Adequacy Evaluation of Fish Intake Parameter used for Human Health Risk Assessment to Derive Freshwater Quality Criteria in Korea (국내담수지역 인체위해성기반 준거치 산정에 활용되는 어류섭취량인자 타당성 평가)

  • Water quality criteria for human health protection are derived based on the human health risk assessment. Water quality criteria in Korean freshwater bodies have been derived according to the equations developed by the US Environmental Protection Agency. The equations include the fish intake parameter, which is very important factor that significantly influences on the criteria derivation. So far, several fish intake values were used in human health risk assessment for water quality standards and effluent standards. However, these values are not consistent and they refer to various sources. Therefore, there is a need to suggest the most appropriate value of fish intake parameter to derive freshwater quality criteria in Korea. In this study, national and international fish intake values were widely collected and evaluated to select the adequate value of fish intake parameter that can be applied in Korea. The USEPA presented fish intake parameter as the 17.5 g/day for general adults and sport fishers and 142.4 g/day for subsistence fishers. In Korean reports, wide range values of 2 to 67.7 g/day were suggested as fish intake value. These values included finfish and shellfish intakes in common but had various habits. This study found that the 52.4 g/day suggested in Korean Exposure Factors Handbook published by the Ministry of Environment in 2007 seemed to be the suitable fish intake parameter to derive the freshwater quality criteria in Korea. The value is based on water corrected intakes of finfish and shellfish present in freshwater and coastal areas. We expect that this report can be useful to select suitable fish intake value in human health risk assessment for establishing freshwater quality standard in Korea.

Bacterial Mutagenicity of some Hot-air Dried Shellfish and Canned Products of some Red-muscle Fish During Storage (열풍건조된 몇 가지 패류와 적색어류 통조림의 저장조건에 따른 돌연변이성 변화)

  • Bacterial mutagenicity of shellfish or canned products of red-muscle fish affected by hot-airdry processings and/or during storages was investigated by Ames preincubation method. Mussel (Mytilus edulis) and surf clam (Tapes japonica) dried under either 40$\circ$C, 50$\circ$C or 60$\circ$C until remained about 25 % water content was stored at 4$\circ$C or 25$\circ$C for 6 months. Commercially available canned product of pacific saury (Cololabis saira) and mackerel saury (Scotnberjaponicus), whose producer or date of manufacture was identical each other, was stored at 25$\circ$C for 6 months. Mutagenic substances from sample (25 g dry weight) were extracted and purified for mutagenicity assay against S. typhitnurium TA98 by the simultaneous distillation extraction (SDE) method (Lee et al. 1955). Mutagenicity of sample was expressed number of revertant per 25 g dry sample. Number of revertant from unheated mussel sample was appeared to be 135, relative to 22 from unheated surf clam sample. Mutagenicity of both shellfish was strongly affected by heat treatment, but a lesser extent by storage conditions, even at 25$\circ$C for 6 months. Revertants of the mussel sample dried at 40$\circ$C, 50$\circ$C, or 60$\circ$C was found to be 227, 779, and 883, respectively. Similar, but lower, mutagenicity was observed from the sample of surf clam dried at 40$\circ$C, 50$\circ$C, or 60$\circ$C than mussel. Mutagenicity of canned pacific saury and mackerel samples was not influenced by storage conditions. Number of revertant from pacific saury was 175 which was relatively higher than 5 from mackerel. Mutagenicity of fresh pacific saury and mackerel samples was significantly higher than that of their counterparts from canned products. These results indicate that mutagenicity of the shellfish was affected by heat treatment (40$\circ$C, 50$\circ$C, or 60$\circ$C) used for drying process, but not much by storage for 6 months at 4$\circ$C or 25$\circ$C and the mutagenicity of canned products of red-muscle fish was not influenced by storage, even at 25$\circ$C for 6 months.

  • The effectiveness of fishery resources enhancement of artificial reef settled in the coast of Cheju Island was determined by using the samples collected by scuaba and caught by the trammel net. The obtained results are summarized as follows; 1. Six types of shellfish were found in the artificial reef area, and the amount of shellfish living in that area was $490{\sim}3,483g/m^2$ according to our research. This shows a high density of shellfish in that area. 2. Ten types of seaweeds were found in the artificial reef area, and the reef living in that area was abundant, with $3,556{\sim}10,550g/m^2$. 3. The amount of a catch of fish in the artificial reef area was 3.5~21.4 times more than in the control area. This result shows the high productivity in that area.

  • Vibrio alginolyticus, a marine fish and shellfish pathogen, has been found at a high frequency around the coastal areas of Korea. Vibrio alginolyticus was purified from various diseased fish, and a V. alginolyticus-specific bacteriophage was isolated from seawater obtained from fish farms located on the west coast of Korea. In a bacterial lysis experiment using the phage and antibiotics, tetracycline, $10^3$ cfu/ml of V. alginolyticus were completely lysed by both the phage and the antibiotic, suggesting that the purified phage in the study could be utilized as an alternative to antibiotics in the control of fish and shellfish diseases caused by V. alginolyticus.

Studies on the Isolation of Vibrio damsela (Vibrio damsela의 분리연구)

  • Authors studied on the isolation of V. damsela from sea water, fish and shellfish at the Keoje Hae keumkang on the southern sea and at Hongdo island and Heucksan island on the western sea of Korea from May to September in 1986. Authors investigated for the isolated strains to bacteriological identification, hemolysis about various erythrocytes and antibiotic susceptibilities. The results obtained were as follows: 1. V. damsela was isolated 14 strains from total 383 specimens; 233 cases of sea water, 40 cases of fish and 110 cases of shellfish, respectively. Eight strains were isolated from sea water and 6 strains were isolated from shellfish. 2. The biochemical characteristics which differentiate it from other Vibrio species were indole negative, ornithine negative, Voges-Proskauer positive, arginine positive, galactose positive, glucose positive, maltose positive, mannose positive, trehalose positive, and growth in nutrient broth with 1% to 6% NaCl. 3. On hemolysis reaction on blood agar media using human, rabbit and guinea pig erythrocytes, human erythrocytes were 11 strain positive, rabbit erythrocytes were 12 strain positive and guinea pig erythrocytes were 13 strain positive. 4. Senistivity test using with chemotherapeutic agents of "BioLab" Microbial Sensitivity Test Discs were generally sensitived to amikacin, ampicillin, cephalothin, chloramphenicol, clindamycin, erythromycin, gentamycin, kanamycin, methicillin, penicillin, streptomycin, tetracycline and tobramycin, respectively, but were resistant to lincomycin.

Vitamin $B_{12}$ Content Using Modified Microbioassay in Some Korean Popular Seaweeds, Fish, Shellfish and Its Products (미생물분석법을 이용하여 한국인이 즐겨 섭취하는 일부 해조류 및 어패류와 그 가공식품의 비타민 $B_{12}$ 함량 분석)

  • There is a limitation to estimate vitamin $B_{12}$ intake due to a lack of data on vitamin $B_{12}$ content in many Korean foods. In this study, vitamin $B_{12}$ content was determined in some seaweeds, fish, and shellfish and their product that are consumed in Korea using a modified microbioassay with Lactobacillus delbruecki ATCC 7830. Dried laver and dried seasoned and toasted laver contained very high levels of vitamin $B_{12}$ (66.8 and $55.2-71.3\;{\mu}g$/100 g, respectively. Sea lettuce and seaweed fulvescene also contained high vitamin $B_{12}$ content of 5.47-9.41 and $6.46-7.20\;{\mu}g$/100 g, respectively, whereas sea mustard and sea tangle contained low levels of vitamin $B_{12}$; vitamin $B_{12}$ was not detected in seaweed fusifome. Pacific saury, trout, sea-bass, or squid contained 12.01, 2.00, 0.49 and $2.33\;{\mu}g$ vitamin $B_{12}$/100 g, respectively. Ochellatus octopus, and naked sand lance contained 0.72-1.43 and $3.68\;{\mu}g$ vitamin $B_{12}$/100 g, respectively. Dried Alaska pollack con-tained $0.19-2.64\;{\mu}g$ vitamin $B_{12}$/100 g. Shellfish such as little neck clam and small ark shellfish contained high levels of vitamin $B_{12}$ of $30.5-40.5\;{\mu}g$/100 g, and mussel and abalone contained 17.71 and $7.82\;{\mu}g$/100 g, respectively. Of unique Korean traditional fermented seafood products, salt-fermented products of squid ($2.91\;{\mu}g$/100 g), clams ($34.31\;{\mu}g$/100 g), Ala-ska pollack roe ($9.98-12.02\;{\mu}g$/100 g), hairtail guts ($4.58\;{\mu}g$/100 g) or small shrimp ($0.58-1.55\;{\mu}g$/100 g), and fish sauce from anchovies ($1.52-1.78\;{\mu}g$/100 mL), sand eel ($0.22-0.24\;{\mu}g$/100 mL) or small shrimp ($0.19-0.78\;{\mu}g$/100 mL) were analyzed. A few commercial brands of flying fish roe ($0.73-1.73\;{\mu}g$/100 g), canned tuna ($0.40\;{\mu}g$/100 g), and fried fish paste ($0.25-0.69\;{\mu}g$/100 g) were also analyzed. In conclusion, vitamin $B_{12}$ content in these foods, chosen considering the Korean food culture, should contribute to improve the present vitamin $B_{12}$ food database. It may be helpful to estimate vitamin $B_{12}$ intake more correctly than before, and provide additional information for dietary education related to vitamin $B_{12}$ and meal management.

Monitoring of residue PBDEs level in human milk and fish & shellfish samples collected from Korea (한국인 모유 및 어패류 중 PBDEs 잔류 레벨 모니터링)

  • Flame retardants are added to prevent catching fire and to slow down the burning process. PBDEs are known to affect thyroid hormones and hormone disruption. The aim of this study was to propose a manual for determination of PBDEs, and investigate the accumulation of PBDEs(BDE-28, 47, 99, 100, 153, 154 and 183) in fish&shellfish and human milk samples. Pre-treatment for PBDEs determination, alkali digestion and L-L(Liquid-Liquid) extraction method could be applied to fish and shellfish. When Multi-layer column was used for cleaning up the sample, 50 mL of hexane and 100 mL of hexane:dichloromethane(9:1) solutions were used for pre- and post-elution, respectively. Activated-carbon column was optimized by a 100 mL of hexane:dichloromethane(3:1). The result of fish, highest concentration was detected in flatfish, 890 pg/g(wet weight). The other side, lowest concentration was detected in pollack, 40 pg/g(wet weight). The result of breast milk, PBDEs was detected 2,580 and 3,600 pg/g(lipid weight) from breast milk of Seoul and Juju, respectively. BDE-153 and 183 were not detected in all samples. There was no difference in PBDEs level was not difference between first and second delivery. In this study, we could find that PBDEs level in Korea is lower than other countries.

Environmental and Antimicrobial Characteristics of Vibrio spp. Isolated from Fish, Shellfish, Seawater and Brackish water samples in Gyeongbuk Eastern Coast (경북 동해안 환경에서 분리된 V. parahaemolyticus 및 V. vulnificus의 생태학적 및 항생제 감수성 특성)

  • This study was carried out to investigated the distribution and characteristics of Vibrio spp. isolated from fish and shellfish, seawater and brackish water samples collected from Pohang, Uljin, Yeongduk and Gyeongju in Gyeongbuk Province from April 2000 to October 2000. Total 155 strains of genus Vibrio were isolated from 439 collected samples, and numbers of isolated strains of V. parahaemolyticus and V. vulnificus were 140 and 15, respectively. The isolation rate from the samples collected in Pohang was the highest as 41.5% (76 strains), and wat the highest as 71.4% (30 strains) in brackish water, and was the highest as 55.7% (34 strains) in August. And the optimal pH, temperature, and NaCl concentration for growth of V. parahaemolyticus and V. cholerae were 8.0, 3$0^{\circ}C$ and 2.0%, respectively. In a resistance test for environmental factors, heat and cold resistants of V. parahaemolyticus were higher than those of V. vulnificus, withstanding for 15 minutes at 6$0^{\circ}C$ and 6 days at -18$^{\circ}C$. The pH range for existence of V. parahaemolyticus and V. vulnificus were 4.5~l1.0 and 4.5~10.0, showing the similar resistance to pH. V. parahaemolyticus and V. vulnificus could grow in media containing up to 10.0% and 7.0% NaCl, respectively, Salt-tolerance of V. parahaemolyticus was higher than that of V. vulnificus. In an antibiotics sensitivity test against 16 strains of V. parahaemolyticus, twelve strains were resistant to ampicillin, eight strains were resistant to cephalothin. one strain was resistant to streptomycin, and one strain was resistant to ticarcillin.

A New Analytical Method for Fluoroquinolones in Fisheries Products by High Performance Liquid Chromatography (HPLC를 이용한 어패류 중의 fluoroquinolone계 항균제의 분석법)

  • Fluoroquinolones are the most common group of antibacterial agents currently used in the Korean aquaculture industry, and use of these agents has been increasing steadily. High performance liquid chromatography (HPLC) with fluorescence detection was used for the simultaneous determination of five fluoroquinolones in fish and shellfish: ofloxacin (OFL), pefloxacin (PEF), norfloxacin (NOR), ciprofloxacin (CIP), and enrofloxacin (ENRO). Fish and shellfish muscle was homogenized, and protein, lipid, and low molecular weight pigments were then excluded from the homogenate. The final eluates were analyzed by HPLC equipped with a Shiseido UG-120 type C18 reverse-phase column ($4.6{\times}250 mm$, $5{\mu}m$) and a fluorescence detector (excitation at 280 nm, emission at 450 nm). The mobile phase was 0.1 M phosphoric acid and acetonitrile solution (91:9, v/v) and tetrahydrofuran (THF) was added to it at a rate of 5 mL per a liter of the mobile phase. Adequate chromatography separation was obtained using the above method. Average recoveries of fortified samples at levels from 0.05 to 0.5 mg/kg were $72.3{\pm}2.5-84.5{\pm}1.2%$ for OFL, $82.7{\pm}3.3- 109.3{\pm}7.5%$ for NOR, $85.3{\pm}6.6-116.0{\pm}7.9%$ for PEF, $76.0{\pm}4.3-109.3{\pm}12.4%$ for CIP, and $78.7{\pm}5.9-100.0{\pm}9.8%$ for ENRO. The limit of detection of OFL was $5{\mu}g/L$, the others were $1{\mu}g/L$. We concluded that the new analytical method was suitable for the determination of fluoroquinolones in fish and shellfish.
