Seasonal Variation in Proximate Composition, Cholesterol and $\alpha$-Tocopherol Content of 12 Species of Korean Fish (12종 어류의 일반성분조성과 콜레스테롤 및 $\alpha$-토코페롤 함량의 계절변화)

  • Seasonal variation in the proximate composition, cholesterol (CHOL) and $\alpha$-tocopherol ($\alpha$-Toc) content of 12 spices of fish muscle, which caught off Tongyeong coast of the Southern Sea (Nam-Hae) from Mar. 1995 to Feb. 1996, was studied. Lipid and moisture content showed remarkable seasonal variation and there were a negative correlation between the both (r= -0,85, p<0.001), while protein and ash content unchanged almost through the sampling period. The lipid content of anchovy, hickoryshad, black sea bream, finespotted flounder sea eel, bastard, and file fish was high in Dec. Oct. Nov. Jan. Jan. Nov, respectively, and low in the season corresponding to their spawning period. However, in case of striped mullet, yellow tail, mackerel, rock fish and red sea bream, there was no correlation between their lipid content and spawning period, and thus these fishes fore considered to be affected more by water temperature and the content of their diet. CHOL content increased In :he season containing a high level of lipid, whereas $\alpha$-Toc content unchanged almost through the sampling period.

Non-Enzymatic Browning Reaction of the Dried File-Fish (건조(乾燥) 말쥐치의 비효소적(非酵素的) 갈변(褐變))

  • A kinetical approach for the non-enzymatic browning reaction of the dried file-fish was studied. The reaction rates revealed a tend to increase with increasing water activity and showed the maximum at $0.75\;a_w$ The activation energies obtained from the Arrhenius plot ranged 12.5 to 16.5 Kcal/mole. From these energies of activation, the $Q_{10}$ values at $45^{\circ}C$ showed 1.9 to 2.3 and both activation energy and $Q_{10}$ values were reduced with increase in $a_w$ Shelf-lives, the time to reach an 0.15 O.D./g solid at which severe brown color change could be de ectable, decreased rapidly as the temperature and water activity increase. A storage study under a square-wave fluctuating temperature condition (at 35 and $55^{\circ}C$ periodically with 7 days interval), the rate constants at all water activities used in the experiments were higher than those at $45^{\circ}C$, the mean temperature of the cycle which affects other kinetic parameters including activation energies, $Q_{10}$ values and finally the shelf-lives. The data obtained from the fluctuating temperaure storage study will be used in prediction of shelf-life. The shelf lives assessed at $25^{\circ}C$ from the accelerated shelf-life tests ranged from 179 daysat $0.75\;a_w$ to 302 days at $0.44\;a_w$.

Annual Reproductive cycle of the File Fish, Thamnaconus modestus, on the Southern Coast of Cheju Island (제주 남부 연안 말쥐치, Thamnaconus modestus의 생식년주기)

  • Annual reproductive cycle of the file fish, Thamnaconus modestus (Gunther), was histologically investigated. Samples were collected monthly in the coastal waters of Chungmun, south of Cheju Island, Korea from July 1997 to June 1999. In males and females of T. modestus GSI values reached the maximum in June and May, respectively. Reproductive cycle could be divided into the following successive stages: in females, growing stage (March to April), mature stage (April to May), spawning stage (May to June), degenerative and resting stage (July to February), and in males, growing stage (January to March), mature stage (April to May), spent stage (May to June), degenerative and resting stage (July to December), respectively. To clarify the spawning cycle of female in T. modestus, some were examined, that is, the weekly changes of GSI, detail developmental stages in the ovary and the weekly changes of sex steroid hormones ($E_2$ and T) levels in plasma during the spawning period. Throughout histological observation of the ovary during the spawning period, T. modestus belonged to an asynchronous and multiple spawner. Changes of plasma $E_2$ and T levels were similar to the changes of GSI and ovary maturity.

Egg Development and Morphological changes of Larvae of the File Fish, Thamnaconus modestus (말쥐치, Thamnaconus modestus의 난발생과정과 부화자어의 형태발달)

  • The egg development and morphological changes of larvae of the file fish, Thamnaconus modestus(Gunther), was investigated by artificial insemination. The fertilized egg was spherical in shape, colorless and adhesive with 0.63~0.67mm in diameter and yolk had 20~30 oil globules of various size (0.04~0.16 mm in diameter). The hatching occured at 53 hours after fertilization in $21.0\sim23.0^{\circ}C$ of water temperature. Hatched larvae measured 2.07~2.20 mm in total length with 5~6+17~18=22~24 myotomes. The 3~4 days after hatching larvae grew to the total length 2.62~2.77 mm, opened mouth and anus. The 5 days after hatching, total length of the larvae was 2.74~3.04 mm, the first dorsal fin was formed at the behind of head. After hatching 12~13 days larvae attained 3.78~3.99 mm in total length. This period formed rudimental dorsal fin and anal fin. Twenty days after hatching, larvae grew to 6.04~6.17mm in total length, at which time the dorsal, anal and caudal fin rays reached 19~20, 18~19 and 9~10, respectively.

Potential Human Risk Assessment of PCBs and OCPs in Edible Fish Collected from the Offshore of Busan (부산 연근해의 해양오염퇴적물과 식용 어류 체내의 PCBs와 유기염소계 농약의 분포 및 잠재적인 인체 위해성평가)

  • Polychlorinated biphenyls (PCBs) and organochlorine pesticides (OCPs) contents in marine sediment and edible fish (4 species) from the estuary near the Yongho wharf in Busan were determined to assess their presence and their potential health risk to the local population through fish consumption was also assessed. Levels of ${\Sigma}PCBs$ and ${\Sigma}DDTs$ in marine sediment were 3.22~197.65 and 1.77~20.27 ng/g dw, respectively. HCHs and endosulfan sulfate concentrations in bottom sediment were 1.42~6.08, 0.56~13.89 ng/g dw, respectively. The bottom sediment in the offshore of the Yongho wharf may be characterized as semi-polluted status with potential adverse marine biological effects in terms of sediment quality guidelines of US NOAA. The mean concentrations of ${\Sigma}PCBs$ in the tissues of olive flounder (Paralichthys olivaceus), Korean rockfish (Sebastes schlegelii), file fish (Stephanolepis cirrhifer) and abbysal searobin (Lepidotrigla abyssalis) were 67.37, 10.20, 48.26, 22.81 ng/g lw, respectively. DDTs and HCHs were also detected in all fish samples. Lifetime cancer risk and target hazard quotient to local residents due to those fish consumption were found to pose negligible cancer and non-cancer risk.

Study on the Midwater Trawl Available in the Korean Waters ( V ) - Opening Efficiency of the Otter Board with a Large Float on the Top - (한국 근해에 있어서의 중층 트로올의 연구 ( V ) - 전개판에 대형 뜸을 달았을 때의 전개성능 -)

  • Near sea trawlers of Korea sometimes catch pelagic fishes like file fish by using midwater trawl gear even though usually catch bottom fish. It is reasonable to use the specific otter board as well as specific net in bottom trawling and in midwater trawling respectively. But, the trawlers are so small ranging 100 to 120GT, 700 to 100ps that it is very complicated to use different otter board for bottom trawling and for midwater trawling. The otter board for bottom trawling. is also used for the midwater trawling without any change even though the net is changed into the specific one. Although the otter board in the midwater trawling should be lighter than that for bottom trawling, to use otter board for bottom trawling directly for the midwater trawling without any change makes the net easily touch the sea bed and also make the horizontal opening of the otter boards be limited owing to the length of warp in the southern sea of Korea, main fishing ground of midwater trawling, which is 100m or so in depth. That is why the otter board for the midwater trawling should be made lighter than that in the bottom trawling, even if temporary. The authors carried out an experiment to achieve this purpose by attaching a large styropol float on the top of the otter board. In this experiment, underwater weight of the otter board was 630kg and buoyancy of the float was 510kg. To determine the depth and horizontal opening of the otter board, two fish finder was used. A transmitter of 50KHz fish finder was set downward through the shoe plate of otter board to determine the elevation of otter board from the sea bed, and a transmitter of 200KHz fish finder was set sideways on the starboard otter board to be able to detect the distance between otter boards. The obtained results can be summarized as follows: 1. The actual towing speed in the experiment varied 1.1 to 1.8 m/sec. 2. The depth of otter board was within 41 to 25m with float on the top and 45 to 26m without float in case of the warp length 100m, whereas the depth 68-44m with float and 74-46m without float in case of the warp length 150m. This fact means that the depth with float was 9-4% shallower than that without float. 3. The horizontal opening between otter boards was within 34-41m with float and 30-38m without float in case of the warp length 100m, whereas the opening was 44-50m with float and 37-46m without float in case of the warp length 150m. This fact means the opening with float was 10% greater than that without float in case of the warp length 100m, and 15% greater in case of the warp length 150m. 4. The horizontal opening between wing tips by using the otter board with float was 1m greater than by without float in case of the warp length 100m, whereas the opening by with float was 2m greater than by without float in case of warp length 150m. From this fact, it can be estimated that the effective opening area of the net mouth by using the otter board with float could be made 10% greater than by without float in case of warp length 100m, whereas the area with float 20% greater than by without float in case of warp length 150m.

  • The ratio of the eatable flesh taken from the filefish and its chemical components were investigated from March 1980 to February 1981 and the results are as follows :1. The ratio of the eatable flesh taken from the fish was low during March to June and high during July to December. 2. The moisture content was $81.4-82.0\%$ during March to June, $77.5-80.1\%$ during July to December and $79.4-81.4\%$ during January to February. 3. The protein content was $15.4-15.7\%$ during March to June, $17.0-19.9\%$ during July to December and $16.1-17.0\%$ during January to February. 4. The Lipid content was $0.31-0.41\%$ during March to January, $0.46-0.84\%$ during July to January and $0.43\%$ during February. 5. The carbohydrate content was $0.3-0.4\%$ during March to July, $0.5-0.7\%$ during August to January and $0.3\%$ during February. In conclusion, the ratios of the eatable flesh taken from the filefish and its protein content were highest during July to December in one year.

Changes in the Microflora of Marine Fishes during Storage by Partial Freezing (해산어(海産魚)의 부분동결(部分凍結)에 의한 Microflora의 변화(變化))

  • Marine fishes, sardine(Sardinops melanosticta), scad kingfish(Caranx equula), horse mackerel(Trachurus japonicus) and file fish(Navodon modestus), were stored for fifty days with partial freezing at$-3.5^{\circ}C$. During the storage, the changes in microflora and volatile nitrogen content was investigated. The fishes exhibited $10^4\;to\;10^6$ of bacterial cells per square centimeter of their skin just before they were submitted to the storage. The bacterial cell number was increased as $10^6\;to\;10^8$ cells as the storage time passed over twenty-two days. Offensive odor which is typical in the spoilage of fishes became strong as increase the bacterial cell numbe. The major isolates among the three hundred strains of bacteria isolated from the fish skins were identified as Pseudomonas I/II, III/IV-H, Vibrio and Moraxella. The same was found in the spoiled fishes, however, Pseudomonas I/II, was predominant on contrast to that of fresh fishes. Pseudomonas III/IV-NH, Flavobacterium, Cytophaga and Micrococcus were also found in early period of storage, but they disappeared as the progress of storage. Nine per cent of isolates were unidentified.

Thermal-Denaturation of File Fish Myofibrillar Protein and Protective Effect of Sucrose, Sorbitol and Amino Acids (말쥐치 근원섬유단백질의 열안정성과 및 가지 첨가제의 영향)

  • Thermal-denaturation of myofibrillar protein of dorsal skeletal muscle from file fish was investigated by measuring denaturation constant($K_D$) and thermodynamic parameters at various temperatures. The protective effects of sucrose, sorbitol and amino acids when added individually or combined were also discussed. The denaturation rate as reflected in inactivation of myofibrillar protein Ca-ATPase was followed the first order reaction. The $K_D$ values at $25^{\circ}C,\;30^{\circ}C,\;and\;35^{\circ}C$ were $19.52{\times}10^{-5},\;112.25{\times}10^{-5},\;and\;247.20{\times}10^{-5}$, respectively. The activation energy of the reaction at $30^{\circ}C$ was 43 kcal/mole. The protective effects of sucrose, sorbitol, glycine, alanine and Na-glutamate were increased with the concentration but the effects of sorbitol and Na-glutamate decreased beyond 1.0 mole. Basic amino acids such as arginine and lysine did not revealed any protective effect on the thermal denaturation. In case of mixed addition, the effects of Na-glutamate to glycine, sorbitol to glycine, and sorbitol to sucrose or sorbitol to Na-glutamate were enhanced 1.2 to 7.0 times as much as that of control (ratio of mixing; 1:1, range of concentration; 0.5 to 1.25 mole). Under the frozen condition at $-20^{\circ}C$, two mixtures such as Na-glutamate to glycine and sorbitol to sucrose apparently revealed the protective effects.

Development of Species-Specific PCR to Determine the Animal Raw Material (종 특이 프라이머를 이용한 동물성 식품원료의 진위 판별법 개발)

  • In this study, the detection method was developed using molecular biological technique to distinguish authenticity of animal raw materials. The genes for distinction of species about animals targeted at Cytochrome c oxidase subunit I (COI), Cytochrome b (Cytb), and 16S ribosomal RNA (16S rRNA) genes in mitochondrial DNA. The species-specific primers were designed by that Polymerase Chain Reaction (PCR) product size was around 200 bp for applying to processed products. The target 24 raw materials were 2 species of domestic animals, 6 species of poultry, 2 species of freshwater fishes, 13 species of marine fishes and 1 species of crustaceans. The results of PCR for Rabbit, Fox, Pheasant, Domestic Pigeon, Rufous Turtle Dove, Quail, Tree Sparrow, Barn Swallow, Catfish, Mandarin Fish, Flying Fish, Mallotus villosus, Pacific Herring, Sand Lance, Japanese Anchovy, Small Yellow Croaker, Halibut, Jacopever, Skate Ray, Ray, File Fish, Sea Bass, Sea Urchin, and Lobster raw materials were confirmed 113 bp ~ 218 bp, respectively. Also, non-specific PCR products were not detected in compare species by species-specific primers. The method using primers developed in this study may be applied to distinguish an authenticity of food materials included animal raw materials for various processed products.
