Temporal Changes in N Assimilation and Metabolite Composition of Nitrate-Affected Tomato Plants

  • The role of inorganic nitrogen assimilation in the production of amino acids, organic acids and soluble sugars is one of the most important biochemical processes in plants, and, in order to achieve normally, nitrate uptake and assimilation is essential. For this reason, the characterization of nitrate assimilation and metabolite composition from leaves, roots and xylem sap of tomato (Solanum lycopersicum) was investigated under different nitrate levels in media. Tomato plants were grown hydroponically in liquid culture under five different nitrate regimes: deficient (0.25 and 0.75 mM $NO_3{^-}$), normal (2.5 mM $NO_3{^-}$) and excessive (5.0 and 10.0 mM $NO_3{^-}$). All samples, leaves, roots and xylem sap, were collected after 7 and 14 days after treatment. The levels of amino acids, soluble sugars and organic acids were significantly decreased by N-deficiency whereas, interestingly, they remained higher in xylem sap as compared with N-normal and -surplus. The N-excessive condition did not exert any significant changes in metabolites composition, and thus their levels were similar with N-normal. The gene expression and enzyme activity of nitrate reductase (NR), nitrite reductase (NIR) and glutamine synthetase (GS) were greatly influenced by nitrate. The data presented here suggest that metabolites, as a signal messenger, existed in xylem sap seem to play a crucial role to acquire nitrate, and, in addition, an increase in ${\alpha}$-ketoglutarate pathway-derived amino acids under N-deficiency may help to better understand plant C/N metabolism.

  • To find out the effects of phosphatic fertilizer application levels on the botanical composition, mineral contents and mineral nutrient deprivation of grasses and the chemical properties of soil, this expriment was arranged as a randomized complete block design with six treatments(0, 50, 100, 150, 200 and 250kg $P_2O_5$/ha) and conducted at hilly land in Gwangju, Gyeonggi Province from September, 1989 to October, 1992. The results obtained are summarized as follows: The botanical composition of grasses without posphatic fertilizer application was poor but with 150kg $P_2O_5$/ha application, remarkably increased in legume of grasses and decreaed in weed. As the phophatic fertilizer levels increase, P, Ca and Mg content of grasses were increased but N and K content tended not to be regular, and Ca/p and K/Ca+ Mg equivalent ratio were lowered. Up to 150kg $P_2O_5$/ha application level, the mineral nutrient content deprived by dry matter yield was increased but with that over application, tend not to be increased. Most of applied mineral elements were accumulated at a subsurface of grassland and as the phosphatic fertilizer application levels increase, the available $P_2O_5$ and exchangeable Mg contents of the soil tended to be remarkably increased and exchangeable K content tended to be decreased, but pH, OM and exchangeable Ca content tended not to be regular.

  • 3요소성분비율(要素成分比率)이 상이(相異)한 복합비료(複合肥料)의 비효를 고추품종(品種) 중원청룡을 사용(使用)하여 포장(圃場)에서 단비와 대비(對比)해서 검토(檢討)한 결과(結果)는 다음과 같다. 가. 고추적과(赤果)의 수량(收量)은 단비구보다 공시(供試)한 복합비료(複合肥料)를 시용(施用)한 경우에 증수(增收)되었는데 통계적(統計的)인 유의차(有意差)는 없었다. 나. 식물분 분석결과(分析結果)는 처리별(處理別)로 아무런 경향(傾向)이 없었으며 수량(收量)과의 상관(相關)도 인정(認定)되지 않았다. 다. 시험후(試驗後) 토양(土壤)의 화학적성질(化學的性質)은 복합비료구(複合肥料區)가 단비구 보다 유효인산함량을 높였고 기타성분(其他成分)은 비종간(肥種間)에 차이(差異)가 없었다.

Botanical Composition, Herbage Production and Plant Mineral Contents as Affected by Application of Chemical Fertilizer and Fermented Sawdust Pig Manure on Cheju Brown Volcanic Ash Pasture Soil

  • This experiment was carried out during the period from September, 1997 to October, 1998 to determine the effect of fermented saw-dust pig manure (FSP) application on the herbage production on a mixed pasture in the Cheju brown volcanic ash soil. Split plot design (main plot: 3 nitrogen application levels of 0, 150 and 300 kgiha; sub plot: 4 pig sawdust manure levels of 0, 3, 6 and 12 tonha) was used. Plant height and dry matter yield increased significantly with an increase of nitrogen and FSP level. There was no difference in the botanical composition of grasses as affected by FSP application level, but herbage yields of grass species were increased by nitrogen application compared to that without nitrogen application. Botanical composition of white clover decreased with an increase of nitrogen application, but increased with an increase of FSP application level. Percentages of weeds were not affected by application rates of chemical nitrogen fertilizer and swine manure in the mixed species pasture. Nitrogen, phosphorus, and potassium contents of species in the pasture significantly increased with increasing application rates of nitrogen fertilizer. In conclusion, it would be an optimum to apply 150 kgha of inorganic chemical fertilizer and plus either 3 or 6 toniha of fermented swine manure with sawdust for optimum production of mixed pasture on Cheju Island. Effects of Short-Term Tillage on Rhizosphere Soil Nitrogen Mineralization and Microbial Community Composition in Double-Cropping Rice Field

  • Soil extracellular enzyme plays a vital role in changing soil nitrogen (N) mineralization of rice field. However, the effects of soil extracellular enzyme activities (EEA) and microbial community composition response to N mineralization of rice field under short-term tillage treatment needed to be further explored. In this study, we investigated the impact of short-term (8-year) tillage practices on rhizosphere soil N transformation rate, soil enzyme activities, soil microbial community structure, and the N mineralization function gene abundances in double-cropping rice field in southern China. The experiment consisted of four tillage treatments: rotary tillage with crop straw input (RT), conventional tillage with crop straw input (CT), no-tillage with crop straw retention (NT), and rotary tillage with all crop straw removed as a control (RTO). The results indicated that the rhizosphere soil N transformation rate in paddy field under the NT and RTO treatments was significantly decreased compared to RT and CT treatments. In comparison to the NT and RTO treatments, soil protease, urease, β-glucosaminidase, and arginase activities were significantly improved by the CT treatment, as were abundances of soil sub, npr, and chiA with CT and RT treatments. Moreover, the overall diversity of soil bacterial communities in NT and RTO treatments was significantly lower than that in RT and CT treatments. Soil chitinolytic and bacterial ureolytic communities were also obviously changed under a combination of tillage and crop straw input practices.

  • Recently the method of basal compound fertilizer (N : P : K = 18.2 : 9.8 : 35.0) Placement has been changed from banding to broadcasting in tobacco cultivation. The effects of the compound fertilizer placement, level of fertilizer and additional urea application on the agronomic characteristics, two chemical compositions and physical Properties of burley tobacco(Nicotiana tabacum L. cv. KB 108) were investigated at Chonju Experiment Station, Korea Ginseng and Tobacco Research Institute in 1995-1996. Six treatments consisted of (1) band (method of fertilizer Placement) + In kg/10a(compound fertilizer) + 0(additional urea applied), (2) broadcast+140+0, (3) broadcast+140+25, (4) band+180+0, (5) broadcast+180+0, and (6) broadcast +180+25. The additional urea was applied at hilling. No significant differences were detected between banding and broadcasting method of compound fertilizer Placement in field 9rowth, wield, organoleptic qualify(price Per kilogram), chemical composition and Physical properties of cured leaf, The increased compound fertilizer by 30%(40k/10a) or the additional urea application by 25kg per 10a produced slightly higher yield than the recommended amount of basal compound fertilizer without additional urea application did. It also increased the total alkaloid content of cured leaf. It is recommended that no more basal fertilizer above the recommended amount and no additional urea application are needed in burley tobacco fertilization, even though the method of basal fertilizer placement being chanced from banding to broadcasting.   • 본(本) 시험(試驗)은 고랭지(高冷地) Timothy 우점초지에서 목초(牧草)의 적정(適正) 예취회수(刈取回數)와 최종(最終) 예취시기(刈取時期)를 구명(究明)하고자 DM 생산량(生産量), 월동후(越冬後) 초기생육특성(初期生育特性), 다음해 1차수량(次收量) 조단백질(粗蛋白質) 및 조섬유(粗纖維) 생산량(生産量)을 조사(調査)하고 그들간(間)의 상관관계(相關關係)를 분석(分析)한 결과(結果)를 요약(要約)하면 다음과 같다. 1. 초기생육(初期生育)이 개시(開始)되는 시기(時期)는 예취회수(刈取回數)가 증가(增加)함에 따라 빨라지는 경향(傾向)이었으나 유의차(有意差)가 없었으며 최종예취일(最終刈取日)에 따른 영향은 일정하지 않았다. 2. 예취회수(刈取回數)가 증가(增加)함에 따라 건물생산량(乾物生産量)은 감소하였으나 최종예취일(最終刈取日)에 따른 차이(差異)는 없었다. 3. 월동후 1 차수량(次收量)은 예취회수(刈取回數)에 따라 현저히 감소하였으나 최종예취일(最終刈取日)에 따른 차이(差異)는 뚜렷하지 않았다. 4. 조단백질(粗蛋白質) 생산량(生産量)은 예취회수(刈取回數)가 증가(增加)할수록 많아졌으나 건물율(乾物率)은 낮아지는 경향이었고 조섬유(粗纖維) 생산량(生産量)은 일정하지 않았다. 5. 건물수량(乾物收量)과 건물율(乾物率)($\gamma=0.78$) 및 다음해 1차수량(次收量)($\gamma=0.82$)간(間)과 건물율(乾物率)과 다음해 1차수량(次收量)($\gamma=0.83$)간(間)에는 정(正)의 상관(相關)이 있었고(p<0.01), 조단백질(粗蛋白質) 생산량(生産量)과 건물율(乾物率)($\gamma=-0.67$) 및 다음해 1차수량(次收量)($\gamma=-0.68$)간(間)에는 부(負)의 상관(相關)이었다(p<0.01). 6. 이상(以上)의 결과(結果)를 종합(綜合)해 볼 때 건물생산(乾物生産)에 있어서는 년간(年間) 예취회수(刈取回數)는 3회(回)호 하고 최종예취일(最終刈取日)은 9월(月) 30일(日)로 하는 것이 적당(適當)하다고 생각되며 조단백질(粗蛋白質) 수량(收量)을 고려한다면 4회(回) 예취(刈取)에 최종예취일(最終刈取日)은 9월(月) 30일(日)로 하는 것이 좋다고 사료(思料)된다.

  • The black soldier fly larvae are able to decompose various organic wastes such as livestock manures and food wastes. We tested whether the quality of the insect derived compost, i.e. larval feces, was comparable to that of a commercial fertilizer. The results show that the chemical composition and the growth rate of cabbages grown on the insect derived compost are virtually identical to those on the commercial fertilizer. Therefore the insect derived compost will be an ideal substitute to commercial fertilizers.

  • To find out the effect of different patterns of nitrogen and potassium distribution on dry matter yield and botanical composition of temperate pastures, a field experiment was conducted with 6 treatments replicated 3 times in a randomized complete block design. It was lasted from September, 1986 to October, 1989 at the Livestock Experiment Station in Suweon. The results obtained are summarized as follows: 1. In early spring, winter hardiness, growth vigour, cold damage and coverage of grasses with 30% dressing of the total amount of N and K20 fertilizer in spring and at the 4th cutting time respectively were better than those of the other treatments. 2. Average dry matter yield for 3 years with heavy dressing in spring and at the 1st cutting time(l1, 187 kg DM/ha) was much more increased by 9% than that of the equally fertilized treatment(l0,24lkg DM/ha). 3. Changes in the botanical composition showed, in general, the same tendency for all treatments except equal N and K20 distribution. However, grassland vegetation with heavy dressing in spring and at the 1st cutting time was changed into relatively good botanical composition with 76.7% grasses, 22.3% legumes and 1.0% herbs at the end of the experiment. 4. By DM yield and botanical composition treatment 3(40-30-15-0-1570) seemed to be an optimal nitrogen and potassium distribution pattern in a temperate pasture.

Preparation and Dissolution Properties of the Eco-glass Fertilizer containing Trace Elements

  • At the previous papers, we showed that K2O-CaO-P2O5 glasses had a solubility in air so that they could be used for glass fertilizer. In this work, we fabricated the Eco-glass fertilizer containing trace elements such as B, Mg, Zn, Fe, Cu, Co, and Mo by a melt-quenching process and the dissolution properties of these glasses were investigated with pH meter and ICP analyzer. In case of the glasses containing the trace elements, effect of trace elements on the dissolutions is not obvious and ignorant the stability of mother glass. Also, the dissolution amounts of each trace elements depend on the mother glass composition and the quantity of each trace element, and mother glasses determined the dissolving velocity of chemical elements.
