Bacillus natto와 B. licheniformis 혼합 Starter로 제초된 청국장의 품질특성 (Quality Characteristics of the Chungkookjang Fermented by the Mixed Culture of Bacillus natto and B. licheniformis)

  • 청국장의 산업적인 대량생산 system에 B. subtilis, B. natto, B. licheniformis 그리고 B. natto와 B. licheniformis의 혼합 균주를 starter로 하여 45시간 동안 청국장을 발효시키면서 각 시험제품의 미생물 생장, 일반성분, 효소활성, 향기성분 등을 분석하고 관능을 평가하였다. 청국장의 수분은 점질물의 형성이 많은 B.subtilis와 B. natto 시험구에서 다소높았으며 pH는 B. licheniformis에서 8.68로 가장 높았고 B. subtilis 에서 8.13으로 가장 낮았다. 미생물의 생장정도는 B. subtilis가 $10^{9}$ CFU/g까지, B. licheniformis는 $10^{8}$ CFU/g까지 생장하였으며 단백질분해효소의 활성은 B. subtilis에서 1.35 unit/g으로 가장 높았고 B. licheniformis에서 0.45 unit/g으로 가장 낮았다. 암모니아태 질소의 함량 은미생물의 생장률 및 단백질분해 효소의 활성에 비례하여 B. subtilis > B. natto> B. licheniformis의 순으로 높은 함량을 나타내었다. 한편, B. natto와 B. licheniformis의 혼합균주를 starter로 하여 제조한 청국장의 일반 품질은 B. natto와 B. licheniformis를 단독 접종한 제품의 평균치에 해당하는 특성을 보였다. 검출된 향기성분의 화학적 구성은 ketones 9종, alcohols 11종, aldehyde 6종, heterocyclic compounds 4종, ester 6종, acids 4종, hydrocarbons 4종, 기타 14종이었으며 균주별로는 B. subtilis에서 25종, B. natto에서 30종, B. licheniformis에서 28종, B. natto와 B. licheniformis를 혼합하여 접종한 청국장에서는 41종의 향기성분이 검출되었다. 유기산의 총량은 B. subtilis를 접종한 청국장에서 3.98%로 가장 높았고 B. natto는 2.24%, B. licheniformis는 1.53%이었으며 B. natto와 B. licheniformis의 혼합 시험구에서 는 1.67%의 함량을 보였다. 분석된 향기성분과 유기산은 starter로 접종된 Bacillus의 종류에 따라 각각 특징적 인 조성을 나타내었다. 관능평가 결과 색, 향, 맛 그리고 전체적인 선호도의 모든 항목에서 B. natto와 B. licheniformis의 혼합 시료가 가장 높은 선호도를 보였고 B. subtilis를 starter로 한 시료의 선호도가 가장 낮았다.

유산균의 혼합배양에 의한 무주스의 가공 (Processing of Radish Juice by Mixed Culture with lactic Acid Bacteria)

  • Leuconostoc mesenteroides와 Lactobacillus brevis, Lactobacillus fermentum을 무주스에 starter로 0.3%(106 cfu/$m\ell$)첨가하여 $25^{\circ}C$에서 7일, 15$^{\circ}C$에서 20일, 5$^{\circ}C$에서 36일동안 숙성하면서 단독, 또는 혼합 배양에 의한 무주스의 가공특성을 조사하였다. 무주스를 $25^{\circ}C$에서 숙성할 경우 유산균수는 혼합배양이 단독배양보다 높게 계수 되었으나 낮은 온도 (15~5$^{\circ}C$)에서는 접종균주에 따라 증감의 차이는 있으나 starter의 첨가효과가 거의 없는 것으로 나타났다. 유산균을 접종한 경우 접종 균주에 따라 다소 차이는 있으나 비휘발성 유기산, 아스코르브산 함량이 접종하지 않은 무주스에 비해 높게 나타났다. 단독 및 혼합배양한 각각의 무주스를 가장 맛있는 상태 (적숙기)에서 차이식별검사를 실시한 결과 군덕내와 생무맛을 제외하곤 5% 수준에서 유의적 차이가 있었으며, 기호도 검사결과 Lactobacillus brevis와 Leu. mesenteroides로 $25^{\circ}C$에서 혼합 배양한 무주스가 가장 높은 점수를 얻었으며, 시중에 유통중인 동치미보다도 높은 점수를 얻어 starter 이용한 무주스의 제조는 동치미의 맛을 지니면서도 품질이 우수하여 상품성이 높을 것으로 기대된다.

한국의 발효식품에 관하여 (Traditional Fermented Food Products in Korea)

  • Fermented foods available in Korea may be classified into four groups, namely, fermented soybean products, fermented cereal products, fermented vegetable products, and fermented fishery products based on raw materials used. The representative fermented foods based on soybean are Kanjang(soysauce), Doenjang(fermented soybean pastes), and Kochujang(red pepper added-fermented soybean paste). Such fermented products are made using Meju(functioning as a starter and prepared by fermentation of steamed soybean mash) as an essential ingredient, and used widely as a soup base and/or in seasoning side-dishes for everyday meals year around. Excepting Sikhae, all fermented products based on rice and other cereal grains are of alcoholic nature. Takju (Makgeolli) used to be made using rice as the major raw material, however, mainly due to the shortage of rice in recent years, other cereals, such as, barely, corn, and wheat flours are also used to replace rice today. Owing to such changes in the raw materials. the popularity of Takju has been somewhat reduced, yet it is still widely consumed in rural areas. Although Chungju is a popular rice wine with superior qualify over Takju, the amount consumed is considerably limited. The highest qualify rice wine, Bupju, in particular, is made by a low temperature fermentation using glutinous rice. Kimchi is an unique fermented vegetable product of long tradition in Korea. Although it was for consumption mainly in winter season serving as a source of vitamins, today it is widely used throughout the year. Except Kkakdugi and Dongchimi, all of the fermented vegetable products contain salted Korean cabbage as an essential item, while they abound in varieties depending on material composition and methods of processing, and also on seasons and localities Next to Kimchi in this category is Kkakdugi made of raddish in popularity and quantity consumed. The four groups of fermented food products described above are reviewed in some detail and evaluated in terms of their nutritional significances, processes and microorganisms involved. and their commercial potentials. Jeotkal (or Jeot) is a name given to all fermented products of fishery origin. A number of Jeot can be prepared by adding salt and allowing fermentation to the raw materials such as shrimp, anchovy octopus, clam, oyster, etc.

Detection of Antibiotic Resistance and Resistance Genes in Enterococci Isolated from Sucuk, a Traditional Turkish Dry-Fermented Sausage

  • The aim of this study was to isolate enterococci in Sucuk, a traditional Turkish dry-fermented sausage and to analyze isolates for their biodiversity, antibiotic resistance patterns and the presence of some antibiotic resistance genes. A total of 60 enterococci strains were isolated from 20 sucuk samples manufactured without using a starter culture and they were identified as E. faecium (73.3%), E. faecalis (11.7%), E. hirae (8.3%), E. durans (3.3%), E. mundtii (1.7%) and E. thailandicus (1.7%). Most of the strains were found resistant to rifampin (51.67%) followed by ciprofloxacin (38.33%), nitrofurantoin (33.33%) and erythromycin (21.67%). All strains were found susceptible to ampicillin. Only E. faecium FYE4 and FYE60 strains displayed susceptibility to all antibiotics. Other strains showed different resistance patterns to antibiotics. E. faecalis was found more resistant to antibiotics than other species. Most of the strains (61.7%) displayed resistance from between two and eight antibiotics. The ermB, ermC, gyrA, tetM, tetL and vanA genes were detected in some strains. A lack of correlation between genotypic and phenotypic analysis for some strains was detected. The results of this study indicated that Sucuk manufactured without using a starter culture is a reservoir of multiple antibiotic resistant enterococci. Consequently, Sucuk is a potential reservoir for the transmission of antibiotic resistance genes from animals to humans.

Assessment of Potential Probiotic and Starter Properties of Pediococcus spp. Isolated from Turkish-Type Fermented Sausages (Sucuk)

  • In this study, the metabolic activities of five strains of Pediococcus spp., in terms of the quantities they produced of lactic acid, hydrogen peroxide, exopolysaccharides, and proteolytic activity, were determined. Lactic acid levels produced by these strains were found to be in the range of 2.5-5.6 mg/ml. All strains produced hydrogen peroxide. The P. pentosaceus Z13P strain produced the maximum amount (0.25 mg/ml) of proteolytic activity. Exopolysaccharide (EPS) production by the Pediococcus strains during growth in MRS (de Man, Rogosa, and Sharpe) medium was in the range 25-64 mg/l. The susceptibility of 10 different antibiotics against these strains was also tested. All strains were found to be resistant to amoxicillin, gentamicin, and vancomycin. Antimicrobial effects of the Pediococcus spp. on pathogens were also determined by an agar diffusion method. All of the strains were able to inhibit L. monocytogenes. The tolerance of the strains to low pH, their resistance to bile salts of strains, and their abilities to autoaggregate and coaggregate with L. monocytogenes were also evaluated.

발효유제품에서 박테리오파지의 특성, 검출과 제어 (The Characteristics, Detection and Control of Bacteriophage in Fermented Dairy Products)

  • This study was to review the classification, detection and control of bacteriophage in fermented dairy products. Bacteriophage has lytic and/or lysogenic life cycles. Epidemiologically speaking, detected major phages are c2, 936 and p335. Among them p335 has been the largest concern in dairy industry. Traditionally, various analytical technologies, such as spot, starter activity, indicator test, ATP measurement and conductimetric analysis, have been used for the phage detection. In recent years, advanced methods such as flow cytometric method, petrifilm, enzyme linked immunosorbent assay (ELISA) and multiflex PCR diagnostic kit have been deveoloped. The phage contamination has been controlled by using heat, high-pressure treatment, and the combinations of heat and pressure, and/or chemical. Also some starter cultures with phage-resistant character have been developed to minimize the concentration of phages in dairy product. Bacteriophage inhibition media such as calcium medium was also mentioned. To prevent the contamination of bacteriophage in dairy industry, further researches on the detection and control of phage, and phage resistant starters are necessary in the future.

고추장에서 분리된 Bacillus subtilis BS16045의 유전체 서열 분석 (Complete genome sequence of Bacillus subtilis BS16045 isolated from Gochujang)

  • 한국 발효식품의 보존성을 높이기 위한 스타터 균주를 얻기 위하여 고추장에서 Bacillus subtilis BS16045를 분리하였다. B. subtilis BS16045에 대한 유전체 분석을 실시하였으며, G+C 비율이 43.6%인 4,165,121 bp 크기의 염기서열을 얻었다. 또한 이 유전체로부터 항진균 및 항균 활성에 연관이 있는 surfactin, kanosamine, bacillaene, plipastatin, subtilosin A, bacilysin 생산 유전자들을 확인하였다. 이러한 결과들을 통해 B. subtilis BS16045는 장류 제조시설에서 유해균의 오염문제를 해결할 수 있는 스타터로 이용될 수 있을 것으로 보인다.

쌀누룩과 수수를 사용한 고체발효 증류주의 제조 특성 (Manufacturing Characteristics of Solid-State Fermentation Distilled Spirits Using Rice Nuruk (Fermented Starter) and Sorghum)

  • In this study, characteristics of solid-state fermentation distilled spirits with nuruk (fermentation starter) and sorghum were investigated. The starch digestion activity was highest in black koji (A. usamii) at 359.15 U/g dry base, white koji (A. luchuensis) at 306.98 and yellow koji (A. oryzae) at 15.31 was followed. The pH of the solid-state fermented mash in yellow, white and black koji showed 5.09, 4.46 and 4.50 respectively with significant differences (p<0.001). The total acid content of white and black koji was 0.73% as citric acid. The alcohol content of mash prepared with yellow, white and black koji was 4.34, 4.24, 3.89% respectively. The contents of reducing sugar showed 3.28, 3.23 and 2.64%. Total sugar were 14.88, 17.84 and 17.60% respectively. The alcohol content of each distillate ranged from 44.3 to 39.9%, and the furfural content in yellow koji was 1.25 times lower than the others.

Characterization of Nonaflatoxigenic Aspergillus flavus/oryzae Strains Isolated from Korean Traditional Soybean Meju

  • Filamentous fungi that could be classified into Aspergillus flavus/oryzae were isolated from traditionally fermented meju commercially available in Korea. The samples were analyzed for aflatoxin B1 and ochratoxin A contamination by HPLC; however, no toxin was detected. In addition, fungal and bacterial metagenomic sequencing were performed to analyze the microbial distribution in the samples. The results revealed that the distribution and abundance of fungi and bacteria differed considerably depending on the production regions and fermentation conditions of the meju samples. Through morphological analysis, ITS region sequencing, and assessment of the aflatoxin-producing ability, a total of 32 A. flavus/oryzae strains were identified. PCR analysis of six regions with a high mutation frequency in the aflatoxin gene cluster (AGC) revealed a total of six types of AGC breaking point patterns. The A. flavus/oryzae strains did not exhibit the high amylase activity detected in the commercial yellow koji strain (starter mold). However, their peptidase and lipase activities were generally higher than that of the koji isolates. We verified the safety of the traditionally fermented meju samples by analyzing the AGC breaking point pattern and the enzyme activities of A. flavus/oryzae strains isolated from the samples. The isolated strains could possibly be used as starter molds for soybean fermentation.
