Expressional Analysis of STAT2 Gene in Rock Bream, Oplegnathus faciatus, Under LPS or Poly I:C Stimulation and Megalocityvirus Infection

  • Rock bream iridovirus (RBIV) is a megalocytivirus widely infected in various fish species in Korea, causing symptoms of acute inflammation and enlargement of spleen. In our previous study, RBIV induced the initial upregulation but later down-regulation of proinflammatory cytokines and IFN1 gene expression. Signal transducers and activators of transcriptions (STAT) are transcription factors involved in the regulation of immune genes including IFNs. This study was conducted to analyse the expression of STAT2. The expressional study of STAT2 gene was performed in head kidney and spleen upon RBIV infection and immune stimulants like LPS or poly I:C in vitro. Consequently, STAT2 gene expression pattern was different in head kidney and spleen as it was significantly up-regulated by LPS from 4 h to 8 h but down-regulated at 24 h while up-regulated by poly I:C at 8 h in head kidney while, in spleen, STAT2 gene expression was down regulated by LPS but significantly up-regulated by poly I:C. Upon RBIV stimulation, STAT2 gene expression was significantly down-regulated by high dose RBIV at 4 h but up-regulated at 8 h and 24 h in head kidney. In spleen cells, it was up-regulated by medium dose RBIV at 4 h and by high dose RBIV at 4 h and 8 h but down regulated later then. In vivo, STAT2 gene expression was not significantly affected by RBIV infection while significant up-regulated by vaccination at day 7 post-vaccination, indicating STAT2 gene can be involved in adaptive immune response in rock bream.

넙치 배아세포에서 tacrolimus에 의한 DNA 손상, 세포사멸 및 염증성 반응에 대한 luthione의 억제 효과 (Inhibitory effect of luthione on tacrolimus-induced DNA damage, apoptosis and inflammatory response in olive flounder natural embryo cells)

  • Tacrolimus, a type of macrolide produced by Streptomyces tsukubaensis, is widely used as an immunosuppressant. However, continuous exposure to tacrolimus causes oxidative stress in normal cells, ultimately inducing cell injury. Therefore, this study investigated whether luthione, a reduced glutathione, could inhibit tacrolimus-induced cytotoxicity in olive flounder (hirame) natural embryo (HINAE) cells. According to the results, luthione significantly inhibited tacrolimus-induced reduction in cell viability in a concentration-dependent manner. Additinally, although luthione unaffected autophagy by tacrolimus, tacrolimus-induced apoptosis was significantly suppressed in the presence of luthione. Luthione also markedly blocked DNA damage in tacrolimus-treated HINAE cells, associated with the inhibition of reactive oxygen species (ROS) generation. Additionally, tacrolimus cytotoxicity in HINAE cells was correlated with increased inflammatory response, also attenuated by luthione. Collectively, these results show that at least luthione protects HINAE cells against tacrolimus-induced DNA damage, apoptosis, and inflammation, but not autophagy, by scavenging ROS. Although additional in-vivo studies are required, this study's results can be used as a basis for utilizing luthione to reduce the toxicity of fish cells caused by excessive immune responses.

Field experiment on effect of butaphosphan and cyanocobalamin complex on the immunity and stress of olive flounder at low temperature

  • In this study, a fish metabolic accelerator (a combination of butaphosphan and cyanocobalamin [BPC]) was injected into the muscle of the olive flounder, Paralichthys olivaceus, to investigate its effect on immunity and stress in fish maintained at low temperatures. A single dose of BPC was injected (100 mg/kg body weight) into the olive flounder, and its immunity and stress were observed after one and two weeks. Immunity tests revealed the presence of lysozyme (LZM), nitroblue tetrazolium (NBT), myeloperoxidase (MPO), anti-protease (AP), glutathione peroxidase (GPx), and total immunoglobulin (TIg). BPC injection was found to increase immunity activity compared to the control group. In particular, there was significantly high GPx activity. There was similarly high activity for MPO and GPx in the first week following the injection, followed by significant differences between the BPC-injected and control groups in the second week. There was a reduced low water-temperature stress response in the BPC-injected fish, as evidenced by the cortisol and glucose levels of the control and BPC groups. Lower levels were also observed in the BPC group than the control group during the second week. Cortisol levels were significantly lower in the BPC group than the control group. Histological examinations were conducted in the first and second weeks after the intramuscular injection of the recommended BPC dose to confirm the safety of BPC in aquaculture. There were no abnormalities observed in any tissue samples. This study confirms that the injection of BPC is safe even when used in a culture situation. BPC helps relieve stress and improves non-specific immune responses (innate immunity) in the olive flounder.

Vibrio균에 대한 뱀장어 (Anguilla japonica)의 면역반응 (Immune Response of the Japanese Eel(Anguilla japonica) to Vibrio anguillarum)

  • 경남 김해 명지 양어장에서 사육중이던 뱀장어(평균체중 20g)에 Vibrio anguillarum vaccine을 주사하여 일정기간이 경과한 뒤 생균을 감염시켜 뱀장어의 면역효과를 조사하였다. 이 실험에서 사용된 vaccine은 $25^{\circ}C$로 조절된 $0.3\%$의 formalin에 24시간 방치하여 사균화시킨 formalin-killed vaccine, $121^{\circ}C$에서 3분 및 15분간 가열한 vaccine등 3종류의 vaccine이다. 이 3종류의 vaccine을 접종했을 때 효과의 차이와 formalin-killed vaccine의 접종 시 온도와 접종농도에 따른 효과를 조사했으며 booster접종의 필요여부도 검토했다. 1984년 12월 4일부터 85년 4월 30일까지 실시한 연구 결과는 다음과 같다. 1. 수온 $15^{\circ}C$에서 $23^{\circ}C$의 범위까지 항체생산에 의한 응집력가는 수온이 높을수록 높게 나타났고, $23^{\circ}C$$28^{\circ}C$에서는 응집력가는 별차이가 없었다. 2. 사균 vaccine 접종양을 $10^7cells/$미로 주사하면 면역능력이 아주 미비하였고, $10^8cells/$미에서 강한 면역능력을 획득했다. 그러나 $10^9cells/$미로 증가시켜도 $10^8cells/$미 때와 유사하였다. 3. $10^8cells/$미를 2회 접종시켰을 때 1회만 접종시켰을 때보다 효과가 훨씬 좋았으나 3회 접종시켰을 때는 2회 접종구와 면역효과의 차이가 없었다. 4. formalin으로 처리된 사균 vaccine이 완전면역을 나타내었으며 $121^{\circ}C$로 열처리한 vaccine의 경우는 면역효과가 약했다.

수산생물의 생산과 관리에 관한 기초연구 : ELISPOT 기법을 이용한 넙치의 항체생성 세포분석 (Study on the Production and Management of Aquatic Animal : Application of ELISPOT-Assay for the Detection of Antibody Secreting Cells in Flounder, Paralichthys olivaceus)

  • 한국 양식산업에서 중요한 어종인 넙치 (Paralichthys olivaceus) 포르말린으로 처리한 E. tarda를 항원으로 하였을 때의 면역반응 분석을 위하여 ELISPOT 기법을 적정화시킨 후 넙치의 각 장기에 있는 총 항체생성세포와 특이 항체생성세포를 계수하는데 응용하고자 하였다. 전신과 비장의 항체생성세포를 2.5시간 이상 96 well plate에 배양하면 충분히 분석이 가능하였다. 그러나 총 또는 특이 항체생성세포 분석을 위하여 과량의 토끼 항 넙치 면역글로불린 또는 E. tarda 항원을 plate에 coating하는 것은 오히려 ELISPOT법의 감도를 감소시키는 것으로 나타났다. ELISPOT법의 특이성은 단백질 합성 억제제인 cycloheximide를 처리한 임파세포에서 총 항체생성세포가 발견되지 않는 것으로서 입증할 수 있었다. 특이 항체생성세포 수의 최대치는 면역 3주째에 나타났으며 이후 계속 빠르게 감소하여 7주째는 거의 발견되지 않았다. 이러한 반응은 신장과 비장에서 유사하게 나타나 임파장기에 따른 차이점은 발견할 수 없었다. 면역 후 2주와 3주 사이에 혈청내 특이 항체량 또한 빠르게 증가하여 ELISPOT법으로 분석된 특이 항체생성세포 수의 변화와 일치함을 발견할 수 있었다. 그러나 증가된 혈청내 특이 항체량이 면역 5주부터 실험 종료 시점까지 계속 높은 수준으로 유지되고 있는 것은 급격한 감소를 보이는 특이 항체생성세포의 동력학적 변화와는 명확히 구별되는 점이었다.

Expression of phospholipase C β1 in olive flounder (Paralichthys olivaceus) following external stress stimulation

  • In this study, to clarify the function of $PoPLC-{\beta}1$, in response to stress challenge, we examined the $PoPLC-{\beta}1$ expression pattern in response to external stress (pathogen-associated molecular pathogen challenge and environmental challenge including temperature and salinity). $PoPLC-{\beta}1$ expression analysis of tissue from olive flounder showed that the messenger RNA (mRNA) was predominantly expressed in the brain, heart, eye, liver, spleen, and stomach. We also tested the mRNA expression of the $PoPLC-{\beta}1$ in the spleen and kidney of olive flounder by RT-PCR and real-time PCR following stimulation with lipopolysaccharide (LPS), concanavalin A (ConA), or polyinosinic:polycytidylic acid (PolyI:C) and compared with the inflammatory cytokines IL-1b and IL-6 in the stimulated flounder tissues. Each of the spleen and kidney and mRNA transcripts of $PoPLC-{\beta}1$ were increased 30- and 10-fold than normal tissue at 1-6 h post injection (HPI) with PolyI:C when the expression of $PoPLC-{\beta}1$ transcript was similar to LPS and ConA. We also tested the expression of $PoPLC-{\beta}1$ in response to temperature and salinity stress. The expression of $PoPLC-{\beta}1$ also was affected by temperature and salinity stress. Our results provide clear evidence that the olive flounder $PLC-{\beta}1$ signal pathways may play a critical role in immune function at the cellular level and in inflammation reactions. In addition, $PLC-{\beta}1$ appears to act as an oxidative-stress suppressor to prevent cell damage in fish.

마늘, Allium sativum이 넙치, Paralichthys olivaceus의 면역반응에 미치는 영향 (Effects of garlic Allium sativum on the immune responses of olive flounder Paralichthys olivaceus)

  • 본 연구에서는 마늘 추출물과 착즙액이 넙치의 비특이적 면역력에 미치는 영향과 투여 방법에 따른 효과를 밝히고자 하였다. 마늘 추출물의 투여는 복강 주사법을, 마늘 착즙액은 침지법을 사용하였다. 마늘 착즙액 침지법은 일정 농도의 착즙액을 해수에 현탁하여 2일 간격으로 3회 침지하였다. 병원균의 공격 시험은 시험어에 마늘 추출물의 일정량을 2일 간격으로 3회 주사한 지2일 후에 실시하였다. 그 결과, 마늘 추출물의 복강 주사 시험구 중 5% 투여구에서 lysozyme의 활성이 가장 유의적인 증가를 나타내었다. 호중구의 수는 1, 5 및 10%를 주사하였을 때 유의적인 증가를 보였다 (p<0.05). SOD activity는 신장에서는 1, 3 및 5% 시험구에서, 간에서는 1 및 5% 시험구가 대조구에 비해 유의적인 증가를 나타내었다 (p<0.05). NBT 환원 시험은 3 및 5% 시험구에서 높은 NBT 환원량을 나타내었다. 마늘 착즙액의 침지 시험에서는 0.25 g/L 시험구에서 체표 점액 lysozyme 활성과 혈청 lysozyme 활성이 가장 높은 유의적인 증가를 하였다. 림프구와 호중구의 수는 0.25, 0.5 및 1.0 g/L 시험구에서 대조구에 비해 유의적인 증가를 보였다 (p<0.05). SOD activity는 신장에서는 모든 시험구에서, 간에서는 0.1, 0.25 및 0.5 g/L 시험구에서 대조구에 비해 유의적인 증가를 나타내었다 (p<0.05). NBT 환원 시험은 0.1, 0.25 및 0.5% 시험구에서 대조구와 비교하여 유의적인 증가를 나타내었다 (p<0.05). 시험구의 생존율은 S. iniae BS10과 E. tarda KE-1의 공격에 대하여 5% 마늘 추출물 복강 투여와 0.25 g/L 마늘 착즙액 침지 시험구의 상대생존율이 가장 높았다. 이상의 결과를 종합하여 볼 때 마늘 추출물과 착즙액이 넙치에 면역능 향상과 어류 질병에 저항성 증강에 효과가 있는 것으로 사료된다.

Changes in Blood Cell Morphology and Number of Red Spotted Grouper, Epinephelus akaara in Response to Thermal Stress

  • Rising of water temperature due to global warming is a great concern to aquaculturists and fishery biologists. Hence, the present study aimed to investigate the effects of high water temperature on juvenile red spotted grouper, Epinephelus akaara based on the evaluation of stress responses in blood. E. akaara juveniles were exposed to different thermal conditions ($25^{\circ}C$, $28^{\circ}C$, $31^{\circ}C$, and $34^{\circ}C$) for 6 weeks following 2 weeks of acclimation at $25^{\circ}C$. Blood cell morphology and number were examined at three sampling points (2, 7, and 42 days) from a total of 180 fish. Major erythrocytic cellular abnormalities (ECA) observed in blood smears of thermally stressed groups ($31^{\circ}C$ and $34^{\circ}C$) after 6 weeks were echinocytes, teardrop-like cells, swollen cells and vacuolated cells. Both red and white blood cell number (RBC and WBC) were significantly (p<0.05) elevated in $31^{\circ}C$ and $34^{\circ}C$ group after 6 weeks thermal exposure. Differential leucocytes number showed significant increases in neutrophil (N) and decreases in lymphocytes (L) in the highest temperature ($34^{\circ}C$). Different N:L ratio was observed at different thermal conditions which can be used as a reliable alternative to measure stress response. Taken together, these results suggest that higher temperature ($31^{\circ}C$ and $34^{\circ}C$) can interfere the immune system of red spotted grouper by altering the blood cell morphology and number.

The Expression Analysis of Complement Component C3 during Early Developmental Stages in Olive Flounder (Paralichthys olivaceus)

  • Fish larvae are immediately exposed to microbes from hatching to maturation of their lymphoid organs, therefore effective innate mechanisms is very important for survival in such an environment. The key component of innate immune system, C3 is central protein of all activation pathways of the complement system, leading to inflammatory reactions, such as opsonisation, chemotaxis, and cell lysis of pathogens. Although, innate mechanisms is essential for survival in the early stage of development, little is known about defence mechanisms. In this study, the alignment analysis showed that amino acid sequence of C3 from olive flounder liver EST homologous to other known C3 sequences with 73-99% identity. Also, we examined the tissue distribution of olive flounder C3 and analyzed expression pattern from the fertilized egg until 28 days post hatching. As a result, olive flounder C3 mRNA was expressed only in the liver and the mRNA level more increased as developmental proceed during the early stage. These results may suggest that olive flounder C3 plays an important function in the early immune response of olive flounder larvae.

External Application of Fermented Olive Flounder (Paralichthys olivaceus) Oil Alleviates Inflammatory Responses in 2,4-Dinitrochlorobenzene-induced Atopic Dermatitis Mouse Model

  • Allergic skin inflammation such as atopic dermatitis (AD) is characterized by edema and infiltration with various inflammatory cells such as mast cells, basophils, eosinophils and T cells. Thymic stromal lymphopoietin (TSLP) is produced mainly by epidermal keratinocytes, as well as dermal fibroblasts and mast cells in the skin lesions of AD. Omega-3 polyunsaturated fatty acids in fish oil can reduce inflammation in allergic patients. Fermentation has a tremendous capacity to transform chemical structures. The antiinflammatory effects of fish oil have been described in many diseases, but the beneficial effects by which fermented olive flounder oil (FOF) modulates the allergic response is poorly understood. In this study, we produced FOF and tested its ability to suppress the various allergic inflammatory responses. The ability of FOF to modulate the immune system was investigated using a mouse model of AD. The FOF-treated group showed significantly decreased immunoglobulin E (IgE) and histamine in serum. Also, the increased TSLP expression was significantly inhibited in the FOF group; the FOF-treated group was not appreciably different from the hydrocort cream treatment group. In addition, FOF treatment resulted in a smaller spleen size with reduced the thickness and length compared to the induction group. Splenocytes from mice treated with FOF produced significantly less IFN-${\gamma}$, IL-4, T-box transcription factor (T-bet) and GATA binding protein 3 (GATA3) expression compared with the induction group. These results suggest that FOF may be effective in treating the allergic symptoms of AD. 5.
