Characteristics of lateral bud development, necrosis and genesis of flower primordium in 'Kyoho' grapevines

  • This study investigated the effective harvest of 'Kyoho' grapevines by examining their characteristics including bud development, necrosis types and flower primordium formation. The size of the axillary bud did not show any difference in the 5th node or more, but it was smaller because it was closer to the base in the 4th node or less. In the 1st node, the rates of main bud necrosis (MBN), accessory bud necrosis (ABN), and whole bud necrosis (WBN) were high, and the rate of flower primordium formation was low, but there was no significant difference in the other nodes. Therefore, it was expected that using other nodes than the 1st node would be advantageous to secure production. Because the growth progresses after sprouting, the main bud necrosis rate increases, showing a very low flower primordium formation rate in March of the following year. Therefore, a method is needed to increase the storage nutrients in the winter and the rate of flower primordium formation after March. This study found that the thickness of the shoots should be less than 8.5 mm between the 3rd and 4th nodes, and the length should be less than 60 cm for nodes up to the 10th node.

Changes in Dormant Phase and Bud Development of 'Fuji' Apple Trees in the Chungju Area of Korea (충주지역에서 '후지' 사과나무의 휴면단계 변화 및 눈 발달)

  • In this study, we investigated the onset and release of endo-dormancy under natural conditions by observing bud break characteristics in 'Fuji' apple trees using water cuttings. Through examinations of bud break rate and days to bud break, we found that the endo-dormancy of 'Fuji' apple tree continues for 70 d from 165 to 255 d after full bloom (DAFB), from late October to early January of the following year. In addition, within 20 d of first bud break, based on a final bud break rate of 60% or more, we able to identify the timing of the changeover from para-dormancy to endo-dormancy, and endo-dormancy to eco-dormancy. Analysis of the chilling requirement during the endo-dormancy period revealed that chilling accumulation up to 255 DAFB to release endo-dormancy amounted to 666 and 517 h based on the CH and Utah models, respectively. Observation of internal changes in the bud during endo-dormancy showed that flower bud differentiation begins from mid-July, and t ime of inflorescence o f the disk f lower is a vailable to f ind. The f lower buds subsequently developed slowly but steadily during endo-dormancy and in the following year in February, the developmental stage of each organ had progressed. Moreover, the flower buds of 'Fuji' apples were mostly healthy during the dormancy period, but some exhibited necrosis of flower primordium, due partial cell damage from the formation of ice crystals rather than a direct effect of the low temperature. Flower buds were formed in both the axillary buds of bourse shoots and terminal buds of spurs, but lower bud differentiation was observed for the terminal buds of spurs at rate of about 65% of total buds, which was directly related to the bud size and shoot diameter.

Effect of $CO_2$ Enrichment on the Differentiation of Multi-shoots and Saponin contents in Tissue culture of Korean ginseng (Panax ginseng C. A. Meyer) (인삼(人蔘) 조직배양(組織培養)에서 $CO_2$처리(處理)가 multi-shoot 분화(分化) 및 사포닌 함량(含量)에 미치는 영향(影響))

  • This experiment was conducted to study the effect of $CO_2$(0, 2, 500, 5, 000, 10, 000ppm) enrichment by enabling ventilation on micropropagation of multi-shoot and on the saponin contents in vitro in Korean ginseng (Panax ginseng C. A. Meyer). Embryo was cultured in Murashige and Skoog medium added 3mg/ l of Indolbutyric acid, Benzyladenin and Gibberellic acid $(GA_3)$, respectively. $CO_2$, enrichment had little effects on the number of adventitious buds and shoots originated from adventitious buds. The ratio of differentiated shoots to adventitious buds were about 50% in $CO_2$, enrichment treatment. The shoots originated from adventitious bud showed more rapid growth and had larger leaf area than the shoots originated from the leaf primordia did. The number of shoot primordia was the highest in 2, 500ppm of $CO_2$ enrichment treatment. On the contrary, 10,000ppm of $CO_2$, enrichment made smaller the number of shoot primordia and ratio of shoots to shoot primordia. The range of shoots differentiated was from shoot primordia were 15. 4 to 23. 9. The rate of dry weight of cultured shoots showed lowest (7. 5%) in control and highest (8. 59%) in 2, 500ppm of $CO_2$, enrichment. Rate of in vitro flower in control was 7.6% and that in 2500ppm of $CO_2$ was about twice (15.7-16.3%) as much as in control. Flower number per a embryo cultured was about 1.2-1.3. In the multi-shoots with callus enriched by 2, 500ppm of $CO_2$, the contents of crude saponin and ginsenosides in multi-shoots alone were higher than in multi-shoots with callus. The characteristics of ginsenosides in multi-shoots were especially the higher content of ginsenoside Rd, Re, and $Rg_1$.
