Research on the Food Habits of Housewives of Korean Students Residing in a Southern Region of Texas, U.S.A. (미국 Texas 일부 지역에 거주하는 한국 유학생 부인들의 식생활습관에 관한 연구)

  • The purpose of this study was to research on the preparing fermented foods and food habits of Korean women in Texas U. S. A. The preparing method of fermented foods and food habits of Korean housewives whose husbands are studying at the various universities in Houston, Texas are surveyed. The analysed results indicated were as follows ; 1. 64.4% of women were in the thirties at their age, those in twenties 29.7%. 75.2% of subject graduated college. Period of residence in America were 52.5% under 3yr, 25.7% over 5yr. The respondents were aged twenties(29.7%) to thirties(64.4%) who have graduated from college(75.2%) or graduate school(24.8%) and have had the residence period of less than three-(52.5%) to more than five years(25.7%). 2. The kind of cuisine chosen for dinner were Korean style food. The most considerable person when preparing menu were husband. 48.51% of those in age were 3∼39yr(p < 0.1). 60.40% of the response graduated college(p < 0.1). The Korean-style foods overwhelmed over western ones in their frequency for dinner(82.2% vs. 17.8%). The person most considered for choosing menu was their husbands as responded by the majority groups of aged thirties(48.5%), college graduates(60.4%) and residence period of less than three years(42.6%). They satisfied comparatively when they prepared the meal (p < 0.1). 55.4% of the response learned cooking method from neighbor, 39.6% of remainder studied from cuisine book, newspaper, magazine, TV. They satisfied comparatively for the preparation of the meal(56.4%) and have learned the new methods of cooking from neighborhoods(55.4%) or cuisine book, newspaper, magazine and TV(39.6%). 3. The frequency of eating-out mostly had once a month. 30.69% of response were 30∼39yr(p < 0.1). 33.66% of subjects lived under 3yr in America. Eating-out once a month occupied 30.7, 33.7, and 20.7% of the selected group of aged thirties, college graduates, and residence periods of less than three years, each respectively. The type of food selecting eating-out were western style. 38.31% of them were 30∼39yr(p < 0.1). 39.60% of them graduated college(p < 0.1). 29.70% of them lived under 3yr in America. For eating-out menu, the western-style foods were most favored by the groups of aged thirties(38.3%), college graduates(39.6%), and residence periods of less than three years(29.7%). 4. 50.5% of subject had beef once three days, 23.8% of the response had pork once two weeks, 27.7% of subject ate chicken once two weeks, 34.7% of the response enjoyed fish once a week. Eating animal foods once three days for beef(50.5%), once two weeks for pork(23.8%) and chicken(27.7%), and once a week for fish(34.7%) were most frequent. 5. Korean traditional fermented food were mostly enjoyed in order of kanjang(91.1 %), kochujang(87.2%), fermented fish(85.1%), deonjang(81.2%), jang-a-chi(76.2%). 81.2% of subject used prepared-food by themselves. 56.4% of response made kimch at home. Among Korean traditional fermented foods, kimchi(56.4%) and prepared side dishes(81.2%) were provided at home while kanjang(91.1%), kochujang(87.2%), fermented fish(85.1%), deonjang(81.2%), and jang-a-chi(76.2%) were mostly purchased from stores.

  • About 75 strains which utilize cholesterol as sole carbon and energy source were isolated from 10 samples of Kimchi and 18 samples of fermented fish food (2 Ojingo-jeots, salt-fermented squid ; 5 Changran-jeots, salt-fermented pollack tripe ; 5 Myungran-jeots, salt-fermented Alaska pollack roe ; 3 Gajami-sikhae-jeots, fermented flat fish ; 2 Gul-jeots, salt-fermented oyster ; a Juneo-jeots, salt-fermented shad). Among them tested, the 3T6-5Mj strain isolated from Changran-jeot showed the highest activity on cholesterol degradation. The optimal composition of medium for the producing cholesterol degradation enzyme by 3T6-5Mj strain was 1.0 g/L NH4NO3, 1.0 g/L K2HPO4, 0.1 g/L MgSO4.7H2O, 1.0 g/L FeSO4.7H2O, 1 g/L NaCl, 5 g/L Trypton, 1 g/L Cholesterol, and 5 g/L Maltose at 30$^{\circ}C$, pH 7.5, and the enzyme production reached a maximum level at 140 hours of cultivation.

  • Moderate halophilic bacteria isolated from several kinds of salted and fermented sea foods (jeotkal) collected from commercial market were identified and investigated on their selt requirements. It was confirmed that the isolates were dominantly moderate halophiles according to their NaCl requirement to grow. And their salt requirements in their growth have been examined for Na, K, Ni and mg ions. Among them, the most dominant and distinctive three strains in protease pro-duction have been examined and two of them identified to be halophilic Flavobacterium sp., and the other one to be halophilic Pseudomonas sp..1'heir optimum growth was observed at 3$0^{\circ}C$ and at 10 percent of NaCl.

Anticancer Effects of Ganjang with Different Aging Periods (숙성 기간에 따른 간장의 항암 효과)

  • Ganjang and doenjang are known as major fermented soy-based foods in Koreans. Current investigations have proved that fermented soybean foods impart anti-cancer, anti-obesity, and anti-diabetic effects. The aim of this study was to evaluate the anti-cancer activities of commercialized soy food, Ganjang, as a function of aging period. The test groups were classified into four time periods-short (under 5 years, S group), mid (under 10 years, M group), long (under 15 years, L group), and eternal (over 15 years, E group). The anti-cancer effects of Ganjang were determined by cell cytotoxicity assay of three types of cancer cell lines and splenocyte proliferation assay. Besides these assays, we also analyzed NK cell activity for cancer immunotherapy. The results show that the anti-cancer effect increased in the S and M period aging groups for all three cancer cell lines. Interestingly, similar to the anti-cancer result, splenocyte proliferation and NK activity showed the highest effect in the S and M groups. In contrast, Japanese ganjang-treated (JG1, JG2) groups and E group showed significantly reduced splenocyte proliferation. Collectively, these results suggest that the short and middle periods of traditional fermented Ganjang might have potential anti-cancer activities.

Safety and Technological Characterization of Staphylococcus xylosus and Staphylococcus pseudoxylosus Isolates from Fermented Soybean Foods of Korea

  • We evaluated the antibiotic susceptibilities, hemolytic activities, and technological properties of 36 Staphylococcus xylosus strains and 49 S. pseudoxylosus strains predominantly isolated from fermented soybean foods from Korea. Most of the strains were sensitive to chloramphenicol, erythromycin, gentamycin, kanamycin, lincomycin, oxacillin, tetracycline, and trimethoprim. However, 23 strains exhibited potential phenotypic acquired resistance to erythromycin, lincomycin, and tetracycline. Based on breakpoint values for staphylococci from the Clinical and Laboratory Standards Institute, >30% of the isolates were resistant to ampicillin and penicillin G, but the population distributions in minimum inhibitory concentration tests were clearly different from those expected for acquired resistance. None of the strains exhibited clear α- or β-hemolytic activity. S. xylosus and S. pseudoxylosus exhibited salt tolerance on agar medium containing 20% and 22% (w/v) NaCl, respectively. S. xylosus and S. pseudoxylosus strains possessed protease and lipase activities, which were affected by the NaCl concentration. Protease activity of S. pseudoxylosus was strain-specific, but lipase activity might be a characteristic of both species. This study confirms the potential of both species for use in high-salt soybean fermentation, but the safety and technological properties of strains must be determined to select suitable starter candidates.

Genomic analysis of WCP30 Phage of Weissella cibaria for Dairy Fermented Foods

  • In this study, we report the morphogenetic analysis and genome sequence of a new WCP30 phage of Weissella cibaria, isolated from a fermented food. Based on its morphology, as observed by transmission electron microscopy, WCP30 phage belongs to the family Siphoviridae. Genomic analysis of WCP30 phage showed that it had a 33,697-bp double-stranded DNA genome with 41.2% G+C content. Bioinformatics analysis of the genome revealed 35 open reading frames. A BLASTN search showed that WCP30 phage had low sequence similarity compared to other phages infecting lactic acid bacteria. This is the first report of the morphological features and complete genome sequence of WCP30 phage, which may be useful for controlling the fermentation of dairy foods.

Bioavailability of Fermented Korean Red Ginseng

  • In an effort to improve ginsenoside bioavailability, the ginsenosides of fermented red ginseng were examined with respect to bioavailability and physiological activity. The results showed that the fermented red ginseng (FRG) had a high level of ginsenoside metabolites. The total ginsenoside contents in non-fermented red ginseng (NFRG) and FRG were 35715.2 ${\mu}g$/mL and 34822.9 ${\mu}g$/mL, respectively. However, RFG had a higher content (14914.3 ${\mu}g$/mL) of ginsenoside metabolites (Rg3, Rg5, Rk1, CK, Rh1, F2, and Rg2) compared to NFRG (5697.9 ${\mu}g$/mL). The skin permeability of RFG was higher than that of NFRG using Franz diffusion cells. Particularly, after 5 hr, the skin permeability of RFG was significantly (p<0.05) higher than that of NFRG. Using everted instestinal sacs of rats, RFG showed a high transport level (10.3 mg of polyphenols/g sac) compared to NFRG (6.67 of mg of polyphenols/g sac) after 1 hr. After oral administration of NFRG and FRG to rats, serum concentrations were determined by HPLC. Peak concentrations of Rk1, Rh1, Rc, and Rg5 were approximately 1.64, 2.35, 1.13, and 1.25-fold higher, respectively, for FRG than for NFRG. Furthermore, Rk1, Rh1, and Rg5 increased more rapidly in the blood by the oral administration of FRG versus NFRG. FRG had dramatically improved bioavailability compared to NFRG as indicated by skin permeation, intestinal permeability, and ginsenoside levels in the blood. The significantly greater bioavailability of FRG may have been due to the transformation of its ginsenosides by fermentation to more easily absorbable forms (ginsenoside metabolites).

The quality characteristics of Jeung-pyun made with different kinds of beans (콩의 종류에 따른 증편의 품질특성)

  • Jeung-pyun, a very popular fermented rice cake consumed in Korea, consists mainly of rice, rice wine (Tak-Ju), and sugar. The effects of addition of different beans on the quality characteristics of the batter and Jeung-pyun were investigated. Six different beans were mixed with the rice flour at levels of 5% and 10% of the rice flour weight, respectively. The addition of Back-tae, Huk-tae, Sori-tae, which are types of soybeans, and black gram significantly increased the batter volume and viscosity. However, the fermented Back-tae (Cheongguk-jang) was not effective at increasing the batter volume and viscosity. The buffering effect of the beans was very significant on the fermented batter, and the decrease in pH of the fermented batter made with beans was less than that of the control batter without beans. Additions of the soybeans and Cheongguk-jang were most effective for the buffering effect in the fermented batter. Although the Back-tae, Huk tae, and Sori-tae were different colors and shapes, they were all soybeans and exhibited similar effects on the Jeung-pyun batter. However, the effects of the beans were not significant on the Jeung-pyun. The volume and moisture content of the Jeung-pyun made with beans were not significantly different from the volume and moisture of the Jeung-pyun made without beans. The above results suggest that the addition of different soybeans, mung beans, and black gram significantly effects on the properties of Jeung-pyun batter, but not Jeung-pyun itself.

Effect of Extracts and Bacteria from Korean Fermented Foods on the Control of Sesame Seed-Borne Fungal Diseases (발효식품 추출물과 미생물을 활용한 참깨 종자전염성 병 방제)

  • In order to control seed-borne diseases, we obtained extracts from commercial fermented food products of Kimchi, Gochujang, Doenjang, Ganjang and Makgeolli and their suppressive effects against seed-borne diseases were studied. In addition, the suppressive effects of bacterial strains isolated from the fermented foods were screened in vitro and in vivo. Among fifty food extracts, twenty food-extracts suppressed more than 92% incidence of seedling rots in vitro and seven food extracts increased 58.3-66.8% of healthy seedling in the greenhouse. Among 218 isolates from the fermented foods, 29 isolates showing high antifungal activity against seven seed-borne fungal pathogens were selected. Among 29 isolates, 13 isolates significantly reduced seedling rot and increased healthy seedlings. Sixteen isolates with high antifungal activity and suppressive effect against sesame seedling rots were identified by 16S rRNA sequencing. Fourteen of sixteen isolates were identified as Bacillus spp. and the other two isolates from Makgeolli were identified as Saccharomyces cerevisiae. It was confirmed that B. amyloliquifaciens was majority in the effective bacterial population of Korean fermented foods. In addition, when the bioformulations of the two selected effective microorganisms, B. amyloliquifacien Gcj2-1 and B. amyloliquifacien Gcj3-1, were prepared in powder forms using bentonite, kaolin, talc and zeolite, talc- and kaolin-bioformulation showed high control efficacy against sesame seed-borne disease, followed by zeolite-bioformulation. Meanwhile control efficacy of each bentonite-bioformulation of B. amyloliquifacien Gcj2-1 and B. amyloliquifacien Gcj3-1 was lower than that of bacterial suspension of them. It was found that the selected effective microorganisms from Korean fermented foods were effective for controlling seed-borne diseases of sesame in vitro and in the greenhouse. We think that Korean fermented food extracts and useful microorganisms isolated from the extract can be used as bio-control agents for suppressing sesame seed-borne diseases based on above described results.

Fatty Acid Composition of Salted and Fermented Sea Foods on the Market (시판젓갈류의 지방산 조성)

  • The fatty acid composition of lipids extracted from 18 kinds of salted and fermented sea foods which have been consumed in Korea were examined. The lipid contents of salted and fermented sea foods showed wide difference (0.8-11.9%) depending on species and portions of raw materials. In fatty acid composition of salted and fermented sea foods prepared with whole fishes, the saturated fatty acid was the most predominant component, and the major fatty acids were 16:0, 18:1, 22:6, 16:1 and 20:5. In case of salted and fermented products prepared with gills, roe or intestines, the polyenoic fatty acid was the main component, the major fatty acids were 18:1, 16:0, 22:6, 20:5, 16:1 and 18:3. In these products the composition ratios of 20:5 and 18:3 were higher, and that of 16:0 and 16:1 were slightly lower as compared with salted and fermented whole fish products. And in case of salted and fermented crustacea and mollusk, the polyenoic fatty acids such a 22:6 and 20:5 were the most predominant component, the abundant fatty acids were 22:6, 16:0, 20:5, 18:1 and 16:1. Judging from results, salted and fermented sea foods were abundant of the highly unsaturated fatty acids such as 20:5, 22:6 inspite of the long fermentation periods.
