High Productivity Ethanol Fermentation Using Flocculant Yeast (응집성 효모에 의한 고생산성 알콜 발효)

  • A tower fermentor equipped with a modified settler was used for ethanol fermentation using highly flocculating yeast, Saccharomyces uvarum. The settler was constructed of glass column divided into two chambers by a funnel shaped divider. Gas was allowed to escape from lower chamber of the settler through a small tube. This design significantly reduced the turbulence in upper chamber of the settler and made it possible to operate at high dilution rate. Using the tower fermentor, the effects of operating conditions such as initial glucose concentration, dilution rate and cell recycle ratio were studied. The maximum ethanol productivity, 64.0 g/l' h was obtained at a dilution rate 1.1 h -1 and a cell recycle ratio 5 with the corresponding ethanol concentration of 58.8 g/l, and cell mass of 88 g/l.

Production of Bioflocculant by Agrobacterium sp. KF-67 (Agrobacterium sp. KF-67에 의한 미생물 응집제 생산)

  • Among 120 microorganisms isolated from soil, KF-67 was the best producer of flocculant and was examined for flocculating ability in the kaolin clay and CaCl2 suspension. KF-67 was identified to be a species belong to the genus Agrobacterium sp. The influence of components of the culture medium for flocculant production by Agrobacterium sp. KF-67 was studied. The favorable carbon and inorganic nitrogen source for production of the flocculant were glucose and NH4NO3 and their addition concentrations were 2% and 0.1%, respectively. Addition of the organic nitrogen such as yeast extract, peptone and inorganic salt such as CaCO3 significantly increased the production of flocculant. These result indicated that the production of flocculant by Agrobacterium sp. was significantly affected by both organic nitrogen and inorganic salt. The components of the optimum culture medium were 2% glucose, 0.1% NH4NO3, 0.01% yeast extract, 0.01% peptone, 0.04% CaCO3, 0.03% NaCl in initial pH 7.5 when cultured with rotary shaker controlled at 3$0^{\circ}C$ and 120 rpm. Under the optimum culture medium, flocculant production was highly improved about 76% than that isolation medium.

  • A bioflocculant producing bacterium for wastewater treatment was isolated and identified as Lactobacillus jensenii. The bioflocculant was supposed as a polysaccharide. Lactobacillus jensenii YW-33 produced the flocculant with highest activity in the medium composed of 2% sucrose, 0.05% tryptone, 0.5% yeast extract, 0.01% K$_{2}$HPO$_{4}$, 0.01% KH$_{2}$PO$_{4}$, 0.01% NaCl, 0.005% MnSO$_{4}$, 5H$_{2}$O and 0.2% Tween 80. The optimal culture pH and temperature were 7.0 and 25$\circ$C, respectively. In the time course of production with jar fermentor, the productivity of the flocculant was increased in proportion to the cell growth and the cultivation time for maximum production was 24 hrs, showing flocculating activity of 1,130 units per ml.
