Anti-cancer effects of fenbendazole on 5-fluorouracil-resistant colorectal cancer cells

  • Benzimidazole anthelmintic agents have been recently repurposed to overcome cancers resistant to conventional therapies. To evaluate the anti-cancer effects of benzimidazole on resistant cells, various cell death pathways were investigated in 5-fluorouracil-resistant colorectal cancer cells. The viability of wild-type and 5-fluorouracil-resistant SNU-C5 colorectal cancer cells was assayed, followed by Western blotting. Flow cytometry assays for cell death and cell cycle was also performed to analyze the anti-cancer effects of benzimidazole. When compared with albendazole, fenbendazole showed higher susceptibility to 5-fluorouracil-resistant SNU-C5 cells and was used in subsequent experiments. Flow cytometry revealed that fenbendazole significantly induces apoptosis as well as cell cycle arrest at G2/M phase on both cells. When compared with wild-type SNU-C5 cells, 5-fluorouracil-resistant SNU-C5 cells showed reduced autophagy, increased ferroptosis and ferroptosis-augmented apoptosis, and less activation of caspase-8 and p53. These results suggest that fenbendazole may be a potential alternative treatment in 5-fluorouracil-resistant cancer cells, and the anticancer activity of fenbendazole does not require p53 in 5-fluorouracil-resistant SNU-C5 cells.

  • 본 논문에서는 청소로봇이 전 청소 영역을 CCPP(Complete Coverage Path Planning)를 이용해 경로를 생성한 후 재 경로계획 시 장애물의 미소한 변화로도 기존에 생성한 전 경로패턴을 바꾸지 않고 수정 할 수 있는 CD(Cell Decomposition)와 FN(Flow Network)을 이용한 CCPP 방식을 제안 하였다. 그리고 제안된 경로 계획에 대해 시뮬레이션으로 결과를 제시하였다.

The Effect of Turbulence Promoters on the Mass Transfer in Ultrafiltration (한외여과에서의 물질전달에 대한 난류촉진물체의 영향)

  • The GR51PP(MWCO 50,000) and GR40PP(MWCO 100,000) membranes manufactured by DDS were used in ultrafiltration of dextran(Mw. : 500,000) solution in flat plate ultrafiltration cell filled with various types of turbulence promoters. The flux improvement by using turbulence promoter was higher in laminar flow region than in turbulent flow region. The maximum improvements of permeate flux were foud as 112% and 50% I laminar flow region and turbulent flow region, respectively. Also, the solute rejection of the ultrafiltration membrane was improved by turbulence promoters and its effect was significant in the high transmembrane pressure and laminar flow region. The smaller the spacer mesh size was used, the higher the flux improved, but the pressure drop in ultrafiltration cell also increased. In laminar flow region, pressure drop by the spacer was negligible, but in turbulent flow region it changed significantly depending upon the mesh size of the spacer and therefore, its mesh size must be baken into account in the design of the process.   • A flow channel shape of PEMFC has an influence on the internal flow uniformity. If the reactant distribution in a flow path is not uniform during operation, both catalyst deactivation and mechanical damage of membrane could occur resulting in decreasing the membrane electrode assembly (MEA) durability. Numerous studies concerning flow design have been conducted to make smooth supply and uniform distribution of reactants in fuel cells. The baffle of flow path could improve fuel cell performance through the forced convection effect. A sub-channel, as an additional air flow path, could increase the reactant concentration and reduce the mass transfer loss via a smooth water discharge. In this study, computational fluid dynamics (CFD) was used to analyze the effect of blocks and sub-channels on the current density and oxygen concentration of the fuel cell. As a result, the limit current density and oxygen concentration at a rear block increased when using blocks and sub-channels in a flow channel. In particular, the current density increased significantly when the sub-channel was placed between two blocks. Also, the sub-channel position was optimized by analyzing the oxygen concentration, and the oxygen concentration was recovered at a rear block in the fuel cell.

  • There is a great demand to manipulate biological cell autonomously since biologist should spend much time to obtain skillful manipulation techniques. For this purpose, we propose a cell chip to control, carry, fix and locate the cell. In this paper, we focus on the cell rotator to rotate individual biological cell based on a micro fluidics technology. The cell rotator consists of injection hole and rotation well to rotate a biological cell properly. Under the variation of flow rate in injection hole, the angular velocity of a biological cell is evaluated to find the feasibility of the proposed rotation method. As a practical experiment, Zebrafish egg is employed. Based on this research, we find the possibility of non-contact rotation way that can highly reduce the damage of the biological cell during manipulation. To realize an autonomous biological cell manipulation, a cell chip with manipulation well and micro channel in this research will be utilized effectively in near future.

Modulatory Effect of BAY11-7082 on CD29-mediated Cell-cell Adhesion in Monocytic U937 Cells (BAY11-7082에 의한 U937 세포의 CD29-매개성 세포간 유착과정 조절 효과)

  • BAY11-7082 was initially found to be an anti-inflammatory drug with NF-${\kappa}B$ inhibitory property. In this study, we evaluated modulatory function of BAY11-7082 on U937 cell-cell adhesion induced by CD29 (${\beta}1$-integrins). BAY11-7082 strongly blocked functional activation of CD29 (${\beta}1$-integrins), as assessed by cell-cell adhesion assay. However, this compound did not block a simple activation of CD29, as assessed by cell-fibronectin adhesion assay. In particular, to understand molecular mechanism of BAY11-7082-mediated inhibition, the regulatory roles of CD29-induced actin cytoskeleton rearrangement under cell-cell adhesion and surface level of CD29 were examined using confocal and flow cytometic analysis. Interestingly, this compound strongly suppressed the molecular association of actin cytoskeleton with CD29 at cell-cell adhesion site. Moreover, BAY11-7082 also diminished surface levels of CD29 as well as its-associated adhesion molecule CD147, but not other adhesion molecules such as CD18 and CD43. Therefore, our data suggest that BAY11-7082 may be involved in regulating immune responses managed by CD29-mediated cell-cell adhesion.

Numerical Investigation of the Discharge Efficiency of a Vanadium Redox Flow Battery with Varying Temperature and Ion Concentration (온도와 이온농도의 변화에 대한 바나듐 레독스 플로우 배터리의 방전 효율에 관한 수치해석)

  • In this study, a numerical simulation of a vanadium redox flow battery was investigated for reactions involving an electrochemical species using comprehensive conservation laws and a kinetic model. For a 3-D geometry of the cell, the distributions of electric potential, vanadium concentration, overpotential, and ohmic loss were calculated. The cell temperature and initial vanadium ion concentration were set as variables. The voltage and electrochemical loss were calculated for each variable. The effects of each variable's impact on the electrochemical performance of a vanadium redox flow battery was numerically analyzed using the calculated overpotential in the electrode and the ohmic loss in the electrolyte phase. The cell temperature increased from $20^{\circ}C$ to $80^{\circ}C$ when the voltage efficiency decreased from 89.34% to 87.29%. The voltage efficiency increased from 88.65% to 89.25% when the vanadium concentration was changed from $1500mol/m^3$ to $3000mol/m^3$.

  • Design of location erea(LA) in a cellular network is to partition the network into clusters of cells so as to minimize the cost of location updating and paging. Most research works dealing with the LA design problem assume that the call. arrival rate and mobile flow rate are fixed parameters which can be estimated independently. In this aspect, most Problems addressed so far are deterministic LA design problems(DLADP), known to be NP hard. The mobile flow and call arrival rate are, however, varying with time and should be treated simultaneously because the call arrival rate in a cell during a day is influenced by the change of a population size of the cell. This Paper Presents a new model on IA design problems considering the time-dependent call arrival and mobile flow rate. The new model becomes a stochastic LA design problem(SLADP) because It takes into account the possibility of paging waiting and blocking caused by the changing call arrival rate and finite paging capacity. Un order to obtain the optimal solution of the LA design problem, the SIADP is transformed Into the DLADP by introducing the utilization factor of paging channels and the problem is solved iteratively until the required paging quality is satisfied. Finally, an illustrative example reflecting the metropolitan area, Seoul, is provided and the optimal partitions of a cell structure are presented.

  • Anodic half-reaction in the $Na^+$ ionic sensors using $Na_2CO_3$ and $MCO_3$ ($M=Cs_2,K_2,Li_2,Ca$) as a sensitive membrane is derived in continuous flow system to explain $CO_2$ sensing characteristics. For various gas-sensitive membranes, a well known overall reaction, $MCO_3\;=\;MO\;+\;CO_2$, cannot be applied for the EMF behaviors of these kinds of sensors. So, the anodic reaction is found to involve $Na_2CO_3$ and $M^{++}$-containing oxide phases by employing the ion exchange reaction at the interface of solid electrolyte and the sensitive membrane to maintain ionic balance in the whole cell. Based on the electrode reaction derived in flow system, differences of cell potentials between continuous and discontinuous flow systems were also discussed. These EMF differences were considerably caused by the partial pressures of oxygen and $CO_2$ as well as irreversible chemical reactions between electrode materials and $CO_2$ atmosphere.

  • Ejector system is a device to transport a low-pressure secondary flow by using a high-pressure primary flow. Ejector system is, in general, composed of a primary nozzle, a mixing section, a casing part for suction of secondary flow and a diffuser. It can induce the secondary flow or affect the secondary chamber pressure by both shear stress and pressure drop which are generated in the primary jet boundary. Ejector system is simple in construction and has no moving parts, so it can not only compress and transport a massive capacity of fluid without trouble, but also has little need for maintenance. Ejectors are widely used in a range of applications such as a turbine-based combined-cycle propulsion system and a high altitude test facility for rocket engine, pressure recovery system, desalination plant and ejector ramjet etc. The primary interest of this study is to set up an applicable model and operating conditions for an ejector in the condition of sonic and subsonic, which can be extended to the hydrogen fuel cell vehicle. Experimental and theoretical investigation on the sonic and subsonic ejectors with a converging-diverging diffuser was carried out. Optimization technique and numerical simulation was adopted for an optimal geometry design and satisfying the required performance at design point of ejector for hydrogen recirculation. Also, some sonic and subsonic ejectors with the function of changing nozzle position were manufactured precisely and tested for the comparison with the calculation results.
