short half life of 30 minutes and an in vivo delivery model must allow for direct and local delivery. The aim of this study was to construct a well-functioning $rhNGF-{\beta}$ adenovirus for the ultimate development of improved method to promote peripheral nerve regeneration with enhanced and extended secretion of hNGF from the injured nerve by injecting $rhNGF-{\beta}$ gene directly into crush-injured LN in rat model. Materials and Methods: $hNGF-{\beta}$ gene was prepared from fetal brain cDNA library and cloned into E1/E3 deleted adenoviral vector which contains green fluorescence protein (GFP) gene as a reporter. After large scale production and purification of $rhNGF-{\beta}$ adenovirus, transfection efficiency and its expression at various cells (primary cultured Schwann cells, HEK293 cells, Schwann cell lines, NIH3T3 and CRH cells) were evaluated by fluorescent microscopy, RT-PCR, ELISA, immunocytochemistry. Furthermore, the function of rhNGF-beta, which was secreted from various cells infected with $rhNGF-{\beta}$ adenovirus, was evaluated using neuritogenesis of PC-12 cells. For in vivo evaluation of efficacy of $rhNGF-{\beta}$ adenovirus, the LNs of 8-week old rats were exposed and crush-injured with a small hemostat for 10 seconds. After the injury, $rhNGF-{\beta}$ adenovirus($2{\mu}l,\;1.5{\times}10^{11}pfu$) or saline was administered into the crushed site in the experimental (n=24) and the control group (n=24), respectively. Sham operation of another group of rats (n=9) was performed without administration of either saline or adenovirus. The taste recovery and the change of fungiform papilla were studied at 1, 2, 3 and 4 weeks. Each of the 6 animals was tested with different solutions (0.1M NaCl, 0.1M sucrose, 0.01M QHCl, or 0.01M HCl) by two-bottle test paradigm and the number of papilla was counted using SEM picture of tongue dorsum. LN was explored at the same interval as taste study and evaluated electro-physiologically (peak voltage and nerve conduction velocity) and histomorphometrically (axon count, myelin thickness). Results: The recombinant adenovirus vector carrying $rhNGF-{\beta}$ was constructed and confirmed by restriction endonuclease analysis and DNA sequence analysis. GFP expression was observed in 90% of $rhNGF-{\beta}$ adenovirus infected cells compared with uninfected cells. Total mRNA isolated from $rhNGF-{\beta}$ adenovirus infected cells showed strong RT-PCR band, however uninfected or LacZ recombinant adenovirus infected cells did not. NGF quantification by ELISA showed a maximal release of $18865.4{\pm}310.9pg/ml$ NGF at the 4th day and stably continued till 14 days by $rhNGF-{\beta}$ adenovirus infected Schwann cells. PC-12 cells exposed to media with $rhNGF-{\beta}$ adenovirus infected Schwann cell revealed at the same level of neurite-extension as the commercial NGF did. $rhNGF-{\beta}$ adenovirus injected experimental groups in comparison to the control group exhibited different taste preference ratio. Salty, sweet and sour taste preference ratio were significantly different after 2 weeks from the beginning of the experiment, which were similar to the sham group, but not to the control group.
미토콘드리아는 산화적 인산화와 연결된 전자전달을 통하여 에너지 생산에 중추적인 역할을 갖는다. 이 외에도 미토콘드리아는 신진대사, 세포자멸, 신호전달 그리고 활성산소 생성 등의 다양한 기능을 수행한다. 따라서, 미토콘드리아의 기능장애는 여러 인체질환에 영향을 준다는 것이 명백하다. 또한, 미토콘드리아 DNA의 돌연변이는 에너지 신진대사에 결함이 있는 여러 유전성 질환의 원인을 제공한다. 불행하게도 아직 이러한 유전성 미토콘드리아 DNA 질환의 치료법은 전무한 상태이다. 이러한 관점에서, 결함 미토콘드리아를 정상 미토콘드리아로 치환하는 최근의 시도는 큰 주목을 받고 있다. 본 연구에서는 녹색형광단백질로 표지된 미토콘드리아를 원심분리에 기반하여 생화학적으로 분리하고, 분리된 미토콘드리아를 동물복제에 쓰이는 미세주입 기법으로 소 난자에 전달 하였다. 이러한 미토콘드리아가 미세주입된 난자에서 단위발생을 유도하여 배반포 단계까지의 초기 발생과정에서 미토콘드리아 미세주입의 영향을 분석하였다. 미토콘드리아에 표지된 녹색형광단백질을 형광현미경으로 분석함으로써 미세주입으로 난자에 전달된 미토콘드리아는 빠르게 세포질에서 분산되고, 이 후 발생되는 딸세포에게 전달됨이 확인되었다. 따라서, 본 연구에서 수행된, 미세주입을 이용한 미토콘드리아의 전달은 최근 활발히 연구되는 미토콘드리아 치환 기법, 유전성 미토콘드리아 DNA 질환 치료법 및 동물복제 등에 유용한 모델로의 기여가 기대된다.
본 연구는 유기농업에서 생물자원으로서 담수 클로렐라의 활용 가능성을 연구하고자 유기농 생태계로부터 담수 녹조류를 분리, 동정하고, 생물학적인 특성을 조사하였다. 조사 지역의 수온은 $12.4{\sim}28.2^{\circ}C$, pH는 6.1~8.5이었다. 담수 녹조류를 분리할 때 고체배양법이 액체배양법보다 오염도가 낮고 분리 빈도가 높았다. 전국 9개 지역, 6개 담수 녹조류 서식처로 부터 총 115개의 균주를 분리하였다. 담수 녹조류의 분리 및 배양을 위해 질소원으로는 $NaNO_3$과 $KNO_3$, 탄소원으로 $Na_2CO_3$를 사용하였고, macro media의 구성 성분 중 $MgSO_4{\cdot}7H_2O$과 $CaCl_2{\cdot}2H_2O$를 분리하여 제조한 BGMM(BG11 Modified Medium)배지를 고안하였다. 담수녹조류는 배양 후 4일째부터 급격히 흡광도가 증가하였고 8일째부터 흡광도가 감소하였다. 공시한 7개의 균주 중 CHK008 균주가 배양 7일째에 가장 높은 흡광도를 보였다. 담수 녹조류 배양에 적합한 BGMM 배지의 pH는 6~7이었고 조사되는 빛이 강할수록 생육이 증가하였으며 5종류 당류 중에서 Glucose와 Galactose를 첨가하였을 때 클로랄라의 생육이 좋았다. 순수 분리한 16개 녹조류 균주의 균총색은 녹색, 진녹색, 연녹색을 나타내었고, 11개의 균주가 형광현미경하에서 강한 형광 빛을 나타내었다. 녹조류 16개 균주의 형태적인 특징을 조사한 결과 C. vulgalis, C. sorokiniana, C. pyrenoidosa, C. kessleri, C. emersonii, and Chlamydomonas sp.의 2개 속 6개종으로 동정되었다. 담수 녹조류의 세포 크기는 종마다 다양한 변이를 보였다. 대부분의 담수 녹조류의 세포형태는 구형이었다. Chlamydomonas sp.는 타원형이었고 Chlorella sorokiniana는 구형과 타원형이 섞여 있었다. 6개 녹조류 종류 중 Chlamydomonas sp.를 제외한 모든 균주는 편모가 없었다. Chlamydomonas sp. 1개 균주와 C. sorokiniana 5개 균주는 세포에서 점질물을 분비하였다.

Liposomes as drug delivery system have proved useful carrier for various disease, including cancer. In addition, perfluorocarbon cored microbubbles are utilized in conjunction with high-intensity focused-ultrasound (HIFU) to enable simultaneous diagnosis and treatment. However, microbubbles generally exhibit lower drug loading efficiency, so the need for the development of a novel liposome-based drug delivery material that can efficiently load and deliver drugs to targeted areas via HIFU. This study aims to develop a liposome-based drug delivery material by introducing a substance that can burst liposomes using ultrasound energy and confirm the ability to target tumors using PET imaging. Liposomes (Lipo-DOX, Lipo-DOX-Au, Lipo-DOX-Au-RGD) were synthesized with gold nanoparticles using an avidin-biotin bond, and doxorubicin was mounted inside by pH gradient method. The size distribution was measured by DLS, and encapsulation efficiency of doxorubicin was analyzed by UV-vis spectrometer. The target specificity and cytotoxicity of liposomes were assessed in vitro by glioblastoma U87mg cells to HIFU treatment and analyzed using CCK-8 assay, and fluorescence microscopy at 6-hour intervals for up to 24 hours. For the in vivo study, U87mg model mouse were injected intravenously with 1.48 MBq of 64Cu-labeled Lipo-DOX-Au and Lipo-DOX-Au-RGD, and PET images were taken at 0, 2, 4, 8, and 24 hours. As a result, the size of liposomes was 108.3 ± 5.0 nm at Lipo-DOX-Au and 94.1 ± 12.2 nm at Lipo-DOX-Au-RGD, and it was observed that doxorubicin was mounted inside the liposome up to 52%. After 6 hours of HIFU treatment, the viability of U87mg cells treated with Lipo-DOX-Au decreased by around 20% compared to Lipo-DOX, and Lipo-DOX-Au-RGD had a higher uptake rate than Lipo-DOX. In vivo study using PET images, it was confirmed that 64Cu-Lipo-DOX-Au-RGD was taken up into the tumor immediately after injection and maintained for up to 4 hours. In this study, drugs released from liposomes-gold nanoparticles via ultrasound and RGD targeting were confirmed by non-invasive imaging. In cell-level experiments, HIFU treatment of gold nanoparticle-coupled liposomes significantly decreased tumor survival, while RGD-liposomes exhibited high tumor targeting and rapid release in vivo imaging. It is expected that the combination of these models with ultrasound is served as an effective drug delivery material with therapeutic outcomes.
청자 투각고리문 의자는 4점 일괄유물로 경기도 개성에서 출토된 것으로 전해진다. 청자 의자는 고려시대 당시 청자 제작기술의 우수성과 화려한 생활상을 보여주는 등 미술사적으로 가치가 높아 보물로 지정 관리되어 왔다. 그러나 일괄유물 중 1점의 경우 과거 수리 복원된 것으로, 처리 재료의 열화, 처리자의 미숙함 등으로 인해 미적가치가 하락되었고, 구조적으로 불안하여 재처리의 필요성이 제기되었다. 처리 전 보존상태를 조사한 결과, 물리적 손상은 전반적으로 제조상 결함부위를 중심으로 인위적 손상이 가중되어 구조적으로 취약한 상태를 보였다. 균열부위 및 탈락된 편은 접합면이 맞지 않고 접착제가 청자 표면에 흐른 자국과 접착제 표면에 분진 등 2차적 오염물이 생겨 변질된 상태였다. 수리복원 상태를 조사하기 위해 자외선과 확대현미경을 이용하여 균열부의 접합 부위의 위치와 범위, 상태를 파악하였다. 적외선분광 분석(FT-IR)과 휴대용 X-선 형광분석을 실시하여 보존처리에 사용한 재료를 분석한 결과, 접착제로 셀룰로오스계 수지와 에폭시계 수지가 사용된 것을 확인하였다. 또한 일부 힘을 받는 접합부위에서는 접합강도를 높이기 위해 접착제에 석고(CaSO4·2H2O) 또는 골분(Ca10 (PO4)6(OH)2)을 첨가한 것을 알 수 있었다. 상태조사 결과를 바탕으로, 유물의 보존처리는 기존 접합된 상태에서 전면 해체하고 물리적으로 취약한 부분을 중심으로 접합·복원을 통해 보강하는데 중점을 두었다. 기존에 사용된 접착제를 제거하고 해체한 결과, 청자 의자는 크게 상부와 하부, 굽다리, 일부 고리 문 등 총 6개 편으로 분리되었다. 해체 후 접합면에 남아 있는 잔류 접착제 및 오염물은 화학적 및 물리적으로 제거하고 스팀세척기로 파단면 세척을 통해 재접합의 효율을 높였다. 유물의 접합은 접합부위와 크기에 따라 접착제를 다르게 적용하였다. 편의 위치만 고정하는 접합부에는 시아노아크릴계 수지 Loctite® 401을 사용하고 구조적으로 안정화시키는 부분에는 가역성을 위해 아크릴계 수지인 Paraloid® B-72 20%(in xylene)로 단면처리한 후 에폭시계 수지 Epo-tek® 301-2를 이용하여 접합하였다. 상·하부 접합 같이 힘을 받는 부위는 Epo-tek® 301-2에 Kaolin을 첨가하여 접합강도를 보강하였다. 연속되는 문양으로 추정 가능한 고리문의 결실 부분은 SN-Sheet로 뼈대를 만들고 Wood epos®로 파손단면을 연결하여 모델링하면서 고리문을 복원하였다. 그 외 접합하면서 생긴 복원 부위는 심미적 및 구조적 안정화를 위해 Wood epos®로 메움처리하였다. 복원부위 및 메움처리한 부분은 추후 전시활용에 있어 이질감이 없도록 색맞춤하였다. 다양한 과학기술을 활용한 조사와 처리과정은 체계적으로 기록하여 보존 관리하는데 기초자료로 활용하도록 하였다.
