Determination of Water-holding Capacity of Porcine Musculature Based on Released Water Method Using Optimal Load

  • The purpose of this study was to evaluate the suitability of the released water (RW) method for measuring water-holding capacity (WHC) by pressure and filter-paper absorption within two thin plastic films. One hundred fifty pork loins with considerable variations in WHC (three pork qualities: PSE, pale, soft, and exudative; RFN, reddish-pink, firm, and nonexudative; and DFD, dark, firm, and dry; 50 pork loins in each pork quality group) were used to measure drip loss (DL)% and RW % after applying different loads (1.5, 2.0, 2.5, 3.0 and 3.5 kg). Percentages of DL and RW were significantly (p<0.05) different among pork quality groups. The RW % of 2.5 kg load sample was significantly (p<0.05) higher than that of 2.0 kg or 1.5 kg load sample. However, there were no significant (p>0.05) differences in RW % among 2.5 kg, 3.0 kg, and 3.5 kg load samples for all three pork quality groups. The RW % was highly and positively correlated (r>0.95; p<0.001) with DL %. The highest correlation was observed between DL % and RW % of 2.5 kg load sample (r=0.97; p<0.001). Among all RW loads within each quality groups, the highest correlation between DL % and RW % was observed for 2.5 kg load sample (r=0.43, 0.66, and 0.46 for PSE, RFN and DFD quality groups, respectively, p<0.001). These results imply that applying 2.5 kg load is the best for determination of RW %. It is recommended as a reference method to assess DL % of pork muscles.

  • The micro-assay methods of vanillin have been shown quite few, though several methods have been introduced for determination. A new sensitive micro-determination of vanillin, applying the yellow color reaction of vanillin and anillin, is shown in this paper. The absorption maximum of the yellow coloring matter, 4-Oxy-3-methoxy-henzal aniline, was 435 mu. The one dimensional ascending paper chromatographic method is applyed for isolation of vanillin from the mixed sample. The microdetection of vanillin is studied only in this paper. Vanillin was detected by the yellow spot on paper trip by the coloring reagent of aniline after several hours paper strip chromatographic at the following condition; paper strip ................ 2.5 X 35cm Whatman Filter Paper No.2 developing Solvent ......... petroleum-benzene-methanol n-butanol-water coloring agent ............. aniline. The Rf-value on petroleum benzene and methanol was 0.63 and that on n-butanol and water was 0.90. The minimum detectable amount of vanillin by this method was 10 micrograms. It is recommendable, if interference substances as aromatic aldehydes present, that the application of this aniline reaction and Foline Denis reaction on the same paper chromatogram is appreciable.

Behavior of Some Metallic Ions in the Process of Ginseng Extracts Preparation (인삼(人蔘) Extract 제조과정(製造過程)중 무기금속(無機金屬)이온의 동태(動態))

  • Some metallic ions such as copper, manganese, iron, zinc and cadmium in ginseng extract, originated from white ginseng, were determined by atomic absorption spectrophotometry. The extracts were prepared with water, 35% ethanol, 50% ethanol or 75% ethanol solution followed by filtration or centrifugation. Greater amounts of metallic ions were found in the solution filtrated by filter paper (Toyo No. 5A) or by filter cloth than in that of centrifugation. The residual amount of metallic ions in the extract decreased with increasing concentration of ethanol. The percentages of each metallic ion in ginseng extracts on the basis of original amount of metallic ions were as follows: Cu, 10.4-31.9%, Fe,5.1-19.0%, Mn,3.0-12.2% Zn,7.4-12.0%, Cd,5.9-11.8%.

  • The yield of cellulase derived from Trichoderma $(O_2-1)$ was remarkably varied with various concentration of ethanol and acetone in purification of the enzyme. In the purification with ethanol of ${\beta}-glucosidase$, the best result was obtained in the concentration of 60% and, of CMCase and of filter paper disintegrating enzyme 80%. And in the purification with acetone of ${\beta}-glucosidase$, filter paper disintegrating enzyme, and CMCase, in the concentration of 60%, 80%, and 90% respectively, was shown the best yield. The activities of crude Cellulase preparation could be seperated into few of fractions by column chromatography with Silica gel, Cellulose powder, and gauze. Most of CMCase, avicelase, and ${\beta}-glucosidase$ were eluted, but most of filter paper disintegrating enzyme and the rest of enzymes mentioned the above were absorbed, and were eluted with water. Therefore, it was considered that CMCase is different from filter paper disintegrating enzyme in properties. The relative activity of CMCase was different from that of avicelase in the peak of elusion part. And it was considered that filter paper disintegrating enzyme and cellulose powder saccharifying enzyme was seperated respectively as absorption part and non absorption part. The auther came to the conclusion that at least there were more than three sorts of cellulase in Trichoderma $(O_2-1)$ cellulase preparation.

  • The aim of this study is to scientifical conservation for the Sin An Ship. The test object is Pinus massoniana and Cunninghamia lanceolata of Sin An Ship's timber .Comtrast object is water soaked fresh wood of Pinus densiflora and Cryptomeria japonica The results were as follows.1. M. C is 280.9% in high degraded Planking board,237.5% in low degraded Planking board,158.6% in fresh wood of Pinus densiflora,222.6% in Bulkhead167.3% in fresh wood of Cryptomeria japonica2. Absorption rate of P.E.G is good at heating condition ($55∼60^{\circ}C$), especially in highdegraded wood.3. The dimensional change of wood during air drying is somewhat shrinked That's 7.22% in low degraded Planking board9.8% in high degraded Planking board and 4.11% in water soaked Pinus densiflora.4. The dimensional change of wood during P.E.G treatment is mostly swollen of wery small quantity.5. Salinity in desalinization tank showed below 50 p.p.m. It's indicate the end of desalinization treatment.6. Filtration with filter paper was possible up to 40% concentration only. A Study on the variations of mechanical and electrical property of epoxy composites due to boiling absorption (비등흡수에 의한 에폭시 복합재료의 기계적 특성 및 전기적 특성 변화에 관한 연구)

  • In this paper, the variable mechanical strength and dielectric breakdown strength of epoxy composites were measured at boiling absorption condition in order to observe the influences of moisture in out door use. Also, in order to improve water resistance of matrix resin, IPN(interpenetrating polymer network) method which had been already reported, was introduced and the influence was investigated. As Adding filter(SiO2) classified by o[phr], 50[phr] and 100[phr] to two kinds of matrix resin, six kinds of specimens were manufactured. As a result, it was confirmed that the moisture absorption rate was increased and mechanical strength and dielectric breakdown strength were degraded with boiling time and filler content increasing. On the other hand, it was confirmed that moisture absorption rates were decreased and the degrading rates of mechanical strength and dielectric breakdown strength were lowered according to improvement of adhesion strength in case of IPN specimens.

A Study on Water Holding Capacity of Fish Meat Paste Products (어육(魚肉) 연제품(煉製品)의 보수력(保水力)에 관(關)한 연구(硏究))

  • It is well known that water holding capacity plays an important role in processing such meat products as frankfurter-type sausage and fish meat paste products as kamaboko and fish sausage. Consumer qualities of meat products, such as appearance, flavor, as well as drip and shrinkage on cooking, depend greatly on the degree of water binding. In this paper, the water holding capacities of fish paste and salt added paste of white corvenia, Argyrosomus argentatus and file fish, Novodon modestus were measured by centrifuging and press method before and after cooking. And the effects of the addition of phosphates and starch to enhance water binding and stabilize gel formation were also discussed. In addition, the experimental conditions which are suitable to determine the water binding of fish meat paste product were suggested. The results were expressed in percent of water absorbed by the filter paper when pressed or released by pressor or centrifuge to the weight of sample. From the results. a proper condition to measure the water holding capacity of fish meat paste was that 3.0 g of sample which was previously added with 10 percent water was centrifuged at 13,400 G or 12,000 rpm for 15 minutes for the centriguging method and for press method, 0.3 g sample with 10 percent of water added was extracted by an oil pressor at $30\;kg/cm^2$ for 1 minute. Water holding capacity of fresh paste of white corvenia was relatively higher than that of file fish and the difference between species of fish was greater than the difference between measurments by two methods. Sodium chloride had a great effect on enhancing the water holding capacity of fish meat paste giving better effect when 3.0 percent of salt was added. Phosphates used except calcium phosphate revealed a certain enhancement in water binding, yielding best effect at 0.3 percent addition, and metaphosphate seemed to be more effective in order. The addition of corn starch, however, appeared to be not so effective for enhancement of water binding in fresh-salt-added fish meat paste but in cooked fish paste which might be attributed to absorption of water by starch grain and swelling during the heating and consequently enforced gel strength of cooked fish paste. And the water holding capacity of cooked fish paste was proportionally related to its gel strength.

Some Quality Changes in Soybean Curd by Addition of Dried Soymilk Residue (건조비지 첨가에 의한 두부품질의 변화)

  • The dried soymilk residue (SMR) was added into soybean water extract in order to investigate its effect on physical characteristics of soybean curd. As the mixing ratio (d. b.) increased, the volume and water holding capacity of the curd was reduced and penetration value was increased. However, the Sag value was rather increased until 20% of mixing ratio reached and then decreased drastically by 30% addition of SMR. The overall physical and sensory quality showed that substitution of soybean with SMR by 10% was recommendable without significant unfavorable change in quality of soybean curd. The proposed method of absorption of unbound water on filter paper was proved to be simple and reliable for measurement of water holding capacity of soybean curd.

Comparison of soybean varieties for physical properties of Tofu (콩 품종별 두부의 물리적 특성의 비교)

  • The physical characteristics of texture, water holding capacity, color and swelling of soybean curd were studied for 14 varieties of soybeans grown in Korea. The relative water holding capacity, measured by moisture absorption method with using filter paper, was high for Millyang-21. Danyeob and Hwangum, low for Suwon-138, -141 and -142. All of the soybean curd prepared showed negative Sag values, which indicates swelling properties of Tofu as it as released from pressed condition. The most swollen variety was Suwon-138 and the least one was Suwon-142. Generally, soybean curd having high moisture content had low valuss in hardness but no significant relationship was found. Elasticity ud cohesiveness showed a little differences except Suwon-138. Other textural characteristics of fracturability, adhesiveness and gumminess showed a wide range in their values among the varieties.

Development and Evaluation of a Badge-type Passive Sampler for the Measurement of Short-term Nitrogen Dioxide in Ambient Air (대기 중 이산화질소의 단기 측정을 위한 뱃지형 passive sampler의 개발 및 평가)

  • The purpose of this study is to develop a badge-type passive sampler for the measurement of short-term nitrogen dioxide and to evaluate its performance. The principle of the method is a colorimetric reaction of nitrogen dioxide with sulfanilic acid, N-1-naphthylethylendiamine, and phosphoric acid. First, it has been shown that the filter paper should be rinsed with ultrapure water and ultrasound, and then dried in a vacuumed desiccator. The concentration and volume of absorption reagent (triethanolamine) were $20\%$ and 100 ${\mu}L$, respectively. The extraction time was determined as 60 min. Second, duplicate measurements (n= 116) were carried out for evaluating the precision of the passive sampler. The relative error and the correlation coefficient between duplicates are $3.4\pm 3.0\%$ and 0.994, respectively. In addition, the $95\%$ confidence interval of intraclass correlation coefficient and the estimated value are 0.992$\sim$0.996 and 0.994, respectively. Third, a paired t-test was carried out for evaluating the accuracy of the passive sampler (n=40). In the result of the test, the $95\%$ confidence interval of the difference was -1.710 ppb <$\gamma$< 0.788 ppb. Finally, the average concentration of blanks, measurement detection limit, limit of detection, and limit of quantification are $2.4\pm 0.4$ ppb, 104 ppb, 3.8 ppb, and 7.0 ppb, respectively.
